Siombak Lake has a unique biological system impacted by high and low tides. Crustaceans (Decapoda) found in this lake are predicted to establish distinctive communities due to their interaction with two biological systems of seawater and freshwater. Therefore, this study aimed to determine the characteristics and distribution of crustacean communities in Siombak Lake, which were influenced by high and low tides. Crustacean samples were taken and observed every month at high and low tides with traps from September 2018 to August 2019. The crustaceans discovered included 5 families and 18 species, with 13 crabs and 5 shrimp. The communities' distribution is extremely dynamic, as seen by the emergence of mangrove crabs at high tide and fiddle crabs during low tide. The crustacean communities are more influenced by tidal ecology, characterized by a higher abundance of individuals at high tide. Furthermore, crustacean density fluctuates throughout the year and reaches the highest in March with 8679 ind/100m2. Penaeid shrimp are always found at high and low tides, dominated by Parapenaeopsis coromandelica, while mangrove crabs are only found at high tide. Therefore, efforts are required to conserve mangrove crabs and enhance their habitats for sustainable usage.

Abstract. Gautan R, Baral N, Sharma HP. 2022. Are white-rumped vultures (Gyps bengalensis) scavengers or predators at a vulture safe feeding site of Nepal?. Biodiversitas 23: 3808-3812. Establishing safe feeding sites has been promoted as a strategy to conserve wild vultures following the population crash in South Asia due to the contamination of natural food by veterinary diclofenac. Several feeding sites have been established in the region, including Nepal and it is important to evaluate their effectiveness. One aspect that is not clear yet is whether such safe feeding sites change the behaviors of wild vultures in South Asia. Here, we report incidents of White-rumped Vultures Gyps bengalensis attacking live animals at the Vulture Safe Feeding Site in Ghachok village in central Nepal. Even though a carcass was available nearby to feed upon, three White-rumped Vultures attacked an adult cow and a calf at the feeding site. The vultures’ attack served as an ancillary cause of the calf’s death in this case. These two incidents of vultures’ attacking live animals at the safe feeding site may allude to changing behaviors and cast doubt on their status of being classified as an obligate scavengers. The findings suggest alteration of vulture behaviors at the safe feeding site. Further research is needed to test the hypothesis of vulture behavior change.

Coconut diversity based on chloroplast Single Nucleotide Amplified Polymorphism (SNAP) and Insertion-Deletion (InDel) markers. Biodiversitas 23: 4073-4081. Indonesia is known as a country rich in biodiversity, and a species with high diversity existing in Indonesia is coconut (Cocos nucifera L.). Although the high diversity of Indonesian coconut is recognized, information about its genetics is limited. The genetic relationship between Indonesian coconut and their maternal inheritance is important and has not been widely studied. This study aimed to analyze the genetic diversity of the Indonesian coconut using chloroplast SNAP and InDel markers, and evaluate their haplotype diversity. Sixty-four coconut accessions were evaluated using ten SNAP primers and five InDel primers based on cpDNA. All of the primers successfully amplified the cpDNA of most evaluated coconut accessions. Based on five InDel markers, six haplotypes were observed in Cocos nucifera. Phylogenetic analysis based on the combined SNAP and InDel markers divided the 64 coconut accessions into three clusters. The SNAP marker analysis results showed that they are less informative for evaluating coconut genetic diversity. On the other hand, the InDel markers are more informative and useful for coconut genetic diversity evaluation. Therefore, InDel markers based on chloroplast genomes has the potential for future coconut genetic variation analysis, evolutionary study, and DNA fingerprinting. The results should contribute to understanding the Indonesian coconut origins and evolution.

Diversity, risk and management feasibility of invasive alien plants in the border zone of Sicike-cike Nature Tourism Park, North Sumatra, Indonesia. Biodiversitas 23: 3156-3165. Invasive alien species (IAS) threaten native ecosystems and biota either directly or indirectly. Once colonizing, invasive plant species tend to dominate a landscape, hindering other native vegetation from growing. Thus, the invasion of alien plant species needs attention moreover in protected areas such as Nature Tourism Park (Taman Wisata Alam/TWA) Sicike-cike, Dairi District, North Sumatra, Indonesia. Besides its importance for nature conservation, TWA Sicike-cike is culturally essential for Pakpak Dairi ethnic community as well as economically for ecotourism. Yet, anecdotal evidence showed the presence of several invasive alien plants around the park. This study aimed to investigate the diversity and composition of invasive alien plants within the vegetation community in TWA Sicike-cike and to assess the risk value and management feasibility of the IAS. Purposive sampling using line transect method was conducted to collect data with a total of 120 observation plots established along 6 transects. Risk value was assessed based on invasiveness, impact and potential distribution. Management feasibility was assessed based on cost, current distribution and persistence. The results showed that Asteraceae was the most dominant family of invasive plants in the studied area with four species. At the species level, Imperata cylindrica was invasive alien plant species with the highest important value index (IVI) of 57.65% and a high-risk value of 235.8, followed by other species including Ageratum conyzoides, Chromolaena odorata, Clidemia hirta and Melastoma malabathricum. In terms of management feasibility, the existence of I. cylindrica is recommended for direct eradication, while other IAS management recommendations, included monitoring and area protection for further invasion by the species.

Genetic diversity based on SSR markers of 30 Aeridinaesub-tribe orchid genetic resources of Indonesian Ornamental Crop Research Institute, Cianjur, Indonesia. Biodiversitas 23: 2943-2956. Advances in molecular biotechnology encourage molecular markers to identify plant diversity to become increasingly common. Using SSR markers, this study will determine the genetic diversity and phylogenetic relationships of 30 accessions from five parental genera of Aeridinaeorchids from the IOCRI genetic resource collection. Molecular characterization of 30 orchid sub-tribes Aeridinaegenetic resources of IOCRI was carried out using 17 polymorphic SSR markers. The resulting binary data was then analyzed using the GenAlEX program to determine Na, Ne, Ho, He, and the PowerMarker V3.25 program to determine the values of MAF, D, and PIC. Molecular characterization using 17 SSR primers generated 240 amplified DNA fragments across 30 accessions, with the fragment sizes ranging from 56-4818 bp. Factorial analysis (PCoA) and phylogenetic tree construction clustered the 30 Aeridinae accessions into three major clusters. Accessions belonging to the Vanda genus are clustered into three clusters (Clusters I, II, and III). The Vascostylisis in Cluster I, along with 12 Vanda accessions. Two accessions of Vanda, two Aranthera, three Arachnis, and one Aerides genera are clustered into Cluster III. This study showed that interspecific hybrids among accessions of Vanda within Cluster I and Cluster II should be possible since they are genetically closely related. Intergeneric hybrids among the accession of Vascostylis and Vanda's genera belonging to Cluster I, should also be possible. Furthermore, intergeneric hybrids among Aerides, Arachnis, Aranthera, and Vanda accessions within Cluster III should also be possible. The generated data from this study should be helpful for future Aeridinae breeding activities intended to generate new hybrid varieties.

Cryopreservation of Gaga’ chicken semen from South Sulawesi, Indonesia with the addition of L-carnitine, hyaluronic acid, sucrose and their combination in diluent. Biodiversitas 23: 3297-3302. Gaga' chicken (Gallus gallus domesticus) is Indonesian germplasm that needs to be preserved with cryopreservation technology. Therefore, this study aims to determine the effect of adding L-Carnitine, hyaluronic acid, and sucrose as well as their combination in a diluent on the quality of Gaga' chicken spermatozoa after freeze-thawing. A completely randomized design was used along with 6 treatments, namely P0, P1, P2, P3, P4, and P5, containing Ringer acetate-egg yolk (RAEY) diluent (P0), RAEY + 0.063 mM hyaluronic acid (P1), RAEY + 1 mM L-carnitine (P2), RAEY + 1 mM sucrose (P3), RAEY + 0.063 mM hyaluronic acid + 1 mM L-carnitine (P4), and RAEY + 0.063 mM hyaluronic acid + 1 mM L-carnitine + 1mM sucrose (P5). Liquid semen was packed in a 0.25 mL straw, equilibrated at 5oC for 2 hours, and placed 3 cm above the surface of liquid nitrogen. It was then immersed in nitrogen for 24 hours and thawed at 60°C for 5 seconds. The results showed that there was a significant difference (P<0.05) in the motility, viability, and kinematic variables of spermatozoa after freeze-thawing. P1, P2, P3, and P4 increased progressive motility and curvilinear velocity, while viability increased with P1, P2, and P4 treatments. Furthermore, the addition of Hyaluronic acid, L-carnitine, or their combination in the RAEY diluent can improve the sperm quality of Gaga' chickens.

Gold mining activities in Indonesia, in many areas, have a negative impact on the environment. Activities that can improve the quality of post-mining land include revegetation activities, which are included in post-mining land reclamation activities. The purpose of this study was to analyze the development of vegetation in post-gold mining revegetation areas. Vegetation data were collected using the plotted line method (natural forest) and the Systematic Sampling method with Random Start (revegetation area). The analysis found that the vegetation structure in the post-gold mining revegetation area aged 3 to 10 years has complete vegetation levels (saplings, saplings, poles, and trees). Compared to natural forest, which only includes 30 tree species and 7 herbaceous, this revegetation region has a greater diversity of species, with 66 tree species and 16 herbaceous. The longer the age of revegetation is not followed by an increase in species and an increase in the value of the species diversity index. However, it is still better than the revegetation at a young age and is closer to the condition of a natural forest. The revegetation area aged 8 and 7 years has a higher number of species than natural forests and other revegetation areas.

ABB), a unique local banana cultivar from Pasuruan, East Java, Indonesia. Biodiversitas 23: 3521-3532. Characterization of local banana (Musa spp.) germplasm is fundamental for identification and provides information on traits that support their sustainable conservation and optimum utilization. This study aims to characterize and discuss the potential and conservation of Pisang Kates,a local cultivar originatingfrom Tutur, Pasuruan, East Java, Indonesia. The observed characteristics include morphology, molecular, agronomy and yield performance, leaf anatomy and stomata, pollen morphology and viability, fruit total soluble solids and nutrients. Results showed that Pisang Kates hasa unique fruit shape ovoid to globose, weight of40-150 g, appear solitary and randomly attached directly to peduncle, like papaya. It was molecularly by Internal Transcribed Spacer barcode confirmed as ABB genotype. It is tolerant to drought and banana bunchy top virus, but susceptible to wilts. Leaf anatomy has three palisades and isodiametric polygonal epidermals. Leaf stomata is amphistomatic, tetracytic on adaxial,and paracytic on abaxial. Pollen unit is monad, very large, subspheroidal, monocolpate aperture, granulum ornamentation, and low viability. Fruit moderately sweet with nutrient contents equal to dessert banana. Pisang Kates has potential as one serving consumer size fruit, ornamental fruit plant, female parent for breeding,and pioneer plant for rehabilitation programs. Both ex-situ and on-farm conservation strategies of this valuable local banana genetic resource are encouraged.

Analysis of the relationship between the ENSO phenomenon, Net Primary Productivity and catches of yellowfin tuna in Sibolga Waters, Indonesia. Biodiversitas 23: 4440-4447. The eastern Indian Ocean includes the western seas off of North Sumatra. They have a high potential for catching fish, which is a measure of their degree of primary production. Numerous factors, including oceanic conditions and the El Niño Southern Oscillation (ENSO) phenomena, frequently have an impact on its potential. Additionally, ENSO has an influence on the availability of nutrients that generate chlorophyll, a component of primary production, as well as water mass. It is also responsible for anomalies of sea surface temperature (SST) that occur in the Pacific Ocean. Therefore, this study aims to determine the effect of the ENSO phenomenon on primary productivity as well as the abundance of yellowfin tuna (Thunnus albacares) in the western waters of North Sumatra. Data were obtained from the Aqua MODIS level 3 satellite for analysis of net primary productivity (NPP) using various parameters, namely Vertically Generalized Production Model (VGPM), Ocean Niño Index (ONI), yellowfin tuna catch, winds from ERDDAP, and currents from AVISO. Its relationship with the abundance of yellowfin tuna, and ENSO was then determined using the Pearson correlation. The VGPM calculation showed that NPP for 2016-2020 ranged from 275.169 mgC/m2 to 994.898 mgC/m2. The study also revealed that the ENSO phenomenon has a weak relationship with primary productivity and the abundance of yellowfin tuna with correlation values of 0.262 and -0.191, respectively.

Abstract. Windiyani IP, Mahfut, Purnomo, Daryono BS. 2022. Morphological variations of superior sugarcane cultivars (Saccharum officinarum) from Lampung, Indonesia. Biodiversitas 23: 4110-4117. Sugarcane (Saccharum officinarum L.) is the main crop for producing sugar and for various industries. The need for sugar is continuously increasing as the population grows and the diverse demands of industrial needs. One of the efforts to increase sugar production is by developing high-yielding sugarcane varieties. In doing so, performing morphological characterization is necessary so that superior sugarcane cultivars can be identified. This study aimed to determine the morphological variations and phenetic relationships of the crosses of sugarcane cultivars of the collections of PT. Gunung Madu Plantation in Lampung, Indonesia. Morphological observation of 15 sugarcane cultivars was carried out on 37 characters covering leaves, leaf midrib, internodes, stems, eyes, and agronomical aspects. The dendrogram was built using the UPGMA cluster using the Gower’s General Similarity Coefficient algorithm, while the PCA analysis was performed using the Euclidean Distance algorithm using MVSP v.3.1 software. The results of the study showed that there were fairly high morphological variations among the sugarcane cultivars observed. The results showed the superior sugarcane cultivars of PT. GMP collections (i.e., GMP 1, GMP 2, GMP 3, GMP 4, GMP 5, GMP 6, and GMP 7) were not in one cluster due to differences between each parent. The principal component analysis of 37 characters showed that the total variation on axis I contributed 27.62% to the variation with an eigenvalue of 0.25, while axis II was 17.9% with an eigenvalue of 0.16. The information about morphological variations in this study suggests that morphological characters as taxonomic evidence are very good for identifying and analyzing local sugarcane cultivar variations and knowing the closeness of their kinship relationships.
