Abstract

The taxonomic position of Psidium cujavillus Burm.f. in relation to P. guajava L. has not been widely agreed upon, so that this research was conducted to clarify it based on morphological characteristics, germination, anatomy, and chromosomes. Observations on the living material of P. cujavillus and P. guajava showed that there were basic similarities in the structure and forms except in segment distance and size of leaves, fruit diameter and seed weight. Both taxa have the same germination processes, and their seedlings morphology are also not much different except in size. Anatomically there were also no significant differences between P. cujavillus and P. guajava. Chromosome size of P. cujavillus and P. guajava do not exceed 2 μm, both has the number of 2n=22. Overall observation showed that there was no significant difference to indicated P. cujavillus as distinct species from P. guajava. Based on this, it is suggested that P. cujavillus should be classified as a infraspecies variation in P. guajava.

Characterization of Local Durian Accession Collected Hortimart Agro Centre, Central Java Using Molecular Markers Inter Simple Sequence Repeats (ISSR). Floribunda 5(7): 267–276. — The lack of morphological markers encourage the use of other markers that are considered more accurate, i.e molecular markers. The purpose of this study is to analyze the genetic diversity as well as determine the identity of the local durian accession collection of Hortimart Agro Centre, Central Java. Leaves of 41 durian accessions were isolated using CTAB method. DNA amplification was conducted with four primers of ISSR PKBT and the results were separated by agarose gel. Data were analyzed using NTSYSpc program version 2: 02. The results showed that DNA amplification using four primers of ISSR BKPT yielded 66 polymorphic bands. The results of the analysis showed that the genetic diversity of local durians of Hortimart collection is high because 100% of polymorphic DNA bands with variations of size 250–1500 bp. No local durian accession was found to have the same DNA100% profile, although four pairs of accessions were known to have a 96% high similarity. Five accessions, namely Petruk 2, Ontoseno, Semar, Arjuno, and Gondomono have specific DNA bands. The only accession of durian apart from the other group is Gondomono because it has only one band on ISSR PKBT 8 with size 750 bp which is not owned by other accession. However, dendrograms showed that no identities that are 100% equal so that eachDNA band profile can be the identity of each durian accession.Keywords: Durio zibethinus, molecular marker, genetic diversity.Ahmad Solikin, Amin Retnoningsih & Enni S. Floribunda 5(7): 267–276. — Keterbatasan ciri morfologi mendorong penggunaan penanda lain yang dipandang lebih akurat, yaitu penanda molekuler. Tujuan penelitian ini adalah menganalisis keanekaragaman genetika sekaligus menentukan identitas aksesi durian lokal koleksi Hortimart Agro Centre, Bawen, Jawa Tengah. Daun 41 aksesi durian diisolasi menggunakan metode CTAB. Amplifikasi DNA dilakukan dengan empat primer ISSR PKBT dan hasilnya dipisahkan melalui gel agarose. Data dianalisis menggunakan program NTSYSpc versi 2:02. Hasil penelitianmenunjukkan bahwa amplifikasi DNA menggunakan empat primer ISSR BKPT menghasilkan 66 pita polimorfik. Hasil analisis menunjukkan bahwa keanekaragaman genetika durian lokal koleksi Hortimart tergolong tinggi karena 100% pita DNA polimorfik dengan variasi ukuran 250–1500 bp. Tidak ditemukan aksesi durian lokal yang memiliki profil DNA100% sama, meskipun empat pasang aksesi diketahui memiliki kemiripan tinggi 96%. Lima aksesi, yakni Petruk 2, Ontoseno, Semar, Arjuno, dan Gondomono memiliki pita DNA spesifik. Satu-satunya aksesi durian yang terpisah dari kelompok lainnya adalah Gondomono karena hanya memiliki satu pita pada ISSR PKBT 8 dengan ukuran 750 bp yang tidak dimiliki aksesi yang lain. Meskipun demikian, dendrogram memperlihatkan tidak ada identitas yang 100% sama sehingga setiap profil pita DNA dapat menjadi identitassetiap aksesi durian.  Kata kunci:  Durio zibethinus, penanda molekuler, keanekaragaman genetika.

Abstract

Ganyong (Canna indica L.) merupakan tanaman pangan berasal dari Amerika Selatan yang tersebar di seluruh kawasan dunia dan banyak dibudidayakan di daerah tropis seperti Indonesia, terutama PulauSumatera. Variasi rimpang pada kultivar di Provinsi Bengkulu belum terdata dengan baik sehingga diperlukan sistem klasifikasi dalam pengelolaan kultivar ganyong. Tujuan penelitian ini adalah untuk mengeksplorasi keanekaragaman tanaman ganyong di Provinsi Bengkulu dan mengelompokkannya berdasarkan karakter morfologi sehingga membantu dalam pengembangan dan pemuliaan kultivar unggul. Data morfologi yang meliputi rimpang, daun, bunga, buah, dan biji diamati dari 57 tanaman sampel hasil eksplorasi. Tanaman ganyong dikelompokkan secara fenetik menggunakan UPGMA berdasarkan 61 karakter morfologi menggunakan program NTSYS. Hasil analisis menghasilkan tujuh kelompok ganyong berdasarkan karakter rimpang, daun, bunga, dan buah. Hasil pengelompokan ganyong di Bengkulu kemudian dibandingkan dengan kultivar yang sudah terdaftar di Asia. Sebanyak lima kelompok ganyong di Bengkulu diindikasikan termasuk ke dalam tipe di Asia. Kelompok I dan III termasuk ke dalam tipe Thai-purple, kelompok II termasuk ke dalam tipe Chinese-purple, sedangkan kelompok IV dan V termasuk ke dalam tipe Japanese-green. Hubungan kekerabatan dianalisis secara kladistik menggunakan program PAUP* berdasarkan karakter morfologi menghasilkan pengelompokan: kelompok tetua (kelompok I, II, III) dan turunan (kelompok IV, V, VI, dan VII).

Floribunda 6(1): 1–11. — Baccaurea angulata (belimbing merah) adalah jenis endemik di Kalimantan yang dimanfaatkan sebagai bahan pangan dan obat tradisional, namun sejauh ini informasi distribusi dan variasinya masih sangat terbatas. Penelitian ini bertujuan memetakan lokasi-lokasi tumbuh pohon B. angulata, menggambarkan variasi morfologi, dan analisis fenetik B. angulata yang tumbuh di Kalimantan dan Pulau Natuna. Eksplorasi B. angulata dilakukan di Pulau Natuna dan  empat provinsi di Kalimantan (Barat, Timur, Tengah dan Selatan). Peta persebaran dibuat dengan program ArcGIS versi 10.3. Pola distribusi dianalisis menggunakan indeks Morishita. Data morfologi batang, daun, bunga, dan buah digunakan untuk analisis fenetik belimbing merah berdasarkan koefisien Simple Matching (SM) dengan metode UPGMA. B. angulata tersebar secara acak di Kalimantan Barat, Tengah dan Pulau Natuna, tetapi memiliki pola penyebaran mengelompok di Kalimantan Selatan dan Timur. Penemuan B. angulata di desa Ceruk dan Limau Manis di Pulau Natuna dengan nama lokal belimbing besi merupakan rekaman baru. Peta sebaran B. angulata di Kalimantan dan Pulau Natuna disediakan. Secara morfologi, B. angulata di Kalimantan dan Pulau Natuna bervariasi pada beberapa ciri yaitu tepi stipula, permukaan tangkai daun, warna tangkai perbungaan betina, dan bentuk buah. Analisis fenetik mengelompokkan seluruh sampel ke dalam tiga kelompok B. angulata berdasarkan 20 ciri morfologi dengan nilai keserupaan 53% sesuai dengan bentuk dan rasa buah. Fruits of Baccaurea angulata are usually utilized as food and local medicine, but up to now the information about its distribution and variation is very scarce. The objectives of the research were to map the distribution, describe the morphological variation, and analyze the phenetics of B. angulata in Natuna Island and Indonesian Borneo, Kalimantan. B. angulata was explored in Natuna Island and four  Provinces of Kalimantan (West, East, Central, and South). The distribution was mapped using ArcGIS version 10.3. The distribution pattern was analyzed using the Morishita index. Morphological data of stem, leaves, flower and fruit were used to analyze the phenetics of B. angulata using Simple Matching coefficient, to construct a dendrogram using UPGMA  method. B. angulata is randomly distributed in the West and Central of Kalimantan, also in Natuna, but it has a clumped pattern in South and East Kalimantan. The presence of B. angulata in the Ceruk and Limau Manis villages in Natuna Island with local name belimbing besi is a new record. B. angulata is morphologically variable in several characters, i.e. stipule margin, petiole surface, pistillate peduncle colour, and fruit shape. Phenetic analysis based on 20 characters grouped all individuals into three groups with similarity index of 53% according to the shape and fruit taste. The  individuals of B. angulata with oval fruits are proposed to constitute a new variety, namely B. angulata Merr. var. globulus Gunawan.Key words: Baccaurea angulata, Kalimantan, Natuna Island, new record, new variety, Phyllanthaceae.

Fitmawati. 2006. Taxonomic Status of Mangifera laurina and its Related Species. Floribunda 3(2): 29-37.-This study is aimed to investigate the taxonomic status of species related to M. laurina, to find out whether they represent a natural species or should be placed in several different species. The result showed that morphological phylogenetic analysis supported the idea for separating three monophyletic species (M. laurina, M. aplanata, M. lalijiwa), considering that M. rubropetala and M. indica are synonym, and defining two independent lineages of sister group (i.e. the clade of M. lalijiwa and the clade consist of M. indica, M. laurina, M. aplanata  and M. rubropetala). Morphologically, M. lalijiwa is characterized by having glomerulate and glabrous inflorescence and tips of petal ridge free of the surface. In contrast, the other species usually posseses non-glomerulate and puberulous inflorescence and base or a half of base of petal ridge at free of the surface. Phylogenetic analysis of E-RAPD was incongruent with that of morphological analysis. However, the molecular phylogeny support whether that M. aplanata is separated from other species and that cultivars of M. lalijiwa are a monophyletic group.

Abstract

Anatomical study on transection of stipe near lamina were carried out on 27 species of Diplazium from Western part Malesia (Java, Sumatra, Kalimantan). The aim of this study is to examine stelar anatomical characters for supporting species delimitation in Diplazium. The leaf-trace shape of Diplazium stipe is varying among species and constant among the adult individuals in a species. Leaf-trace shapes of Diplazium can be classified into five main types: (1) uninterrupted V-shaped, (2) interrupted V-shaped, (3) uninterrupted U-shaped, (4) interrupted Ushaped, and (5) W-shaped. Each type diversifies into some different derivative forms that enable to determine a species among closely related species. Therefore the leaf-trace shapes are important diagnostic character which support species delimitation in Diplazium.
