The detection of plantaricin-encoding genes and amino acid profile in Lactobacillus plantarum AKK30 isolated from Indonesian native chicken. Biodiversitas 21: 5792-5799. Plantaricin-encoding genes of lactic acid bacteria (LAB) Lactobacillus plantarum AKK30 which was isolated from Indonesian native chicken has been characterized using DNA sequencing technology. In order to substitute antibiotics for chickens and enhance probiotics in their gut, characterization of the indigenous probiotics in the chicken should be conducted. A potential probiotic in native chicken gut is Lactobacillus plantarum AKK30. Genes that harbor the bacteriocin biosynthesis of the strain were reported for the first time. The bacterium has diverse plantaricin genes, i.e., plnA, plnEF, plnJ, plnK and plnO. According to the dendrogram, the five plantaricin genes were closely related to plnA, plnE, plnF, plnJ, plnK and plnO genes from L. plantarum WCFS1, L. plantarum C11, and L. plantarum V90. Intriguingly, besides various plantaricin-encoding genes, another important probiotic property in L. plantarum AKK30 is cholesterol-lowering activity. Amino acid profiling also indicated proteolysis activity and probiotic potential, for instance, GABA production. Thus, L. plantarum AKK30 may be added in chicken broiler’s diet to provide probiotic benefits.

Multivariate analysis unravels genetic diversity and relationship between agronomic traits, protein, and dietary fiber in yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.). Biodiversitas 21: 5662-5671. Yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.) is a vegetable legume that possesses the highest amount of protein and total dietary fiber (TDF) content among other vegetables. However, protein and TDF content diversity are found among yardlong bean genotypes. Genetic improvement to produce cultivars with high yield and improved nutritional content is required. This study aimed to analyze genetic diversity and identify the relationship between important agronomic traits from 21 yardlong bean genotypes assessed by multivariate statistical analysis. The experiment was arranged in alpha design with three replicates, each consist of six blocks. Six pod-related important agronomic traits, protein, and TDF content were observed in this study. Total pod weight, pod weight, and pod length were found to be negatively correlated with protein and TDF content based on correlation and principal component analysis (PCA). Although negatively correlated, several variables possess a large and positive direct effect on protein and TDF content, such as the number of pod, pod weight, and days to flower. D2 analysis and Tocher’s method classify genotypes used in this study into five clusters with diverse traits value. Information from this study laid a foundation for the improvement of important agronomic traits as well as nutritional content in yardlong bean.

Abstract. Novianti V, Indradewa D, Maryani, Rachmawati D. 2020. Selection of local swamp rice cultivars from Kalimantan (Indonesia) tolerant to iron stress during vegetative stage. Biodiversitas 21: 5595-5606. Kalimantan with the large swamp area is a potential region in Indonesia to develop local rice cultivars with high tolerance to iron toxicity in tidal lowlands. This research was conducted to select iron toxicity-tolerant in local Kalimantan-rice cultivars, namely: Amas (AMS), Pandan Ungu (PDU), Kambang (KMB), Suatek Merah (SM), Siam Unus Mayang (SUM), Siam Unus Kuning (SUK), and Siam 11 Panjang (S11P). As control, two rice cultivars: Ciherang (iron toxicity-susceptible) and Inpara 5 (iron toxicity-tolerant) were also used. The seeds were grown hydroponically for 35 DAP (days after planting) in Yoshida nutrient solution containing 0 ppm (control), 100 ppm (low), and 400 ppm (high) FeS2 as pyrite treatments. Morphological analysis (plant height, leaf length and width, and leaves number) showed that PDU and KMB cultivars have better characters for tolerating iron toxicity. The highest level of chlorophyll and carotenoid contents during iron toxicity treatment was observed in KMB cultivar. AMS cultivar had the highest proline level under high iron stress. Antioxidant enzyme analysis showed that AMS, PDU, and KMB cultivars had a high percentage increase in the activity of superoxide dismutase (SOD), hydrogen peroxide (H2O2), and ascorbate peroxidase (APX) enzymes when exposed to high pyrite conditions. Based on morphophysiological characteristics, biochemical component, and enzymatic antioxidant activity, it can be concluded that AMS, PDU, and KMB cultivars are potential candidates as iron toxicity-tolerant rice.

Garut using in vitro culture methods. Biodiversitas 21: 5845-5849. Several efforts have been made to preserve Citrus reticulata var. Tawangmangu and Citrus reticulata var. Garut as indigenous Indonesian mandarin cultivars, including in vitro tissue culture methods. This study aimed to determine growth differences of the stem explants of C. reticulata var. Tawangmangu and C. reticulata var. Garut,which planted on the same Murashige and Skoog (MS) media. The treatment groups were derived from different explants, grown in 4 separate culture bottles for 35 days at 25°C. The observed parameters included the percentage of explants indicating callus development browned-colored explants, and the contaminated explants. Among C. reticulata var. Tawangmangu explants, 23.53% indicated callus development, 29.42% were browned-colored explants, and 0% indicated contamination. In contrast, among the C. reticulata var. Garut explants, 0% indicated callus development, 7.14% brown-colored, and 7.14% indicated contamination. The stems explants from C. reticulata var. Tawangmangu showed a tendency to develop calluses, but the explants of C. reticulata var. Garut was able to support the growth of shoots. C. reticulata var. Tawangmangu and Garut mandarin stem explants showed differences of shoot growth because physiological conditions varied according to the variety.

Diversity of mollusks (bivalves and gastropods) in degraded mangrove ecosystems of Kolaka District, Southeast Sulawesi, Indonesia. Biodiversitas 21: 5884-5892. Mollusks are one of the mangrove organisms whose classes are bivalves and gastropods. It plays an important role in mangrove and marine ecosystems as filter feeders, predators, and herbivores. This study aims to knows the diversity and abundance of mollusks (bivalves and gastropods) in several mangrove ecosystems in the Kolaka coastline as well as the similarity of these locations. This study was conducted in mangrove ecosystems of Induha Village, Mangolo Village, Tahoa Village, and Towua Village of Kolaka District, Southeast Sulawesi, Indonesia, from July to August 2019. This is a transect method stretched along a 100 m line perpendicularly from the seaward. The size of the mollusks subplot was 1 m2 and placed along the line transect. Each line transect comprises 10 subplots. To analyze the diversity index, evenness index, and its abundance, Kaleida Graph 4.0 version was used. This research indicates that the mollusks consist of 4 families of bivalves with 6 species and 10 families of gastropods with 182 species. It also found out 23 species of mollusks scattered to each location. The molluscan species which spread in all four mangrove ecosystems were Terebralia sulcata, Nerita planospira, and Batillaria multiformis. In Induha, the mollusks species were Anadara notabilis and Drupella margariticola. Meanwhile, Saccostrea cucullata, Pirenella incisa, Clithon oualaniensis, and Clithon pulchellum were only found in Towua. The diversity index of bivalves in each location was categorized as low diversity index category, as well as gastropods were categorized as medium diversity index. The highest diversity index of gastropods was in Induha (H' = 1.96). It was supported by the good mangrove ecosystem for mollusks' habitat. The lowest diversity index of gastropods was in Towua (1.41). This research depicts that three kinds of species with high abundance rate whose rates were >1 ind/m2are located in two different locations namely; P. incisa (3.9 ind/m2) and S. cucullata (3.2 ind/m2) in Towua and followed by B. multiformis (2 ind/m2) and Isognomon ephippium (1.2 ind/m2) in Mangolo.

Abstract. Akwu N, Naidoo Y, Singh M, Thimmegowda SC, Nundkumar N, Lin J. 2020. Isolation of lupeol from Grewia lasiocarpa stem bark: Antibacterial, antioxidant, and cytotoxicity activities. Biodiversitas 21: 5684-5691. Lupeol a pentacyclic triterpenoid, was extracted from the stem bark of Grewia lasiocarpa. Stem bark was macerated with chloroform (CHCl3) at 25 ± 2°C, and then purified using a silica gel column. The mobile phase was hexane (100%) and hexane: ethyl acetate (80:20) as the mobile phase. The structure of lupeol was identified by 1HNMR, and 13C NMR spectral data, compared with reported data. The antibacterial, antioxidant and cytotoxicity activities of lupeol were evaluated using agar-well diffusion against six clinic bacteria isolates, namely Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853), Klebsiella pneumoniae (ATCC 314588), and Salmonella typhimurium (ATCC 14026) Gram-positive bacteria: Staphylococcus aureus (ATCC 25923), methicillin-resistant Staphylococcus aureus (MRSA) (ATCC BAA-1683). The antioxidants assays were performed using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP). Cytotoxicity activity was carried out by MTT (3-(4,5-dimethylthiazol)-2,5-diphenyl tetrazolium bromide) assay was carried out against HEK293 (human embryonic kidney), HeLa (cervical carcinoma), and MCF-7 (breast adenocarcinoma) cell lines. The concentration range of lupeol was 240-15 µg/mL. Lupeol at the concentration of 240 and 120 µg/mL have no antibacterial activity against Methicillin-resistant Staphylococcus aureus (MRSA) (ATCC BAA-1683). The antioxidant activity was dose-dependent, while cytotoxic activities were observed in HEK293, HeLa, and MCF-7 cells. This study was the first report on the extraction, isolation, and biological activities of lupeol, from the stem bark of G. lasiocarpa.

Comparison of Agathis philippinensis resin tapping and collection practices in three selected sites in the Philippines. Biodiversitas 21: 5595-5604. This study documented and compared almaciga (Agathis philippinensis Warb.) resin tapping and collection practices in three sites namely Mt. Hamiguitan, Governor Generoso, Davao Oriental; Mt. Mantalingahan, Brooke’s Point, southern Palawan; and Puerto Princesa Subterranean River National Park, Marufinas, central Palawan. Resin tappers, mostly male belonging to a local organization, were interviewed, and tapping sites were visited. Tapping and collecting A. philippinensis resin is one of the supplemental income sources of the respondents from the three sites. Market influences, and how tappers learned the trade could partly explain the similarities and differences in tapping practices. Discrepancies in selecting trees to be tapped, tools used, type of incisions made on the bark, and how resin was harvested were noted. Differences in resin appearance across sites were observed, and the existence of uncollected and wasted resins clinging to trees or sprawling at its base was found in all sites. Removing dirt from the portion of the stem to be tapped, covering the bark incision with plastic for protection, and contriving handmade resin baskets from plant parts available in the site were some practices found to be worth replicating. On the other hand, there were damaged or dying trees due to the intensity of tapping, the lack of tapping skills, and limited attention given to the health of trees. There is a need for strengthening tappers’ organizations for increased bargaining power to secure better resin price and policy support to prioritize indigenous peoples in tapping into their ancestral domain. Tappers can benefit from cross-visits and other learning opportunities for sharing lessons and experiences to improve tapping practices for better resin quality and higher prices.

Biodiversitas 21: 5632-5639. Oil palm is one of the plantation commodities which has an important role in Indonesia’s economy. Indonesia is the largest producer and exporter of oil palm in the world with its export value capable of supplying 55.78% of global consumption. Rats are wild animals that are important pests for human life, both in agriculture plantations and urban areas. Rats can damage crops at various stages of plant growth. This study was aimed to determine the species abundance of rats in oil palm plantations, calculate the severity of crop damage due to rats infestation. Three blocks of mature plants were used in OC and OH plantation blocks were used to calculated, crop damage caused by rats, Identify the species of rats, and estimated rats abundance. The results of these studies showed there were four species of rats found in the oil palm plantation, namely Rattus tiomanicus, R. tanezumi, Rattus sp, and Maxomys hellwaldii. However, only R. tiomanicus and R. tanezumi were dominant species. There were significant differences in the number of, sex, and body weight between R. tiomanicus and R. tanezumi. The OC 12 block had the lowest infestation intensity than the other blocks since it was well maintained. The OH 02 block had the highest infestation intensity, infestation area, and abundance due to a lack of maintenance.

Biodiversitas 21: 5605-5613. This study aims to determine the biomass of reef shark and fish in South Morotai Waters in North Maluku, Indonesia. The Audible Stationary Count and Underwater Visual Census method were used to collect the data of reef sharks and fish. Seven and one site locations of coral reef and shark ecosystems were surveyed and, the data were analyzed using regression analysis to obtain a correlation between the variables. The three reef sharks found were Carcharhinus melanopterus, Triaenodon obesus, and Carcharhinus amblyrhynchos. Biomass estimation of reef sharks and fish ranged from 0.59 to 19.97 kg/ha and 30.95 kg/ha to 49.92 kg/ha, consisting of 8 families. In the area of aggregations, both species were found in 7 sites, and the population of reef shark amounted to 86.96 kg/ha and fish was around 55.705 kg/ha, consisting of 9 families. There was a positive relationship between the biomass of reef shark and fish in South Morotai waters, and the index of determination was 0.8043, showing that the biomass of reef shark was influenced by that of fish. These results indicated the importance of biomass in determining the functional composition and diversity of reef shark and fish.

An assessment of the reproduction, predation, and nesting behavior of Sulawesi Masked-owl (Tyto rosenbergii) in oil palm plantation: A case study of West and Central Sulawesi, Indonesia. Biodiversitas 21: 5685-5689. The quality and quantity of yield in palm plantations are reduced due to inevitable factors such as pests, diseases, and weeds. Furthermore, owls (Tytonidae) play a role as potential predators to control rats, the major pest. Therefore, this study aims to observe the reproduction patterns, predation, and nesting behavior of Tyto rosenbergii. To achieve this, direct observation and the installation of a camera trap in the nest box was adopted. Furthermore, a predation test was carried out by feeding the owls with rats daily in a cage measuring 4m x 8m x 6m. The results showed that the number of eggs produced ranged from 2 to 6, with an average of 3.85. Also, the Percentage of hatching ranges between 0-100% with an average of 80.77%, and that of chicks that survive to the fully feathered is 51%. In addition, T. rosenbergii was able to prey on 1-4 rats per night, and its activity mostly occurs outside the cage than inside, except when they had a reproductive season.
