Abstract

The stufy on the initiation and proliferation of somatic embryos from papaya (C. papaya L.) zygotic embryo explants was undertaken. Best results regarding initiation of embryogenic callus formation was obtained with 90 days after pollination (DAP) zygotic embryos. The medium were liquid and solid MS (0.5) medium containing glutamine (2.7 mM), 2,4-D (9 µM) and sucrose (170 mM) at 6 and 10 weeks after the start of the cultures. In this condition 90% explains produced embryogenic callus formation. The best medium for proliferation of somatic embryos was a liquid MS (0.5) medium containing glutamine (2.7 mM), sucrose (170 mM) in the presence of 2,4-D (9 µM). Using this medium, somatic embryos proliferated three times higher within two weeks.

Abstract

Estimation of cashewnut production in Yogyakarta Special District was very important in coming period, as well as with cost planning, income and marketing. This paper proposed one mathematical estimation method for estimating of cashewnut production in Yogyakarta Special District. The result shows that cashewnut production in that district would be 0.62 percent increasing every year on 1996-2001 period. Cashewnut production on 2001 was estimated to reach 315.62 ton. The correlation between estimating production (Y) with year to-t (X) on 1996-2001 had equation: Ln Y = 5.7055 + 0.0196 Ln X (r=0.74) Key word: Anacardium accidentale, L., planning area vast, productivity.

Abstract

The objective of the research was to investigate the effect of sewage application on growth and production of stevia, a sweetening alternative producing plant, as compared with manure application. Sewage is a by product of antibiotics processing of PT SBFI Sewage and manure application each consisted of 4 level, i.e. 0.0, 0.5, 1.0 and 1.5 kg per polybag. Sewage application has resulted in significant increase in tree high, leaf number, shoot number and root and shoot dry weight as compared with control. Manure application has resulted in significant increase in growth and production as compared with control. There was no interaction between sewage and manure application on all parameter observed

Abstract

Glasshouse experiment was conducted to study the effect of mycorrhizae inoculation and it's interaction with P supply on P leaf content and quality of oil palm seedling. Factorial experiment was arranged in Randomized Block Design with three replications. The first factor were mycorrhizae inoculation : without inoculation, mixed mycorrhizae, Acaulospora sp, Glomus manihotis, Glomus sp.1, Glomus sp. 2, Glomus sp. 3, The second factor were level of Roch Phosphate fertilizer : 0 g/seedling, 2 g/seedling, 4 g/seedling and 6 g/seedling. Mycorrhizae inoculation affected the P leaf content and percentage of infection significantly, Glomus sp decreased P leaf content. Acaulospora sp. gave the best quality of seedling, but Glomus sp. 3 gave the bad one. The Rock Phosphate application did not affect on P leaf content absorption efficiency and inoculation percentage due to high soil phosphorus content.

Abstract

This experiment was aimed to study the effect of biofertilizer on growth of young Robusta coffee. New planting of Robusta coffee BP 42 x BP 358 was used with Randomized Complete Block Design and 3 replications. Treatments were fertilization as follows : P1 (20 g EMAS + 50 % recognized dosage of inorganic fertilizer (d.a.p.a.)), P2 (10 ml EM 4 + 50 % d.a.p.a.), P3 j (25 g OST + 50 % d.a.p.a.), P4 (5 kg manure + 50 % d a.p.a.) and P5 (d.a.p.a., 23 g Urea, 28 g SP36 and 16 g KCI). The results showed that sum couple, length and sum node of plagiotrophic stems were significantly different from 2 till 6 month after treatment, but it were not significantly different for height and stem diameter: Manure + 50 % d.a.p.a. was the best responses for all variables. EMAS + 50 % d.a.p.a. and EM 4 + 50 % d.a.p.a. had responses as good as inorganic fertilizer: Whereas, OST+ 50 % d.a.p.a. had the lowest responses for all variables.

Abstract

The objective of the experiment was to study the effect of lAA and GA3 on vegetative growth of Dendrobium orchids. The experiment was carried out using 10 months-old orchid plants, and consists of two treatments, one with lAA of 0, 30, 60, and 90 ppm and the other with GA3 of and 100 ppm. The experiment was conducted at Baranang Siang Research Centre of Bogor: Agriculture University. Each treatment was replicated three times. Data were taken on number of leave, height, leaf size, number of roots, and number of pseudobulbs. Application of IAA up to 90 ppm increased the number of leaves, leaf size, height, number of roots and number of pseudobulbs. Application of 100 ppm GA 3 increased number of leaves and number of roots. Application of 90 ppm IAA combined with 100 ppm GA3 significantly increased number of roots and leaf size of orchid plants.

Abstract

The objective of the experiment was to observe the effect of application of paclobutrazol as growth retardant and hidogen sianamida which can act as dormancy break on growth and flowering of Mango cv. Arumanis. Experiment was conducted at IPB Experiment Station, Tajur, Bogor (250 m above sea level), from April to August 1996. Three-year-old of Mango cv. Arumanis trees were used for this experiment. Trees were planted in the field with spacing of 5 m x 5 m. Experiment using Randomized Block Design, consisted of two factors. First factor was dosage of paclobutrazol i.e. 0 ppm as control and 1000 ppm. Second factor was application time of hidrogen sianamida i.e. no treatment (control), 15 days after paclubutrazol application( HSP), 30 HSP and 60 HSP. Each treatment was replicated six times using one tree for each replication. Observation was conducted once a week on the sprouting bud number, shoot length flowering time, cluster number, and cluster size. Paclubutrazol application significantly played a role in off seson flower induction of mango Arumanis, flower bud sprouting occurred on 61-71 days after paclubutrazol treatment. Percentage of flowering on treated trees was 83.3-100%, while no flowering was observed on the kontrol trees. More-over, paclobutrazol application significantly inhibited vegetatif growth through decreasing total bud sprouted and shortening shoot length. Time of hidrogen sianamida application tended to increase total of bud sprouted To generatif growth, time of hidrogen sianamida application tended to increase cluster number and significantly increase cluster length. Application time of hydrogen sianamida 60 HSP showed vegetatif and generatif growth better as compared with other treatments.

Abstract

The isolation and regeneration of potato (Solanum tuberosum L) protoplasts have been carried out. Mesophyl cell protoplast were isolated from two dihaploid cultivars of potato BF 15 and SVP 10 leaves used four different enzymes solution. Protoplast were cultured onto four different cultures media to increase plating efficiency. Calli were then transferred to ten different regeneration media. Using cellulose RS 0.5 % and pectolyase Y-23 0.05 % protoplast yield of both cultivars were improved. Medium VKM  supplemented with 0.2 mg/l 2, 4-D, 1.0 mg/l NAA and 0.5 mg/l zeatin or 2iP were increase recovery of colonies from protoplast up to 5.9%. Regeneration medium containing zeatin did always produce more shoots than those of 2iP. Genotypes dependant regeneration frequencies have also been showed in this experiments

Abstract

The objectives of this experiment were to evaluate the possibility of using quick lime for extraction of mangosteen seed and the effects on its viability; The experiment was done in Seed Science and Technology Laboratory, Greenhouse, Bogor Agriculture University from November 1997 until February 1998. The materials were taken from smallholder plantation in Jasinga, Bogor. The treatments were arranged factorial in a group randomized block design with two factors. The first factor was 6 levels of quick lime concentration (K) : 0 g/l, 10 g/l, 15 g/l, 20 g/l, 25 g/l, 30 g/l and the second factor was 4 levels of soak periods: 0 minute, 10 minutes, 20 minutes, 30 minutes. The experiments had 24 treatments combination with 4 replicates so there were 72 experiment units. The data show that seed extraction using quick lime gave statistically significant different results compared with seed extraction without using quick lime and so the soak periods compared with no soak. Interaction between 20 g/l quick lime concentration and 30 minutes soak period gave the best seed emergence (96 %). 20 g/l quick lime concentration save higher vigour than other concentration as measured from its speed of germination, spontaneous growth, and root length. The soak periods increased seed moisture content 30 minutes soak period resulted higher viability and vigour than other soak periods as measured from its normal seed dry weigh, spontaneous growth, epycotyl length and root length.

Abstract

The objective of the experiment was to test the effectiveness of several postharvest treatments of mango at different storage temperatures. Mango cv. Arumanis at 80 % maturity was used in the experiment. A factorial experiment consisted of 2 factors was used. The two factors were storage temperature (24 - 25°C and 18 -19°C) and postharvest treatments (spermidine 1 mM; CaCl2 3 %, plastic wrapping, waxing, and control). Result of the experiment showed that plastic wrapping was the best in inhibiting weight loss, the increase of total soluble solid, ratio of TSS / acidity, the decrease of acidity. However; it also gave off-flavor. Storage at 18 -19°C was effective in inhibiting the increase of softening, total soluble solid, and the decrease of acidity
