Diversity and potential of herbaceous plants as mercury (Hg) hyperaccumulators in small-scale gold mining sites in Pancurendang, Banyumas, Indonesia. Biodiversitas 24: 3378-3386. Plants resistant to mercury can be used as an alternative to processing gold mining waste with the phytoremediation method. The study aimed to inventory herbaceous plant species in artisanal and small-scale gold mining (ASGM) sites in Pancurendang Village (Banyumas, Central Java, Indonesia), measure mercury concentrations in plants and root zones, and analyze the potential of herbaceous plants as mercury (Hg) hyperaccumulators. Herbaceous plants were cataloged by purposive sampling with multiple quadrats of 2m x 2m in 14 locations and then analyzed to obtain density, frequency, dominance, importance value, and diversity index. Biomass and Hg concentrations were measured from selected herbaceous plants and soil samples of the root zone. The Biological Accumulation Coefficient (BAC) was calculated to determine the plants' potential as Hg hyperaccumulators. Plant inventory identified 54 herbaceous plant species belonging to 26 families, with Paspalum conjugatum P.J.Bergius being the dominant species. In the root zones, Hg was found at 11-73 ppm and 7.6-85.36 ppm in 19 species. Ipomea aquatica Forssk. accumulated the highest concentration (85.36 ppm) and absorbed the greatest amount of Hg (5.25 mg). Based on their BACs, the nineteen species were categorized into moderate to high (hyper) accumulators, with Plectranthus sp. having the highest BAC of 4.54.

Abstract. Prabowo TA, Bintara S, Yusiati LM, Sitaresmi PI, Widayati. 2023. Evaluation Deoxyribonucleic acid (DNA) fragmentation of local Indonesian cattle frozen sperm using Halomax method. Biodiversitas 24: 2225-2230. Deoxyribonucleic acid (DNA) is a molecular chain containing genetic material that determines the quality of each individual produced. DNA damage in cattle can affect fertility and miscarriage of embryos. There is no simple research on DNA damage in local Indonesian cattle (Bos indicus Linnaeus 1758); therefore, miscarriage and infertility cases in Indonesia are still high, one of which can be attributed to sperm DNA damage. This study aimed to evaluate DNA damage in the frozen semen of local Indonesian cattle and suggested SDF (Sperm DNA Fragmentation) inclusion screening as the main requirement for frozen semen. The materials used in this study were 54 straws of frozen semen from nine males from three different types of local cattle, including Bali, Madura, and Aceh. Each type consisted of three bulls, each with six replicates. Sperm-Bos-Halomax was used to examine sperm DNA damage. Data were analyzed using one-way analysis of variance (ANOVA). Aceh cattle had the highest post average compared to Bali and Madura at 63.33±2.43%. Abnormality of Balinese cattle had the lowest average compared to Aceh and Madura at 8.83±1.04%. Evaluation of sperm DNA damage in local cattle using the Sperm-Bos-Halomax® method showed that the highest average DNA damage was 1.72±0.62% in Aceh, and the lowest was 1.67±0.66% in Madura cattle. The sperm DNA damage in Bali cattle was 1.67±0.70%. The sperm DNA quality in the frozen semen of the local cattle in this study was good, but it is still necessary to determine SDF as the main standard in frozen semen.

The tongue morphology of Pteropus vampyrus from Timor Island, Indonesia: New insights from scanning electron and light microscopic studies. Biodiversitas 24: 3512-3518. The large flying fox (Pteropus vampyrus Linnaeus, 1758) is a Southeast Asian megabat species includes with frugivorous bats. The tongue plays a pivotal role in taking, chewing, and swallowing food. The structure of the bat tongue hampers considerable variation, mainly in the papilla. Variations occur owing to the feeding habits, environment, and adaptation of bats to their environments. The aim of this study was to clarify the morphological structure of the tongue of P. vampyrus obtained from the island of Timor, East Nusa Tenggara, Indonesia, by using Scanning Electron Microscope (SEM) and Light Microscopy(LM). This study included six adult bats regardless of sex. Macroscopically, the tongue of P. vampyrus consists of three parts: the apex, corpus, and radix. SEM and LM confirmed that the apex presents filiform papillae of several subtypes, including scale-like filiform, giant trifid, and small crown-like papillae. In addition, the apex features fungiform and transitional papillae between the giant trifid and small crown-like papillae. Furthermore, the corpus consists of filiform papillae (leaf-like filiform and large crown-like papillae) and fungiform papillae. The radix consists of filiform papillae (long conical, leaf-like filiform, and short conical papillae), fungiform papillae, and three V-shaped circumvallate papillae pointing to the larynx.

Utilizing DNA barcoding approach to study the diversity of the blue swimming crab from Tuban District, East Java, Indonesia. Biodiversitas 24: 4731-4737. Blue swimming crabs are one of the economically significant fisheries resources in Indonesia. However, Indonesia's blue swimming crab fisheries still depend on wild catches that lead to a non-target species harvest, especially the cryptic swimming crabs. This study aims to investigate the genetic diversity of blue swimming crab specimens and other cryptic species using a molecular approach (DNA Barcoding). The samples were caught from the waters of Tuban District, East Java, Indonesia, using traditional foldable traps or "bubu lipat". This study utilized the Polymerase Chain Reaction (PCR) method targeting the Cytochrome c Oxidase Subunit I (COI) gene. The sequences were then compared to the NCBI database to get the closest species based on the sequences. 21 out of 23 samples were identified as Portunus pelagicus, and two samples were identified as Charybdis feriata with percentage of similarity ranging from 99.68% to 100%. The phylogenetic trees were constructed using Neighbor Joining and Maximum Likelihood method with sequences from Scylla serrataas an outgroup. The genetic distance between P. pelagicus and C. feriata was 16%, whereas the genetic distance between P. pelagicus and C. feriata to the outgroup was 20%. The molecular method is crucial for species identification and population stock assessment. This information helps in determining stock boundaries, estimating population sizes, and understanding the genetic health of populations. Genetic data will help the government to have a better management of the fisheries activities in Indonesia by providing the diversity and population data.

Antibacterial and anti-inflammatory potential of three sea cucumber species from Tukuran, Zamboanga del Sur, Mindanao, Philippines. Biodiversitas 24: 2527-2535. The marine environment is a rich source of potential antibacterial and anti-inflammatory agents. This study investigated three sea cucumber species' antibacterial and anti-inflammatory activity from Tukuran, Zamboanga del Sur, Philippines. The methanol: ethyl acetate crude extracts of Holothuria scabra, Stichopus sp., and Holothuria atra were tested for their antibacterial activity against gram-negative (Pseudomonas aeruginosa, Escherichia coli) and gram-positive (Staphylococcus aureus, Bacillus subtilis) bacteria using modified microtiter plate method. Results revealed that sea cucumber extracts have MIC values of 0.0008-0.0944 mg/mL. One-way ANOVA results revealed a p value <0.05, and Tukey’s pairwise result confirmed that the antibacterial activity of H. scabra extract is not comparable with Ciprofloxacin (p value <0.05). However, Stichopus sp., and H. atra (1) and H. atra (2) revealed a comparable antibacterial activity with Ciprofloxacin (p value >0.05). The crude extracts were also evaluated for in vitro anti-inflammatory activity using the albumin denaturation method. The results showed a percentage inhibition within the range from 44.50-49.69%, 44.74-51.44%, 31.82-49.48%, and 49.16-52.15% inhibition at 200-, 100-, 50-, and 25 ppm, respectively. One-way ANOVA and Tukey’s pairwise result revealed that the sea cucumber extracts showed a comparable percent inhibition with the positive controls, Celecoxib and Diclofenac sodium, at 200 ppm. This study revealed sea cucumber crude extracts as a potential antibacterial and anti-inflammatory agent.

Diversity, carbon stock and associated biota of seagrass beds in Central Tapanuli District, North Sumatra, Indonesia. Biodiversitas 24: 2080-2087. Various human activities emit carbon dioxide which drives global warming and climate change. Seagrass ecosystems can absorb and store enormous quantities of carbon, hence reducing carbon emissions. This research aimed to assess the diversity, vegetation cover, carbon stock and associated biota of seagrass beds in Central Tapanuli District, North Sumatra, Indonesia. Purposive sampling method was employed with data collection used the line transect quadrant method. Carbon stock was measured using the loss of ignition method. There were four species of seagrass in the studied area, namely Enhalus acoroides (L.f.) Royle, Thalassia hemprichii (Ehrenb. ex Solms) Asch., Cymodocea rotundata Asch. & Schweinf. and Halodule uninervis (Forssk.) Boiss. with a density ranging from 23-164 stands/m2. Seagrass cover ranged from 3.87%-34.37%. Associated biota (fish and crustaceans) at each observation station were Siganus javus Linnaeus 1766, Siganus canaliculatus Park 1797, Siganus vermiculatus Valenciennes 1835, Colomesus psittacus Bloch & Schneider 1801, Epinephelus fuscoguttatus Forsskål 1775, Terapon jarbua Forsskål 1775, Ambassis dussumieri Cuvier 1828, Cryptocentrus leptocephalus Bleeker 1876, Leiognathus robustus Sparks & Dunlap 2004, Portunus armatus A.Milne-Edwards 1861, Rhinolambrus pelagicus Rüppell 1830 and Hippolyte obliquimanus Dana 1852. The total carbon stock of the top substrate (leaves) was 12.8 tons, while carbon bottom substrate (rhizomes and roots) was 12.9 tons, resulting in a total carbon stock of 25.7 tons. The analysis of water quality parameters indicated that the studied area had suitable environmental conditions for seagrass growth. The results of this study add new information regarding seagrass ecology of the western region of Indonesia, especially Sumatra.

Biodiversitas 24: 2261-2271. The study of species diversity by survey method and traditional utilization by interviewing local philosophers, folk healers, elders, and villagers with semi-structured interviews of bamboos (Poaceae) in the Phu Thai Ethnic Group in four provinces including Kalasin, Mukdahan, Nakhon Phanom, and Sakon Nakhon Provinces, in the Northeast of Thailand was conducted from May 2019 to August 2021. In addition, the aim of this research is to study the species diversity and traditional utilization of Bamboo (Poaceae) in the Phu Thai Ethnic Group in Nakhon Phanom, Sakon Nakhon, Kalasin, and Mukdahan Provinces, in Northeastern Thailand. Survey Five genera, and 13 species including Bambusa bambos (L.) Voss, B. beecheyana Munro, B. burmanica Gamble, B. nutans Wall. ex Munro, B. spinosa Roxb., B. vulgaris Schrad. ex J.C.Wendl., Dendrocalamus asper (Schult. & Schult.f.) Backer ex K. Heyne, D. giganteus Munro, D. sinicus L.C. Chia & J.L.Sun, Gigantochloa albociliata (Munro) Kurz, Thyrsostachys siamensis Gamble, ×Thyrsocalamus liang Sungkaew & W.L. Goh, and Vietnamosasa pusilla (A. Chev. & A. Camus) T.Q. Nguyen were found. The genus with the highest species is Bambusa, with seven species, followed by Dendrocalamus (3 species). One species per genus was found in the other three genera: Gigantochloa, Thyrsostachys, and Vietnamosasa. Vernacular name, distribution, ecology, and phenology are present here. The traditional utilization of bamboos in the Phu Thai Ethnic Group in these four provinces indicated that bamboo was used as food for 13 species from five genera. While six species in three genera were used for medicinal purposes, 13 species from five genera were used as equipment, appliances, and housing materials, seven species from four genera were used as ornamental plants, and 13 species from five genera were used in ritualistic ceremonies. The data on species diversity and traditional utilization of bamboos in the Phu Thai Ethnic Group in these four provinces in Northeastern Thailand were newly reported.

Abstract. Ambarwati A, Santoso B, Sofyan A. 2023. Phylogenetic analysis of Streptomyces producing antimicrobial agent isolated from Kukup Beach Sand, Yogyakarta, Indonesia. Biodiversitas 24: 2374-2383. Microorganisms such as bacteria, fungi, and Streptomyces are sources of bioactive compounds. Streptomyces is known as the largest antibiotic-producing genus. This study aimed to determine the antimicrobial activity of Streptomyces isolates and analyze the relationship between Streptomyces isolates based on the 16S rRNA gene sequence. The Streptomyces isolates were screened for their antimicrobial activity based on their ability to inhibit test bacteria growth using the agar block method. The selected isolates that showed antimicrobial activity were molecularly characterized using 16S rRNA gene sequence analysis. The result showed 32 Streptomyces isolates successfully isolated on SCA and Raffinosa Histidine Agar media. Among 32 isolates, it was observed that five isolates demonstrated the ability to inhibit the test bacteria growth on 9 to 25 mm of inhibition zone diameters. The BRI-18 isolate showed the highest antimicrobial activity inhibiting Bacillus subtilis G. FNCC 0060 on 25 mm of inhibition zone diameter (strong inhibition category). Based on 16S rRNA sequences, it was known that five isolates belonged to Streptomyces. BLAST analysis on the ARA-5 isolate revealed that it was closely related to Streptomyces griseoincarnatus strain JCM 4381 with 99.62% sequence similarities. The AR3-29 isolate was a sister clade identical to Streptomyces rochei strain NRRL B-1559 with a 99.35% similarity level. The BRI-18 and BRI-19 isolates were the most similar to Streptomyces fradiae strain NBRC 12773 on 96.87% and 95.15% index similarity, respectively, whereas the BRI-35 isolate demonstrated the highest similarity to Streptomyces zhihengii strain YIM T102 (97.02%). The research demonstrated that Streptomyces isolates from Kukup beach sand showed the potential as an antimicrobial agent.

Abstract. Handayanta E, Hadi RF. 2023. Screening the anthelmintic effects of leaves and fruits extracts from various Acacia species as a potential feed for small-ruminant animals. Biodiversitas 24: 3140-3144. This study aimed to determine the anthelmintic properties of leaves and fruits obtained from various Acaciaspecies against Haemonchus contortus Rudolphi, 1803. Both leaf and fruit samples were allocated into five different groups: negative control (NC), 0.9% of NaCl solution; positive control (PC), albendazole solution; and solutions containing 12% concentration of each Acacia auriculiformis A.Cunn. ex Benth.(AA), Acacia mangium Willd.(AM), and Acacia crassicarpa A.Cunn. ex Benth.(AC) powder. The added concentration was set at 12%, and each treatment group was brought to incubation in 6 different time parameters (1, 2, 3, 4, 5, and 6 hours) against H. contortus. Each Acacia leaves extract did not significantly differ in eliminating H. contortus, unless for AM under 4 hours of incubation (p<0.05), wherein at the final incubation time, all Acacia-treated samples displayed complete mortality. Meanwhile, treatment with Acacia fruits during 6-hour incubations led to a removal of 76.66-83.33 % H. contortus population, in which Albendazole-treated groups showed complete mortality. Thus, this study suggested the leaves from Acacia are a potential animal feed as anthelmintic against H. contortus parasites. However, further studies investigating the potential mechanism by which various Acacia species present anthelmintic activity are necessary.

Short Communication: Dacini tribe's fruit fly species in Depok (Indonesia) with special reference to the abundance of orchard fly, Bactrocera dorsalis, for fruit pest controlling. Biodiversitas 24: 2447-2457. Knowledge of diversity and regularly monitoring pest insects in fields is crucial in crop protection and is key to effective pest control management. Our research group aims to map the species diversity of the Dacini tribe's fruit flies in Depok (Indonesia) using lure fruit fly traps and rearing fruit fly larvae from infected fruits. This preliminary study result is important for our next study to control fruit flies using tropical fruit extract as a natural lure. Four fruit flies were obtained from infected fruit purchased from a traditional fruit seller: Bactrocera dorsalis (Hendel 1912) and Bactrocera albistrigata (Meijere 1911) from Syzygium aqueum (Burm.fil.) Alston fruit and Bactrocera fuscitibia (Drew & Hancock 1994) and Bactrocera carambolae (Drew & Hancock 1994) from arum manis (Mangifera indica L.) fruit. Meanwhile, three fruit fly species were caught in lure traps using methyl eugenol: B. dorsalis, Bactrocera umbrosa (Fabricius 1805), and Bactrocera musae (Tryon 1927). Detailed morphological descriptions were provided for each species obtained. The B. dorsalis was the most abundant, while B. musae were the least abundant. This study's total number of individuals was 6,329 for B. dorsalis, 485 for B. umbrosa, and 34 for B. musae. The B. dorsalis is widely distributed in many tropical areas of Asia-Pacific and is considered an invasive species in Africa. Moreover, it is well known as one of the most important pest species within the Tephritidae family. While the remaining obtained fruit fly species have also been reported as pests, it is interesting to note that B. fuscitibia was reported as a non-pest species but was obtained from an infected mango fruit in this preliminary study. The B. musae was also reported for the first time in Depok, West Java Province, Indonesia.
