Detection of transovarial transmission of dengue virus in Aedes spp. (Diptera: Culicidae) from Brontokusuman Village, Yogyakarta, Indonesia. Biodiversitas 20: 2061-2067. Dengue Hemorrhagic Fever (DHF) is an infectious vector-borne disease. It is caused by dengue virus infection through the vector of Aedes spp. Dengue virus can be transmitted transovarially from infected female mosquitoes to their eggs. The presence of transovarial transmission allows maintenance of serotypes of dengue virus in nature. Detection of transovarial transmissions can be used to predict disease outbreaks as an effort to control the dengue virus. Dengue control is generally carried out through control of dengue vectors (mosquitoes) with environmental management, the use of chemical agents (insecticides) and biological control. This study was conducted to determine the existence of transovarial (vertical) transmission, factors affecting it and the resistance status of Aedes spp. against cypermethrin insecticide (pyrethroid) in Brontokusuman Village, Mergangsan Sub-district of Yogyakarta, Indonesia. The study was conducted from September 2018 to February 2019 with larvae sampling in 100 houses. The adult Aedes spp. mosquitoes were used as the sample of immunohistochemical methods and bioassay to get the mosquito resistance status against cypermethrin. The transovarial transmission of dengue virus in Aedes spp. from Brontokusuman Village was found in 3 of 100 samples of Aedes spp. mosquitoes, and the TTI value (Transovarial Transmission Index) was 3%. The transovarial transmission is influenced by the density of the infected vectors, the condition of their eggs and the number of dengue cases. The bioassay test results showed that the resistance status of Aedes spp. can be presumed resistance against cypermethrin insecticide.

Abstract. Bidyaleima L, Kishor R, Sharma GJ. 2019. Chromosome numbers, RAPD and ISSR profiles of six Zingiber species found in Manipur, India. Biodiversitas 20: 1389-1397. The present investigation was done to assess the cytological information and genetic relationships among six species of Zingiber, viz., Zingiber capitatum, Z. kangleipakense, Z. kerrii, Z. montanum, Z. officinale and Z. zerumbet found in Manipur, North-East India. The somatic chromosome numbers observed were 2n = 22 for Zingiber capitatum, Z. kerrii, Z. montanum, Z. officinale and Z. zerumbet, whereas Z. kangleipakense showed tetraploidy with 2n = 44. The somatic chromosome numbers of Zingiber kangleipakense and Z. kerrii are reported for the first time. Randomly amplified polymorphic DNA (RAPD) and inter-specific sequence repeat (ISSR) molecular markers were employed to reveal the genetic relationships among the six species. The pair-wise Jaccard genetic similarity varied from 0.518 to 0.658 for RAPD and from 0.436 to 0.682 for ISSR data. Dendrogram derived from the combined data of RAPD and ISSR clustered the six members into two groups. The detected polymorphism level represents high genetic distance at the inter-species level, and introduces RAPD and ISSR as efficient markers for the assessment of genetic relatedness in Zingiber. Our results may provide useful information for application in breeding, conservation and utilization.

Abstract. Efin A, Atmowidi T, Prawasti TS. 2019. Short Communication:  Morphological Characteristics and Morphometric of Stingless Bee (Apidae:  Hymenoptera) from Banten Province, Indonesia. Biodiversitas 20:  1693-1698. Tetragonula (Meliponini:  Apidae) belong to stingless bees that characterized by complex communication, permanent colonies with division of castes, i.e., queen, males, and workers. This paper described morphological characters and morphometric study of stingless bee from Pandeglang, Banten Province, Indonesia. Local people (Sundanese) known as the specimen examined is “teuweul omas” that have different in nest entrance characteristics compare to nest of Tetragonula laeviceps found commonly. In average, specimens examined were 4.445±0.072 mm body length, 1.911±0.019 mm head width, mesoscutum has distinct hair bands separated by glabrous interspaces area, with plumose frontal hairs, and black metasoma, legs, and hairs on the frontal. Male genitalia of stingless bee examined are long gonostylus and slender with sparse hairs at apex, penis valve is very robust, tapering at the apex and shorter than gonostylus. Based on morphological characteristics and morphometric measurements, we identified the specimen examined is Tetragonula cf. laeviceps.

Abstract. Wasis B, Saharjo BH, Putra EI. 2019. Impacts of peat fire on soil flora and fauna, soil properties and environmental damage in Riau Province, Indonesia. Biodiversitas 20: 1770-1775. Land fires set by communities can result in the death of flora and fauna, damage the soil, and cause smoke disasters. In this study, we focused on the burnt community land in Riau Province, where we conducted a soil analysis through purposive sampling to highlight the effect of the fires on the environment. Our findings showed that fires caused a decrease of peat soil thickness of 10-15 cm, and resulted in 100% mortality of soil flora and fauna. We found a significant increase in the Ca and soil pH after a fire, but total microorganisms, total fungi, and soil respiration are found decreased. Our results strongly showed that the soil and environmental damage of burnt peat soil on community land exceeded the minimum criteria to be classified as "environmental damage" as defined by Indonesian Government Regulation No. 4/2001 (PP No 4/2001) for the parameters of bulk density, porosity, pH, organic C, N, total microorganisms, total fungi, and respiration.

Diversity and characterization of entomopathogenic fungi from rhizosphere of maize plants as potential biological control agents. Biodiversitas 20: 1435-1441. The diversity and characters of entomopathogenic fungi in soil are influenced by cultivation techniques. This study aims at determining the characters and pathogenicity of entomopathogenic fungi in maize plant. Materials were extracted from maize plant rhizosphere at different planting system (monoculture, polyculture, and monoculture of corn on formerly oil palm plantation or replanting) in Nagari Koto Baru, Luhak Nan Duo Subdistrict, West Pasaman District, and West Sumatra Province. Insect bait and series dilution were used to carry out the isolation, while Tenebrio molitor larvae were used for the pathogenicity test. Parameters observed include macroscopic and microscopic characteristics, and pathogenicity of entomopathogenic fungi. The result shows that there are three types of entomopathogenic fungi with various features. After identification, it was known that Aspergillus and Metarizium were obtained from mono, polyculture and by replanting crops, while Beauveria was found from the rhizosphere of corn grown in polyculture with pathogenicity identified as one of the high causes of death to larvae T. molitor with LT50 ranging from 48.03-48.48 hours. Abstract. Husodo T, Shanida SS, Febrianto P, Pujianto MP, Megantara EN. 2019. Mammalian diversity in West Java, Indonesia. Biodiversitas 20: 1846-1858. Protected forests in West Java are wider than conservation forests, whereas mammalian diversity in protected forests is as high as mammalian diversity in conservation forests. Mammals in protected forests are not protected by regional protection regulations, while anthropogenic factors in Java are quite high. This is possible that mammals who have high conservation status will experience local extinction. This study aims to determine (i) the composition of mammalian species and (ii) the species that are always found in studies of mammalian diversity in West Java. The study was conducted through a qualitative approach by combining several methods such as interview, camera trapping, sign survey, observation and transect, and collapsible traps. Mammalia in West Java found 54 species, 21 families, and nine orders with details of three species of Artiodactyla, 12 species of Carnivores, seven species of Chiroptera, one species of Dermoptera, one species of Euphotyphla, one species of Pholidota, five species of Primates, 21 species of Rodentia, and three species of Scandentia. The species that are always found in all study locations are nine species, including wild boar, Javan leopard, leopard cat, Javan mongoose, oriental small-clawed otter, Asian palm civet, grizzled leaf monkey, Javan langur, and Javan tree shrew. The results presented here may facilitate improvements in the mammalian diversity database, especially in the non-conservation area.

Biodiversitas 20: 1243-1254. The cultivation of upland rice in acid soils faces aluminum (Al) toxicity. Development of Al-tolerant rice could be one of the solutions to overcome the problem. Marker-assisted breeding to develop Al-tolerant rice requires at least a molecular marker for foreground selection. Snpb11 is a molecular marker developed from the nucleotide differences in a specific allele between Al-tolerant and sensitive rice. Snpb11 has never been used as a molecular marker in rice. Therefore this study aimed to examine the correlation among Snpb11 marker, root growth, and physiological characters under Al stress in upland rice. We used physiological characters and the Snpb11 marker to justify the Al tolerance level in several upland rice varieties. We found that physiological characters, i.e.: primary root length, total root length, chlorophyll, and carotenoid content showed positive correlation to Snpb11. Conversely, root malondialdehyde content, which represents the level of lipid peroxidation showed a negative correlation to Snpb11. There is evidence that the Snpb11 highly correlated with primary and total root length characters, which are the Al tolerance parameters used in rice. Therefore, Snpb11 markers can be used to distinguish the Al tolerance level in upland rice.

Catharanthus roseus (L.) is an important medicinal plant of the family Apocynaceae used to treat many of the diseases. C. roseus also possess good antioxidant potential. The purpose of this research was 1) to determine the variability of C. roseus based on the morphological, anatomical characters and chlorophyll contents, 2) to know the differences in morphological, anatomical characteristics and chlorophyll contents of C. roseus found in Banyumas Regency. The research used a survey method, with a purposive sampling technique. Preparation of leaf for anatomical analysis was based on paraffin method. The results showed that C. roseus in Banyumas Regency were grouped into eight varieties. The morphological characters observed consisted of 21 characters, 8 quantitative characters, and 13 qualitative characters. Most of the quantitative morphological and anatomical characters were significantly diverse (p <0.05) except for petiole length, corolla number, stomata width and number of trichomata. The eight varieties of C. roseus have different chlorophyll a, chlorophyll b, and total chlorophyll contents but their anthocyanin contents were not significantly different.

Abstract. Hastuti AR, Lumbanbatu DTF, Wardiatno Y. 2019. The presence of microplastics in the digestive tract of commercial fishes off Pantai Indah Kapuk coast, Jakarta, Indonesia. Biodiversitas 20: 1233-1242. Microplastics in fishes have been reported in many studies due to their threat to marine fishes and human health. But only a few data exist on commercial fish for human consumption in Indonesia. This study revealed the presence of microplastics in the digestive tract of 9 commercial species collected from six sites along the Pantai Indah Kapuk coast. Ingested microplastics were identified using gut content analysis. Microplastic particles were counted and categorized by type, color, and size. 169 of 174 (97.13%) of examined fish had microplastics. A total of 2063 microplastic particles were collected with the average number of particles per individual of 12.21 ± 9.76. The highest number (20.0 ± 8.0 particles individual-1) was found in Sardinella fimbriata and the lowest one (4.9 ± 4.7 particles individual-1) was found in Oreochromis mossambicus. Majority of ingested particles were fibers (89.63%), followed by fragments (6.24%), films (4.13%) and no pellets were observed. In terms of color, the most abundant were transparent particles (79.20%), followed by blue (7.03%), red (3.54%), black (2.86%), green (2.71%), and others which were found in low number. The highest number of fibers by size was <20-100 µm (55.03%), films were 100 - 1000 µm (33.93%), and fragments were <100 µm (25.25%). Microplastics ingested per individual in each species were not correlated to total body length, total body weight, digestive tract length, digestive content weight, mouth height, and mouth length.

Abstract. Tan WA, Wijaya I, Purwadaria T. 2019. Bioprospecting of polyhydroxyalkanoates-producing bacteria from Indonesian marine environment. Biodiversitas 20: 1309-1315. Polyhydroxyalkanoates (PHA) are potential alternates to conventional synthetic plastics. PHA production in bacteria involves PHA synthase gene encoded by phaC. In this study, we isolated PHA-producing bacteria from the coastline and 1 mile from the coastline of three beaches in Indonesia. Further phaC detection and characterization of PHA production were conducted. The isolates were subjected to phylogenetic analysis based on 16S rDNA. Red Nile staining on minimal agar revealed that twenty-three isolates showed orange fluorescent, which indicated that they accumulated PHA in their cells. PCR detection showed the presence of PHA synthase class I-encoding gene phaC in twelve isolates. One representative amplicon was sequenced to verify its identity, in which it shared 86% similarity with the PHA synthase class I-encoding gene from an uncultured bacterium. Interestingly, the production of PHA in isolate ST.PA.75, which was closely related to Vibrio sp., was 2.1-fold higher than that in the Ralstonia eutropha JMP134 control. Three isolates showed similarity with bacterial genera and/or species for which PHA producing phenotypes had never been described before TP.SWC.20, which was closely related to Microbacterium arborescens, as well as TP.SWC.33 and TP.SWC.85, which were similar to Psychrobacter spp. Phylogenetic analysis showed that the PHA producing isolates were clustered into three phyla: ?-Proteobacteria, Actinomycetes, and Bacilli. A majority of the isolates (75%) were related to ?-Proteobacteria. In this study, we uncovered diverse novel promising strains for use in the production of PHA as a more environmentally-friendly alternative to hydrocarbon-based plastics.
