Biodiversitas 22: 4750-4757. Genotype-by-environment interaction (GEI) causes differences in the productivity of rice cultivars in agroforestry systems. For this reason, the stability of rice cultivars is an important aspect that should be considered before a cultivar is recommended to farmers. Superior genotypes and ideal environments are commonly identified using two statistical models, namely, genotype–genotype-by-environment biplot (GGE-biplot) and empirical best linear unbiased prediction (EBLUP). In this study, 15 rice cultivars were evaluated in terms of their productivity and stability in three soil types (Lithic Haplusterts, Ustic Epiaquerts, and Vertic Haplustalfs) in an agroforestry system with kayu putih (Melaleuca cajuputi) in 2019 and 2020 at the Menggoran Forest Resort, Playen Forest Section, Yogyakarta Forest Management District, Indonesia. The cultivars were treated as random effects to select and obtain the EBLUP of the best cultivars in each soil type. The EBLUP revealed that Situ Patenggang showed the highest yields of 4.887 and 5.456 tons ha?1 in Lithic Haplusterts and Vertic Haplustalfs, respectively. GM 28 exhibited the highest yield of 6.492 tons ha?1 in Ustic Epiaquerts. Ciherang, GM 2, GM 8, GM 11, GM 28, Inpari 6 Jete, Inpari 33, IR-64, and Way Apo Buru were classified as stable and fairly stable cultivars, whereas the other cultivars were unstable. Therefore, rice cultivars with high yields in specific soil types should be selected.

Effect of legume varieties and fermentation time of tempe using usar inoculum on the inhibitory activity of angiotensin I-converting enzyme. Biodiversitas 22: 5262-5267. The effect of three varieties of legumes, namely, velvet bean (Mucuna prurient), lima bean (Phasaelus lunatus L.), and pigeon pea (Cajanus cajan), on the activity of angiotensin I-converting enzyme (ACE) inhibitors was studied during tempe fermentation. Fermentation is one way to produce bioactive peptides. Furthermore, the effect of inoculum usar on tempe fermentation was studied. Hydrophobic amino acids are essential in binding firmly to the active site of ACE, and the research showed that pigeon pea had higher levels of hydrophobic amino acids compared with soybeans. The legumes affected the protease activity, peptide content, degree of hydrolysis, and production of ACE inhibitory (ACEI) activity during tempe fermentation. High protein levels and peptide content did not guarantee a high ACE inhibitor level. The mixed culture of usar caused no effect on the production of ACE inhibitors compared with a single culture of Raprima. Legume beans that reached optimal fermentation fast produced high levels of ACE inhibitors. Of the three types of legumes tested in the study, lima beans had the highest ACEI activity during fermentation.

Novel carbonatogenic bacterial strain isolated from limestone quarry in East Java, Indonesia to improve concrete performance. Biodiversitas 22: 3890-3898. Carbonatogenic bacteria can precipitate CaCO3 in the form of calcite, aragonite, or vaterite. Calcite has the potential to be applied for strengthening concrete structures. This research aims to explore several new bacterial strains that can precipitate calcium carbonate leading to produce calcite and could be useful for strengthening concrete structures. Soil and stalactite samples were taken from a well-known limestone quarry in East Java, Indonesia. The isolated bacteria species were identified using 16S rRNA gene sequences. CaCO3 crystal properties were characterized using X-Ray Diffraction and Scanning Electron Microscopy. Six novels isolated CaCO3 precipitating bacterial strains; Bacillus huizhouensis JA1; B. galactosidilyticus JB3; B. niacini AK4, B. lentus SU1, Lysinibacillus macroides JB2, and Sporosarcina soli JA4 were successfully isolated and have the potential to enhance concrete strength. All isolates were able to produce CaCO3 in calcite form except B. galactosidilyticus JB3. The experimental concrete with the addition of bacterial cells showed higher compressive strength and maximum load compared to control concrete and met the requirements for building construction so that it could be applied for building structure materials.

Morphological and molecular identifications of three native arbuscular mycorrhizal fungi isolated from the rhizosphere of Elaeis guineensis and Jatropha curcas in Indonesia. Biodiversitas 22: 4940-4947. Molecular analysis has been widely used to provide more accurate identification within arbuscular mycorrhizal fungi (AMF) species than identification based on morphology. However, morphological analysis is essential for a basic preliminary of classification studies. Therefore, a study is needed to complete the identification of AMF isolates through morphological and molecular analyses. This research used three AMF isolates, namely MV 5, MV 17, and MV 18, which were isolated from Indonesian agricultural land. Spore-based taxonomy (shape, size, color, ornamentation, PVLG, and Melzer’s reaction) and fungal colonization on roots of maize trap plants were employed for the morphological studies. AMF species identification was performed using molecular analysis through nested-Polymerase Chain Reaction (PCR) to amplify a fragment of SSU rRNA followed by sequencing and phylogenetic tree construction. Morphological analysis showed that MV 5 had spores borne from the neck of the sporiferous saccule, MV 17 was found to have a bulbous suspensor without a germination shield, and MV 18 had spores borne from subtending hyphae. The SSUR rRNA analysis revealed that MV 5, MV 15, and MV 18 were identified as Acaulospora longula, Gigaspora margarita, and Glomus etunicatum, respectively. Both morphological and molecular methods demonstrated reliable and consistent results that complement AMF taxonomy studies.

Screening of some Indonesian medicinal plant extracts for anti quorum sensing activity to prevent Aeromonas hydrophila infection in Oreochromis niloticus. Biodiversitas 22: 3517-3522. Aeromonas hydrophila disease is a threat to Indonesia's aquaculture and fishing industries because of its results in mass deaths. Efforts to treat and prevent A. hydrophila infection in fish have so far been limited to the use of antibiotics. The use of these antibiotics can result in bacterial resistance as well as issues related to fish quality and environment. Another option is to target the bacterial quorum sensing (QS) system, a bacterial intercellular communication system that uses signal molecules, regulates the expression of A. hydrophila virulence factors such as exoprotease enzymes, elastase, and biofilms. The expression of A. hydrophila virulence factor can be inhibited by using QS inhibitor compounds, preventing infection from starting. The purpose of this study was to screen some Indonesian medicinal plant extracts for anti-QS activity and then use the selected extract to prevent A. hydrophila infection in Tilapia fish (Oreochromis niloticus). With the administration of extract, QS inhibition was observed in vitro based on the production of violacein in Chromobacterium violaceum; enzyme exoprotease, elastase, and the formation biofilm in A. hydrophila. Tilapia fishes were fed with extract supplementation and then infected with A. hydrophila in an in vivo infection challenge test. These test fishes were divided into six treatment groups given a different concentration of extract in feed (Healthy fish with a concentration of 0 g/100 g of feed as control; infected fish with a concentration of 0 g/100 g of feed; infected fish with a concentration of 0.2 g/100 g of feed; infected fish with a concentration 0.4 g/100 g of feed; infected fish with a concentration of 0.6 g/100 g of feed; infected fish with a concentration of 0.8 g/100 g of feed). Fish were reared for 2 weeks with the frequency of feeding 3 times a day. The variables observed included fish survival rate, behavior (swimming, agility, movement, and frequency of operculum opening), and morphology of the test fish. Data analysis of virulence factors production, fish survival rate, fish appetite, and frequency of operculum opening was carried out using the ANOVA variant test, continued with a 5% DMRT test. Others were analyzed descriptively. Of all the extracts studied, 4 extracts showed inhibition of the production of the violaceum pigment in C. violaceum. Muntingia calabura leaves extract was chosen for further testing due to its ease of availability and safety. The extract was significantly decreased the production of A. hydrophila caseinolytic and elastolytic protease, but had no effect on biofilm formation. In vivo challenge test with A. hydrophila showed better survival rate and fish condition in fish groups that were supplemented with M. calabura extract.

Abstract. Saensouk S, Boonma T, Saensouk P. 2021. A new species and a new record of Curcuma subgen. Curcuma (Zingiberaceae) from Northern Thailand. Biodiversitas 22: 3617-3626. Recent fieldwork to study the species diversity of the Zingiberaceae family in Thailand and preparing a revision of the Curcuma genus for the Flora of Thailand, an undescribed species of Curcuma uses as traditional medicinal for more than two decades well known in the Thai vernacular name “Rang-Jued” was found, this vernacular name is also used to call the other two medicinal plants belonging to different families which are Thunbergia laurifolia Lindl. (Acanthaceae), and Crotalaria spectabilis Roth (Fabaceae), thus some villagers are named Curcuma rangjued as “Rang-Jued-Khamin” to communicate specifically identify species in order to use correct species for each utilization. While Thunbergia laurifolia was called "Rang-Jued-Thao", which "Thao" means "vine" refers to its habit and Crotalaria spectabilis was called "Rang-Jued-Ton" which "Ton" means "stem" refers to its erect stems. After comparing this undescribed species of Curcuma with its allies species, we found that this species did not match with any existing taxa, thus described with detailed illustrations, photographs, summaries for its distribution, ecology in the name of Curcuma rangjued Saensouk & Boonma and followed by a new record of C. cordata Wall. for Thailand which in the past has been determined as a synonym of C. petiolata Roxb. but now it is back to a recognized name once again. Both species in this study were found distributed in the northern region of Thailand and belong to Curcuma subgenus Curcuma, their description along with ecology, phenology, and a revised key to 28 species of Curcuma subgenus Curcuma in Thailand are also present for facilitating identification.

Abstract. Haneda NF, Supriatna AH, Shabrina H, Istikorini Y, Siregar UJ, Wahyudi I. 2021. Chemical characteristics of Falcataria moluccana wood infested by Boktor stem borer (Xystrocera festiva). Biodiversitas 22: 4203-4208. Boktor (Xystrocera festiva Thoms.) is the most destructive and detrimental pest in Falcata plantations in Indonesia. It attacks the inner bark of sapwood when the tree reaches 3-years-old. This study aimed to analyze the chemical characteristics of Falcata woods, which were infested with Boktor in trees aged 3 and 4 years, compared to the healthy tree at 2 years old. The primary chemical components and type of extractive compounds were analyzed using standard procedures. The results showed a decrease in hemicellulose and an increase in lignin content of infected wood. This condition occurs because Boktor larvae prefer hemicellulose rather than lignin, which is related to the presence of enzymes in the digestive system of the larvae. The decreasing extractive contents as trees grow and undergo age-related shifting metabolism can be alleged as the start of Boktor infestation in 3-years-old trees. Boktor infestation also altering the wood chemical components by degrading the lignin that produced 4-vinyl-syringol. The isoeugenol compound is thought to prevent Boktor infestation in the early period of tree growth.

Fruit flies (Diptera: Tephritidae) are pests of several horticultural crops that can reduce the quality and quantity of fruit production. Information on fruit flies in Lombok Island, Indonesia, is still limited. However, it is predicted to have a high diversity of fruit flies because this island belongs to the Wallacea region. The aims of this research was to study the diversity and species composition of fruit flies in different habitat types in Lombok Island. The research was carried out on two habitat types, i.e., tropical rainforest and orchard with each habitat type consisted of three different sites that located spread across Lombok Island as replication. A sampling of fruit flies was conducted using parapheromone traps from March to June 2020. Twenty-two species and 210,267 individual fruit flies were collected from all locations during the study period. The most dominant species were Bactrocera carambolae, Bactrocera limbifera, Zeugodacus caudatus, and Bactrocera dorsalis. Based on the ANOVA, habitat types significantly affected the abundance but not the species richness of fruit flies. The visualization results obtained from the NMDS ordination indicated a difference in the species composition of fruit flies between the two habitats. In conclusion, habitat types are an essential factor in shaping the community of fruit flies in Lombok Island.

Abstract. Dimara PA, Purwanto RH, Sunarta S, Wardhana W. 2021. The spatial distribution of sago palm landscape Sentani watershed in Jayapura District, Papua Province, Indonesia. Biodiversitas 22: 3811-3820. Sago palm is one of the starch sources used as local food in Papua, therefore this research aims to identify the supporting environment for the plant to grow by utilizing spatial data. The methods used were Spatial Analysis and Field Survey, where the first employed satellite imagery of Quickbird in 2012 and Landsat 8 in 2020 to distinguish between sago and non-sago palm landscape. In the process, five parameters were used, consisting of covering land elevation, slope gradient, soil type, rainfall as well as the optimal distance from the river and lake. The result showed the sago palm landscape in Sentani Watershed lies in the elevation of 0-450 m asl, while its largest habitat which lies between 0-100 m asl covering an area of 4,385.63 is found in a flat slope covering an area of 2,941.99 ha and in a very steep slope that spreads out over 41.92 ha. Generally, in Sentani Watershed, the plant grows in Mediterranean soil possessing thick solum with pH 5.0-7.0 and medium to great soil erodibility. Moreover, the largest habitat experiences a precipitation rate of 1,750 mm yr-1 covering a total of 6,846.24 ha, while the Doyo River has the largest sago palm landscape compared to other rivers.

Abstract. Devkota RP, Subedi JR, Wagley K. 2021. Prevalence of gastrointestinal parasites in equines of Mustang District, Nepal. Biodiversitas 22: 3958-3963. The study was conducted from March 2018 to November 2018 in four villages of Mustang district to find out the prevalence of gastrointestinal parasites in equines (horses and mules). Fecal samples collected from 175 randomly selected equines (105 from horses and 70 from mules) were preserved in 2.5% potassium dichromate and microscopically examined using concentration techniques to detect ova, larvae, and oocysts of parasites. The overall prevalence of gastrointestinal parasites was found to be 84.57% with an incidence rate of 81.90% and 88.57% in horses and mules respectively. The prevalence of gastrointestinal parasites between horses and mules did not vary significantly (p>0.05). Also no significant association (p>0.05) between the prevalence rate of parasites and study areas was observed. Among identified parasites in horses, Strongyles showed the highest prevalence (68.57%) followed by Strongyloides spp. (23.80%), and Parascaris equorum (14.28%) and in mules Strongyles showed the highest prevalence (80.0%) followed by Parascaris equorum (35.71%), Strongyloides spp. (28.57%), and Dictyocaulus spp. (5.71%). Prevalence of gastrointestinal parasites showed no significant variation (p>0.05) with respect to age and feeding status. However, the prevalence rate significantly varied (p<0.05) with respect to parameters like infection status (single and mixed), housing condition and health status. The present study revealed a higher prevalence rate of gastrointestinal parasites due to poor husbandry practices and lack of adequate veterinary services. Therefore, improvement in housing and feeding system, awareness regarding effective regular deworming, and establishment of veterinary hospitals in the study areas were recommended.
