Abstract

ABSTRACT Gelatin from catfish (Pangasius hypophthalmus) skin had been produced by Research Center for Marine and Fisheries Product Processing and Biotechnology. To determine the safety of thegelatin, a subchronic in vivo toxicity study had been done on mice. The study used male mice (Mus musculus) with 20–30 g body weight each. Gelatin was given orally by gavage once a day. Mice were divided into 4 groups of treatment. The doses given were 0 (negative control) 1.5; 3 and 6% of gelatin which was equal to 0, 12, 24 and 48 mg/g of mice body weight. The study had been conducted for 4 weeks and continued for 2 weeks of recovery phase. Blood serum condition, i.e. Glutamic Oxaloacetic Transaminase (GOT), Glutamic Pyruvic Transaminase (GPT), creatinine, albumine and Blood Urea Nitrogen (BUN) and organ (liver, kidney, stomach) damage were investigated. The results showed that gelatin at the highest dose (48 mg/g bw of mice) gave asignificant effect on GOT level at the second week of treatment. Furthermore, there was no significant effect of treatments on organ damage level.

Abstract

ABSTRACTThe important thing considered in the processing of fish mince meat based products is gel forming ability, which is affected by additives applied in the processing of the products. This research was aimed at studying the effect of TGase and salt addition on the physical and sensory properties of restructured product from Priacanthus macracanthus. Salt was added into minced meat at the concentration of 0, 1 and 2%, TGase with the concentration of 0; 0.3; 0.6 and 1%. The meat dough was then filled into plastic tubes and heated at 30oC for an hour before being heated at 90oC. The restructured meat was then evaluated its sensory properties texture profile (hardness,chewiness, gumminess, cohesiveness, springiness and breaking force, deformation/distance, gel strength), and its microscopic observation under the scanning electron microscope. The result showed that addition of salt as well as TGase gave significant effect on the sensory properties related to texture, appearance and brightness; and physical properties ofthe restructured products espescially gumminess and breaking force. Based on the sensory score, addition of 2% salt wasenough to produce gel which met with panelist preference, however based on the physical/texture properties addition of 2% salt and 0.3% TGase was needed to increase the gel properties. At this treatment combination, the gel strength produced was 3,235 g cm.

Abstract

ABSTRACTMarine-derived fungi have proven to be a rich source of cytotoxic compounds for the development of new anti cancer drugs. The aims of this research were to: 1) screen cytotoxic activity of marine fungi from Indonesian waters, 2) indentify marine fungus that produced thatproduced the most active cytotoxic compound and 3) investigate inhibition concentration 50 (IC50) value of cytotoxic compound. The fungi were isolated from marine organism collected fromWakatobi Marine National Park-South East Sulawesi, Binuangeun Beach-Banten, Manado watersNorth Sulawesi and Kepulauan Seribu Marine National Park-Jakarta. Liquid cultures of the fungiwere carried out in Malt Extract Broth and Soluble Starch Soytone medium for 4 weeks at 27–28oC without shaking. Molecular identification of fungus was conducted through PCR amplificatin usingprimers of ITS1 and ITS4 primer. Cytotoxic activity of the extract was tested by using MTT (3-(4.4-dimethylthiazol-2-yl)-2.5-diphenyl-tetrazolium bromide) method. The MTT test showed that MFW39strain exhibited the strongest cytotoxic activity. Molecular identification revealed that MFW39 marine fungus was similar to Emericella nidulans with precent identity of 99%. Mycelium and broth extract of MFW39 fungus inhibited the growth of T47D cell with IC 50 values of 21.9 and 169.3 µg/mL, respectively. Further research will be focus on to the strain of MFW39 marine fungi.

Abstract

Penelitian ini bertujuan untuk mengevaluasi pengaruh penambahan Pediococcus acidilactici F-11 sebagai kultur starter terhadap kualitas rusip melalui proses fermentasi. Rusip teri dibuat melalui fermentasi tanpa dan dengan penambahan starter dengan variasi penggaraman 10, 15, dan 20% dan masing-masing ditambah gula merah 10% dari berat ikan  pada suhu kamar (30 ± 2°C) selama 12 hari. Parameter yang diamati meliputi parameter mikrobiologi (ALT, total bakteri asam laktat (BAL), dan total coliform), kimiawi (pH, total asam, TVB, kadar air, dan kadar garam), dan sensori. Hasil penelitian menunjukkan bahwa penambahan Pediococcus acidilactici F-11 dapat meningkatkan total BAL dan menurunkan total coliform produk rusip yang dihasilkan. Selain itu dapat mempersingkat waktu fermentasi dari 12 hari menjadi 9 hari dan menghasilkan produk rusip dengan sifat sensori yang lebih disukai terutama pada perlakuan penggaraman 15%. Produk rusip yang dihasilkan memiliki total BAL lebih tinggi yaitu sebesar 7,47 log, total coliform lebih rendah yaitu 3,34 log daripada rusip tanpa starter serta memiliki rasa dan tekstur yang lebih disukai.

Abstract

ABSTRACTStudy on carrageenan extraction using filter press with variation ratio of seaweed and extraction medium (water), temperature of precipitation, potassium chloride concentration has beenconducted. Ratio of seaweed and water used were 1:20, 1:30 and 1:40 (w/v), temperature of precipitation were varied at 15 and 30°C, and potassium chloride concentration used were 1 and 1.5%. Parameters observed on the carrageenan were yield, viscosity, gel strength, moisture content, ash content, sulfate content and acid insoluble ash. Result showed that the best treatment was ratio seaweed : water = 1:20, temperature of precipitation at 30°C and potassium chlorideconcentration of 1%, with quality of carrageenan i.e. gel strength 1897.14 (g/cm2), viscosity 145 (cPs), ash content 29.59%, acid insoluble ash 0.83%, sulfate content 18.36%, yield 31.77% andmoisture content 9.73%. The carrageenan quality has met with the standard established by Food Agriculture Organization (FAO), Food Chemical Codex (FCC) and European Economic Community(EEC).

Abstract

ABSTRACT: Research on the correlation of sensory (Demerit Point Score), chemical, and microbiological parameters during deterioration process of fresh tilapia stored in ice has been conducted. Live tilapia was killed by hypothermia method (immersing in ice water) and subsequently arranged in insulated box filled with flake ice with the ratio of ice and fish of 2:1 (w/w). The ice in boxes was checked and melted ice was replaced every day. The box was stored at ambient temperature andobservation on freshness changes of tilapia was done every 3 days for sensory (Demerit Point Scores/DPS), chemical (TVB, K value) and microbiological (TPC) qualities until the fish was rejected by panelists. Results of the experiment showed that after 15 days storage, fresh tilapia was still accepted by panelists, however, after being stored for 18 days, tilapia was unfit for human consumption with the value of DPS 25.9, TVB 48 mgN%, K value 76.4% and TPC 5.0 x 105 cfu. It was shown that DPS correlated well with TVB as well as K value but did not correlate well with total bacteria.

Abstract

ABSTRACTResearch on the oil contents of microalgae i.e. Botryococcus braunii, Nannochloropsisand Spirulina platensisof different cultivation time have been done at Research Center for Marine and Fishery Product Processing and Biotechnology(RCMFPPB) Laboratory, Slipi, Jakarta. The three species of microalgae were cultivated outdoor in 100 L of seawater medium of 20 ppt salinity using sun light intensity and continuous aerations. Experiments were conducted in three replicates.Observations on the cell growth were carried out every 2 days and the biomass were harvested on day 5, 9 and 15 and sun-dried. Oil were extracted from the dry biomass using hexane. The highestcell density was reached by S. platensiswith 8.46 log cell/mL on day 13, while the highest growth rate was shown by S. platensiswith growth rate (k) = 9.40 on day 3. The highest yield of oil was obtained from B. brauniion day 9 which was 14.90%.

Abstract

ABSTRACT Agar-agar is widely used as an additive for food products. However, the application is still limited due to its low functional properties. Increasing functional properties of agar-agar was conducted by the addition of gums because they have synergism effect with phycocoloid. Research was conducted by formulation of agar-agar with other gums i.e. Arabic gum, guar gum, locust bean gum (LBG) and konjac. The ratios of agar-agar and gums in the experiment were 1:3, 1:1 and 3:1. Native agar-agar without addition of gums was used a control. Observations done to evaluate their functional properties were gel strength, elasticity, syneresis, viscosity, melting point, and gelling point. Results showed that the addition of LBG and konjac significantly increased agar-agar gel strength. LBG and konjac showed synergism effect on increasing agar-agar gel strenght. Gel strength of agar-agar added with konjac at a ratio of 1:1 increased from 493 g/cm 2 to2011 g/cm 2and added with LBG with ratio of 1:1 increased from 493 g/cm 2to 864 g/cm 2. Elasticity of agar-agar added with guar gum at a ratio of 3:1 increased from 45 mm to 47.90 mm. Guar gum, LBG and konjac showed synergism effect on increasing agar-agar viscosity. Viscosity of agaragar added with guar gum, LBG and konjac at a ratio of 1:3 increased from 101 cPs to 1880, 1610 and 5380 cPs respectively. The melting point of agar-agar added with arabic gum at ratio 1:3 of decreased from 56 o C to 48o C.

Abstract

ABSTRACTBreast cancer is the second mostly found disease after cervix cancer to Indonesian woman. Research on the effect of Ulva fasciata Delile and Turbinaria decurrensBory ethyl acetate extract feeding on the Adenocarcinoma mammaecell proliferation on C3H mice has beenconducted. Thirty experimental mice were devided into 6 treatment groups, i.e.: K-I, negative control; K-II, solvent control; K-III, given U. fasciata ethyl acetate extract dose of 41.32 mg/20 g BW (body weight); K-IV, given U. fasciataethyl acetate extract dose of 82.64 mg/20 g BW; K-V, given T. decurrensethyl acetate extract dose of 24.29 mg/20 g BW; and K-VI, given T. decurrens ethyl acetate extract dose of 48.59 mg/20 g BW. All treatments were given during 21 days. The mice body weight and tumor volume were measured and collected subsequently at every two days. Tumor doubling time was calculated at the end of research. Results showed that there wasa highly tumor growth inhibition on the K-IV shown by the high doubling time value. Tumor growth inhibition decreased according to the sequence as follows: K-IV (U. fasciata dose of 82.64 mg/20g BW), K-V (T. decurrensdose of 24.29 mg/20 g BW), K-VI (T. decurrensdose of 48.59 mg/20 g BW) which equal to K-II (solvent control), K-III (U. fasciatadose of 41.32 mg/20 g BW), K-I(negative control).
