Abstract

One of the most sensitive and selective instrumentation techniques for the analysis of pesticide residues in vegetables at a high level is High Performance Liquid Chromatography (HPLC). To obtain optimum result of measurement, optimization and validation of measurement method is needed. The purpose of this research is to optimize and validate the determination of chlorpyrifos concentration by HPLC method, so it can be used for analysis of chlorpyrifos residues in samples of cabbage vegetables taken from some traditional markets in North Sulawesi. Optimized chromatographic parameters are the composition of mobile phase of water and acetonitrile, mobile phase flow rate, and the volume of the sample injection. While the validated measurement parameters include precision, linearity, detection limits, and quantitation limits. Based on the results obtained using Zorbax Eclipse Plus C18 column, the optimum condition of HPLC was achieved at λmax of 289 nm, mobile phase composition of water : acetonitrile was 10:90, mobile phase flow rate of 0.70 mL/min, and sample injection volume of 15 μL. The result of validation of analytical method shows that analytic performance of determination of chlorpyrifos with HPLC method is very good, it is indicated by measurement parameter such as precision shown with coefficient of variation (CV) was 0,2086%, linearity indicated by correlation coefficient (r) was 0.9963, the detection limit (LOD) of 0.67 ppm, and the quantitative limit (LOQ) of 2.24 ppm, which all meet the requirements. The results of this study indicate that this method can be used for analysis of chlorpyrifos residues in cabbage vegetable samples.

Abstract

Hard water is a term known for water with high content of Calcium (Ca) and Magnesium (Mg) mineral. This type of water determines social-environment quality as well as people health in an area. Water with high degree of hardness could harm people health as it deteriorates people kidneys. The residence of Dusun Gelaran 01, Bejiharjo, Karangmojo, Gunungkidul use a 10-to-20 meter wells to obtain ground water for drinking and other household needs. The soil of Dusun Gelaran 01 is dominated by the red mediterranean association and black grumusol with limestone, so that the ground water has high amount of Ca and Mg minerals. Throughout the study, it can be determined the level of water hardness of 40 samples of Dusun Gelaran 01 well water by complexometry titration method. Out of all the samples tested 6 (15%) samples were hard water, and 34 (85%) samples were very hard water. The highest levels of total hardness was 490,2 mg/L. Total hardness levels have not exceeded the threshold according to Regulation of the Ministry of Health Republic of Indonesia No. 492/MENKES/PER/IV/2010.

Abstract

One of the plants that are used to regular treatment is behind the wind (Mallotus sp.). Traditionally this is used for the relief of pain and increase stamina by way of boiling the seven pieces of young leaves and drink once a day in the morning. This research aims to know the acute toxicity of ethanol extracts of leaves behind the wind (Mallotus sp) against larvae of Artemia salina Leach BSLT method. The research method used is qualitative analysis of chemical compound and Brine Shrimp Lethality Test (BSLT) method with post-test-only control group design. The results of research on ethanol extracts of leaves behind the wind showed the value of LC50 of 32.061 ppm, so can be said of ethanol extracts of leaves behind the wind on this experiment of acute toxicity has on the larvae of Artemia salina Brine Shrimp method using Leach Lethality Test (BSLT).

Abstract

The development of functional food today mainly focuses on founding food that rich in nutrition and also provide health benefit for human consumption. At this point the combination of prebiotic and probiotic food that refer to food with health benefit to help digestible function become increasing. This research was done by literature study and laboratory experiment with 5 medium and 4 replications in order to found the possibility to produce fructooligosaccharide from Aspergillus niger fermentation in some typical foodstuffs from North Sulawesi i.e pisang goroho (Musa acuminate), sweet potato (Ipomeae batatas), coconut water, and palm sugar. Statistical analysis of the results shown that the Aspergillus niger tend to grow in 4 of 5 medium which indicate the possibility of fructooligosaccharide in those medium. Further studies are needed in order to know the production of fructooligossaccharide based on its medium.

Abstract

The purpose of this research is to search for compound inhibitor endocytosis by using Molecular Modeling program. Screening is performed on approximately 800,000 compounds derived from several commercial companies. From the results of virtual screening obtained 320 hits, and of this number only 175 compounds that can be obtained to test its biological activity. Biological tests showed that there were 7 compounds with moderate endocytosis inhibition capability (IC50 = 50-170 μM) to good inhibition (IC50 <50 μM). The compounds are: compound 1 (IC50 = 42 ± 4 μM), compound 2 (IC50 = 83 ± 8 μM), compound 3 (IC50 = 135 ± 58 μM), compound 4 (IC50 = 151 ± 18 μM) compound 5 (IC50 = 144 ± 18 μM), compound 6 (IC50 = 13 ± 4 μM), and compound 7 (IC50 ~ 170 μM).

Abstract

The purpose of this study was to identify the total phenolic compound extract of pumpkin fruit strands. Pumpkin is a food rich in vitamin A and C, minerals, and phenolic compounds. Pumpkin plant as a minor food commodity, it is very rich in bioactive compounds that act as antioxidant very useful for human health. Strands in yellow fruit processing is often used as waste. This research was conducted by using the method of laboratory analysis through stages, determination and sampling, sample preparation, analysis of phenolic compound content in extract determined by Folin-Ciocalteu method and phenolic content calculation. The results showed that yellow pumpkin strands had total phenolic content 167,85 mg / kg in ethanol extract and 15,20 mg / kg in Petroleum ether extract. The data showed that the highest total phenolic content was found in the ethanol extract 167,85 mg / kg. This is because polar ethanol solvent can dissolve the polar compound.

Abstract

Cadmium (Cd) is one of the most important non-essential heavy metals, but can continue to increase in the environment. Bioaccumulative cadmium poisoning may cause kidney disease, stomach disorders, brittle bones, haemoglobin decline, and tooth pigmentation. Therefore, an analytical method for determining cadmium content should be developed. Methods for the determination of cadmium metal content that has been widely used is the method of AAS and ICP-AES method. With comparable sensitivity and selectivity, electrochemical methods such as voltammetry have been widely used as an interesting method for determining cadmium content, the instrumentation in this method is simpler than that of AAS and ICP-AES. The purpose of this study was to develop a method of voltammetry in determining cadmium using graphite pencil as a working electrode and parameter analysis of this method for cadmium quantification. The results of this study indicate that the voltammetric method using graphite pencil electrode can detect cadmium (II), with detection limit of 0.8 ppm, quantification limit of 2.8 ppm, linearity range from 2.8 to 4.8 ppm, and an accuracy of 0.21 -0.58%.

Abstract

The purpose of this study was to measure the levels of protein, fat, fiber, carbohydrates, water and ash, in jumpy flour from pumpkin. Pumpkin strands are part of the pumpkin fruit where the pumpkin seeds are attached to the inside of the fruit flesh. In processing pumpkin as a food, strands often used as waste or waste and is considered as a fruit that is not useful. Pumpkin plants as a minor food commodity, it turns out, are very rich in bioactive compounds which act as anti-oxidants which are very useful for human health, including spikes that have not been maximized until now due to the lack of scientific information on nutrient content and bioactive compounds in pumpkin fruit jots. This research was carried out using laboratory analysis methods with the following stages, determination and sampling, sample preparation, protein content analysis of the gunning method, fat content, fiber content, carbohydrate content and water and ash content by oven drying method. The results of the study showed that jumpy flour from pumpkin had protein content of 1.09%, fat of 5.15%, fiber of 1.27%, carbohydrate of 73.38%, water of 14.68% and ash of 4.45%. The results of this study concluded that pumpkin fruit pumpkin flour can be used as food fortification material to increase nutritional value.

Abstract

The MCM-41 synthesis has been performed starting from the dissolution stage of cetyltrimethylammonium bromide (CTAB) as a pore mold, then added tetramethylammonium hydroxide (TMAOH) and sodium sulfate salt (K2SO4) to improve the stability of the material. The crystallization process is carried out by adding sodium silicate and sodium aluminate dripwise into the solution. Decrease in pH of the solution was done by adding sulfuric acid (H2SO4 50%). The hydrothermal process is then carried out at 90°C for 36 hours, then washed, dried and calcined at 540°C. MCM-41 synthesis results were characterized using XRD, FTIR and TEM. The results showed that MCM-41 was successfully synthesized with a high degree of crystallography with 48Å lattice parameter size and had a uniform hexagonal pore structure.

Abstract

A research on physical and chemical parameters on clean water in Lalumpe village, Kombi district has been done. . Water sampling is done by purposive sampling method. Samples were taken 2 times and then analyzed the physical and chemical parameters in the laboratory of Balai Teknik Kesehatan Lingkungan dan Pemberantasan Penyakit Menular Kelas 1 Manado. From the research results, clean water samples are categorized well and feasible to use because all parameters both physics and chemistry are analyzed below the threshold quality standard required based on the standard of environmental health standard for water media for sanitary hygiene purposes
