Abstract

This study was designed to identify the effects of nicotine on the skeletal and internal organ abnormalities of mice prenatally. Seventy two female mice strain Balb/C were used. Pregnant mice on gestation days (GD) 8, 10, and 12 was injected intraperitoneally with 3, 6, and 12 mg/kg BW nicotine respectively. Control nice was injected by aguabidestylata. On GD 18, and all of pregnant mice were sacrificed, and a half of living fetuses were immersed in Bouin’s solution to observe their internal organs, while another half were immersed in 95% ethanol to observed sekeletal malformation using alizarin red S staining. Results were analyzed using SPSS program version 11 for personal computer.The results showed that nicotine caused Skeletal malformation, especially in the sternum when administered on GD 8 and 10, in doses of 3 and 6 mg/kg BW, delayed of ossification was observed in the supraoccipital and sacrocaudal bones on GD 10 and 12 and doses of 12 mg/kg BW, while in the extremitas the malformation was observed in the proximal and medial phalanx both in the fore and hind limb especially on GD 10 and 12, and doses of 6 and 12 mg/kg BW. Nicotine was effects on the brain development especially hydrocephalus was observed on GD 8 and doses 10 and 12 mg/kg BW. Another internal malformation such as ectopic renalis was observed on GD 8 and doses of 3 and 6 mg/kg BW. These finding suggested that nicotine has caused skeletal malformation, brain malformatin and ectopic renalis, when administered in mice prenatally.

Abstract

Peperomia pellucida L. Kunth herbs (Herba Suruhan) is one of the alternative for inflamation therapy. In this study, albino rats were devided into seven groups. The first three groups was administered orally the extract of Suruhan herbs 1500 mg/Kg body weight, 2000 mg/Kg body weight and 2500 mg/Kg body weight. The next three groups were given Indomethacine solutions with the doses of 13.5 mg/Kg body weight, 15.8 mg/Kg body weight and 15 mg/Kg body weight, and the last groups (control) were given 1% of PGA suspension alone. Fifty microliter of Carrageen solution of 1.5% was injected subcutaneously to all rats in those group.The volume of the foot paw of albino male rat injected with carrageen, was measured and recorded for 4 hours. The number of leucocyte and the number of differential count were observed every two hour, namely at the fourth, sixth and eight hours following carrageen injection. There were significant differences in the antiinflamation effect using plethysmometer and the number of leucocyte counts between control and all the treatment group. For the differential count, not all the results showed significant differences.

Abstract

Curcuma zedoaria, familia Zingiberaceae, has been used for along time as an alternative healing. active compounds within it are coneol, comphene, borneol, camphor, zedoarin, resin, gum, and curcumin. Curcumin is an antioxidan that can slow cell damages caused by toxicant. The purpose of this research was to find out the role of Curcuma zedoaria’s rhizome for minimizing damage of microanatomical structure of liver caused by alcoholic toxicant. Samples in the research were 24 mice (Mus musculus) Swiss Webster. They were grouped randomly into 8 groups. Each group was treated as follow. Group A (control group) was given fresh water. Group B was given whisky. Group C was given 500 mg Curcuma zedoaria/kg body weight. Group D was given 750 mg Curcuma zedoaria/kg body weight. Group E was given Curcuma zedoaria/kg body weight. Group F was given whisky and, 2 hours later, 500 mg Curcumazedoaria/kg body weight. Group G was given whisky and, 2 hours later, 750 mg Curcuma zedoaria/kg body weight. The last group was given whisky and, 2 hours later, 1000 mg Curcuma zedoaria/kg body weight. The treatment was carried out during 8 days. A microanatomy slide of liver was made by using parafin method and Hematoxylin-Eosin dying. Data was analyzed by using descriptive method, comparing between the control group and the experiment groups.The result of the research showed that the powder of the Curcuma zedoaria’s rhizome has been useful in minimizing damage of hepar microanatomy structure of the mice affected by alcohol. The effective dosage was 1000 mg/kg body weight.

Abstract

Small GTPases are monomeric guanine nucleotide binding proteins with molecular weight of 21 to 30 kD. Plants have plant-specific small GTPases, termed Rop (Rho GTPases from plants) which serve as signaling proteins in plant signal transduction. To have better understanding on nodulation process in legume, which Rop proteins that play a dominant role in nodule formation would be determined. Therefore, the expression of five Medicago truncatula Rop GTPase genes (MtRops) in young nodules were studied in this experiment. RT-PCR method was used to examine the expression of MtRops. The MtRops expression in inoculated M. truncatula plants were compared with those in uninoculated plants. Results of this studies showed that the expression of MtRop5 and MtRop6 were significantly induced in young nodules, it was suggesting that these Rops may have important role during nodule formation.

Abstract

Phanerochaete chrysosporium was known as white rot mold which could biodegrade persistent organic pollutants. In this research the ability of the mold in biodegrading textile dye direct orange S and rhemazol yellow, which contained naphtol functional group, was studied. This research included characterization of the waste water and determination of optimum concentration of the waste water for biodegradation. The optimum concentration for biodegradation was determined by measuring the radial growth of the mold in agar medium containing various concentrations of textile dye. The final stage was the application of the mold for biodegrading the textile dye in aerobic batch reactor using the selected concentration. This research showed that P. chrysosporium could grow satisfactorily in minimum medium containing synthetic textile wastewater in various concentrations. The highest colour removal efficiency of 93% was achieved in 3 days in the waste water with highest color intensity (absorbance value l520 = 4.00), and 48% in the waste water with absorbance value l520 = 1.22. COD removal efficiency of 87% was achieved within 12 days in the wastewater of 30% concentration, and 83% in the waste water of 100% concentration. The pH values decreased to 3.23 in the waste water with lower dye concentration and to 3.42 in the higher color intensity, from the initial values of 5.0-6.0. This research concluded that P. chrysosporium was capable to biodegrade naphtol textile dye with reasonably high efficiency.

Abstract

Seventeen slow loris, Nycticebus coucang were examined for the endoparasitic occurrence, four are alive and the others are cadavers. Eggs, embryonated eggs and larvae of Enterobius sp. (Nematode: Oxyuridae ) were found on the fecal examination. The adult and mature helminth of Syphacia sp. (Nematode: Oxyuridae) and Rictularia sp. (Nematode: Rictularidae), a species of the tape worm and species of the hook worm were found on the necropsy of thirteen cadavers.

Abstract

The study of secondary succession was carried out on Segara Anakan mangrove forest, since the secondary succession has been the significant problem in the mangrove forest Segara Anakan as the impact of illegal cutting and the neglected fishpond activities. Side-by-side comparisons method and quadrat sampling method were employed in this study. There were 3 areas of abandoned fishpond with different levels of succession. The sample stands were determinated subyectievely and the quadrat sample were put at random. Ordination method was used to see the correlation between vegetation pattern using environment parameters observed. The result of this study indicated that the important value on the stratum of mangrove herb was succession process. The ordination pattern of mangrove was not related to environment factors. In general, some spesies: Avicennia alba, Acanthus ilicifolius, Paspalum sp., Fimbristylis alboviridis, Bulbostylis puberula had the important role in the process of secondary succession.

Abstract

This research was an effort to reveal the effect of fruit juice of ‘labu siam’ [Sechium edule (Jacq.) Swartz] on total cholesterol and total glyceride of mice serum and also to reveal the secondary metabolites content of the juice. In this research, four groups of mice each consist of 8 was used. First group ws given normal food, the second group was given hyperlipidemic inducer. The third group was given hyperlipidemic inducer and fruit juice at dosage 0.546 mg/kg b.w. The third group was given hyperlipidemic inducer and fruit juice at dosage 1.092 mg/g b.w. The exogenic and endogenic hyperlipidemic inducers used were cholesterol suspension and propyl thiouracyl 0.01%, espectively. Both the inducer and the juice were given every day orally for three weeks. At the last day, the mice were fasted for 12-24 hours and the blood was collected intracardially. Assay of cholesterol and triglyceride in the sera was carried out using the reagent kits.Statistical analysis shows that the administration of the fruit juice of ‘labu siam’ at dosage 0.546 mg/g and 1.092 mg/g b.w. can decrease the triglyceride concentration in the sera. The dose 0.546 mg/g decreases the total cholesterol concentration, but the dose 1.092 mg/g did not show the sera cholesterol decreasing activity. Phytochemical screening for the constituents of the juice indicated that the juice contains saponin glycosides.

Abstract

Dextranase enzyme has been purified and characterized from Arthrobacter sp. B7. This enzyme was purified from the culture supernatant of Arthrobacter sp. B7 by procedure of native PAGE. The molecular size of the enzyme was estimated 72,5 kDa by SDS-PAGE. The N-terminal amino acid sequence of this enzyme determined using Edman degradation techniques were APVTADVGNLHT. SDS-PAGE and native-PAGE analysis revealed that the enzyme molecule consisted of one sub-unit.

Abstract

This research was conducted in automotive workshop of VEDC Malang and the samples were taken at the automotive machine and automotive electric workshops. The parameters which examined were the Pb concentration in the mouse blood (Mus musculus) in 2 and 3 months years old. The measurement was run for 5 times every 10 days, and the result was mean value of the increasing rate of Pb concentration in the mouse blood. The aim of this research was to determine the increasing rate of Pb concentration in the mouse blood which has the relation with different working area, different age, and also duration of exposure. Based on the data of laboratory test, research continued by T-test analysis, Simple regression test, and correlation, and also General Linear Model. By doing statistic analysis with the different working area, duration of explanation, and different age.Result of this research showed that there were the differences of rapid increasing of Pb concentration in both workshop. Generally the age which influence so much for that (in the mouse body) and have the relation with the body stamina. If the mouse body absorb amount of Pb constantly, so the Pb concentration in it will increase more and more and it cause the decreasing of the body height.
