Abstract

α-amilase is one of digestive enzyme that hydrolize starch to maltose by α-glukosidase and degradation to form a glucose and continue with blood adsorption through villi of small intestine. Consomption of acarbose drug is one of ways for diabetic treatment to inhibit the activity of α-amilase. Tamarind leaves (Tamarindus indica L) herbal benalu api (Scurrula Sp) and herbal putri malu (Mimosa pudica D) regularly used as medical plant with activity of antidiabetic medicine. The aims of this studies was to analyze the potency of three medical plant with form of infusa and ethanol extract from fresh or dried plant to inhibit activity of α-amilase and akarbose used as positive control. Inhibition potency of sample against activity of α-amilase were determine base on maltose produced by of starch hydrolysis α-amilase to reduction dinitrosalicylic acid become 3-amino-5-nitrosalicylic acid and the absorbance were measured with spektrofotometer at 530 nm. Resulted of percentage inhibition against activity of α-amilase were herbal infusa from dried benalu api 85.58 ± 2,93%, infusa of fresh putri malu 87.40 ± 1,81%, and the dried 98,85 ± 0,66%. These results did not significancy different with inhibition of akarbose 93.89 + 0,02%. Infusa herbal of dried benalu api, fresh and dried putri malu were potential cover for acarbose to used as alternative medicine.

Abstract

The study was aimed to gasp inventory an expired plant mangrove species characteristic respectively collection specimen land us function BTEX and heavy metal at growth environment. The study held from April through November 2007. Design with two classes, Rank I : survey activities and explored in land for quantity and identification, Rank II : Analysis totality solid matter petroleum level BTEX and heavy metal situ by research in location. The results of mangrove identification (main component, supporting component, and associated mangrove) showed there are 11 families and 25 species. BTEX parameter from soil sample was detected benzene intensive contamination on Andil river (16.30 ppm), Porangan river (81.30 ppm), Gombal river (56.90 ppm), Kacar river (36.90 ppm) but it was not detected on Krondo river. Toluene intensive contamination Porangan river (824 ppm) and Kacar river (896 ppm). BTEX parameter from water sample was detected benzene intensive contamination on Andil river (24.30 ppm), Porangan river (8.13 ppm), Gombal river (20.30 ppm), Kacar river (8.13 ppm), Krondo river (8.13 ppm), and Toluene contaminate slightly. Ethylbenzene and xilene were not detected by gas chromatography. Soil medium showed that contamination mainly Pb on Gombal river (5.98 mg/Kg), Kacar river (7.24 mg/Kg) and Krondo river (6.43 mg/Kg), where are Andil river and Porangan river were only slightly contaminated. The metal occurred on Cu for all, except Andil river (6.22 mg/Kg), whereas Zn and Cr did not contaminate, while Ag and Cd were not detected. Water medium was not contaminated by Pb, Cd, Cr, and Zn, furthermore Ag and Cu were not detected

Abstract

An investigation on purification and characterization of lipase enzyme production Aspergillus oryzae from copra by fermentation of oliveoil has been carried out. This enzyme can be produced by fermenting olive oil in a medium containing Aspergillus oryzae. Crude enzyme isobtained by centrifuging the medium cultures containing Aspergillus oryzae at 3500 rpm for 30 minutes and than adding 0.2 M borat buffer(pH 8.2). Enzyme activity was determined from paranitrophenol as product of lipase catalysis of paranitrophenylbutirat (0.2 M) assubstrate measured by the Vorderwulbecke method. Prepurification process was by ammonium sulphate fractionation. Precipitation60-80% ammonium sulphate produced maximum activity of enzyme. Purification by Q sepharosa FF and sephadex G-75 collumchromatography produced four and three fractions with purifity of 12.85 and 20.25 times than crude enzyme respectively. Characterizationof this enzyme showed optimum condition at pH 8.2, temperature at 350C, the Km value at 0.046 M, and Vmaks is 1.926 μmol/menit and themolecular weight at 40.7 kDa.

Abstract

This research was conducted in the Experimental Field, Faculty of Agriculture, Riau Islamic University and SoilLaboratory, BPTP Riau Province from August to December 2007. The research aim to study the effect of Cu2+, Zn2+,Fe3+ ameliorant addition in peat to N, P, K uptake and rice yield of various varieties. The experimental with CompletelyRandomized Design for Factorial 4 x 4 were used. The first factor was ameliorant addition, consists of four levels(Cu2+; Zn2+; Fe3+) and second factor based on varieties, consists of four levels (PB-42, IR-64, Ciherang, Cisantana).The result of research indicates that: (1) Interactian effect of treatment was significant to N, P, K uptake, productivestem, weight of dried seed, and weight of 1000 seeds, (2) The best treatment was Cu2+ ameliorant on IR-64 withuptake of N ( 254,0 mg/plant), P (32,8 mg/plant), K (76,0 mg/plant), productive stem (30,0 stems/plant), weight ofdried seed (63,9 g/plant), weight of 1000 seeds (23,7 g).

Abstract

Ujung Kulon National Park is one of conservation area in Banten Province. A research on bryophytes that grow onpandanus substrat in this area have been done at March to April 2008. The surrounding areas methode used onthis research. In the collections found 9 species usually growth on bark of Pandanus dubius. Two species wasreported as the important species, because one species (Fissidens teysmanianum Doz. & Molk) is endemicspecies in Java and the other (Calymperes cougiense Besch.) as abundant species in Malesia and it is only foundin Polynesia region.

Abstract

The chitosan from shell of crabs (Portunus sanginolentus L.) waste was produced from deproteinization, demineralization, depigmentation,and deacetylation steps. This process was obtained 22.66% chitosan which identified by FTIR spectrophotometry and had 65.47% degree ofdeacetylation. This chitosan was used to adsorb methylene blue dye which fit to the Langmuir isotherm with 0.0312 l mg -1 equilibriumconstant of adsorption (K) and 4.20 mg g -1 maximum capacity of adsorption (q m).

Abstract

Potatos cyst nematode (Globodera rostochiensis Woll.) in Indonesia has been find for the first time in Malang EastJava. This nematode suppress potato growth and caused yield looses up to 70.63%. Examination the ability of soilfungus Fusarium oxysporum TR1, F. solani TR2, F. oxysporum KT1, F. chlamydosporum KT2, F. oxysporum SM1,Paecilomyces lilacinus SM3, and F. chlamydosporum SM4 dalam pengendalian G. rostochiensis againsit G.rostochiensis on susceptible potato has been carried in out the green house, using Randomized Block Design withfour replication. Results showed that all of the seven fungi had the capacity to parasite G. rostochiensis. Inoculationof P. lilacinus SM3, F. chlamydosporum SM4, F. oxysporum KT1, and F. oxysporum SM1 decreased female populationto 58.56%, 59.09%, 60.76%, and 61.01% respectively compared to that without inoculation. While the other fungionly decrease female population up to 50%. The impact of male population reduction was the enhancement oftuber weight of potato. Adult male nematode was not found in this experiment.

Abstract

Proteus mirabilis is opportunistic and nosocomial pathogen that usually found in clinical specimen from patientswith catheter. The pathogenic mechanism of the bacteria are not fully elucidated especially its potential activity ofthe protein as hemaglutinin and adhesion molecule. The aim of this study is to evaluate the role of 35 kDa outermembrane protein from P. mirabilis. After identification, bacterial isolate of OMP fraction 12,5% SDS-PAGE wereused to isolate OMP followed by hemaglutinin test and invitro adhesion test. The study showed that the 35 kDa OMPof P. mirabilis was a hemaglutinin protein that could agglutinate mice erythrocytes, rabbit erythrocytes, and humangroup O erythrocytes. Hemaglutination test were negative on erythrocytes human blood group A,B, and AB. The 35kDa OMP was also adhesion protein showed by its activity to adhere to the rabbit vesica urinaria epithel receptor.The increase dose of 35 kDa OMP will decrease the amount of P. mirabilis bacteria to adhere to rabbit vesicaurinaria epithel (p< 0,05 ).

Abstract

Model selection in neural networks can be guided by statistical procedures, such as hypothesis tests, informationcriteria and cross validation. Taking a statistical perspective is especially important for nonparametric models likeneural networks, because the reason for applying them is the lack of knowledge about an adequate functionalform. Many researchers have developed model selection strategies for neural networks which are based onstatistical concepts. In this paper, we focused on the model evaluation by implementing statistical significancetest. We used Wald-test to evaluate the relevance of parameters in the networks for classification problem.Parameters with no significance influence on any of the network outputs have to be removed. In general, theresults show that Wald-test work properly to determine significance of each weight from the selected model. Anempirical study by using Iris data yields all parameters in the network are significance, except bias at the firstoutput neuron.

Abstract

The aim of research was to find the molecular characteristics of probiotic bacteria in the digestive tract of grouper fish (Chromileptes altivelis) based on 16S ribosomal DNA technique. The bacteria were isolated from stomach and intestine of the fish and were analized in the Marine Microbiology Laboratory of fishery and marine Science Faculty of Riau University. The bacterial DNA were isolated by using a PCR (polymerase Chain Reaction) and was conducted in the biotechnology laboratory of Diponegoro University, Semarang. The purified DNA was sequenced in the BPPTTangerang. Result shown that 6 bacterial species may be potential as probiotic. There were Bacillus velesensis strain CR-11, Vibio alginolyticus A3G-2, Bacillus cereus site2S, uncultured bacterium clone BB3S16S-17, Bacillus subtillus strain CICC10066, and Bacillus flexus strain LF-3. these bacteria grow well at pH 2 and this indicated one of probiotic bacteria characteristics.
