Abstract. Talebi SM, Sheidai M, Ariyanejad F, Matsyura A. 2019. Stem anatomical study of some Iranian Marrubium L. species. Biodiversitas 20: 2589-2595. Marrubium is one of problematic genera of Lamiaceae family which distributed in different parts of Iran with 11 species. These species have been used in folk medicine for treatment of different disorders. In the present study, we used stem anatomical characteristics of six Marrubium species for taxonomical treatment of the genus and delimitation of these species. Based on the geographical distribution, one to three populations were selected from each species. Stem hand transverses were double-stained and studied using Light Microscopy. In total, sixteen qualitative and quantitative anatomical traits were studied. Anatomical data were analyzed using MVSP and SPSS software. Stems in transverse section were quadrangle, with a continuous ring of vascular tissue. The anatomical characteristics varied among the species, while the ANOVA test revealed significant variations for some of them. The studied species and their populations were divided into two groups in the UPGMA dendrogram of anatomical data. Populations of each species did not cluster together and were scattered in these groups, except those for M. cuneatum, which were clustered in only one group. Furthermore, these groups were clearly observed in PCA plot. According to CA-joined plot, each group or population had distinct anatomical feature (s) which was useful in identification of them. In some cases, the clustering pattern agreed with previous molecular and morphological studies, while in several times populations clustering did not agree with traditional classification of species as Flora of Iran and Flora Iranica. It seems that some infraspecific ranks must be redefined.

Biodiversitas 20: 3011-3017. Submergence affects plant growth and fitness in natural and agricultural ecosystem. Plant survival to submergence is strongly influenced by the water level and duration of submergence. The objective of this study was to determine the survivalysiological and root anatomical changes of two rice cultivars seedlings to submergence The experiment used completely randomized design with three factors, cultivar (IR-64’ and ‘Inpara 5’), depth and duration of submergence. Depth of submergence consisted of 3 levels: without, partial and complete submergence. Fourteen days old seedlings were treated for 5, 10 and 15 days of submergence. The observed parameters were seedling survival, chlorophyll content, soluble sugar and starch content, plant biomass, and root anatomical structure. The results showed that seedlings survival of ‘IR-64’ and ‘Inpara 5’ decreased by increasing depth and duration of submergence. Increasing depth of submergence decreased chlorophyll content, soluble sugar content and plant biomass. Anatomically, depth and duration of submergence showed a greater increased in root cortex thickness, aerenchyma tissue diameter and the percentage of cortex to be aerenchyma of rice ‘IR-64’ and ‘Inpara 5’. Fifteen days of complete submergence significantly decreased seedlings survival and increased the diameter of aerenchyma in both ‘IR-64’ and ‘Inpara 5’.

Abstract. Fitriana N, Susandarini R. 2019. Short Communication: Morphology and taxonomic relationships of shallot (Allium cepa L. group aggregatum) cultivars from Indonesia. Biodiversitas 20: 2809-2814. Shallot is an important vegetable crop commodity in Indonesia, and its ranks third in terms of cultivation area after chili (Capsicum annuum L.) and cabbage (Brassica oleracea L.). There are a number of superior cultivars of this species that have been studied in terms of their morpho-agronomic traits, but their morphological variability and its implication to taxonomic relationships have not been studied in depth. This study aimed at documenting morphological variability and taxonomic relationships of 12 superior shallot cultivars using numerical taxonomic approaches. Sixteen morphological characters from leaves and bulbs were used in cluster analysis and principal component analysis. The results revealed that twelve shallot cultivars from Indonesia showed significant variability in leaf and bulb morphology, particularly in quantitative characteristics as indicated from ANOVA. The grouping of cultivars and their taxonomic relationships generated from cluster analysis and principal component analysis revealed the role of bulb morphology as determining characteristics in the grouping of cultivars. This study suggested the importance of bulb morphology in the characterization of shallot cultivars.

Abstract. Yasmin Y, Fitri L, Fauziah, Wasliyah F. 2019. Isolation of bacteria from Apis cerana hive, their antibacterial potency and cytotoxicity. Biodiversitas 20: 2733-2738. This study aimed to identify bacteria from Apis cerana hive; to determine their antibacterial activity, and cytotoxic effect of hive extract as well as bacterial extract. Identification of bacteria isolated from the hive was carried out based on morphological and biochemical characters. The antibacterial assay of beehive bacteria isolates was done by disk diffusion method against Staphylococcus aureus and Escherichia coli. Toxicity test of beehive extract and beehive bacterial extract were carried out using Brine Shrimp Lethality Test (BSLT) with six final concentrations (32 ppm, 64 ppm, 126 ppm, 250 ppm, 500 ppm, and 1000 ppm). The LC50 value was determined by probit analysis using SPSS. Bacterial isolation showed there were four bacterial isolates from A. cerana hive were the genus Bacillus (BSL1, and BSL3), the genus Micrococcus (BSL2) and the genus Neisseria (BSL4). The antibacterial assay showed that BSL4 isolate has the highest antibacterial activity against S. aureus with the diameter of the inhibitory zone was 32.6 mm. Toxicity test showed that A. cerana hive extract and bacterial ethanol extract from A. cerana hive had LC50 value of 67,744 ppm and 86.985 ppm respectively and categorized as toxic.

Abstract. Daryono BS, Ramlah, Pabendon MB. 2020. Local food diversification of foxtail millet (Setaria italica) cultivars in West Sulawesi, Indonesia: A case study of diversity and local culture. Biodiversitas 21: 67-73. Foxtail millet (Setaria italica (L.) P.Beauv.) is one of the non-rice cereal crops that has long been domesticated in the world, including West Sulawesi-Indonesia as an alternative food crop instead of rice on local culture. Traditional millet cultivars may become an indispensable part of the local culture and traditions of millet-growing people across Asia over many generations. The existence of this germplasm in West Sulawesi Province, Indonesia has been observed morphologically and showed a close relationship with local culture. A total of six traditional millet cultivars (Tarreang) with different morphological characteristics have been found in this region and still preserved sustainably by the local farmers along with their local culture and traditions. The tradition of Sayyang pattu’duq may become a good example of this relationship. In this tradition, some traditional food is made by the local people like porridge tarreang, jelly tarreang, buras tarreang, jepa golla mamea, jepa anjoroi, dodol tarreang, also widely presented and used as a symbol in important ceremonies, such as celebrations in the month of Muharram, completed Al-Qur'an for children, weddings, births or show the baby's. This information may be useful as guidance for conservation and millet breeding programs in the future.

Abstract. Authors. 2019. Growth and fosE gene expression in inulin-containing medium of two strains of Lactobacillus casei originated from the human intestinal tract. Biodiversitas 20: 2758-2763. Inulin is a fructooligosaccharide consisting of a fructose monomer with ?- (1,2) glycosidic linkage. The human colon cannot degrade inulin because it lacks the enzyme that degrades ?- (1,2) glycosidic linkage. Lactobacillus casei strain AP is able to grow in inulin, L. casei strain AF grows gradually. The ability to grow in inulin is proposed by the fosE gene, which hydrolyzes ?- (1,2) glycosidic linkage. The present study aimed to detect and measure fosE gene expression in inulin. Extracellular and intracellular inulin and fructose concentrations were calculated using high-performance liquid chromatography. The reduction of inulin concentration in L. casei strain AP was higher than that in L. casei strain AF. Extracellular fructose concentration was lower in L. casei strain AP because fructose from inulin degradation was transported into the cell. No fructose was detected inside Lactobacillus cells. The fosE gene was detected in L. casei strains AP and AF with a length of 229 bp. fosE gene expression was up-regulated 5.9-fold in L. casei strain AP and 1.7-fold in L. casei strain AF and was three times higher in L. casei strain AP than that in L. casei strain AF, suggesting that the former metabolizes inulin better than the latter.

Short Communication: Isolation and characterization of thermophilic actinobacteria as proteolytic enzyme producer from Ie Seuum Hot Spring, Aceh Besar, Indonesia. Biodiversitas 20: 2802-2808. Proteases are enzymes who catalyze the breakdown of peptide bonds in proteins. This enzyme could be produced from thermophilic bacteria that are able to grow at temperatures of 45-80ºC and are stable to heat. The samples were collected at Ie Seu'um hot spring, Aceh Besar. This study aimed to isolate, to characterize and to determine actinobacteria that were capable of producing protease enzymes. The sampling method in this study was conducted by purposive sampling at temperatures of 50, 60 and 70ºC. Isolation of thermophilic actinobacteria was carried out in Humic Acid Vitamin B (HV) Agar medium and morphological characterization was carried out in Yeast Malt Agar (YMA), Yeast Starch Agar (YSA), Oatmeal Agar (OA) media. Microscopic characterization and measurement of clear zone diameter formed were carried out in skim milk medium. The results showed that one actinobacteria isolate was obtained at a temperature of 50ºC and five isolates at a temperature of 60ºC, meanwhile, no actinobacteria could be obtained at a temperature of 70ºC. A total of 4 isolates obtained were able to produce protease enzymes. The highest Proteolytic Index (IP) value was obtained from IS01 which was 3.8.

Abstract. Bareke T. 2019. Diversity and genetic potential of various morphological traits among common bean (Phaseolus vulgaris, Fabaceae) landraces. Biodiversitas 20: 3237-3245. The study was undertaken to identify the morphological diversity and genetic potential among landraces of common bean (Phaseolus vulgaris L.) at field conditions of Adama District, Ethiopia. Twenty four common bean landraces from different agroecological zones were collected and grown in three replications. Data of eight agromorphological descriptors were collected using IBPGR protocol. Accordingly, Zale wehy was significantly different (p<0.05) from all landraces with the highest mean number of pods per plant. On the other hand, the mean number of seeds per pod was significantly (p<0.05) higher for Dobole followed by Dima, Kalibushay and Adi tikko while Logoma dume has the lowest number of seeds per pod. The average number of seeds per pod had inverse relationship with seed size. The highest diversity was found in number of seed per plant among landraces while the lowest diversity was found in number of seed per pod. Based on morphological traits similarities among 24 landraces were classified into 4 major categories. Heritability was the highest for seed length (0.97) followed by number of seed per pod (0.94) and seed per plants (0.69) and the lowest (0.1) for height of plant and the rest found between these values. The variability observed in seed length, number of seeds per pod, number of seeds per plant and number of pods per plant which could be utilized in variety improvement programs. Therefore, crop breeders could give emphasis to these traits of landraces to monitor and use for the breeding program, future production and for other domestic purposes.

Abstract. Listihani, Hidayat SH, Wiyono S, Damayanti TA. 2019. Characteristic of Tobacco mosaic virus isolated from cucumber and tobacco collected from East Java, Indonesia. Biodiversitas 20: 2937-2942. Tobacco mosaic virus (TMV) is a newly emerging virus infecting cucumbers in Java, Indonesia. The basic characters of the TMV isolated from cucumber need further study to investigate its differences with that from tobacco. Thus, the research aimed to study the character of both isolates based on their biological, symptomatology and nucleic acid of coat protein (CP) gene properties. The TMV isolates from both cucumber and tobacco were able to infect similar indicator plants with differing symptom expressions, especially on eggplant. Homology of nucleotide and amino acid of coat protein gene among isolates were about 90.3% and 91.0%, and homology to other isolates was about 87.6 to 93.8% and 89.3 to 96.8%, respectively. There were 17 amino acid differences in the CP gene which is presumed to differentiate those two isolates. Phylogenetic analysis CP gene sequences compared with corresponding isolates in GenBank showed the two isolates separated in different clades. The cucumber isolate from Kediri is closely related to tomato isolate from China in clade I, while tobacco isolate from Jember closely related to tobacco isolate from China in clade IIindicating the existence of two differ TMV variants. It will increase the difficulty to manage TMV in the fields.

Abstract. Budiantoro A, Retnaningdyah C, Hakim L, Leksono AS. 2019. Characteristics of olive ridley sea turtle (Lepidochelys olivacea) nesting beaches and hatcheries in Bantul, Yogyakarta, Indonesia. Biodiversitas 20: 3119-3125. We analyzed the ecological conditions of the beaches in Bantul, Java, Indonesia, in supporting the olive ridley sea turtle emergences and nesting. The study was performed at four beaches in Bantul, namely Pelangi, Samas, Goa Cemara, and Pandansimo. We collected data in areas where olive ridley sea turtle nests, such as beach width, slope, sand size, the percentage of magnetic mineral, daily temperature fluctuations, hatchlings rates, and vegetation. The results showed that Pelangi Beach was broader and steeper (31.26 m; slope 5.10°) than the rest (p<0.05). There was no difference in the sand size (diameter of 0.99 mm). The magnetic mineral content in sand ranged from 51.68-87.50%, while the daily temperature ranged from 24.3-31.0°C. The average percentage of hatchlings from 2012 to 2018 was 60.89%. Vegetation on the four beaches is almost the same, which was shrimp pine trees, runny grass, thistle, papyrus, and pandanus thorns. The average pH level of the nest hatchery was 7, supporting the hatchling embryology process. Ecologically, Pelangi Beach is the best turtle landing site in Bantul. Furthermore, transferring olive ridley sea turtle eggs to seminatural nests is recommended due to the quite high hatching rate.
