Abstract

Sponge is one of the marine biota that known have a high bioactivity potential and contain secondarymetabolites that can be used as antibiotic and medicinal materials. The aim of this research was tobioactivity assay of hexa-tetra contane compound contained in Callyspongia pseudoreticulata.Antibacterial tested has been carried out for Ralstonia solanacearum in potato plant. The resultsshowed that 400 mg/ml of compound has the best inhibitory diameter of “Penyakit Layu” bacterial(Ralstonia solanacearum) was 21.33 mm and significantly different to other concentration.

Abstract

Analysis of histamine content in processed bullet tuna (Auxis thazard) stew flesh can be carried out by Spectrophotometry method, using p- phenildiazonium sulfonate reagent. A qualitative, visual comparison of colour intensity of samples with reference colour scale of the standard solution concentration can be used to determine levels of histamine without the aid of a spectrophotometer. The histamine content is determined quantitatively by the standard curve regression equation (y= 0.003x–0.062) with determination coefficient value (R2= 0.938). The results showed that histamine content inprocessed bullet tuna (Auxis thazard) obtained: 46.9964; 40.3093; 34.4348; 28.9900 and 24.8862 mg/100g fish, at 0.5; 1.0; 1.5; 2.0 and 2.5% concentration of NaCl, respectively.

Abstract

Bioethanol can be produced through glucose fermentation using saccharomyces cerevisiae. Was done to make coconut water contains a small amount of carbohydrate. A researcher had taken research was done to make bioethanol from coconut water waste. The result of the research shows that coconut water fermentation with yeast has maximum speed after 70 minutes of incubation. The fermentation solution was then distilled. Bioethanol purity was about 76-80% after HPLC analysis.

Abstract

Trimyristin isolation from the nutmeg and synthesis of methylester using heterogen catalyst has been carried out in several stages, included isolation of nutmeg oil, trimyristin isolation and synthesis of methylester. The synthesis was started by isolation of nutmeg oil with steam distillation for 8 hours. It yields was 308 mL of nutmeg oil. The isolation trimyristin from nutmeg with maseration using chloroform yields trimirystin 17 g (11%), structure elusidation of these products was analized by FTIR and 1H-NMR. The synthesis methylester using CaO catalyst and methanol yields 1,78 g (89,02%) of methyl myristate. Product purity was tested by GC and structure elusidation was analized by FTIR and MS.

Abstract

The research of utilitasion of beef tallow into biodiesel with heterogenous (CaO) catalyst has been conducted. Biodiesel production initially by esterification reaction with catalyst H2SO4 1% (toward oil and methanol weight) with molar ratio oil : methanol (1:9). After esterification process, trygliceride separated from methanol and continued by transesterification step with molar ratio 1:12 (oils and methanol) and CaO catalyst 11% wt. Esterification and transesterification process carried out for 2 hours and 6 hours respectively at 65 ℃. Composition of the biodiesel was analyzed by 1HNMR, Gas Chromatography-Mass Spectrometry (GC-MS) and their physical properties analyzed by ASTM method. Result of biodiesel conversion is 93.99% while based on GC-MS analysis showed that mayor component of biodiesel were mixture of methyl esters with methyl octadecanoat as the major coumpound (40.65%). Based on data from ASTM, biodiesel produced from beef tallow have qualified as diesel fuel.

Abstract

Sagoo plant (Metroxylon sp.) numerous in eastern Indonesia, such as Maluku and Papua. Sagoo starch containing amylose and amylopectin. Sagoo starch hydrolysis can be carried out by enzymes, such as glucoamylase or often called amiloglukosidase or α-1 ,4-glukanoglukohidrolase, an extracellular enzyme capable hydrolyzes α-1, 4 on amylose and α-1, 6 in amylopectin. The enzymatic hydrolysis of starch can be reviewed in the kinetics of the Michaelis-Menten equation and Lineweaver-Burk equation. The aims of this study to determine the kinetic parameters (Vmax and Km) of glucoamylase on hydrolysis of sagoo starch. Glucoamylase has an optimum temperature of 50 oC and pH optimum 6.5. At optimum conditions, based on Michaelis-Menten equation, the value of Vmax is 1.09 U/mL/min and Km value is 0.82. Based on Lineweaver-Burk equation, the value of Vmax was 1.03 U/ mL/min and Km value was 0.84%.

Abstract

5,5,7,12,12,14-hexamethyl-1,4,8,11-tetraazacyclodeca-7,14-dienium bromide disolvate ((Me6N4H4)Br2.2H2O) is a 14-membered deprotonated tetraaza macrocyclic salt. Its neutral analogues and their complexes were prepared by templation method under refluxed condition since it was discovered by Curtis in 1961. However, the present diprotonated tetraaza salts react with many metallic salts in both methanolic and aqueous solutions. The complexation involved deprotonation of the diagonally opposite protonated amines and the type of metal salt and solvents also play an important role in the final complex formation. The complexation of (Me6N4H4)Br2.2H2O with copper acetate in methanol and aqueous solutions gave the expected complex of [Cu((Me6N4H2)Br]Br.2H2O. Pseudo-first order condition for the complexation reaction in water was established.

Abstract

The papain enzyme was isolated from the latex of papaya and it was purified by ammonium sulfat fractionation and dialysis methods. Determination of papain enzyme activity showed that the highest activity was found in the 80% fraction with a specific activity of 10.5797 x 105 U/g. Papain concentration after Bradford method was 53 mg/mL. Molecular mass of papain was detected to be 21.6 kDa after SDS-PAGE analysis. Characterization of papain showed that the optimum activity of papain was found at pH 8.0 with a specific activity of 14.07031 x 10 U/g, and at temperature 60 °C with a specific activity of 14.39755 x 10 U/g.

Abstract

The research to determine histamine content in bullet of tuna (Auxis rochei) with variation of tamarind (Tamarindus indica L.) extract concentrations and time of marinate has been done. The histamine content is determined quantitatively by using ultraviolet-visible spectrophotometer with the calibration curve method. The result showed that histamine content in bullet tuna marinated in tamarind extract with varied concentrations of 5%, 10%, 15%, and 20% within 10 minutes, are 37.4561, 35.3041, 23.8778, and 22.1186 mg/100 g respectively. While, the concentrations of histamine in bullet tuna marinated in 20% of tamarind extract with variation of 10, 20, 30, 40, and 50 minutes, are 22.2965, 19.7544, 17.6925, 11.5865, and 8.0396 mg/100 g respectively. A qualitatively, visual comparison of colour intensity of samples with reference colour scale of the standard solution concentration can be used to determine concentrations of histamine without the aid of a spectrophotometer.

Abstract

The study on production of activated carbon from cacao shell (Theobroma cacao) was conducted by added 4 M HCl as activating agent. The results show that acidity of solid carbon active before activated with adsorption weight is 3.2865x10-3 mol.g-1 and after activated is 3.5052x10-3 mol.g-1. Active carbon result is used for the application at adsorption Cd with some variantion, that are optimum concentration 20 ppm and optimum time 80 min. Cd adsorption from active carbon was analyzed by AAS. The results are coeficient correlation (r2) 100 % followed Freundlich isotherm with KF value was 9.994 and n was -0.982.
