Squalen Bulletin of Marine and Fisheries Postharvest and Biotechnology

Squalen publishes original and innovative research to provide readers with the latest research, knowledge, emerging technologies, postharvest, processing and preservation, food safety and environment, biotechnology and bio-discovery of marine and fisheries. The key focus of the research should be on marine and fishery and the manuscript should include a fundamental discussion of the research findings and their significance. Manuscripts that simply report data without providing a detailed interpretation of the results are unlikely to be accepted for publication in the journal.

Abstract

Rumput laut merupakan tumbuhan laut yang berpotensi sebagai sumber pangan dan obat-obatan. Tumbuhan ini mengandung polisakarida yang banyak digunakan sebagai bahan pangan. Rumput laut juga kaya akan senyawa yang bermanfaat bagi kesehatan diantaranya yaitu pigmen, yang dihasilkan oleh rumput laut merah (Rhodophyceae), rumput laut hijau (Chlorophyceae), dan rumput laut coklat (Phaeophyceae). Setiap jenis rumput laut mempunyai pigmen yang spesifik dengan komposisi yang berbeda. Beberapa hasil penelitian menunjukkan bahwa kandungan dan komposisi pigmen rumput laut diantaranya terdapat pada jenis Kappaphycus alvarezii, Caulerpa sp., dan Sargassum sp. Pada Kappaphycus alvarezii, terdapat beberapa jenis pigmen yaitu karoten (0,947%), turunan klorofil (16,418%), klorofil a (74,920%), xantofil (7,715%). Caulerpa sp. mengandung karoten (0,294 %), turunan klorofil (18,731%), klorofil a (26,817%), klorofil b (12,906%), dan xantofil (29,758%). Sargassum sp. mengandung b-karoten (1,49%), fukoxantin (20,95 %), klorofil a (52,82%), klorofil c (1,05 %),  turunan klorofil (15,23%) serta xantofil (8,46%).

Abstract

Chemical investigation of the endophytic fungus Botryosphaeria australis isolated from Avicennia marina originally from Hainan Province, P.R. China, yielded a new compound botryosphaenin (1), from the class of napthoquinone, together with 5 known compounds, botryosterpene (2) and 5-hydroxy-2,7-dimethoxynaphthalene-1,4-dione (3) and its derivatives, 6-ethyl-5-hydroxy-2,7-dimethoxynaphthalene-1,4-dione (4), O-methylaspmenone (5), O-methylasparvenone (6) and 5-(carboxymethyl)-7-hydroxy-1,4a-dimethyl-6-methylene decahydron aphthalene-1-carboxylic acid (7). Their structures were determined on the basis of spectroscopic methods including 1D (1H, 13C, and DEPT) and 2D (COSY, HMQC, HMBC, and ROESY) NMR experiments and by mass spectroscopic measurements The new compounds, 1 showed activity against the bacterial pathogens Staphylococcus aureus, several Streptococcus species and Bacillus subtilis, but also against the eukaryotic cell lines THP-1 (human leukemia monocyte) and BALB/3T3 (mouse embryonic fibroblast).

Abstract

 The strain LBF-005 from marine bacteria have already isolated and screened for mannanase degrading enzyme in submerged fermentation process. This strain was further identified by using 16S rRNA showed that bacterium is belong to Bacillus subtilis that could produce mannanase with activity around 9.5 U/mL. The optimum pH and temperature for the activity of crude enzyme for mannanase was 6.0 and 50 oC. Cloning of mannanase gene from B. subtilis was conducted using six primers set designed based on the homology analysis conserve region several mannanase from bacteria (Bacillus sp.) glycosyl hydrolase (GH) family 26. Optimization of PCR conditions was performed by gradient PCR to obtained PCR product of b-mannanase gene. The PCR product was obtained by third primer combination and was estimated to be around 972-bp. Analysis of the nucleotide sequence showed that sequences has similarity with mananase gene from other Bacillus sp., such as the B. subtilis strain WLY-12, B. subtilis strain WL-8, B. subtilis strain CICC 10260, B.subtilis strain CD-25, B.subtilis strain G1, and Bacillus sp. SWU60 were about 98%, 98%, 98%, 98%, 97% and 95%, respectively. The next research will to obtain a whole gene encoding β-mannanase by using in vitro cloning method, characterization of recombinant mannanase and application this enzyme for mannooligosaccharides production.

Abstract

Food infection and intoxication of fish products are common due to consumption of foodexposed by either pathogenic or toxin promoter bacteria. A number of pathogenic bacteria havebeen identified related to fish and seafood such as Vibrio parahaemolyticusand other Vibrios,Escherichia coli, Aeromonas spp., Salmonellaspp., Staphylococcus aureus, Listeriamonocytogenes, Clostridium botulinum, C. perfringens, and Shigellaspp. However, isolationand identification of pathogenic bacteria from fish and seafood are often difficult due to the highnumber of contaminating and indigenous bacteria, while numbers of pathogenic bacteria arerelatively low. In the last 3 decades, several molecular methods have been developed and adoptedas official standard to supplement classical method, such as polymerase chain reaction (PCR)and its applications. Besides its limitation, molecular based identification offers advantages asroutine or screening analytical protocol for pathogenic bacteria in fish and seafood products.

Abstract

Hydrocarbons are one of the main sources of acute and chronic stressors in marine environmentthat are potentially damaging the ecosystem if not properly overcame. As an attempt to restore theenvironment, microbial degradation is a logical solution owing to its low cost and environmentalfriendliness. Screening of microbes with bioremediation activities can be performed in vitrobymeans of high throughput screening (HTS) and/or in silicovia docking method. The latter haspractical advantages over the first in terms of time and cost. In this review, the use of virtualscreening is demonstrated to analyse the specificity of cytochrome-c peroxidase (CCP) enzymefrom hydrocarbonoclastic bacteria Marinobacter hydrocarbonoclasticus. Result showed thatCCP is a decent receptor for simple aromatic hydrocarbons. Despite previous reports on thealkane degradation activities of M. hydrocarbonoclasticus, this result demonstrates a newperspective on its potential to bioremediate low molecular weight polycyclic aromatic hydrocarbon(PAH) with moderate activity.

Abstract

A research related to the production of  alkali treated cottonii (ATC) had been carried out in order to study the effect of chopping step and drying techniques on the quality of ATC produced. Four treatments were applied in the experiment, namely was copping before sun drying (treatment A); chopping after sun drying (B); chopping before mechanical drying (C); and chopping after mechanical drying (D). The quality parameters of ATC measured were gel strength, moisture content, viscosity, yield, and whiteness. The results showed that the quality of ATC was significantly affected by chopping step and drying technique, especially in ATC gel strength, viscosity and yield. However, the effect of chopping step and drying technique was insignificant to ATC moisture content and whiteness. Chopping seaweed before drying resulted in higher gel strength of the ATC but lower in yield, while chopping after drying tended to result in lower gel strength but higher viscosity and yield.

Abstract

Spons merupakan hewan multiseluler yang paling sederhana sekaligus merupakan sumber senyawa bioaktif yang sangat berguna di dalam bidang farmasi. Senyawa-senyawa baru yang sangat beragam sudah banyak yang diisolasi dari spons. Kesulitan utama dalam memproduksi senyawa obat baru dari spons adalah kurang mencukupinya jumlah senyawa bioaktif yang dihasilkan spons untuk keperluan uji preklinis dan klinis karena rendemen yang dihasilkan sangat rendah. Artikel ini merupakan tinjauan terhadap metode kultur sel spons untuk memproduksi senyawa bioaktif  dalam rangka mengatasi masalah suplai senyawa bioaktif yang diuji.  Produksi senyawa avarol dari kultur primmorph spons Dysidea avara dibahas dalam review ini.  Avarol dilaporkan  memiliki bioaktivitas yang kuat sebagai substansi antitumor, antibakteri, dan antivirus. Model pendekatan melalui kultur primmorph dapat digunakan sebagai cara yang menjanjikan  untuk memproduksi senyawa bioaktif dari spons.

Abstract

This research was aimed to isolate marine derived-fungi which associated with macro algae and sponge from Indrayanti Beach, Jogjakarta and to screen the antitumor (T47D and HeLa) bioactivity of the fungi extracts. Three solid media of MEA (malt extract agar), GPY (glucose peptone yeast) and MFM (minimal fungi medium) were used as isolation medium. Each of the pure fungi isolates was then cultivated in 100 ml of liquid medium for 4 weeks at room temperature (27-28°C) in static conditions. The antitumor activity of the fungi extracts were tested against breast tumor cells (T47D) and cervical cancer cells (HeLa) using Thiazolyl Blue Tetrazolium Blue (MTT) assay method. A number of 21 isolates of fungi were isolated from 4 macro algae and 1 sponge samples. The identification of fungi isolate was conducted using combination of molecular approach (ITS1-5.8S-ITS4 DNA regions) and macro-micro morphological characteristics. Among those 21 marine fungi species isolated, MFGK-21 extract showed the best anti-servical tumor (HeLa) with an IC50 value of 240.1 µg/ml and MFGK-27 extract showed the best anti-breast tumor (T47D) with an IC50 value of 59.6 µg/ml. The MFGK-21 fungi isolate was identified as Penicillium steckii, while the MFGK-27 fungi isolate was identified as Aspergillus sydowii.

Abstract

Study aimed to investigate the utilization of shrimp shell powder in the production of lindur fruit-potato simulation chips, which was assessed using proximate composition, texture performance and sensory evaluation parameters. Three different ratios of lindur fruit-potato flour (w/w) (30:70, 40:60 and 50:50), as well as different concentrations of shrimp shell powder (0, 1, 3 and 5%) were used in the formula. The results showed that lindur fruit-potato simulation chips enriched with shrimp shell powder had 3.22-4.42% moisture content, 3.33-4.94% ash content, 3.77-5.83% protein content, 14.59-19.04% fat content, 71.06-76.34% carbohydrate content and 341.4-530.9 g/cm2 hardness. Ratio lindur fruit flour and potato flour of 40:60 as well as 3% of shrimp shell powder was chosen as the best treatment since the formula produced lower hardness, higher protein content and the most crispy and tasty chips preferred by panelists.
