Abstract

Semen used for artificial insemination (AI) can be prepared in different ways, fresh extended fresh or chilled, or frozen-thawed (FT). The aim of the experiment was to evaluate the use of preserved semen to inseminate the oestrus mares. Nineteen mares were used in this research. The mares were synchronized with double injection of PGF2? 14 days apart. The follicle size was monitored using ultrasound scanner during the third day of oestrus, and 2500 IU hCG was administered at the same time. The AI was conducted 35 hours after hCG injection with total motile sperm 200x106 for chilled semen and 250-300x106 for frozen semen. The result demonstrated that the response of the oestrus with double injection of PGF2? was 73.7%. The conception rate (CR) was 14.3% (1/7) with frozen semen and 42.9% (3/7) with chilled semen. It is concluded that AI with chilled semen resulted higher conception rate than frozen semen.Key words: mare, synchronization, oestrus, ovulation, artificial insemination dose, conception rate

Abstract

The aims of this experiment were to identify carbohydrases in rumen liquor of cattle, and to determine optimum temperature and pH of enzymes, as well as to evaluate the effect of metal ions and chemical substances on enzymes activity. Optimum precipitation of the enzymes from local and imported cattle was reached at the concentration of 60% and 70% of ammonium sulphate, respectively. The enzymes (xylanase, mannanase, and amylase) had optimum temperature  in the range of 50-60 °C, except for cellulase  from imported cattle  which had optimum temperature at 39 oC, optimum pH of the enzymes were 6-7 except for cellulase was at pH 4. The enzymes mostly needed methal ions as activators. EDTA and β-mercaptoethanol inhibited activity of cellulase from rumen liquor of local cattle, while enzymes from rumen liquor of imported cattle EDTA inhibited xylanase activity and β-mercaptoethanol inhibited mannanase and amylase activity.  Activity of cellulase in rumen liquor of local cattle was higher than those of imported cattle. It is concluded that rumen liquor of imported and local cattles from abbatoir contained cellulase, xylanase, mannase and amylase that most of them had optimum temperature and pH at 50-60 oC and 6-7, respectively, and they needed methal ions as activators.   Key words: cattle, rumen liquor, abattoir, enzymes, characterization

Abstract

Coleus blumei is a herbal plant used in the traditional medicine in Indonesia to expel the intestinal worm infections. Previous in vitro study showed that ethanol extract had the strongest anticestode activity compared to chloroform, hexane and aquaous extracts. The aim of the study was to evaluate the effectivity of anticestode of ethanol leaves extract againts Hymenolepis microstoma infections in mice. The plant extract was tested against H. microstoma infections in the single doses of 250, 500, 1000, and 2000 mg kg-1 body weight. Dose were administered to H. microstoma infected mice for 3 consecutive days. The efficacy of the leave extract was determined in terms of eggs per gram of feces (EPG) and worms reduction at necropsy. The results showed that the efficacy of leaves extract was dose dependent. The maximum efficacy of leave extract was observed with 2000 mg/kg dose reducing the EPG and worm counts by 55.46%-69.75% and  63.83% respectively.  The standard anticestodal drug, praziquantel at 25 mg/kg single dose revealed 100% reduction in both of EPG and worm counts. The study suggests that the leaves extract of C. blumei possesses significant anticestodal efficacy and supports its use in traditional medicine. Abstract

The present study was designed to investigate the efficacy of Bacillus circulans and Bacillus sp. as probiotic during realimentation period on performance of broiler chickens. Three hundred of 7 days-old unsexed broiler chicken of Cobb commercial strain were used in the experiment. The treatments were: control ration was fed ad libitum (P-0), ration with energy restriction at 1.5xBW0.67EM kcal/day without (P-1) and with (P-2) probiotic supplementation during realimentation period, and ration with energy restriction at 2.25xBW0.67EM kcal/day without (P-3) and with (P-4) probiotic supplementation during realimentation period. Feed restriction was applied from 7 to 14 days of age. The experiment was arranged as a completely randomized design with 5 treatments and 6 replications. The results showed that feed intake and body weight gain of chickens submitted to early feed restriction on P-1, P-2, P-3 and P-4 were significantly lower (P < 0.05) than that of chicken fed control diet. During realimentation period, body weight gain of chicken given P-1 and P-2 was lower (P < 0.05) than those obtained from P-3, P-4 and P-0 from 14 to 28 days of age. Compensatory growth only occur during the 28 to 35 days of age on P-4 and had similar performance with control at age of 42 days. It was concluded that energy restriction 2.25xBW0.67EM kcal/day for 7 days (7 to 14 days of age) with supplementation of probiotic B. circulans and Bacillus sp. (P-4) during realimentation period showed similar compensatory growth and body weight, but production index and feed cost better than control.   Key words: feed restriction,  probiotic, realimentation

Abstract

The aim of this study was to determine the effective Cr (chromium) level for Ganoderma lucidum growth in solid state fermentation of oil palm by-product. Treatments were combination of Cr level (0, 500, 1000, 1500, 2000, 25000, and 3000 ppm) and fermentation time (0, 2, 4, 6, and 8 weeks).  The treatments were allocated in a factorial 7x5 of complete randomized design with four replications. Inoculant of G. lucidum was grown in potato dextrosa agar (PDA) medium for 10 days and then innoculated to substrate which have been autoclaved and mixed with CrCl3.6H2O. The moisture of substrate was maintained at 65%. Growth media of G. lucidum was diluted with aquades and the supernatant was analysed for its Cr content.  The result showed that the addition of Cr up to 3000 ppm into the medium stimulated the G. lucidum growth in all experimental condition. The Cr ions were incorporated into the media and G. lucidum cells during fermentation. Incorporation of chromium by G. lucidum was higher in oil palm by-product substrate with 3000 ppm Cr than the others. It is concluded that Cr can be incorporated into the G. lucidum cells during fermentation. The effective level of Cr for G. lucidum growth was 3000 ppm with efficiency of Cr incorporation 68.23% in 8 weeks fermentation and chromium in protein of fermentation product was 12.01%. Abstract

The aim of this experiment was to evaluate the nutrient quality of cassava-based materials silage with cattle rumen liquor enzymes and Leuconostoc mesenteroides as poultry feed. The cassava material was hydrolyzed with cattle rumen liquor enzyme and incubated for 24 hours. The hydrolyzed product was added L. mesenteroides and ensiled in mini silo for 30 days. The experiment was designed in completely randomized design with 15 treatments and 3 replications. The result showed that temperature of cassava-based silage ranged from 26 to 30 oC. The flavor was sour and fresh fragrant and changed in color. Addition of cattle rumen liquor enzyme and L. mesenteroides bacteria significantly affected (P < 0.05) pH (3.73-4.86), dry matter(30.14%-43.28%), cyanide (86.71%-96.50%) and crude fiber content (0.78%-5.05%), but gave a fluctuate effect on protein content (-1.92%-2.39%). However, the treatment didnot affect dry matter losses (1.20%-2.66%). It is concluded that nutrient quality of cassava-based silage improved when it was added with cattle rumen liquor enzymes and L. mesenteroides by decreasing crude fiber and cyanide content. The best silage quality was obtained on tuber substrate and it increased protein KDUO (peel+leaves+tuber+tapioca waste) silage.   Key words: cassava-based silage, cattle rumen liquor enzymes, Leuconostoc mesenteroides, nutrient quality

Abstract

A study was carried out to evaluate the effect of earth worm meal (Lumbricus rubellus) added into feed additive (FA) on performance and histopathology of broilers which were infected with 1 ml/bird containing 3x108 pure culture of Salmonella pullorum orally. Broilers were tested their antibody to S. pullorum by agglutination test before the chickens were infected at 22 days of age. Treatment consisted of control without FA (R0), control with antibiotic (R1), FA contain 25% earth worm meal/EWM (R2), FA contain 50% EWM (R3), FA contain 75% EWM (R4), and FA contain 100% EWM (R5) which were periodically fed to broilers every three days at seventh days before and 10 days after infection. Treatments were arranged on completely randomized design with five treatments and four replications of four broilers each. Broilers given FA had significantly (P < 0.05) increased body weight gain and improved feed conversion ratio. Broiler treated with FA containing 25% EWM (R2) improved feed utilization compared to R1, R3, and control. Histopathology analysis showed that broilers treated with FA could reduce viscera's necrosis, especially in the liver and intestine. Lesions found in broilers which were infected S. pullorum, could be reduced by FA treatment. It is concluded that EWM can be used as feed additive to improve broilers performance.   Key words: feed additive, Lumbricus rubellus, Salmonella pullorum, broilers, histopathology

Abstract

A field experiment using Indigofera sp. was conducted at the Farm Research Station of Faculty of Animal Science, Bogor Agricultural University, Darmaga Campus, during 2008-2009. The objectives of this study were to identify the effect of defoliation time on herbage dry-matter production, protein, fiber contents, and in vitro digestibility of different vegetative parts of Indigofera sp.  Block randomized design comprising three levels of defoliation time at first regrowth after pruning (38, 68, and 88 days) with 3 replications were used in this experiment. Pruning was done 3 months after transplanted into the experimental plots. First defoliation was conducted after the plant had been pruned. Herbage was derived from different vegetative parts, i.e: leaves of branch base and all parts of shoot tips. The results revealed significant effect of defoliation time on dry matter (DM) production of both branch base and shoot tip herbages. Crude fiber (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF) and in vitro organic matter digestibility (IVOMD) of branch base were influenced significantly by defoliation time, except crude protein (CP) and in vitro dry matter digestibility (IVDMD). Defoliation time significantly affected CP, CF, NDF, ADF, IVDMD and IVOMD of herbage derived from shoot tips.   Key words: Indigofera, defoliation time, herbage quality, herbage yield

Abstract

Leucaena (Leucena leucocephala) is a feed source which has high protein and carotenoid, but its utilization as feed was limited by the presence of mimosine (β-N-(3-Hydroxy-4 pyridone)-α amino propenoic acid as a toxin. Experiment was carried out using completely randomized design with 4 detoxification treatments and 5 replications. Detoxification treatments of leucaena leaf consisted of (1) non treated leucaena leaf, (2) steamed heating at 70 oC for 15 minutes, (3)  dry heating at 70 oC for 12 h, (4) soaking with fresh water for 12 h, (5) spray with NaOH 5%, incubated  for 12 h. Variables measured were mimosine and xanthophylls contents. Data were analyzed using analysis of variance and duncan multiple range test was further done to test the significant differences between means. The results showed that soaking leucaena leaf with fresh water for 12 h gave the highest reduction for both mimosine and xanthophylls contents and steamed heating at 70oC for 15 minutes had the highest xanthophylls content post detoxification. It was concluded that steamed heating at 70oC for 15 minutes was the best detoxification methods for mimosine in leucaena leaf.   Key words:  mimosine, Leucaena leucocephala, physical and chemical detoxification

Abstract

Sheep farming is a potential livestock commodity for agricultural development in the Country, but most of the farmers are not able to manage the farm effectively, technically or economically. The aims of this study were: (1) to determine production factors that would influence the sheep production, (2) to analyze sheep farming production efficiency. This study held in Cibunian Village, Pamijahan Sub District, Bogor Regency, West Java. The data was analyzed using Cobb-Douglas production function for regression analysis. The result showed that there were four significant variables. The variables which have significant effect to the production are sum of ewe (X1), roughages consumption (X2), labour employed (X3), and farmer experience in sheep farm (D2). Based on elasticity analysis can be said that technically the number of ewe was already efficient but not economically, because the ratio MVP/MIC of ewe was 1.15 so that farmer should increase the use of this production factor. The roughages consumption was already technically efficient, but economically it was not efficient, because the ratio MVP/MIC of roughages was less than one. It means that the use of this production factor needs to be reduced.   Keywords: sheep farm, Cobb-Douglas production function, technical efficiency, economical efficiency
