Abstract

Ethanolic extract of mango’s leaves variety of bapang (Mangifera indica L. ‘Bapang’) has been known to contains mangiferin which has antidiabetic activity in patients with diabetes mellitus. This study aimed to obtain a formulation of tablets of ethanolic extract mango leaves with variety of bapang with good physical characteristic and know the effect of variations concentration of guar gum on the properties of mucoadhesive. Tablets were prepared using wet granulation method varying levels of guar gum as a bioadhesive matrix. Tablets were tested physical characteristics and mucoadhesive strength in vitro by using a rotating cylinder and observed within 8 hours. The results showed that the tablets of ethanolic extract mango leaves with variety of bapang have good physical characteristics. The result of mucoadhesive in vitro test showed that all formulas have good mucoadhesive strength. Mucoadhesive strength increased with the increase in the concentration of guar gum. The result of this research showed that the tablets of formula 4 with the highest concentration of guar gum showed the best mucoadhesive strength where they could be attach of 7 ± 1 hour.

Abstract

Cyclodextrin glycosyltransferase (CGTase) is an extracellular enzyme which is produced by some bacteria and archae that converts starch into cyclodextrin (CD). This enzyme has a high commercial value because of its product (CD) is widely used in several industries, such as pharmaceutical, chemistry, cosmetic, food, textile and also for environment protection. In Indonesia, there are still a few bacteria as CGTase producer were discovered and observed. The aim of this research was to find bacteria as CGTase producer from soil of corn farm land in Sumedang, West Java, and further characterized the isolate in produce CGTase. Horikoshi agar media was used as screening media. An alkalophylic bacteria as CGTase producer, JS-1 was successfully gained from this process. This isolate was characterized in the term of Gram staining, oxygen demand, motility and catalase test. The characterization results showed that JS-1 is a bacillus, Gram positive, facultative anaerob, motil and catalase positive. JS-1 isolate was also further characterized in order to determine optimum incubation period and temperature for CGTase production. The results were showed that JS-1 isolate was produced CGTase in maximum level if incubated for 48h at 37 °C.

Abstract

Ketumpangan air (Peperomia pellucida (L.) Kunth), an Indonesian wild plant, is commonly found in every Indonesian region with moist shady place. People usually consume ketumpangan air as a fresh vegetables. Empirically, ketumpangan air is consumed to treat fever, stomach disease, kidney disorders, drug abscesses, boils acne, headaches, pain in arthritic and rheumatic gout, or other external treatment. In this research, we tested the acute toxicity of ethanol extract from ketumpangan air using experimental animals mice DDY strain. We also tested the Brine Shrimp Lethality Test (BSLT) using Artemia salina Leach larva shrimp. The main aim of this reseach was to determine the LD50 and LC50 values of ethanol extract from Ketumpangan air. The results showed that the ethanol extract was categorized as ‘mild toxic’. The calculation of LD50 with Reed and Munich method showed that the LD50 value for male and female group was 15.13 g/ kg and 11.87 g/kg respectively. The BSLT result showed that the LC50 values of Ketumpangan air was 81.28 ppm.

Abstract

Catechin is one of bioactive compounds in gambier. Result of preliminary studies showed that catechin is active as an antioxidant and antibacterial, thus it was potential as an oral nutraceutical for prevention of dental plaque. In present study, preparation of the oral nutraceutical in the form of mouthwash was performed by variating content of catechin in the extract as formulation parameter. The contents of catechin in the mouthwash solution are 0.01-0.1% (w/v). The mouthwash products were analyzed for pH, antioxidant and antibacterial activities, also particle size. The results showed that mouthwash with catechin content of 0.01-0.08% (w/v) produced the nanoscale of the particle size of mouthwash.

Abstract

Chemotherapy is one of the breast cancer treatments. However, there are several adverse effects with mild to high severity related to this treatment. This study was made to evaluate adverse effects which occurred in breast cancer patients who are still in their chemotherapy cycle. The evaluation consists of assessment of adverse effects associated with chemotherapy regimens and its dosage, causality assessment, and preventability assessment. This study was done from December 2012 until March 2013 concurrently. The data was collected directly from the patients by doing the interview. The data was also collected from the patient’s medical record and nurse’s daily documentation. Clinically, FAC caused more adverse effects than other chemotherapy regimens. However, in the 6% of excess dosage, there were no different acute adverse effects. The most occurred adverse effect was alopecia with 81% percentage. Causality assessment with probability scale Naranjo and WHO criteria showed that 97.3% and 99.2% were ‘probable’. The preventability assessment which based on Thornton- Schumock criteria showed that only nausea and vomit which were ‘definitely preventable’.

Abstract

Listeria monocytogenes is a Gram positive rod-shape bacteria. Other foodborne pathogens, mostly, may cause typical symptoms of gastroenteritis. Unlike these microorganisms, Listeria monocytogenes infection may result in more serious symptoms, usually take in the form of meningitis or septicaemia. People with vulnerable immune system are in a bigger risk of being exposed. Furthermore, the bacterium is also severer than any other bacteria found in the food, with more resistant to heat, drying, or salty environment. This research was purposed to identify and characterize a sample named LM 25722248, which was isolated from fresh food factory in a city of United Kingdom. The isolate was characterized as a Gram positive bacterium, showed tumbling motility, and gave positive result for catalase test and haemolysin assay. Moreover, benefiting from molecular technology, it was possible to reveal that the isolate was Listeria monocytogenes.

Abstract

Epidemiologic studies of dietary marine n-3 fatty acids and risk of colorectal cancer have been inconsistent. The study was conducted to understand the inhibitory effect of fish oil in mice with colorectal preneoplasia induced by azoxymethane (AOM) and dextran sodium sulfate (DSS). In this study, Balb/c mice was induced by AOM 10 mg/kg body weight followed by administration of 1 % DSS during a week. Fish oil administrated orally at a dose of 1.5, 3, and 6 mg per day. Histopathological examination of the colon tissue (hematoxylin-eosin staining) was done by counting the number of inflammation, and hyperplasia foci, the number of mitosis epithelial cells, and the scores of dysplasia in ten visual fields. In the second month, decreasing in the number of inflammation foci occurred between the control and low dose groups with medium and high dose groups (p<0,05). While in the third and fourth month, decreasing in the number of inflammation foci were observed in all treatment group (p<0.001). The number of mitotic colonic crypt epithelial cells was statistically significant between control group and treatment groups, initially observed since the second month. In the second month to the fourth month, generally increasing of doses of fish oil has decreased the number of hyperplasia foci. Our research indicates that fish oil also inhibits the occurrence of dysplasia. In the third and fourth month, dysplasia was found in the control group. Dysplasia was found only in the fouth month in the treatment group.

Abstract

Acne can be caused by bacteria, such as Propionibacterium acnes and Staphylococcus epidermidis. Red ginger (Zingiber officinale Rosc. var. rubrum) has been studied for its antibacterial activity. The aims of this research was to determine the minimum inhibitory concentration of red ginger rhizome ethanol extract against P. acnes and S. epidermidis bacteria and the effectiveness when the extraxt formulated into gel. The simplicia was extracted by soxhlet using 96% ethanol. Diffusion method was used to measure the minimum inhibitory concentration (MIC) of extract. Afterwards, the extract was forrmulated into gel by using three variation of HPMC 4000 and carbopol 934 as gel base with ratio of 70:30 (Formula I), 50:50 (Formula II) and 30:70 (Formula III). Gel evaluation included organoleptic, spreadability, adhesive property, pH and antibacterial effectiveness. The data was analyzed by R-Commander program 2.14.1 Version. The results showed MIC values are 0.45% against P. acnes and 0.5% against S.epidermidis. The results of evaluation for 30 days of storage showed that the gels had stable pH and adhesion, homogenous, whereas the spreadablity of gels increased. Antibacterial evaluation showed that formula I give the best effectiveness with 16,11 mm zone of inhibition against P. acnes and 14,00 mm against S.epidermidis.

Abstract

In attempt to develop Indonesian traditional medicine (Jamu) to become standardize herbal, it is need to examine the extract that can guarantee its efficacy, quality and safety. Brotowali stem (Tinospora crispa (L.) Miers.) known has antidiabetic effect and no toxic activity. In this research, the quality and α-glukosidase inhibitor activity of ethanolic extract of brotowali stem has been determined. In the quality test were determined specific parameters (identity, organoleptic, extractable matter), non specific parameters (loss on drying, water content, total ash content, solvent residue, heavy metal, and microbial contamination), quantification of markers content (total flavonoid content with spectrophotometric UV-Vis and assay of apigenin content with densitometric method). The research showed that the extract was fulfiled the requirement, total flavonoid content was 0.52% and apigenin content was 0.03635%. The result of α-glukosidase inhibitor activity test gave IC50 of 237.26 ppm.

Abstract

This research was aimed to determine biologic activities of n-hexane, ethylacetate, ethanol and water extracts of lelutung tokak (Tabernaemontana macrocarpa Jack.) stem as antioxidant and anticancer. The result of this research showed that four extracts of T. Macrocarpa Jack. stem have antioxidation and anticancer activity. The antioxidant activity test with brine shrimp lethality test (BSLT) showed the LC50 value of n-hexane, ethylacetate, ethanol and water extracts of lelutung tokak stem were 567.89; 119.34; 120.56 and 156.44 ppm respectively; with DPPH method were 653.54; 48.10; 53.322 and 121.02 ppm, respectively. The anticancer test result showed that the IC50 of ethylacetate and ethanol extracts were 6.04 and 7.14 ppm, respectively. The phytochemical analysis result showed that four extracts of stem of T. macrocarpa Jack. preliminary contained alkaloid, flavonoid, saponin and tanin compounds. It was concluded that ethylacetate and ethanol extracts of stem of T. macrocarpa Jack. possess a powerful anticancer and antioxidation activities.
