Abstract

Vegetable oil can be produced from a perennial plant such as coconut (Cocos nucifera L.). There are few techniques for coconut oil extraction, such as physical, chemical, and fermentative processes. The fermentation process uses microbial inoculum as starter. Ground coconut meat was soaked in warm water, than squeezed several times to get coconut milk. After being allowed to stand for 4-5 hours, it separated into two layers, cream and skim. Starter was prepared from a mixture of milk and coconut water (1:9, v/v) which enriched with 2% tomato extract, 0.5% urea, and 1.0% molasses and then preincubated for 5 days under agitation. Starter with different concentration (1.0; 2.5; 5.0; and 10%) were added onto coconut milk and allowed to be fermented for over night. The extracting oil was analyzed for further experiment, especially, on its antibacterial activity. The maximum yield of 23% was achieved by using 2.5% starter. Total protein, fat, FFA, and cholesterol content of the fermented coconut oil were 0.05%, 96.45%, 0.29%, and 0.008%, respectively. The gas chromatogram showed that this oil contained high lauric acid (46.20%), and 13.94% miristic, 5.97% palmitic, 9.00% palmitoleic, and 19.73% stearic acid, respectively.© 2008 Jurusan Biologi FMIPA UNS SurakartaKey words: coconut oil, fermentation, antibacterial activity, biochemistry analysis.

Abstract

Amorphophallus titanum Becc. (Araceae), is known by titan arum (western) or bunga bangkai (Indonesian), is the bulkiest inflorescence in the world. The species is valued for its unique character of their inflorescence. The species has been overexploited and is considered as vulnerable plant species. The present study is carried out to assess genetic diversity and to estimate genetic relationship among 22 accessions collected from two different populations in Sumatra using Random Amplified Polymorphic DNA (RAPD). Nine RAPD primers generated 143 scorable bands which 137 (95.84%) of them were polymorphic. Clustering analysis was performed based on RAPD profiles using the UPGMA method. The range of genetic dissimilarity value among accessions was from 0.14 to 0.59, while the range of genetic dissimilarity among populations was from 0.67 and 0.77. Abstract

RAPD-PCR method and isozyme analysis were used to obtain information of genetic relationship among cucumber varieties. Such information is urgently utilized to support plant breeding program of cucumber. Research was done at the Biotechnology Laboratory of Plant Pest and Diseases, Faculty of Agriculture of Brawijaya University, Malang and Molecular Biology Laboratory, Faculty of Matemathic and Natural Sciences of Brawijaya University, Malang. DNA isolation was done using CTAB method by additional NaCl modification. Sixteen primers from operon were employed to amplify DNA genome by RAPD-PCR. Two enzymes, Esterase and AAT were chosen for isozyme analysis. Clad 97 Program was used for analyzing the data and results in data grouping based on proximity value. Cluster analysis based on isozyme data indicated that there was an adequate lower genetic variation in cucumber, where seven of nine tested varieties showed proximity value of 1.00. Eleven of sixteen primers in RAPD-PCR analysis produced DNA bands. Relativity analysis by using RAPD-PCR method showed high enough of genetic variation. Relativity analysis by using both methods showed that variety 07 was the furthermost. Abstract

The aim of this study was to undstand chemical properties of different rattan species traded in Central Sulawesi Province. The chemical of rattan were tested according to SII.1657-85 for wich character of holoselulosa, SII.0443-81 for wich character of -selulosa, SII.0528-81 for wich character of lignin, and SII.1292.85 for wich character of silica. The study indicated that chemical properties of rattan varied amongst the species. The differences are supposed to have important criteria in identifying different species of rattan species traded in Central Sulawesi. Based on the result the highest quality of rattan is Calamus inops and the lowest is Calamus ornatus var. celebicus (lambang).© 2008 Jurusan Biologi FMIPA UNS SurakartaKey words: chemical, rattan, traded.

Abstract

Sandalwood (Santalum album L.) is one of the economic plants that are threatened rare. Belu regency is a part of East Nusa Tenggara Province was known as center of natural distribution of sandalwood. In 2005 was found some mother seed trees at several locations in Belu are Suriklulik, Lelowae, Biau, Uabau, Haitimuk, Alas and Tialai. For research propose every location was chosen only one mother seed tree that was marked with capital letters as followed ; A is mother seed tree from Suriklulik, B from Lelowae, C from Biau, D from Uabau, E from Haitimuk, F from Alas and G from Tialai, In this research was studied the germination of sandalwood seeds from severalmother seed trees was mentioned above and some aspects of seedling growth include model of seedling growth and correlation between height and number of leaves growth during 5 months. This research was carried out in paranet house during 5 months and seedling was planted in polybag. Research was designed follow the Completely Randomized Design with 4 replications. The result of this study as follow: The mother seed trees of sandalwood influenced germination process and seedling growth especially on height and number leavesof seedlings. Seedling growth from each mother seed trees follow models of logistic growth with high value of coefficient determination about > 0,83, and be found also, close correlation between growth of seedling height with growth of seedling leaves was marked by high value of coefficient correlation about >0.85.Several mother seed trees was marked with capital letter E, D, C, A and G are better than F and B.© 2008 Jurusan Biologi FMIPA UNS SurakartaKey words: Santalum album L., mother seed trees, germination, seedling growth, Belu.

Abstract

Starch of ganyong is one of the sources of ethanol which is able to be produced by acid hydrolysis and fermentation process. It had high concentration of carbohydrate that is 80%, so it could produce glucose highly within acid hydrolysis process. The result showed that the optimal amount of reducing sugar had been produced by nitrate acid 7% (dextrose equivalent, DE = 28.4). Nevertheless, type and concentration of acid had no significantly correlation to reducing sugar yielded. The total amount of glucose had correlation to amount of ethanol, in fermentation process. The optimal amount of ethanol was yielded from 4.81% of glucose and it produced about 4.84% ethanol. The more amount of glucose was yielded the more ethanol was produced. Abstract

Styphelia abnormis (Sond.) J.J. Smith is a member of family Epacridaceae that grows in East Waigeo Nature Reserve (Cagar Alam Waigeo Timur). An inventory was done using 30 sampling plots of 2 x 2 m2. The results were: density 2.375 per m2 and presence 90%. This species grew as a shrub on ultramafic soil in open hilly areas at 50-80 above sea level (a sl). Abstract

Sulasih Talang Nature Reserve is a conservation area in West Sumatra Province, which contains high diversity of plants including Araliaceae. Araliaceae mostly consist of attractive plants which have potentiallity as ornamental plants. The aim of this research is to record the diversity of Araliaceae in Sulasih Talang Nature Reserve. The result of the study indicates that there are, species of Araliaceae found in Sulasih Talang Nature Reserve, which are : Arthrophyllum diversifolium, Brassaiopsis glomerulata, Brassaiopsis sumatrana var. variaefolia, Macropanax dispermus, Schefflera elliptica, S. farinosa, S. lucescens, S. scandens, Schefflera sp., and Trevesia beccarii, in which Bogor Botanic Garden collected 6 species of them, i.e.: Brassaiopsis glomerulata, Brassaiopsis sumatrana var. variaefolia, Macropanax dispermus, Schefflera elliptica, S. scandens, Schefflera sp. Abstract

The objective of this study was to found on composition and structure of vegetation of upstream Cianjur Watersheds at the montane zone Mt. Gede Pangrango National Park. Research sampling conducted by using plot method placed purposive random sampling at research location with increase elevation equal to 300 m from sea level. Every 300 m elevation level, were taken 10 sample plots so that up to 1600- 2400 (montane zone) totalize plot were 30. Measured parameter cover density, frequency, dominancy, Important Value Index (IVI), Diversity Index (H'), and Evenness (E). The subdividing pattern plot had been done through cluster analysis. Result of research indicatethat species puspa (Schima wallichii) and rasamala (Altingia excelsa) were two species with highest Important Value Index, so that both species considered to be dominant the community. The species diversity index (H’) equal to 3.78 categorized to high, but the evenness value (E) was categorized to low (1.26). The phenomena happened because the distribution of species was heterogeneous. The Cluster Analysis indicated that there were three sub-groups which can be categorized as sub-zone in sub-montane zone were the sub-zone lower, middle and high.© 2008 Jurusan Biologi FMIPA UNS SurakartaKey words: structure and composition, vegetation, montane zone, Mt. Abstract

This research was conducted in the forest of Selayar Island and in the Nature Reserve on forest of Patunuang South Sulawesi. The study sites were located at 0-250 m above sea level on four locations. Data being collected were the distribution, type of feeds, nest sites, habitat utilization and of type. During the observation, we found at least 10 nest sites which were distributed on three locations. The tarsier consume mainly insect, and it isn’t collected.
