Biodiversitas 23: 5469-5480. Developing drought-tolerant genotypes are required to face drought condition that often occurs. The study aimed was to assess the growth, biomass, yield traits, and gas exchange of fifteen curly pepper (Capsicum annuum L.) genotypes under normal and drought-stress conditions and to determine the key traits as indicators for selecting and identifying the drought-tolerant curly pepper genotypes. Fifteen curly pepper genotypes were cultivated under normal and drought-stress conditions. Twenty-one morphological and physiological traits were recorded in the reproductive phase. The data obtained were subjected to analysis of variance, Principal Component Analysis (PCA), and UPGMA dendrogram cluster analysis. This study showed that drought stress conditions significantly reduced plant growth, biomass, yield characters, photosynthesis rate, stomatal conductance, transpiration rate, and mesophyll conductance compared to normal conditions. Based on PCA analysis, stomatal limitation, mesophyll and stomatal conductances, root length, photosynthesis rate, fruit weight per plant, chlorophyll b, number of fruit, and transpiration rate were established as indicators for screening drought tolerant genotypes. The UIN-70 and UIN-65 were highly drought-tolerant genotypes according to the MFVD index. UPGMA dendrogram displayed clustering of the curly pepper genotypes consistent with the genotype's cluster based on the MFVD index. This study's findings can be utilized to improve curly pepper's drought tolerance in the future.

Biodiversitas 23: 6547-6556. Jabon (Neolamarckia spp.) is one of the fast-growing trees with high economic value for industrial plantations. Its monoculture planting system is faced with certain problems, such as the leaf spot and leaf blight diseases caused by Pestalotiopsis sp. Therefore, the aim of this research was to describe the morphology of Pestalotiopsis species through macroscopic and microscopic observations, followed by the identification of fungus using DNA barcoding causing leaf spot and leaf blight on the tree. A total of 11 Pestalotiopsis isolates were used for this research. The morphology was identified by observing mycelium colonies and fungal conidia, while DNA barcoding was initiated after DNA isolation using cetyltrimethylammonium bromide (CTAB) method, amplification with ITS 1 and 4 primers, followed by sequencing process. The results of DNA barcoding showed that 5 species of Pestalotiopsis, namely Pseudopestalotiopsis theae, Pestalotiopsis microspora, Pestalotiopsis palmarum, Neopestalotiopsis clavispora, and Pestalotiopsis virgatula were found associated with leaf spot and blight disease. All the five species were further classified into 3 clades using phylogeny according to their median cells and apical appendage shape. The clade X consisted of P. palmarum, N. clavispora, and P. virgatula with versicolorous median cells, whereas clade Y included P. microspora with concolorous median cell, and clade Z consisted Ps. theae with concolorous median cell and knobbed apical appendage.

Abstract.  Aristya GR, Dyatama GR, Zuyyina C, Maulina NTA, Arif MF, Musthofa A, Kasiamdari RS. 2023. Relationship analysis of FaPYR1 and FaCHS genes encoding fruit ripening of three species of strawberries (Fragaria spp.) fruit. Biodiversitas 24: 87-97. The difference in the phenotypes characters and gene expressionof strawberries (Fragaria spp.) is influenced by changes in hormone signaling mechanisms to receptors due to the adaptation to various types of environments. Abscisic acid (ABA) is one of the phytohormones that plays a role in the growth and development of strawberries, response to environmental stress, and fruit ripening. One of the ABA receptors is the PYR1 protein which is encoded by FaPYR1 gene. ABA also plays a role in inducing the FaCHS gene associated with pigmentation in strawberry fruit. This study aimed to analyze the relationship between FaPYR1 and FaCHS gene sequences in three strawberry species encoding fruit ripening. The FaPYR1 (585-633  bp) and FaCHS (90-93  bp) genes from three strawberries species namely Fragaria moschata, Fragaria vesca L. 'Californica', and Fragariax ananassa 'Festival' were successfully amplified. The results of the gene family relationship analysis of FaPYR1 gene sequences showed a close relationship with the comparative three species to GenBank, with a genetic distance value of 0.01-0.02. Meanwhile, FaCHS gene sequences from three species of strawberries showed a close relationship with the comparable FaCHS gene sequence from GenBank, with a genetic distance of 0.1-0.2.

Genetic variability for micronutrients under an aerobic condition in local landraces of rice from different agroecologicalregions of Karnataka, India. Biodiversitas 24: 1-10.Genetic variability is a fundamental reference for the innovative development of breeding programs to improve varieties and desirable traits. This study aimed to assess genetic variability for micronutrients (iron and zinc) and protein content under an aerobic condition in local landraces of rice (Oryza sativa L.) from different agroecological regions of Karnataka, India. Fifty-fivelocal landraces of ricefrom India were studied.High phenotypic and genotypic coefficient of variation was observed for grain yield per plant and moderate for the remaining traits. Heritability was high for most of the traits observed while moderatefor harvest index and grain yield plant-1. The genetic advance (GA) was high for the majority of yield and its attributing traits. GA as mean was high for grain yield per plant. Based on D2value, 50 genotypes were grouped into 8 clusters. Clusters VI and VIII are the largest and had accessions with the highest protein and zinc content. Marabatta, Jeerige Batta, Neerguli, Mugadh Sughanda, and Kesarnellu genotypes contained high grain protein and zinc content, whereas Naga Batta genotype has high iron content. Among the superior nutrient landraces, Neerguli, Jeerige Batta, DBN, Kesarnellu, and Doddiga genotypes had high grain yield plant-1.

Phenolic and flavonoid content with agro morphological characters of 12 accessions of Justica gendarussa grew in Indonesia. Biodiversitas 23: 5101-5107. Justicia gendarussa Burm. f is a medicinal plant used in traditional and modern medicine. This study was conducted to determine the agro morphological characters, phenolic and flavonoid content of 12 accessions of J. gendarussa plants obtained from various locations in Indonesia and then grown in the same conditions and places. The agro morphological characteristics include plant height, number of leaves, number of branches, wet weight, and dry weight of leaves and stems. Total phenolic content (TPC) and total flavonoid content (TFC) were analyzed by spectrophotometric method. The results showed significant differences in the characters of 12 accessions of J. gendarussa (p<0.05). Accessions of BK4 (Bekasi 4) and KP (Kejayan Pasuruan) had the best agro morphological characteristics. Accessions of BK4 (Bekasi 4) and BG (Bogor) had the best growth rates. The highest TPC value was obtained from TG (Trenggalek) accession (0.752 mg GAE/g DW) and the highest TFC (1.899 mg QE/g DW from LK (Langkat) accession. The best accessions based on agro morphological characters were BK4 (Bekasi 4) and KP (Kejayan Pasuruan), and based on the content of polyphenols were TG (Trenggalek) and LK (Langkat). The accessions of BK4, BG, TG, and LK could be further developed into superior varieties of J. gendarussa.

Isolation and molecular identification of agarwood-inducing fungi and their virulence test using Aquilaria sp. seedlings. Biodiversitas 24: 140-148. Agarwood is a lump of fragrant resin with a dark color and distinctive aroma from secondary metabolite products that accumulate in wood fibers produced from the genus Aquilaria and Gyrinops, a plant defense mechanism against damage caused by wounds and infection of specific fungal pathogens. The three main factors in producing agarwood sapwood are the presence of plants that have the potential to produce agarwood (family Thymelaceae) as a host, a supportive environment, and the presence of specific fungal pathogens. The isolation, identification, and virulence testing of local isolates must be made to get the correct agarwood-inducing pathogens without posing potential environmental problems. A total of five explants of Aquilaria sp. branches producing agarwood from the study site were used. This study aims to isolate local strains of fungi associated with agarwood formation from North Aceh regency, Aceh province, Indonesia, figure out their virulence against and ability to induce agarwood from seedlings of target trees, and determine their species identity. The experiments comprised isolation, macroscopic and microscopic identification, virulence tests, and species identification based on ITS sequences. The study resulted in five local Aceh isolates (LAI-1- LAI-5). Four isolates (LAI-1 - LAI-4) can induce agarwood formation, and oneisolate (LAI-5) is a white rot fungus. The ITS sequence analysis showed that LAI-1 - LAI-4 were Fusarium solani, while two colonies taken from LAI-5 isolate were identified as Ceriporia lacerataor Irpex laceratus (LAI-5 colony#1), and Daldinia eschscholtzii (LAI-5 colony #3). Based on these findings, the LA-5 isolate was probably a mixture of the two white rot fungus isolates.

Biodiversitas 24: 492-497. Over-exploitation of important indigenous plant species impact negatively on their population structure. Unmonitored and unsustainable harvesting from plant ecosystems may inhibit plant recovery. The aim of the study was to investigate the population structure of Spirostachys africana in a communal area of Ha-Matsa Village in Limpopo Province, South Africa. Eleven transects of 100m x 10m were used to sample ecological parameter data from 66 plant specimens of S. africana species. Transects were subjectively placed within the population of S. africana. Data on plant height size classes and basal stem diameter size classes exhibited an almost Bell-shaped population curve. The population of S. africana did not have many seedlings, with the Crown health status data revealing a population with generally moderately damaged crowns (24%), while 19% of individuals had healthy crowns. The concern is the recorded 9% of dead crowns. Although the population looks great from a distance, the sampled ecological data revealed an unviable population. The Spirostachys africana population of Ha-Matsa Village does not have more individuals in the small size classes of both height and basal stem diameter. Conservation practice implies that if anthropogenic activities are not properly monitored, there may not be enough young individuals to support the population of S. africana into the future. Therefore, this study aimed to improve the assessment and the effects of harvesting on the S. africana species population. This study also suggests important information and guides ecologists and conservationists on sustainable harvesting practices and offtakes of S. africana.

Biodiversitas 23: 5806-5813. Salmonella spp. are bacteria that causes enteric fever. Salmonella infection is a serious burden in both developed and developing countries, including Indonesia. Because of infrastructure limitation, Salmonella spp. infection treatment mostly based on clinical observation without proper identification of the causative bacteria. Although the clinical manifestation of the diseases is difficult to distinguished, it is believed that Salmonella spp. diversity is high in the community. This work aims to analyze the diversity of Salmonella spp. and their susceptibility against antibiotics. The Salmonella spp clinical isolates obtained from three clinical laboratories were subjected to cultivation, phenotypic characterization, and further biochemical and molecular characterization. Biochemical properties of strains were analyzed, whereas protein profiles were analyzed using the SDS-PAGE method. The molecular characterization was carried out by sequencing of 16S rRNA encoded gene. Numeric-phenetic clustering analysis of the 18 Salmonella spp clinical isolates showed that, they were allocated to two large clusters, namely Cluster I and Cluster II. Reconstruction of the phylogeny tree of the 18 clinical isolates and the reference strains showed that all of them belonged to one clade, namely the Salmonella enterica subsp. Enterica. However, it was recognized also six sub-clades. The antibiotic susceptibility pattern of Salmonella spp. revealed that practically all antibiotics (80%) had a high proportion of susceptibility, except for cefazoline, gentamicin, and amikacin, which have zero susceptibility. Numeric-phenetic clustering analysis combined with phylogeny tree reconstruction showed the diversity of Salmonella spp. circulating in Yogyakarta with alarming signal of certain antibiotic resistance.

Abstract. Sitanggang NDH, Zuhud EAM, Masy’ud B, Soekmadi. 2022. Ethnobotany of the Toba Batak Ethnic Community in Samosir District, North Sumatra, Indonesia. Biodiversitas 23: 6114-6118. The Toba Batak ethnic community is one of the ethnic groups in Indonesia that is believed to have unique characteristics in terms of ethnobotanical aspects. The objectives of this study were to identify the diversity of plant species that were used and collected by the Toba Batak ethnic community, analyze their Cultural Significance Index and determine the economic potential of plant species of high cultural value. The survey was conducted in four villages near Samosir Botanical Gardens and Aek Natonang Lake. We collected data through field observations and interviews with key respondents of chiefs, healers, homemakers and youth as users. We analyzed the data to determine the number of plant species collected and used by the Toba Batak ethnic community, to calculate the cultural importance of the plant species, and to determine the economic potential of the plant species. The result showed that the community collected and used 146 plant species from 53 families. We distinguished it into three categories, food (66 species), medicine (67 species) and traditional rituals (13 species). Twenty plant species had the highest cultural value (ICS > 30). Nine species had the highest cultural value, i.e., ginger (Zingiber officinale) (54), rice (Oryza sativa) (50), garlic (Allium sativum) and turmeric (Curcuma longa) 48), mango (Mangifera indica) and sweet potato (Ipomoea batatas) (40), and lime (Citrus aurantifolia), kaffir lime (Citrus hystrix) and rambutan (Nephelium lappaceum) (39). Six of the nine plant species with high cultural value were considered potential and priority economic commodities. These species were ginger, rice, garlic, turmeric, mango and sweet potato.

Abstract. Saadah S, Tulandi SM, Rohman RA. 2022. Phytochemical and gas chromatography-mass spectrometry profiling of two plant parts of Sandoricum koetjape. Biodiversitas 23: 6199-6207. Sandoricum koetjape has been used in traditional Indonesian medicine for centuries. In Indonesia, the stem and leaves were used to treat helminthiasis, cough, stomachache, diarrhea, bloating, leucorrhoea, colic, and fever. To ascertain the phytochemical constituents of each plant part, and provide a sufficient basis for clinical application, so Gas Chromatography-Mass Spectrometry (GC/MS) analysis of stems and leaves was established. Phytochemical screening showed that stems and leaves contained alkaloids, flavonoids, quinone, triterpenoids, and tannin. Total phenolics in the leaves are higher than in the stem. Saponin and steroids were not detected. The GC/MS analysis indicated that the leaf extract contained more identified compounds (30) than the stem extract (28). Both S. koetjape samples contained 1H-Cycloprop[e]azulen-7-ol. The major compounds of the stem extract of S. koetjape are Fonenol (12.89%), 1H-Cyclopenta[1,3]cyclopropa[1,2]benzene (12.54%), and 1H-Cycloprop[e]azulen-7-ol (11.70%), while the major compounds in leaf methanol extract were 14,15-didehydro-Cyclodecacyclotetradecene (23.71%), 1H-Cycloprop[e]azulen-7-ol (17.04%), and Solanesol (8.34%). The two main components of the variable can account for 50.92% of the total variance, according to Principal Component Analysis (PCA) (PC1 is 21.19% and PC2 is 29.74%). These results indicate that the two components effectively categorize samples with distinct characteristics. The groups were divided based on the type of sample used, namely stem and leaf samples. According to the loading plot, 1H Cyclopenta[1,3]cyclopropa[1,2]benzene and Fonenol are two components that are very effective in categorizing samples based on their type. Both metabolites are higher in the stem than in the leaf. However, the metabolites activities of S. koetjape stem and leaves extracts could be revealed by further studies using in silico and in vitro approaches.
