Abstract

This study aims to develop a HOTS-based test instrument in measuring students' critical thinking skills on the matter of effort and energy. This research is a development research or Research and Development using the ADDIE model, namely Analysis, Design, Development, Implementation and evaluation. The subjects in this study were students of class X SMAS Gema Buwana. In the content validation test by the validator the question is classified as a valid instrument. While the validity of the items obtained 9 questions are said to be valid or feasible. The reliability of the test on the questions belongs to the reliable category with a very high reliability interpretation with a result of 0.879. To test the level of difficulty of the questions obtained 1 question with easy criteria, 2 questions with difficult criteria and 6 questions with moderate criteria. While the differentiating power of the questions obtained 3 questions with improved criteria and 6 questions with accepted criteria. The effectiveness of the distractor results in 5 answer choices with rejected criteria and 31 answer choices with accepted criteria. Based on the results of the development carried out, it can be concluded that the HOTS-based open multiple choice answering test instrument is said to be feasible to measure students' critical thinking skills.

Abstract

Chitinolytic bacteria have the ability to produce chitinase enzymes, so they have a role in degrading chitin into chitin derivative compounds. The constituent components of chitin are used by various microorganisms capable of producing chitinase enzymes. One such microorganism is bacteria. Chitinolytic bacteria are known to be able to control the growth of pathogenic fungi in plants. Fungal cell walls are composed of various complex compounds including chitin, glucans, mannans and proteins. Based on the composition of these constituents, the enzymes that can hydrolyze the fungal cell wall also vary, namely chitinase, glucanase, protease and mananase. The synergy of these hydrolase enzymes can have a good pathogenic effect in lysing the cell walls of pathogenic fungi. This study aims to determine the potential of chitinolytic bacteria as biological control agents for plant pathogenic fungi. The research was conducted at the Microbiology Laboratory, University of North Sumatra. The research method used is descriptive qualitative. The chitinolytic bacteria used were derived from shrimp shell waste. There were 10 isolates of chitinolytic bacteria tested, namely NR02, NR03, NR04, NR05, NR06, NR07, NR08, NR09, NR10, NR11 and PU01. The results showed that isolates of chitinolytic bacteria from shrimp waste were able to inhibit the fungus Fusarium oxysporum.

Abstract

Bacteria are microorganisms that are abundant in the environment. Isolation of bacteria has been carried out from various sources of water, air and soil. Soil is a material that is rich in nutrients needed by various organisms including bacteria, which are capable of producing secondary metabolites that can be utilized in various fields. Different sources of isolates will produce different characteristics of the bacteria. This research aims to determine the type and characterization of bacteria originating from burnt waste soil. The research was conducted using a qualitative descriptive method. The soil sample used is burnt waste soil. The results showed that 6 bacterial isolates from burnt waste soil had different characteristics, namely TBS01, TBS02, TBS03, TBS04, TBS05, TBS06. Microscopic identification results showed that the bacterial isolates TBS01 and TBS02 were gram-negative bacteria and the other isolates included gram-positive bacteria. Differences in morphological and microscopic characteristics indicate different types of bacterial isolates.

Abstract

This study aims to determine the difference in learning outcomes and the magnitude of the differences in student learning outcomes using the Talking Stick and Snowball Throwing learning models on the excretion system material in class VIII SMP Swasta Bina Guna Tanah Jawa. This research is a type of quasi-experimental research that is given different treatment in each class. The research sample was taken using a saturated sampling technique (census) which consisted of 2 classes, namely class VIII-1 as experiment 1 with 32 students using the Talking Stick learning model, while class VIII-2 into experiment 2 with 32 students using the Snowball Throwing learning model, total of sample is 64 students. Data analysis was carried out by finding the mean (X ), standard deviation (S) and hypothesis testing (t test) at a significance level of = 0.05. From the results of the study, the average value of post-test experiment 1 was 86.56 and experimental 2 was 85.93 with a difference in learning outcomes of 0.63. From the results of hypothesis testing (t test) obtained tcount (1.708) t table (1.669) at the significance level = 0.05 and degrees of freedom (dk) = 32, then Ho is rejected and Ha is accepted, meaning that there are differences in student learning outcomes using the model Talking Stick and Snowball Throwing lessons.

Abstract

This research aims to determine the maximum concentration of Maleat Anhidrida in grafting into the LLDPE chain with the help of Benzoyl Peroxide as an initiator. Mixing is done by the Blending method using an internal mixing tool. The titration results showed that the maximum percentage of grafting degree occurred at a concentration of 9% Maleat Anhidrida. The formation of new peaks at wave numbers 1707 and 1267cm-1 proves that grafting has taken place into the LLDPE monomer.

Abstract

Breadfruit plants have been used as traditional medicine. Parts of medicinal plants are latex, roots, bark and leaves which are used to treat fractures, sprains, diarrhea, abdominal pain, dysentery, infectious diseases, headaches, hypertension, diabetes, asthma, hepatitis, and inflammation. This study aims to determine secondary metabolites and characterization of breadfruit leaves. The research method was carried out experimentally with extraction by maceration method, and phytochemical screening on simplicia and extracts for identification of secondary metabolites. The secondary metabolites of simplicia and extracts showed the presence of flavonoid compounds, saponins and tannins. Characterization showed the presence of water content of 7.32%, water soluble extract and ethanol content of 12.29% and 8.60%, total ash content and insoluble in acid were 8.50% and 2.34%, respectively. The conclusion of the study is that the secondary metabolites of breadfruit leaves have the potential as medicinal ingredients and the simplicia characterization has met the simplicia standard.

Abstract

Acid mine drainage (AMD) formed from open pit mining can cause environmental impacts if discharged directly into the waters because it has a low pH and contains heavy metals. Attempts to neutralize the pH and absorb heavy metals in AMD can be carried out by means of phytoremediation using Eleocharis dulcis (Burm.f.) Trin. Ex Henschel. This study aims to determine the activity of PO, PPO, and CAT on the roots of E. dulcis in AMD phytoremediation and evaluate the success of AMD phytoremediation. Sampling using convenience sampling method. Measurement of metal content of Fe and Mn with atomic absorption spectrophotometer (AAS). Protein extraction using refrigerated centrifugation, measurement of total protein content with BSA, and activity of PO, PPO and CAT using a uv-vis spectrophotometer. Based on research that has been done, E. dulcis is able to increase the pH from 5.6 to 7.7 and reduce Fe levels from 7.88 mg/l to 0.25 mg/l, Mn 1.71 mg/l to 0.75 mg /l. The physiological response shown by E. dulcis in AMD phytoremediation was an increase in antioxidant enzyme activity PO 178.98 U/mg protein/minute, PPO 388.96 U/mg protein/minute and CAT 184.34 U/mg protein/minute. Based on research, E. dulcis is a plant that is tolerant to pH stress and heavy metals, so it has the potential to be used in AMD phytoremediation.
