Cytochrome-b mitochondrial DNA characteristic from non-invasive samples of wild population Javan Banteng (Bos javanicus d’Alton, 1823). Biodiversitas 20: 350-355. Javan banteng (Bos javanicus javanicus) is one of the protected big mammals in Indonesia, due to its population decline. The main populations of Javan banteng are currently in Ujung Kulon National Park (UKNP), Alas Purwo National Park (APNP), Meru Betiri National Park (MBNP) and Baluran National Park (BNP). These four remaining populations are fragmented with wide geographical distances. Population reduction and fragmentation between populations can promote depletion of genetic characters. This study aim to examine the genetic characteristics of Javan banteng based on cytochrome b region of mitochondrial DNA. Genetic materials were obtained by a non-invasive method (feces) from four national parks in Java.The sequencing results were aligned with cytochrome b mitochondrial DNA sequences from the other banteng (B. j. lowi and B. j. birmanicus) from the GeneBank sequence database. The results from this study revealed that the genetic distance of Javan banteng between UKNP, APNP, and BNP were 0, while the genetic distance between these populations with MBNP was 0.005. Moreover, the population of MBNP has only one haplotype that does not exist in the other populations. Based on these results, there should be caution in conducting genetic infusion of Javan banteng in their natural populations, especially in Meru Betiri National Park.

Biodiversitas 20: 356-363. Qualitative and quantitative variations of biochemical compounds in plant organs is one of the causes leading to infestation by a certain pest which prefers to feed on particular genotypes than other genotypes. Objectives of this study is to find out the relationship of selected agronomical features and seed biochemical contents of soybean genotypes with their tolerance to Nezara viridula and also the preference of N. viridula to these genotypes. A total of 50 soybean germplasm were grown at Muneng research station during the dry season of 2016. The variability of the tested germplasm was reflected in the diversity of agronomic characters and seed biochemical contents. There was a significant positive correlation among the studied seed biochemical parameters, namely protein, methionine and cysteine contents. The protein and cysteine contents did not relate to N. viridula preference, but it was related to soybean tolerance against N. viridula. It was indicated by the insignificant positive correlation of protein and cysteine contents with the number of unfilled pods and the negative correlation of protein and cysteine contents with the ratio of number of unfilled pods to the total number of pods. Methionine content increased N. viridula preference, but it had no effect on soybean tolerance. Five genotypes, namely MLGG 0164, MLGG 0195, MLGG 0103, MLGG 0896 and MLGG 0196 were selected based on high seed biochemical contents which can be used as gene sources in soybean breeding programs. Among these five genotypes, MLGG 0195 had high protein, methionine and cysteine contents, but it was not tolerant to N. viridula. Hence, this genotype should be cultivated by controlling N. viridula optimally. MLGG 0196 had the highest methionine content, relatively high cysteine content, and N. viridula tolerance. This genotype can be used to improve seed biochemical content and plant tolerance against N. viridula.

Hemelda NM, Safitri R, Suhandono S. 2019. Genetic diversity of Ralstonia solanacearum, A phytopathogenic bacterium infecting horticultural plants in Java, Indonesia. Biodiversitas 20: 364-372. Ralstonia solanacearum is a phytopathogenic bacterium causing bacterial wilt disease which has been reported to infect many important plants in Indonesia. However, less study has been done on its genetic diversity in Indonesia. This study aimed to investigate the genetic diversity of R. solanacearum virulent strains isolated from Java Island. BOX-PCR fingerprint was carried out to investigate the genetic pattern of R. solanacearum isolates, while phylogenetic analysis of mutS gene was performed to determine the genetic diversity of R. solanacearum isolates. A total of 21 isolates was obtained from four crop species: twenty isolates identified as phylotype I while one isolate identified as phylotype II. Based on BOX-PCR, most of the isolates clustered according to their original provinces, indicating site-dependent distribution pattern. However, BOX-PCR also detected site-contaminations indicated by the similar genetic patterns found in two provinces. Phylogenetic analysis of mutS gene discovered that most of the phylotype I isolate showed 100% similarity to each other, while phylotype II isolate belonged to phylotype IIB and showed 100% similarity to IPO1609 strain. Pathogenicity and biovar test were confirmed that the phylotype IIB isolate belonged to the R3bv2 strain which was adapted in low temperature. This study provided the first description about genetic diversity patterns of R. solanacearum strains in Java Island and revealed new challenges related to how to prevent contamination of R. solanacearum from one province to another, as well as the phylotype IIB strain infection in a highland area in Indonesia.

Gac (Momordica cochinchinesis) plays important roles in Vietnamese cuisine and medicine. It has high nutrient and medicinal values. Demand of gac fruit as ingredient for food processing and raw material for health industry requires accelerated gac cultivation. However, gac quality is variable due to the uncertain available germplasm and its management as breeding stock. Identification of gac cultivars and landraces in Vietnam is mainly based on personal experience relying on morphological traits, thus the conservation and breeding have low impact on productivity and quality of product. Recently, the development of molecular markers has been useful in identifying specific plant(s). In this study, genetic richness and relativeness of 14 gac accessions collected from different provinces in Southern Vietnam were evaluated by using 10 Inter-Simple Sequence Repeats (ISSR) markers. Results revealed large variation in genetic background of studied gac accessions. Seven DNA-based markers with potential to differentiate gac genotypes were recorded. The results provide molecular biological information for classification, identification plant origins, breeding and conserving programs of gac in Vietnam.

Genetic and phenotypic diversity, heritability, and correlation between the quantitative characters on 30 sweet potato germplasms in Lampung, Indonesia. Biodiversitas 20: 380-386. Local food commodities such as sweet potato is an alternative rice substitute food which has high nutritional content. Estimating the value of genetic diversity, heritability and correlation between quantitative characters with the weight of large storage root per plant, is needed in the selection program for the development of quality sweet potato varieties. The purpose of this study was to find out information about genetic diversity, heritability, and correlation between quantitative characters with the weight of large storage root in 30 local Lampung sweet potato germplasm, introduction and national superior genotypes. The study was carried out in the Politeknik Negeri Lampung experimental garden from September 2017 to January 2018. Using 30 genotypes of sweet potato germplasm. The study was arranged in a randomized block design (RBD) with two replications. The results showed that all characters (weight of large storage root, number of large storage root, storage root length, storage root diameter, vines length, segment length, vines diameter, and weight of vines) had a value of genetic diversity, whereas wide phenotypic diversity and has a high value of heritability. Genetic factors more influence all characters in this study compared to environmental factors. The results of the correlation analysis showed that the characters of the weight of large storage root, storage root length, and storage root diameter were positively correlated with the weight of the storage root. Whereas, the length of vines is negatively correlated with the weight of large storage. The character of the segment length, vines diameter and weight of vines did not correlate with the weight of large storage root. The character of the segment length, vines diameter and weight of vines did not correlate with the weight of large storage root. Cluster analysis results from 30 sweet potato genotypes were grouped into 13 clusters. Based on similarity of character and provenance. The germplasm of 30 Sweet potato genotypes found in Politeknik Negeri Lampung can be selected and crossed to create the desired superior characters.

Relative condition factor, length-weight relationship, and growth of three-spotted flounder, Pseudorhombus triocellatus (Bloch & Schneider, 1801) from Parangipettai Coast, India. Biodiversitas 20: 373-379. The three-spotted flounder, Pseudorhombus triocellatus is usually caught as trawl by-catch from Parangipettai coastal waters but regrettably, limited information on its age structure and growth is available. Hence, the present study aims to understand some basic parameters of its biology and growth. A total of 759 specimens (360 males and 399 females) ranging from 4 cm to 13 cm in total length (TL) and 5.26-35.41 g in weight were collected from the trawl catches of Parangipettai and Mudasalodai landing center during January–December 2015. The estimated growth coefficient (b) value for males (2.797) was greater than 3 and for females (3.023) was less than 3. The relative condition factor (Kn) values of females were higher than males except during February, June, and September. The values of asymptotic length (L?) obtained in this study using various methods didn’t vary significantly (P>0.05). The most appropriate values of asymptotic length (L?) for males were 137.11 mm and for females were 141.75 mm. The most appropriate values of growth rate (K) for males were 0.397 yr-1 and for females were 0.410 yr-1. The estimated length at age 0 (t0) for males and females were 0.197 mm and 0.292 mm, respectively. The longevity (tmax) of males and females P. triocellatus were 2.04 years and 2.24 years, respectively. The result of this study provides basic information for future studies.

Abstract. Anita S, Haryono, Wahyudewantoro G. 2019. Nutritional component of Barbonymus balleroides: A wild fresh water fish from Indonesia. Biodiversitas 20: 581-588. Barbonymus balleroides is a native species of fish from Indonesia that threatens by overfishing activity and habitat damage whereas little is known about its nutritional information. This study aimed to evaluate the nutritional content of wild barb fish that was collected from two different sites in Indonesia. Fishes were collected from Serayu and Cipunagara River. The body part of fishes was grouped and weighted to identify the ratio. Proximate composition was determined for flesh and byproduct parts. Flesh parts of fish were then examined for mineral content, fatty acid, and amino acid profile. Barb fish samples mainly form Cipunagara River contain high protein compare to several Indonesian freshwater fishes that commonly consumed. Flesh samples from both fish group were the heaviest part followed by its edible byproduct that often wasted. The amount of moisture and protein were higher in the flesh parts while byproduct showed higher content of fat and ash. Both samples contained a dominant amount of macro elements K and P. Fatty acid profile of flesh samples showed essential fatty acid that is good for human health. Amino acid profile showed the most dominant amino acid found in both samples was glutamic acid.

Short Communication: Population and stand structure of Cinnamomum sintoc in the Low Land Forest of Mount Ciremai National Park, West Java, Indonesia. Biodiversitas 20: 1042-1047. Cinnamomum sintoc (C. sintoc) is one of the anthelmintic and anti-inflammatory medicinal plants which produces essential oils from its roots, skin, and leaves. This study aimed to analyze the population and stand structure of C. sintoc in Mount Ciremai National Park, West Java, Indonesia. Data collection was carried out using plots/ strips method. The plots were purposively placed and made at each altitude and direction of the slope. The research recorded 804 individual trees from each growth edge. Individual density between slope directions and altitudes was not significantly different for all diameter classes, but was significantly different between slope directions of stratum B (P = 0.001) and E (P = 0.012). These results indicate that sintoc population is spread evenly and can be cultivated at various slope and altitude directions and has no preference for the two topographic variables.

Ratnaningrum YWN, Kurniawan A. 2019. Floral structure and genetical differences of sandalwood variants in Gunung Sewu (Java, Indonesia), and its effects on breeding systems and reproductive ability. Biodiversitas 20: 393-404. Our preliminary studies reported that the failure on rehabilitation program of sandalwood, an endangered endemic species in Indonesia, was caused by low viability and survival due to reproductive failure. New sandalwood landraces in Gunung Sewu Geopark, Java island consist of three variants (YBF, refers to "yellow big flower"; RBF, "red big flower"; and RSF, "red small flower") differed by floral structures. This study was made on three sandalwood variants grew in four landraces representing landscape zones in Gunung Sewu, from April to September 2017 flowering season. This advanced study was aimed to estimate the differences in floral structures and genetic diversity among variants, and their effects on breeding systems and reproductive ability. Floral organ measurements were made on each variant. Isoenzyme analysis was conducted to estimate the genetic diversity of each variant and in each site. Mating systems were estimated by Index of Incompatibility (ISI) and Cruden's Out Crossing Index (OCI) methods. Reproductive ability was measured by counting Pollination Effectiveness, Reproductive Success and seed viability. Results found that six loci were polymorphic in most of sites and variants, with exception for Petir and Bejiharjo sites and YBF variant. Observed heterozygosity varied with sites but was similar among variants. Some of diversity existed among both sites and variants. The OCI value scored more than 3 for all variants, indicating an outbreeding mating system. RSF showed higher OCI value compared to both RBF and YBF. Bleberan and Nglanggeran, the outcrossed and completely self-incompatible populations (ISI = 0), failed to produce selfed seeds. In such highly outcrossing, self-incompatible populations, the highest seed set was gained from intraspecific-crossed pollination. Contrastly, the inbreeding and self-compatible populations (ISI = 3 to ?), Petir and Bejiharjo, tended to alter its matting system to be more inbreeding. Reproductive ability differed by sites but was similar among variants.

Choosa-Nga P, Sangwanit U, Kaewgrajang T. 2019. The Arbuscular Mycorrhizal Fungi’s diversity in Fabaceous trees species of Northeastern Thailand. Biodiversitas 20: 405-412. The species diversity of arbuscular mycorrhizal (AM) fungi, found in three Fabaceous tree species, namely, Dalbergia cochinchinensis, Pterocarpus macrocarpus and Xylia xylocarpa, was studied. Sixty rhizosphere soil samples were collected from the tree species plantations, and AM fungal spores were extracted and identified using their morphological characteristics. The spore density and species diversity of the fungi associated with these Fabaceous tree species was assessed. The 11 AM fungi morphospecies identified were Acaulospora (3 species), Glomus (6 species), Sclerocystis (1 species) and Scutellospora (1 species). Meanwhile, three species remained unidentified, which Unknown sp.2 was frequently found in all the three of plantations. The Shannon-Weiner diversity index of the fungal species ranged from 1.40 to 1.88, while the Evenness index ranged from 0.58 to 0.76. The diversity and Evenness index was the highest in the D. cochinchinensis plantation and lowest in the X. xylocarpa plantation. These differences may be due to differences in soil characteristics, including moisture, % organic matter, and nutrient composition. The species composition of AM fungi in the P. macrocarpus plantation was not similar with the D. cochinchinensis and X. xylocarpa plantations. It was indicated that different plant species were hosts to different AM fungal communities. Therefore, further studies are required to select the appropriate AM fungal species which have a positive effect on the growth of the three Fabaceous species when we would like to do the inoculating programs.
