Abstract

Cabai merupakan salah satu komoditas hortikultura bernilai ekonomi tinggi di Indonesia. Cabai lokal Karo berbatang ungu merupakan cabai yang memiliki keunggulan produksi dan pertumbuhan vegetatif tinggi, namun sangat rentan terhadap penyakit virus keriting kuning oleh begomovirus. Salah satu upaya pengendalian begomovirus ialah dengan perakitan genotipe baru yang tahan melalui induksi iradiasi sinar gamma. Penelitian bertujuan mengkaji pengaruh iradiasi sinar gamma pada benih cabai lokal Karo berbatang ungu terinfeksi begomovirus untuk mendapatkan calon genotipe mutan yang tahan. Benih dari tanaman cabai terinfeksi begomovirus diberi perlakuan iradiasi sinar gamma dengan taraf dosis 150, 200, dan 250 Gy untuk mendapatkan tanaman mutan 1 (M1). Benih sehat dan benih berasal dari tanaman sakit tanpa perlakuan digunakan sebagai tanaman kontrol. Parameter yang diamati meliputi insidensi dan keparahan penyakit, AUDPC dan deteksi begomovirus pada tanaman M1 dan benih generasi kedua (M2). Perlakuan iradiasi sinar gamma taraf dosis 150 Gy pada benih cabai terinfeksi begomovirus menunjukkan rata-rata insidensi, keparahan penyakit, dan AUDPC paling rendah dibandingkan dengan perlakuan lainnya pada tanaman mutan M1 dan M2. Berdasarkan deteksi dengan PCR pada tanaman mutan M2, dari perlakuan dosis 150 Gy didapatkan 7 calon genotipe tahan begomovirus terbanyak di antara perlakuan lainnya.

Abstract

Potensi keanekaragaman hayati flora di Indonesia belum dimanfaatkan secara maksimal sebagai pestisida nabati yang ramah lingkungan, dan mengurangi penggunaan pestisida sintetis pada tanaman. Daun Mansoa alliacea dan Allamanda cathartica memiliki potensi sebagai pestisida nabati karena secara in vitro mampu menghambat Athelia rolfsii, penyebab penyakit busuk batang pada tanaman kedelai. Penelitian dilakukan pada skala lapangan untuk menguji formula aplikasi ekstrak daun M.allicea dan A. cathartica yang berpengaruh terhadap perkembangan penyakit busuk batang, pertumbuhan dan produktivitas tanaman kedelai. Tahapan penelitian meliputi: ekstraksi, pembuatan formula dengan mencampurkan ekstrak daun M.allicea dan A. cathartica, aplikasi formula pada tanaman kedelai, pengamatan, dan pemanenan. Formula ekstrak 1.5% daun M. alliacea dan A. cathartica direkomendasikan untuk diaplikasikan karena dapat menekan penyakit busuk batang, meningkatkan pertumbuhan tanaman dan hasil panen di lapangan.

Abstract

Biological Control of Bacterial Grain Rot Disease Caused by Burkholderia glumae Using ActinomycetesBacterial grain rot disease is an important disease in rice plants caused by the bacterium Burkholderia glumae and causes yield loss up to 70%. In general, it is recommended to control this disease by seed treatment using various biological agents, including actinomycetes which produce antibacterial bioactive compounds. The aim of this study was to obtain actinomycetes strains that have the potential to control bacterial grain rot caused by B. glumae. The stages of the research included isolating and screening actinomycetes as potential biological agents against B. glumae, testing the phytotoxicity of actinomycetes on rice seeds, and testing the effectiveness of actinomycetes in controlling B. glumae on rice in the nursery phase. There were 40 actinomycetes isolated from rice plants and the rhizosphere and 17 of them had no potential as pathogens for plants or mammals. The suppression potency of the actinomycetes based on antibiosis test yielded 7 actinomycetes strains that were able to suppress the development of B. glumae and 5 strains had an inhibition zone of more than 2 mm, were not phytotoxic to rice plants and had the ability to promote plant growth. This actinomycete strain affects the growth of B. glumae and also suppresses the degree of its infection. Actinomycetes ST1, ST27, and BT23 strains were effective in suppressing bacterial grain rot in the nursery phase.

Abstract

Character Diversity of Black Rot Bacterial Strains (Xanthomonas campestris pv. campestris) on Cabbage against Mixture of Active Ingredients Azoxystrobin and Diphenoconazole Xanthomonas campestris pv. campestris is a bacterium that causes black rot on cabbage. Farmers still rely on the synthetic pesticides azoxystrobin and difenoconazole to control diseases in cabbage which are feared to have an impact on the diversity of X. campestris pv. campestris. The objective of the research was to obtain genetic and phenotypic diversity data on X. campestris pv. campestris, as well as obtaining data on the resistance response of these bacterial isolates to a mixture of the active ingredients azoxystrobin and difenoconazole. The study consisted of five stages (1) sampling on land with black rot symptoms; (2) isolation of bacteria using tissue implant technique; (3) selection of isolates by testing properties of Gram, hypersensitivity, starch hydrolysis, and pathogenicity; (4) identification using specific and universal 16S rRNA primers; and (5) analysis of genotypic diversity by in silico RFLP method and phenotypic diversity by measuring EPS weight. The results of molecular characterization and identification obtained five isolates of X. campestris pv. campestris (CLT01, CDA08, SDA02, SDA22, and SDA26). The results of the analysis of genotypic diversity showed that the five isolates had genetic diversity based on the cutting of the 16S rRNA gene DNA fragment, while phenotypically indicated different Inhibition concentration (IC) values. SDA22 isolate had the highest IC50 value and different genetic diversity compared to other X. campestris pv. campestris. The use of synthetic pesticides azoxystrobin and difenoconazole continuously for a long time is feared to have an impact on pathogenic microbes such as X. campestris pv. campestris. So that other control alternatives are needed so that there is no resistance to these pathogenic microbes.

Abstract

Yellow mosaic disease was reported for the first time in Indonesia in 2008. Its infection on yard long bean caused significant yield loss. Mungbean yellow mosaic India virus (MYMIV), member of genus Begomovirus was identified as the main causal agent. Specific and accurate detection is important for disease monitoring as part of disease management strategy. The aim of this study was to construct specific primer pairs for quick and robust detection of MYMIV using polymerase chain reaction method. A pair of primers MY1/MY2 was designed in this study to amplify part of MYMIV coat protein. In silico and in vitro test showed that MY1/MY2 primers specifically amplified MYMIV.
