Abstract

Experimental activities is conduct in several stages, that is : somaclonal variation induction incastor oil in vitro culture through the use of PEG and selected plantlet is reproduce in vitro. In vitroselection procedure is done by planting cotyledon explants from castor oil seedlings age 10-14 daysin nutrient medium MS+BAP+IBA. Seed sterilization is done using Tween 20, Benlate, 90% alcohol,50% chloroc and sterile water.After callus is grown, callus will be sub-cultured into nutrient mediumMS+BAP with PEG addition. Selected callus will be transferred into nutrient medium MS + potassiumpantotenat + active coal until it shows root growth.Result of experiment has shown that the higher PEG concentration will suppress thegrowth of castor oil callus. In PEG concentration 15% the mortality rate of castor oil callus (LD) is64,29% and for PEG concentration 20% the mortality rate of castor oil callus is 92,86%. For thenext phase is in vitro selection process through treatment of 15% PEG toward all seven accessions.Result of this experiment has revealed that average mortality rate of callus in 15% PEG treatment is87,64%. HS-49 has shown the lowest percentage rate of response (79%) in 15% PEG treatment,while the highest percentage rate of response occurs for SP-8 (93%). Callus that succeeding growthfrom PEG selection medium will be reproduce in regeneration medium until bud is formed. Buddingconstruct percentage is range between 28,57% (accession HS-49 and SP-38) until 37,03% (accessionSM-35). Average budding construct percentage from selected callus in regeneration medium is31,45%. For the next stage of experiment (second year), those bud will be sub-cultured into rootingmedium until forming root and become complete plantlet ready to acclimatize.

Abstract

PENGARUH KONSENTRASI SUKROSA DENGAN KRIOPROTEKTAN DIMETHYL SULFOXIDE TERHADAP KUALITAS TELUR IKAN MAS (Cyprinus carpio Linn.) PADA PROSES KRIOPRESERVASIEffect of Sucrose Concentration with Dimethyl sulfoxide cryoprotectant on the Quality Egg Goldfish (Cyprinus carpio Linn.) This study aimed to examine the effect of the concentration of sucrose solution as an extender with cryoprotectant Dimethyl sulfoxide (DMSO) on the quality of fish eggs Mas (Cyprinus carpio Linn.) In the cryopreservation process, so that this study can help in the process of genetic pemulyaan parent and improved seed. The design used in this study using a completely randomized design (CRD), consisting of 4 treatments with one control, each treatment was held repetitions 5 times, so there are 25 experimental units. Treatment of sucrose concentration used was 0.5 M, 1 M, 1.5 M, and 2 M to 1 M. Testing cryoprotectant DMSO hypothize using Fingerprint Test Range. The results showed that the quality of the eggs that are morphologically goldfish in a normal condition and deserves to be the process of cryopreservation, fertilization measurement results carp eggs without treatment (control) was 80.06% and the hatching rate of 70.10%. Carp egg fertilization levels highest in treatment B 1 M sucrose concentration of 71.60%, while the lowest fertilization treatment D at a concentration of 2 M sucrose is 31.14%. Hatching or hatching highest rate at treatment B 1 M sucrose concentration of 69.89%, the lowest rate in hatching treatment D 2 M sucrose concentration of 53.78%. Sucrose is known to meet the needs of the physical and chemical energy for metabolic processes during egg storage processes besides sucrose also acts as a cryoprotectant that is not penetration into the egg cells of fish.Keywords: Sucrosa, Cryopreservation oosit fish, DMSO.ABSTRAKPenelitian ini dilakukan di Laboratorium Departemen Perikanan Terpadu Pertanian Fakultas Peternakan, Universitas Muhammadiyah Malang. Penelitian ini bertujuan untuk menguji pengaruh konsentrasi larutan sukrosa sebagai extender dengan krioprotektan Dimethyl sulfoxide (DMSO) pada kualitas ikan telur Mas (Cyprinus carpio Linn.) Dalam proses kriopreservasi, sehingga penelitian ini dapat membantu dalam proses genetik pemulyaan induk dan benih unggul. Rancangan yang digunakan dalam penelitian ini menggunakan Rancangan Acak Lengkap (RAL), terdiri dari 4 perlakuan dengan 1 kontrol, masing-masing perlakuan diadakan pengulangan sebanyak 5 kali, sehingga terdapat 25 unit percobaan. Perlakuan konsentrasi sukrosa yang digunakan adalah 0,5 M, 1 M, 1,5 M, dan dengan krioprotektan DMSO 1 M, Perlakuan D dengan konsentrasi sukrosa 2 M dengan krioprotektan DMSO 1 M. Pengujian hipotesi menggunakan Uji Sidik Ragam. Hasil penelitian menunjukan bahwa bahwa secara morfologis kualitas telur ikan mas dalam kondisi normal dan layak untuk dilakukan proses kriopreservasi, hasil pengukuran fertilisasi telur ikan mas tanpa perlakukan (kontrol) sebesar 80,06% dan hatching rate sebesar 70,10%. Tingkat fertiliasi telur ikan mas tertinggi pada perlakuan B konsentrasi sukrosa 1 M sebesar 71,60%, sedangkan fertilisasi terendah pada perlakuan D konsentrasi sukrosa 2 M yaitu 31,14%. Daya tetas atau hatching rate tertinggi pada perlakuan B konsentrasi sukrosa 1 M sebesar 69,89%, dengan hatching rate terendah pada perlakuan D konsentrasi sukrosa 2 M sebesar 53,78%. Sukrosa diketahui mampu memenuhi kebutuhan fisik dan kimiawi energi untuk proses metabolisme telur pada saat proses penyimpanan selain itu sukrosa juga berperan sebagai krioprotektan yang bersifat tidak penetrasi kedalam sel telur ikan.Kata Kunci : Sukrosa, Kriopreservasi oosit ikan, DMSO.

Abstract

Methane capture is very helpful in the development of garbage recycling system to be producedinto bio gas as a substitute for gasoline (petrol), which lately has been difficult to find.This study aimed to design a methane capture waste, low cost and good performance made ofplastic polyethilene for a small scale. This research design produces methane capture-based plasticspolyethilene with the following specifications: biodigester with a total volume of 11 m3, 8.8 m3 ofwet volume, processing time 40 days, fulling material 220 kg / day, total area 18 m2, and has a gasreservoir with dimensions of 4.6 m height, diameter of 0.954 m, the effective volume of 2.5 m3.

Antifertility men who have used the form of injections, namely testosterone, it is necessary antifertility of medicinal plants revealed that beluntas (Pluchea indica). Beluntas antifertility can be used as a drug in mice (stage pre-clinical trials) (Susetyarini, 2011). This study is a continuation of the tannins that can lower sperm fertilization potential of rat (Susetyarini, 2010). Assessment of leaf tannin in influencing consentration of beluntas spermatozoa has not been done, it is necessary to study leaf tannins beluntas against consentration of spermatozoa. Consentration of  spermatozoa role in the fertilization process. This study aims to prove the leaf tannin beluntas against spermatozoa consentration white male rats. The research used the experimental treatment control group (without giving tannin), giving tannin treatment groups with as many as 0.8 ml to a white male adult rats were repeated 3 times. Giving time for 60 days. Observations at the time treatment was stopped (0 days); discharged after treatment, rats were left for 7 days, 14 days, and 21 days. Each observation time and was taken off the white rat epididymis and giving spermatozoa. Data were analyzed using ANOVA and Duncan?s test further. Results of data analysis showed that p <0.05 means giving tannin beluntas shown to decrease sperm concentration. Highest concentration of spermatozoa when white male rats were left for 14 days after treatment was stopped Concentration lows while white male rats were left for 7 days after treatment was stoppedKey word: Tannis leaf belintas,consentration of spermatozoa, white ratABSTRAKAntifertilitas dari obat tradisional belum terungkap. Antifertilitas pria yang telah menggunakan bentuk suntikan, yaitu testosteron, maka hal ini memerlukan antifertility tanaman obat yang terungkap dari beluntas (Pluchea indica). Beluntas antifertilitas dapat digunakan sebagai obat pada tikus (tahap uji coba pra-klinis) (Susetyarini, 2011). Penelitian ini merupakan kelanjutan dari tanin yang dapat menurunkan potensi pembuahan sperma tikus (Susetyarini, 2010). Penilaian tannin daun dalam mempengaruhi konsentrasi spermatozoa beluntas belum dilakukan, maka perlu untuk mempelajari tanin daun beluntas terhadap konsentrasi spermatozoa. Peran konsentrasi spermatozoa dalam proses pembuahan. Penelitian ini bertujuan untuk membuktikan daun beluntas tannin terhadap spermatozoa konsentrasi tikus putih jantan. Penelitian ini menggunakan kelompok kontrol perlakuan eksperimental (tanpa memberikan tannin), memberikan kelompok perlakuan tannin dengan sebanyak 0,8 ml ke tikus jantan dewasa putih diulang 3 kali. Memberikan waktu selama 60 hari. Pengamatan pada waktu pengobatan dihentikan (0 hari); habis setelah pengobatan, tikus yang tersisa selama 7 hari, 14 hari, dan 21 hari. Setiap kali observasi dan diambil dari epididimis tikus putih dan memberikan spermatozoa. Data dianalisis lebih lanjut dengan menggunakan tes ANOVA dan Duncan. Hasil analisis data menunjukkan bahwa p <0,05 tannin pada beluntas terbukti menurunkan konsentrasi sperma. Konsentrasi tertinggi spermatozoa ketika tikus putih jantan yang tersisa selama 14 hari setelah pengobatan dihentikan Konsentrasi terendah ketika tikus putih jantan yang tersisa selama 7 hari setelah pengobatan dihentikan.Kata kunci: daun beluntas Tanin, konsentrasi spermatozoa, tikus putih

Abstract

Critical situation by vehicle of fast moment at turning is vehicle skidding and rolling. Both the things caused by centrifugal force. Vehicle roll to be marked upraisedly its one or two wheel of surface of road.             This research is done by measurement of torsion stiffness from one of  bar model of stabilizer and its influence to stabilizer rolling stiffness after attached at vehicle. Level of this stabilizer rolling stiffness will be seen its influence to level of angle of rolling vehicle.             From result of this research, installation of stabilizer influence at inclination of vehicle body. Angle of rolling vehicle after attached by stabilizer smaller than before attached. This condition caused difference of maximum of turning speed. After attached by stabilizer higger than before attached. Influence of installation of stabilizer to stability of rolling vehicle of this 4 wheel road take its case study at Toyota Kijang LX in road level off with turning radius 50 m. Maximum of turning speed before attached by stabilizer is 64,87 km/hour while after attached  become 69,66 km/hour ( increase 7,4 %)

Land ground used in this research is obtained from farm of Mount of Pandermanof check 227 Malang RPH Oro-Oro Ombo BKPH Pujon KPH.This Research executed at month, moon of June 2011. Data obtained to be calculated andreconciled with standart (measuring rod) is made available, way of this used for determination offunction of area of forest and determination type of appropriate crop. Perception conducted to landground drainage, floods danger, pH land ground, effective deepness, salinitas, tekstur land;ground,ramp, dry month, rainfall of per year, mean of annual temperature and mount erosion danger.Result of this research indicate that region of ex- fire of Mount of Panderman own ramp 45%,type of land ground andosol, intensity of rain 14,73 mm/rainday, drainage of land ground, danger oflight floods, pH of land ground 5,7, salinitas 0,03 mmhos/cm, tekstur land ground of dusty clay,effective deepness 120 cm, dry month (< 75 mm) 6, pouring rain/year 1390 mm, mean of annualtemperature 23,47 0C and mount danger of erosion. From data is above region of ex- fire Mount ofmade compatible Panderman and defended as protected forest (PF) with crop of acacia and bamboo.Keyword : Analyse according to farm, farm of ex- fire.

Abstract

Village Donowarih Karangploso District of Malang Regency is one of the areas that havetrouble and waste management in the central (dumped in landfills / TPA) where the existing landfillis very far from the village Donowarih with a distance of approximately more dari15 miles by roadup kontour conditions down so it can be concluded is not likely to be transported to a landfill. Sothat the Village Donowarih require a landfill area (TPA) that is close and be able to serve the communityin the surrounding area.With topography at the foot of the mountain Arjuno the Landfill to be built so the potential topollute the environment, especially water and soil in areas that have a height below the surfaceDonowarih village, so it becomes very important that a comprehensive landfill design which focuseson health aspects of the environment.With reference to kaedah comprehensive planning Landfill is expected to have DonowarihVillage Landfill that meet environmental health standards are accompanied by a simple application ofwaste regulations for the village community Donowarih so it will get additional revenue through therecycling process that accumulates.

Raya Tlogomas No 246 MalangEmail: diding.suhardi@gmail.comABSTRACTWith the increasingly shrinking reserves of fuel oil as an energy source, whereas energy needs increase, then the man demanded more creative in finding and developing environmentally friendly technology efficient in energy consumption. In the field of lighting (lighting), developing lighting technology since the invention of the incandescent lamp, TL, SL, and now LED (Light Emitting Diode). Economically, the price is still expensive LED lights, but LED bulbs have good prospects in the future, considering the LED lights have several advantages over other types, one of which is age more than 30,000 hours.? Solar power plants (solar cells) attractive used considering solar energy is a renewable energy source that is clean and available in a number of very many in Indonesia, the price of solar cells began to decline, operation and easy installation.?The use of solar cells made of copper oxide (CuO) and zinc oxide (ZnO) with H2SO4 dielectric that has been found by researchers, will be used as a source of energy for lighting this LED.?  During this LED lighting system that uses electrical energy source PLTS not use control system voltage, current, short-circuit safety and cooling LED lights, so batrai easily damaged and LED short-lived due to excessive heat, to avoid some of these then made a series of system LED lighting controller types.? After doing some research, obtained LED lamp lighting system with solar cells and their source backup batrai can work normally with time, voltage, current, power and charging process, with a series of independent light controller solar-powered LED lighting. Have been made and tested, and the ignition timing control system outages appropriate natural lighting, system voltage between 2 volts and 7 volts to batrai, the current system is between 0.5 Amp and 10 Amp to batrai, cut-off interference and LED cooling.Keywood : Controller, LED, Solar Powered.ABSTRAKDengan semakin menyusutnya jumlah cadangan bahan bakar minyak sebagai sumber energi, sedangkan kebutuhan energi semakin meningkat, maka manusia dituntut lebih kreatif dalam menemukan dan mengembangkan teknologi ramah lingkungan yang efisien dalam konsumsi energi. Di bidang penerangan (lighting), teknologi lampu berkembang sejak ditemukannya lampu pijar, TL, SL dan kini lampu LED (Light Emitting Diode). Secara ekonomis harga lampu LED masih mahal, tetapi lampu LED mempunyai prospek yang baik di masa depan, mengingat LED mempunyai beberapa kelebihan dibanding lampu jenis lain, salah satunya adalah usia lebih dari 30.000 jam. Pembangkit listrik tenaga surya (sel surya) menarik digunakan mengingat energi surya adalah sumber energi terbarukan yang bersih dan tersedia dalam jumlah sangat banyak di Indonesia, harga sel surya mulai menurun, pengoperasian dan instalasinya mudah dilakukan. Penggunaan sel surya berbahan tembaga oksida (CuO) dan seng oksida (ZnO) dengan dielektrikum H2SO4 yang telah ditemukan oleh peneliti, akan digunakan sebagai sumber energi bagi lampu penerangan LED ini. Selama ini sistem penerangan LED yang menggunakan sumber energi listrik PLTS belum menggunakan sistem kontrol tegangan, arus, pengaman hubung singkat dan pendingin lampu LED, sehingga batrai cepat rusak dan lampu LED berumur pendek karena panas yang berlebihan, untuk menghindari beberapa hal tersebut maka dibuatlah rangkaian sistem pengendali lampu penerangan jenis LED. Setelah dilakukan beberapa penelitian, didapatkan sistem penerangan lampu LED dengan sumber sel surya beserta batrai cadangan dapat bekerja dengan normal sesuai waktu, tegangan, arus, daya dan proses pengisian, dengan rangkaian pengendali lampu penerangan LED independent bertenaga surya. Telah dibuat dan diuji, sistem kendali waktu penyalaan dan pemadaman sesuai pencahayaan alam, tegangan sistem antara 2 Volt dan 7 Volt ke batrai, arus sistem antara 0,5 Amp dan 10 Amp ke batrai, cut-off gangguan dan pendingin LED.Kata kunci : Controller, LED, Bertenaga Surya

Contained in the waste leachate has elements glokosa (C6H12O6), Amino Acid and Ammonia (NH3), which will help the growth of anaerobic  bacteria  that  produce  methane  (CH4)  and  carbon  diaoksida  (CO2).  Humidity  will  be reduced as the discharge of garbage in garbage leachate resulting decline in productivity of anaerobic bacteria, thus maintaining levels of humidity (moisture content) is very important in stabilizing the productivity of anaerobic bacteria.In  the  humidity  of  59.85%  productivity  waste  anaerobic  bacteria  produce  landfill  gas  in stagnant in the  range  of  less  than  10%  CH4.  With  a  circulation  process  of  garbage  leachate  to maintain moisture of 67.25% are able to stabilize the production  of landfill  gases, especially CH4 concentration more than 25%. Leachate circulation periodically able to stabilize the moisture content of waste, from which the waste water content, it makes the productivity of stable anaerobic bacteria produce methane gas in concentrations between 20% -25% is quite a power plant of energy.Renewable energy was from waste to be more realistic to be implemented in full scale in place of final processing (TPA) / Landfill.  So the need for energy for the benefit of the power plant could help meet the electricity needs of the community.Keywords: bioreactor anaerobic, leachate circulation
