Biodiversitas 23: 3950-3959. The yellow-crested cockatoo (Cacatua sulphurea abbotti, Oberholser 1917) is found only on Masakambing Island, Indonesia. Its conservation efforts are now facing hardship due to massive exploitation of their habitat, and stakeholder contribution is essential. The study was conducted on Masakambing Island using a sequential exploratory mixed method design. The results show that based on a ladder of participation, central government, local government, and non-government organization (NGO) are at the second top of the ladder (co-learning), while the community group is at the lower level (cooperation) and the local community at the lowest (compliance ladder). Limited knowledge, resources, and group characteristics contribute to the failure to be at the top level. Meanwhile, the local government, NGO, and community groups achieve a large participation dimension. The central government reaches medium coverage, while the local community has the lowest coverage. Therefore, it is necessary to get support from all stakeholders and put the local community as the main actor using a pragmatic approach. The lack of consideration from the views of older people and community members may add to the complexity of defining financial exploitation, which can result in problems in detection, disclosure, and underreporting of incidences.

Biodiversitas 22: 3671-3676. There were several speculations regarding the main purpose of sea turtle cloacal fluid, such as lubrication during egg deposition, which could help reduce egg speed as they are dropped in nesting sand, or contain antimicrobial properties to protect their eggs from pathogenic microorganisms. However, the exact purpose of this fluid which is mucous-like in consistency remains unknown. This study aimed to identify antimicrobial properties in the cloacal fluid of olive ridley sea turtles. Cloacal fluid samples were collected during nesting season in Banyuwangi City coastal areas, East Java Province, Indonesia. Moreover, the Kirby-Bauer diffusion disk method recorded antimicrobial properties and tested against several bacterial colonies collected from sand in natural nesting chambers. This preliminary study also concluded that antimicrobial properties were recorded when presented against several bacteria colonies from natural nesting sands namely Escherichia coli, Salmonella spp., and Bacillus spp. Furthermore, the present study should be expanded to include other bacteria and fungi colonies. In addition, results from the present study could be used to develop methods in creating antibiotics similar to those found naturally in natural nesting areas to prevent microorganism contamination and globally increase hatchling success, which has a greater impact to support sea turtle conservation efforts as endangered species.

Bird diversity and ethno-ornithological knowledge of local people in Ciletuh-Palabuhanratu Geopark, Sukabumi, West Java, Indonesia. Biodiversitas 22: 3409-3422. Birds have ecological and social functions, and are common to be used as bioindicators of environmental changes. Birds are very important consideration when establishing conservation areas, including geopark which has uniqueness in terms of geological, biological and cultural diversities. This study aimed to investigate the bird diversity in Ciletuh-Palabuhanratu Geopark, West Java, Indonesia and as well as to reveal the local knowledge on bird species and folk classification, functions and utilization of birds among the village people. Mixed methods of quantitative and qualitative approaches were applied in this study, while some techniques, including semi-structured interviews, structured interviews, and bird observation on transect and spotlighting were employed to collect the primary data. In total, there were 121 bird species of 47 families of which 88 species were recorded during the field observation and 79 species were documented through the interviews with respondents. Among them, 12 species are listed as having high conservation status under IUCN, 17 species are listed as Appendix II (CITES), and 21 species are listed by Regulation of the Minister of Environment and Forestry of the Republic Indonesia No. P 106/2018 on Protected Plant and Animal Species. The local community has profound knowledge of birds species, folk classification, ecological, socio-cultural, and socio-economic functions. In the past, the village people had traditional beliefs and myths on birds, resulting in some sacred birds had been traditionally conserved. Nowadays, these are no longer strongly existed, and consequently, many birds have been hunted by using different methods of hunting birds, both traditional and modern for various purposes, including for consumption, bird keeping, and trading to obtain cash. Therefore, bird conservation efforts that involve the community are really needed in this area as the strategy of sustainable management of Ciletuh-Palabuhanratu Geopark.

Biodiversitas 22: 3829-3837. Agroforestry is an agricultural system that many people use in mountainous areas. Some experts have proven that this system can become the mitigation strategy in volcano areas. This study analyzes the risk and hazard perception of agroforestry adopters and non-adopters in the Mt. Merapi prone area. 139 agroforestry adopters and 130 non-adopters were randomly taken from four different areas in Mt. Merapi. Results show that both adopters and non-adopters show different perceptions regarding hazards and risk. The effectiveness of agroforestry practice for mitigation strategies needs to be studied more. The awareness of hazards and risks in the Mt. Merapi prone area is essential to save more lives during the eruption. The challenge is how to persuade agroforestry adopters and non-adopters to flee when the big eruption comes. Even though they flee, they still insist on returning home to take care of their livestock, farm, and protect properties. Mitigation education is needed; also, the infrastructure is important in supporting the mitigation efforts.

Biodiversitas 22: 4731-4739. Pepper yellow leaf curl disease (PYLCD) caused by infection of Begomovirus is a serious threat to pepper production worldwide. Identification of the resistance gene analogs (RGA) and resilience response of pepper against PYLCD is needed especially for selection resistance genotype. Evaluation of resistance response involving 28 pepper genotypes was carried out through transmission of Begomovirus using whitefly (Bemisia tabaci) as vector. The result showed that IPB C12 and F4-012328-6-3 were potential resistance genotypes. A total of 15 R gene analogs (CaRGA) containing NBS motif, namely CARGA1 to CARGA15, were identified by degenerated PCR amplification and database mining. The alignment of deduced amino acid sequence revealed conservation of subdomains Ploop (GKTT), kinase2 (LVVLDDV), RNBSB/kinase3 (IILTTR) and GLPL. BLASTp analysis indicated that 15 RGA showed high homology at deduced amino acid level with R gene identified such as whitefly resistance protein Mi-1.2, Pvr 9 gene for potyvirus, Begomovirus resistance protein, TRGA15 and RGA 13 for putative late blight resistance. Phylogenetic analysis exhibited that isolated sequences distinguished into CNL-NBS groups. These pepper RGA could be considered as candidate sequences of resistance genes.

Genetic diversity of eucalypts for germplasm conservation in Forest Area with the Special Purpose of Mount Bromo, Karanganyar, Indonesia. Biodiversitas 22: 4223-4235. As a repository of a gene pool, eucalypts germplasm enriches biodiversity, maintains ecosystem sustainability, and aids in conservation. Therefore, this study aims to analyze the genetic diversity of eucalypts (Corymbia and Eucalyptus) for the development of germplasm conservation in Forest Area with the Special Purpose (KHDTK) Bromo Forest, Karanganyar, Indonesia. In this study, 14 simple sequence repeat (SSR) markers were used to assess the genetic diversity among 20 accessions (Corymbia and 5 Eucalyptus species) from Central and West Java. Subsequently, the genetic parameters were measured and a phylogenetic tree was constructed. The result showed that the SSR markers have high variability, although they belong to different genera. Furthermore, the genetic diversity showed 49 alleles with an average of 3 alleles per locus, while the polymorphism information content (PIC) values were 0.55. There were 4 SSR markers (EMBRA13, EMBRA8, EMCRC11, and EMBRA2) with high PIC value, while the gene diversity (He) of Corymbia and 5 Eucalyptus showed a low level of genetic diversity. The genetic relationship and population structure were divided into genera Corymbia and Eucalyptus. For further application, the eucalypt cultivated in the KHDTK Bromo Forest can contribute as a reference set and 14 SSR markers as a potential marker in combination with morphological characterization to generate a database for germplasm management and conservation.

Abstract. Nurhayati PA, Affandi M, Nurinsiyah AS. 2021. Diversity and abundance of terrestrial Gastropods on the slopes of Mount Arjuna-Welirang, East Java, Indonesia. Biodiversitas 22: 4193-4202. Before this study, the latest list of land snail species from Mt. Arjuna-Welirang was published in 1952. The current study aims to update the list of terrestrial Gastropods in the area, focusing on Sumber Brantas Arboretum and Raden Soerjo Forest Park Conservation Area (Tahura), East Java, Indonesia. Live snails and dead shells were collected from 24 10 × 10 m2 sampling plots to determine species richness, composition, and abundance. Sampling was carried out by combining visual observation (direct collection) and sorting-sieving soil-leaf litter samples. Total 646 specimens belonging to eight families and 13 species were collected, all new records to the area. Thus, the number of species known from Mt. Arjuna-Welirang was increased to 16 species from nine families. The diversity index of the Mt. Arjuna-Welirang slope was 1.1, and the dominance index was 0.54. The Tahura had a higher diversity index than the Arboretum, at 2.0 and 0.38, respectively, but the dominance index for Tahura was lower (0.18 and 0.85, respectively). This was due to the dominance of Bradybaena similaris, an invasive species in the latter area. Less canopy cover and more human activity in the Arboretum are factors that might cause this discrepancy. Many land snail species are sensitive to desiccation and habitat disturbance. Therefore, minimizing habitat changes and human activities is necessary to maintain or increase native land snail species diversity.

Abstract. Polapa FS, Werorilangi S, Ali SM, Jompa J. 2021. Physiological responses of scleractinian corals in marginal habitat. Biodiversitas 22: 4011-4018. This study aims to analyze physiological differences in corals in marginal habitats. Under different conditions, the production/respiration (P/R) ratio and photobiology of various coral genera were compared. Samples were taken from three coral reef zones representing typical reef habitats and from the mangrove ecosystem as a marginal habitat. Surveys revealed two coral genera surviving in extreme conditions (marginal habitat). The P/R ratio measurements indicated that corals living in the mangrove ecosystem tend to be heterotrophic. This was supported by observations of colonies with tentacles extended from the corallites. Furthermore, Porites living in the mangrove habitat consume more O2 directly than saving it for other purposes, such as growth. The genus Dipsastraea exhibited elevated zooxanthellae density in the mangrove ecosystem, whereas Porites exhibited similar densities in both ecosystems.

Biodiversitas 22: 4923-4929. The doubled haploid (DH) technology when integrated with induced mutation could accelerate development of local rice cultivars with several desirable traits. Anthers from the first generation of mutant (M1) plants were utilized for DH production. However, the effectiveness of inducing and fixing mutation requires detailed evaluation with the help of molecular techniques for its accuracy and reliability than that of morphological or biochemical assessments. The objective of this research was to develop and detect genetic variation of DH plants derived from anther culture of M1 plants. Seeds of local red rice cultivar, Cempo Abang, were treated with four gamma-ray doses (0, 100, 200, 300 Gy) and planted to produce the M1 plants. Panicles at the booting stage of the M1 plants were collected and used as donor anthers. The present study suggested that gamma irradiation treatments indirectly could increase callus formation. However, it also reduced the number of callus producing plantlets due to higher frequency of brown calli. Eleven spontaneous DH plants obtained in the study along with parental plants were assessed to determine the occurrence of genetic variation using six RAPD primers. The RAPD primers generated 51 bands, of which 34 alleles or 66.7% were polymorphic with an average of 5.6 polymorphic alleles per primer. The genetic similarity among parent and 11 DH lines based on Jaccard’s similarity index ranged from 0.622 to 0.902. The DNA polymorphism among the DH plants demonstrated the effect of gamma irradiation to create genetic variation. Therefore, this method could be used as an alternative for rice breeding programs especially to develop preferred traits in the local rice cultivars.

Abstract. Jariyapong M, Roongtawanreongsri S, Romyen A. Somboonsuke B, 2021. Growth prediction of sago palm (Metroxilon sagu) in Thailand using the Linear Mixed-effect model. Biodiversitas 22: 5293-5301. A living sago palm is a potential resource for carbon sequestration in peat swamp. It can provide benefits for agriculturists and environmental sustainability. This study’s aim is to create a growth prediction of sago palm size at individual ages using a linear mixed-effect model. The sample palms were selected from 43 sago palm parents which have been re-measured and recorded at intervals of 2-20 years. The experimental plot was on deep peat soil (with peat depth at over 2.5 m) at Phru Todaeng Swamp Forest, Southern Thailand. To find the best model for growth prediction, we used a traditional linear model (Model 1 and 4) and linear mixed-effect models (Model 2, 3, 5 and 6) to generate the relationship between age, diameter, and height. The best model was selected based on considering the smallest Akaike’s Information Criteria (AIC), the Bayes Information Criteria (BIC), and the absolute mean error (AME). The results showed the smallest values of AIC, BIC, and AME in Model 3 which produced the best model for predicting the total overall growth in sago palm. The age and diameter under the independent variable were statistically significant (? = 0.01); these two factors influenced the sago palm height rate. Hence, this model can be used to predict the overall growth of sago palms, which is useful for biomass estimation and calculating the carbon sequestration of planted sago palms. The carbon sequestration in living sago palm can be compared with other commercial crops for future benefit. This approach can lead to a future solution for wetland management and land-use changes.
