Chemical compound isolated from antioxidant active extract of endophytic fungus Cladosporium tenuissimum in Swietenia mahagoni leaf stalks. Biodiversitas 20: 2645-2650. Swietenia mahagoni L. Jacq is a medicinal plant that widely used as an antimicrobial, anti-inflammatory, antimutagenic, anticancer, antitumor, and antidiabetic. Several bioactive compounds have been reported to be associated with this plant that supports its treatment activities. Flavonoid content of the S. mahagoni has antioxidant activity. Therefore the plant is widely used to treat degenerative diseases caused by free radical activity in the body. Scientists have proven that the medicinal plants have a symbiotic relationship with the number of microorganisms that can produce bioactive compounds. One of the symbiotic microorganisms is endophytic fungi believed to be associated with every plant on earth. In this paper, we report a secondary metabolite compound from endophytic fungi isolated from S. mahagoni. The endophytic fungi (DMG1-DMG4) were isolated from leaf stalks of S. mahagoni. Antioxidant activity of endophytic fungi extracts was tested by 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The chemical compound from the active extract was isolated by the chromatographic method, and the structure was elucidated by spectroscopic methods including UV, IR, NMR 1D (1H-NMR and 13C-NMR), NMR 2D (HMQC and HMBC). Antioxidant activity test showed the endophytic fungus (DMG3) has the highest activity. The chemical compound from DMG3 was identified as 5-hydroxy-2-oxo-2H-piran-4-yl) methyl acetate). The phylogenetic method used to molecular analysis of DMG3 confirmed the fungus as Cladosporium tenuissimum.

Prevalence and intensity of Trypanosoma sp. in wild swamp eels (Synbranchus bengalensis) marketed in Surabaya, Indonesia. Biodiversitas 20: 3262-3268. Trypanosoma sp. is parasitic protozoa, which can infect not only aquatic organisms but also humans. As the parasite considered a zoonosis disease, there has been a lot of concern about the presence of this parasite in aquaculture commodities. This research aimed to detect and determine the prevalence and intensity of Trypanosoma sp. infection in wild-caught swamp eels (Synbranchus bengalensis) marketed in Surabaya. A total of sixty swamp eels with 47.30±4.69 cm in length were collected from two different locations, Ambengan and Karah, Surabaya, East Java, Indonesia. The swamp eels were transported alive in two aerated plastic bags to the laboratory. The observed parameters were prevalence and intensity of Trypanosoma sp. in the eels’ blood, total erythrocyte counts and total differential leucocyte count (monocytes, lymphocytes, basophils, eosinophil, and neutrophil). The result showed that 7 of 30 (23%) wild-caught swam eels obtained from Ambengan and 9 of 30 eels (30%) collected from Karah were infected by Trypanosoma sp. The intensity of the parasite in eels collected from both locations was considered as moderate, 12.6 parasites/eel (Ambengan) and 5.9 parasites/eel (Karah). Additionally, hematology analysis indicated total erythrocytes count of blood in the infected eels from both locations were significantly lower than total erythrocytes of blood in the non-infected eels, p<0.05. Furthermore, hematology analysis indicated that the numbers of monocyte, lymphocyte, and neutrophil were significantly different than that of non-infected eels, P<0.05. Meanwhile, there was no significant difference in cell counts of basophils and eosinophil in the blood of infected and non-infected eels, p>0,05. These results demonstrate that swamp eels marketed in Surabaya were infected by Trypanosoma sp.

Abstract. Wahyudi AT, Priyanto JA, Fijrina HN, Mariastuti HD, Nawangsih AA. 2019. Streptomyces spp. from rhizosphere soil of maize with potential as plant growth promoter. Biodiversitas 20: 2547-2553. Actinomycete is one group of rhizobacteria that plays an important role as a plant growth promoter. This study was aimed to evaluate the potential of Actinomycetes isolated from maize rhizosphere in promoting plant growth in vitro including their ability to produce IAA, promote maize sprout growth, solubilize phosphate, and grow in N-free medium. Thirty isolates have been isolated from maize rhizosphere using a spread plate method. All 30 isolates were probably not pathogenic to plants as tested by hypersensitivity reaction test on tobacco leaves. Based on the colorimetric assay, 30 isolates (100%) were able to produce IAA with concentrations ranging from 1.05 to 26.89 ppm. The highest concentration of IAA (26.89 ppm) produced by ARJ 21 and the lowest IAA concentration (1.05 ppm) produced by ARJ 12. By using the Ragdoll method, it showed that 9 isolates (30%) were able to promote maize sprout growth significantly on five growth parameters including primary root length, stem length, number of lateral roots, wet weight and dry weight. Twenty-one isolates (70%) were capable of solubilizing phosphate in Pikovskaya medium containing tricalcium phosphate. Also, 30 isolates (100%) were able to grow on N-free medium, suggesting their ability to fix nitrogen. Based on 16S-rRNA, five potential isolates with plant growth-promoting properties were highly similar to Streptomyces spp. Based on their potential characters, these Actinomycetes isolates have the potential to be further developed as a biofertilizer agent for sustainable maize farming.

Mineral levels of Bali cattle (Bos javanicus) from four different types of land in different rearing areas in Bali, Nusa Penida, and Sumbawa Islands (Indonesia). Biodiversitas 20: 2931-2936. This study aims to prove that the mineral level of Bali cattle in serum is dependent on the origin and raising habitat. Serum samples were collected from three islands in Indonesia, i.e. Bali, Nusa Penida, and Sumbawa. Samples from Bali were further classified into four different areas (i.e. agroforestry, perennial plantation, horticultural plantation, and rice field). Blood samples were collected in the jugular vein without anticoagulant. The contents of macrominerals (Ca, Mg, Na, K, P) and microminerals (Fe, Cu, Zn, Co, Mn) were measured using the flame method in atomic absorption spectrophotometer. The results showed that the level of phosphor (P) macromineral and all microminerals under study were influenced by the origin of cattle. The macrominerals are statistically varied in different cattle raising environments. The same is also valid for all microminerals, with an exception of Fe. All mineral levels under study were statistically equal in male and female Bali cattle. Phosphor macromineral and all microminerals levels were origin and environment-dependent. Supplementation of deficient minerals in a certain area and land type is recommended to improve the performance of Bali cattle.

Abstract. Chaiphongpachara T, Sumruayphol S. 2019. Geometric morphometrics as a tool for three species identification of the firefly (Coleoptera: Lampyridae) in Thailand. Biodiversitas 20: 2388-2395.  Firefly is an insect belonging to the Lampyridae family that is important to the ecosystem, generates income from tourism in the area and act as a biological control for snail, an intermediate hosts of trematode parasites. Some types of the firefly, it is difficult to distinguish the species using standard methods because of similarities in morphology while the external characteristics of specimens are damaged during collection, presenting an obstacle to species identification. This study used geometric morphometrics (GM) for identification of firefly species. Samples collection was conducted in the rainy season from August to October 2015 in three districts of Samut Songkham province, Thailand. A total of 200 and 157 fireflies from 3 genera and four species, including Luciola aquatilis Thancharoen, Pteroptyx valida Olivier, Pteroptyx malaccae Gorham and Pyrocoelia praetexta Olivier were used for landmark-based and outline-based GM analysis, respectively. The results showed that both types of GM analysis produced similar results, size variation between two firefly pairs was not statistically significant, including female L. aquatilis with male P. praetexta, and female P. valida with male P. valida. Both landmark-based and outline-based GM methods can distinguish the shape for each type of firefly with high accuracy, especially the outline-based GM method. This result was supported by reclassification scores. According to our findings, the GM can be used to separate species of firefly with high efficiency. Therefore, this GM method represents one way of overcoming difficulties with firefly identification and can support future studies of fireflies.

Polymorphism of myostatin gene and its association with body weight traits in a hybrid of GAMA chicken (Gallus gallus domesticus Linn. 1758). Biodiversitas 20: 3207-3212. An experiment was conducted to detect SNP of the myostatin gene and its association with the body weight of hybrid chicken crossbreed from F1 Kamper and BC1 Broiler. Four F1 Kamper hens were crossbred with BC1 Broiler rooster. Day old chick (DOC) hatched were maintained for 49 days with body weight measurement every seven days. The blood samples from 49 days old chicken were taken for DNA isolation by Chelex 5% method and then amplification of the myostatin gene. PCR products were sequenced, and sequence alignment was performed using Clustal Omega to obtain SNP. The SNP obtained was analyzed by the Pearson correlation test with the body weight of forty nine-days-old chickens. The body weight of the hybrid chicken is higher than of Pelung chicken but lower than the Broiler. There are 7 SNPs in myostatin gene exons included 2 Adenine insertions, 1 Guanine deletion, and four substitutions (C2244G, G2283A, T4842G, G7378T) that yield nine haplotypes. Six haplotypes had different protein sequences with Myostatin protein, while three haplotypes were identical to Myostatin protein. The correlation analysis showed that there was a strong positive correlation (r = 0.736) between normal Myostatin protein and mutant to chicken body weight at 49-days-old. Adenine insertion to nucleotide 2099-2100 of myostatin gene had a very strong positive correlation (r = 0.800) to 49-days-old chicken body weight, although T4842G substitution had a strong negative relationship (r = -0.773) to 49-days-old chicken body weight. Adenine insertion to nucleotide 2099-2100 of myostatin gene could be a genetic marker of heavier body weight of the hybrid chicken.

Short communication: Anatomical changes in the roots, rhizomes, and leaves of seagrass (Cymodocea serrulata) in response to lead. Biodiversitas 20: 2583-2588. Runoff of heavy metals as a result of urban and industrial development is a potential threat for seagrass populations in the coast. The objectives of this study were to study the anatomical changes in the tissues of roots, rhizomes, and leaves of seagrass Cymodocea serrulata in response to treatment with different concentrations of lead (Pb) for different time durations. This experiment used heavy metal Pb from a solution of Pb (NO3)2-with a concentration of 0 ppm, 5 ppm, 10 ppm, and 15 ppm and the treatment period extended up to 4 weeks with 3 replications. Analysis of changes in anatomical features showed that exodermis and endodermis cells in the roots thickened as lead concentration increased. The air spaces in the root cortex and rhizome also widened. Thickening of cell walls occurred in the epidermis and endodermis of rhizome. Likewise, in the leaves, thickening occurred in the upper and lower cuticle and also the upper and lower epidermis. In general, changes in anatomical features of root, rhizome, and leaves were observed in response to increasing lead concentrations. The results showed that C. serrulata developed some level of tolerance to heavy metals, especially lead.

Pesisir cattle is one of Indonesia's local cattle descended from Bos indicus which has small body characteristics. It was assumed due to crossbreeding between pesisir cattle and other cattle. Therefore this study aims to study the origin of pesisir cattle based on the D-loop mtDNA region and to find out the phylogenetic relationships between pesisir cattle and other cattle. Cattle blood samples were taken in two locations, Krui Way and Tanah Rekah. Amplification of the mitochondrial gene using the primary pair of collections of Dr. Achmad Farajallah, namely AF22 (forward) 5'-GCGTACGCAATCTTACGATCA-3' and AF23 (reverse) 5'-ATGCAGTTAAGTCCAGCTAC-3'. Reconstruction of phylogenetic trees was analyzed using the Neighbor-Joining Method based on Kimura 2 Parameters. The results of the mtDNA analysis in the D-loop region showed that there were two broods in pesisir cattle, namely Bos indicus and Bos javanicus. The closeness between these species is due to the close geographical distance between Bos indicus originating from Way Krui, Lampung and Bos javanicus from Java.

Abstract. Nahdi MS, Kurniawan AP. 2019. The diversity and ethnobotanical study of medicinal plants in the southern slope of Mount Merapi, Yogyakarta, Indonesia. Biodiversitas 20: 2279-2287. Medicinal plants have been widely used by local communities in Indonesia due to its effectiveness as medicine and its low cost. In addition, the scientific and technological development of medicinal plants has been developed rapidly. This research aims to conduct inventory of medicinal plants recorded in the ecosystem of the southern slope of Mount Merapi, Yogyakarta and to reveal the local knowledge on their uses and traditional healing method applied. We used a combination of qualitative and quantitative methods. The data of local knowledge on medicinal plants and traditional medical practices when using them were collected through in-depth and semi-structured interview with local informants selected through purposive and snowball sampling. The results showed that the abundance of medicinal plant species at the study area was very high during the rainy season, while during the dry season, some of the plant species were found in the yard and open land. A total of 82 species from 40 families were identified as medicinal plants and used by the community through various processing and for various medical treatments. The information was inherited from previous generations through counseling and training. The most favorable way to use medicinal plants was by consuming the leaf directly. Among all identified medicinal plants, betel leaves (Piper betle L) had the highest importance value (43.6%), followed by turmeric (Curcuma longa L) (38.5%), and soursop (Annona muricata L) (33.3%). Kelor (Moringa pterygosperma, Gaertn) had the most significant usage value (0,6), followed by lemongrass (Cymbopogon citratus (DC) Stapf) and ginger (Curcuma xanthorrhiza Roxb) with the same value (0.3). The medicinal plants had also co-benefits including to encourage communication as well as to building relationship among community members.

Abstract. Saputro TB, Prayogo Y, Rohman FL, Alami NH. 2019. The virulence improvement of Beauveria bassiana in infecting Cylas formicarius modulated by various chitin based compounds. Biodiversitas 20: 2486-2493. Sweet potato (Ipomoea batatas L.) is one of alternative carbohydrate source with an important role in food production, animal feed industries, and as raw materials for other derivatives. However, severe losses are constantly occurred in its production due to a notorious pest known as sweet potato weevil (Cylas formicarius). The chemical insecticides commonly used to treat the pest, but have not been efficient in eliminating C.formicarius. Beauveria bassiana is well known as a biological control agent, has been identified to effectively eliminate the pest. The aim of this research is to observe the optimum concentration of several chitin-based compounds in improving the virulence of B. bassiana. The research was conducted by the addition of chitin from various sources - insects, crustaceans and mollusk shells in a growth medium of B. Bassiana. Moreover, conidia produced by B.bassiana was exposed to C.formicarius to investigate the mortality rate of this insect. The results showed four different characteristics of colonies - velvety, wooly, pellicular, and farinaceous. The 1% chitin from Tellina sp, (P9), was found to be the best treatment which increased the growth of the colony diameter attaining 6.7cm in 20 days, increased the conidia viability that reaches 93.5%. In addition, the highest mortality percentage of C. formicarius was 91.67% in P9 at 6 days after infection. Overall, this research gave new potential sources of chitin that can be applied in improving the virulence of B. Bassiana in eliminating C. formicarius.
