Abstract

Klebsiella pneumoniae is one of coliform bacteria which causes human and mammalian diseases. The bacteria dominate in dairy cow milk which has been infected by mastitis and has resistent on antibiotic. Vaccination is one of aims to prevent the diseases. Nuclear technique could be used to have a vaccine candidate.This research was conducted to get inactivated K. pneumoniae by gamma irradiation and heat inactivated as vaccine candidate. The experiments were done by determination of inactivated doses, protein content, and protein profile. analysis. K. pneumoniae could be inactivated using gamma rays by doses higher than 600 Gy. Neither irradiation nor heat (65 oC) influenced the K. pneumoniae total protein content The intensity of protein profile in gamma ray inactivated was higher than heat inactivated. There were 35, 36, and 60 kDa protein which were diagnosed as antigen protein. It could be concluded that inactivated by gamma irradiation K. pneumoniae could be chosen as a vaccine candidate and as a model for other bacterial vaccine.

Abstract

Lactic Acid Bacteria (LAB) has long been used in fermented milk because its stability, unique taste and texture and also for its health benents that are provided by the probiotic. There are many fermented dairy products in the market nowadays that declared containing living culture or as probiotics. The aim of this study was to identify LAB from fermented milk product by Visual inspection on plate culture for their morphology, as well by molecular technique performing PCR and DNA sequencing methods. Morphology and growth incubation time of each isolate was observed. A pair of primer that is highly speciiic for LAB targeting the gene encoding for 168 rRNA, which represents sequences of region 677 to 693 of E. coli numbering has been used for PCR identiiication in this study. A total of nineteen isolates were obtained from 10 fermented milk samples that were grown on de Mann Rogosa Sharpe Agar (MRSA) media. Nine out of 19 isolates which represent each of the samples were selected. Genomic DNA extracted from those 9 isolates were used as template for PCR. The results showed that 8 amplicons were positively identitied as LAB, but only three identities indicated similar species as declared, while 5 others did not.

Abstract

Cancer therapy using chemotherapeutic agents associated with many adversed side effects, including immunosupressant. The use of immunostimulator together with chemotherapeutic agent (co-chemotherapy) can be the alternative method to solve that problem. Ganoderma lucidum (Ling Zhi) has been reported to have immunostimulatory activity. The aim of this research was to evaluate the immunostimulatory activity of water extract of G. Zucidum by determining the relative CD8+ T lymphocyte cell percentage in rats induced by doxorubicin. Extraction of plant material was carried out by infusion method. Sprague Dawley female rats were divided into six groups, they were doxorubicin as control, commercial product as comparing control, 100 mg/kgBW and 450 mg/kgBW extract treatment, extract control, and without treatment control. Relative CD8+ T lymphocyte cell percentages of blood samples were obtained by flow cytometry by using Multiset program. The data were analyzed statistically using paired sample t test and one way ANOVA continued by Post Hoc test. The result showed that the water extract of G. lucidum increased the relative CD8+ T lymphocyte cell percentage in rats induced by doxorubicin. The water extract of G. lucidum is promising to be developed as co-chemotherapy immunostimulatory agent.

Abstract

Hydroxyl octadecadienoic acid (HODE) is an oxidized aliphatic fatty acid from the oxylipins group, which shows bioactivity and can be produced by Curvularia lunata BioMCC FE-00283 by submerged fermentation. Glucose, monosodium glutamate, and corn oil were media components that showed significant effect toward HODE production by C. lunata BioMCC FE-00283. Composition optimization of these three medium components was carried out by Response surface methodology (RSM). A full factorial central composite design (CCD) was used to explain the interaction effect between media components. Maximum HODE production as predicted by the quadratic model have chromatogram area of 4540000, which was verified experimentally to be 1091338 ± 152489. Eventhough experimental veriiication showed lower result compared to the model prediction, however it was higher compared to the HODE production using basal media with unoptimized composition of carbon, nitrogen, and inducer which only have chromatogram area of 295045 ± 71200. The verification also showed that optimization increased HODE production by 3.7 fold compared to that produced by the unoptimized media.

Abstract

Antibiotic UK 3A had been isolated as a minor component from the mycelium of Streptomyces sp. 517-0. It was elucidated as a nine-member dilactone ring derivative. To produce an analog of UK-3A, i.e. hydroxypicolinil dioctyl glutamic ester, was synthesized from L glutamic acid and octanol, and the product was then reacted with 3-hydroxypicolinic acid and dioctyl glutamic ester. The tinal product was identified by 1H and 13C FT NMR, FT-IR spectrophotometry, and LC-MS. The it was tested for cytotoxic activity against P388 Murine Leukemia cells. The results showed that the 3-hydroxypicolinyl dioctyl glutamic ester has higher inhibition activity against P388 leukemia cells with IC50 value of 9.80 µg/mL than the activity of UK 3A with IC50 value of 38.4 µg/mL.

Abstract

Parasitic plant [Dendrophthoe pentandra (L.) Miq.] on the masisin [Rhodomyrtus tomentosa (Aiton) Hassk.] was traditionally used for cancer treatment in Central Kalimantan. Maceration of dried Dendrophthoe pentandra using ethanol followed by ethyl acetate produced ethanol and ethyl acetate extracts which exhibit inhibitory activity against leukemia L1210 cells with IC50 of 17.3 and 20.9 µg/mL, respectively. Fractionation of ethyl acetate extract by column chromatography yielded 7 fractions (F1~F7). Inhibitory activity test on fractions F1~F7 against leukemia L1210 cells showed inhibitory activity with IC50 0f 19.9, 19.0, 20.7, 16.0, 13.7, 12.5, and 17.4 µg/mL, respectively. During evaporation of F5 by rotary evaporator, a yellowish precipitate was observed and identified as quercitrin (quercetin 3-O-rhamnoside). Inhibitory activity test on quercitrin against leukemia L1210 cell showed that the crystal has an IC50 of 14.7 µg/mL. Separation of F7 by semipreparative HPLC reverse phase column yielded isolate A and B which exhibited inhibitory activity on leukemia L1210 cells with IC50 of 15.0 and 14.4 µg/mL, respectively. Subsequent solvent partition of ethanol extract into ethyl acetate-water and separation of ethyl acetatesoluble portion by column chromatography produced 7 fractions (F8~F14). Further separation of F14 by column chromatography yielded quercitrin and isolate C which has not been identified yet.

Abstract

Endophytic fungi Kabatiella caulivora var B, have been isolated from Alyxia reinwardtii which was collected at Purwodadi Botanical Garden, Pasuruan, East Java. The objective of this present work is to asses the antimicrobial activity from extracts of the endophytic fungi Kabatiella caulivora var B against Bacillus subtilis, Escherichia coli, Salmonella thypi, Pseudomonas aeruginosa, Staphylocooccus aureus and Candida albicans. Disc diffusion method was conducted in this project with two concentration extracts (10 mg/disc and 5 mg/disc). The results showed that the methanol and ethyl acetate extracts of Kabatiella caulivora var B both have antimicrobial activity against all microbial tested, but the methanol extract showed negative activity against Staphylocooccus aureus and Candida albicans at concentration 5 mg. The discovery of endophytic fungi that produces bioactives metabolites has opened a promising new approach to obtain drugs on a commercial scale from microbes that grow inside plants.

Abstract

Transdermal transport was an alternate to propranolol HCl delivery to overcome the low bioavailability by oral route. Kinetics of transdermal transport can be predicted by software WinSAAM. The aim of this research was to know kinetics of propranolol HCl transdermal transport with the present of enhancer oleic acid, propylene glycol and iontophoretics. Propranolol HCl transdermal transport was examined through the hairless rat as membrane on the vertical diffusion cell in the in vitro permeation. Enhancement methode used was oleic acid in propylene glycol and iontophoretics at varying concentrations. The donor phase contained 5 mg/mL propranolol HCl in citrate buffer, and the acceptor phase contained phosphate buffer saline at pH 7.4. Results of propranolol HCl transdermal transport analyzed by WinSAAM software. Parameters of transdermal transport were the rate of mass transfer from donor compartement to skin (Ka), available dose to transport (AD), and the rate of mass transfer from skin to acceptor compartement (KR).The results indicated that propranolol HCl transdermal transport with the present of enhancer can be explained by three compartement model and first order kinetics. Theoretically, value of AD influenced by oleic acid, interaction of oleic acid-iontophoretics and interaction of propylene glycol-iontophoretics. Value of Ka inliuenced by iontophoretics and interaction of oleic acid-propylene glycol-iontophoretics. Value of KR induenced by iontophoretics.

Abstract

Goat milk containing many amino acids and fatty acids that can be used as an emollient and humectant in skin liquid soap. The aim of this study is to investigate the effect of variation in cocamide DEA concentration on a face liquid soap stability. Face liquid soap formulae containing 10% goat milk and different concentration of cocamide DEA (3%, 4%, 5%, and 6%) were made by dissolving and mixing methods. The goat milk liquid soap products were evaluated and tested for their stability. Stability were justified from the colour, smell, viscosity, pH, density, and homogenity. The result showed that higher cocamide DEA concentration would produce better soap stability consecutively. The goat milk liquid soap containing 6% of cocamide DEA is the best formula investigated.

Abstract

Four xanthones and two benzophenones from the stem bark of kandis gajah (Garcinia griffithii) by extraction and chromatographic methods have been isolated. The six compounds were determined as: 1,7-dihydroxyxanthone (1), 1,6,7-trihydroxyxanthone (2), 1,6-dihydroxy-3-methoxy-4,7-di-(3-methylbut-2-enyl)xanthone (3), 1,5-dihydroxy-3,6-di-methoxy-2,7-di-(-3-methylbut-2-enyl)xanthone (4), guttipherone I (5), and isoxanthochymol (6). All of the isolated compounds were submitted to antioxidant activity assay using three methods: (i) free radical scavenging method, (ii) xanthine-xanthine oxidase (XO) method, and (iii) nitroblue tetrazolium xanthine oxidase (NBT/ XO). Guttipherone I (5) and isoxanthochymol (6) have strong antioxidant activity (IC50 ≤10.2 µg/mL), 1,6,7-trihydroxyxanthone (2) and 1,6-dihydroxy-3-methoxy-4,7-di-(3-methylbut-2-enyl)xanthone (3) as active compounds (IC50 ≤ 100 µg/mL), while 1,7-dihydroxyxanthone (1) and 1,5-dihydroxy-3,6-dimethoxy-2,7-di-(3-methylbut-2-enyl)xanthone (4) were inactive (IC50 > 100 µg/mL). Base on the data, antioxidant activity of isolated compounds was inhuenced by dihydroxyl groups at ortho position, number of hydroxyl group, and hydroxyl group at para position to C carbonyl at heterocyclic.
