Abstract

Abstract. Laksmiani NPL. 2019. Ethanolic extract of mangosteen (Garcinia mangostana) pericarp as sensitivity enhancer of doxorubicin on MCF-7 cells by inhibiting P-glycoprotein. Nusantara Bioscience 11: 49-55. Generally, doxorubicin is used in combination with other anticancer agents. Reduction side effects tend to be better in combination use than the use of single doxorubicin. Therefore the development of anticancer agents with low side effects and combination agents that decrease the side effects of doxorubicin still need to be pursued. The use of doxorubicin also shows a phenomenon of resistance due to over-expression of P-glycoprotein (Pgp). The research was conducted to evaluate the cytotoxic effect of lower dose doxorubicin combine with ethanolic extract of mangosteen pericarp (EEMP) on MCF-7 cells and determine the mechanism of alpha (a)-mangostin as the active compound in EEMP that contribute increasing sensitivity of doxorubicin on MCF-7 through Pgp by in silico. The cytotoxic activity was observed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Molecular docking with 4.2 autodock program was used to evaluate the interaction of a-mangostin with Pgp. EEMP has IC50 of 54 ?g/mL on MCF-7 cells while for doxorubicin has IC50 of 1392 nM. Exposure EEMP enhanced cytotoxic effect of doxorubicin on MCF-7 cells. Solely doxorubicin 750 nM treatment gave cell viability percentage 94.98%, but when combined with EEMP 22.5 ?g/mL, the cell viability percentage was reduced to 16.91%. This shows that EEMP potent to be used as a combination agent for doxorubicin chemotherapy. The in silico assay demonstrate that a-mangostin has an affinity for Pgp with binding energy of -6.13 kcal/mol. Keywords: EEMP, doxorubicin, MCF-7, Pgp, combination

Abstract

Abstract. Murningsih T, Praptiwi, Liana, Fathoni A. 2019. TLC profiling and antioxidant activity of phenolic compound from Sterculia oblongata bark extract. Nusantara Bioscience 11: 44-48. In the present study, we investigated the total phenolic content and in- vitro antioxidant activity of Sterculia oblongata. Total phenolic content (TPC) of the methanol extract was examined by Folin-Ciocalteu reagent and gallic acid as standard. The in-vitro antioxidant activity was performed by DPPH free radical scavenging activity and ?-carotene bleaching assay. The result showed that the TPC of methanol extract was the highest (P<0.05) compared to other extracts. There is a positive and significantly very high Pearson’s correlation between antioxidant activity index (AAI) and TPC (r = 0.996). A negative and significantly high Pearson’s correlation was obtained between IC50 value and TPC (r = -0.885). The result of ?-carotene bleaching assay showed that the rate of the decreased antioxidant activity of S.oblongata extract was lower than that of ?-carotene/linoleic acid. The results suggest that the extract of S. oblongata can be used as a natural antioxidant. However, further investigation to isolate and identify the chemical compounds responsible for antioxidant was needed

Abstract

Solikin. 2018. Effect of nodes position on the growth and yield of stem cutting of Sambiloto (Andrographis paniculata). Nusantara Bioscience 10: 226-231. Plant propagation by cuttings can be done to produce relatively uniform quality of plant yield such as sambiloto (Andrographis paniculata (Burm.f.) ex Nees). The study aimed to determine the effect of nodes position on the growth and yield of stem cutting of sambiloto. The experiment was conducted in Purwodadi Botanic Garden in February-May 2017. The study used randomized block design with the treatment of nodes potition i.e. top, middle and base of stem cuttings. Each treatment was replicated three times. The results showed that the top stem cutting produced the highest cutting life, leaf area, leaf dry weight, root and generative organs dry weight. In contrast, the base stem cutting produced the lowest live cutting and plant growth.

The phytochemistry of Cymbopogon winterianus essential oil from Lombok Island, Indonesia and its antifungal activity against phytopathogenic fungi. Nusantara Bioscience 10: 232-239. Cymbopogon winterianus Jowitt ex Bor [Sereh Wangi] is a plant producing essential oil that has been extensively used in perfumery, mosquito repellent, medicinal, and cosmetic industry. In Indonesia, the accession of C. winterianus plant had been collected from natural forest in Sembalun, Lombok, West Nusa Tenggara (NTB) Province. However, the phytochemical properties and its biological potential of this accession have never been reported. The aim of this study was to determine the phytochemical content of essential oil of C. winterianus from Lombok and to explore its potential as a bioantifungal against several phytopathogen fungi. The essential oils were extracted from fresh stem and leaf of C. winterianus by hydrodistillation. The chemical composition of them was determined by chromatography-mass spectrometry (GC-MS). A total of 26 and 20 compounds were obtained from the stem and leaf oil with the dominant compounds of essential oil in stem were Torreyol (17.89%), Oxirane, octyl- (15.84%), (-)-Isopulegol (14.44%), Isopulegol 2 (12.18%), and Geranyl Acetate (5.89%) while in leaf were (-)-Isopulegol (22.69%), Oxirane, octyl- (16.64%), Cyclohexanol, 5-methyl-2-(1-methylethenyl)- (14.21%), "KW3 AUS Epiglobulol” (9.47%), and Cyclohexene, 1-methyl-4-(1-methylethenyl)- (7.14%). The presence of these contents as dominant chemical compounds in S. winterianus was reported for the first time. The highest content of oils was identified as monoterpene (38.01% and 45.78% in stem and leaf oil) and sesquiterpene ( 27.67% and 17.78 % in stem and Leaf oil). The biological study showed that leaf essential of C. winterianus expressed positive antifungal activity against Fusarium solani, Aspergillus niger, dan Cladosporium sp.

Huang CH, Adeni DSA, Johnny Q, Vincent M. 2018. Production of bioethanol from sago hampas via Simultaneous Saccharification and Fermentation (SSF). Nusantara Bioscience 10: 240-245. Sago hampas is an inexpensive, renewable and abundant agro-industrial residue that can be exploited to produce bioethanol. In this study, ethanol production was performed via simultaneous saccharification and fermentation (SSF) on fresh sago hampas at 2.5%, 5.0% and 7.5% (w/v) feedstock loadings with the aid of amylolytic enzymes, cellulolytic enzymes and Saccharomyces cerevisiae, under anaerobic condition for five days with a constant agitation of 150 rpm and ambient temperature. Results obtained indicated that SSF with 5.0% (w/v) sago hampas loading produced the highest ethanol yield at 17.79 g/L (79.65% Theoretical Ethanol Yield, TEY), while SSF using 2.5% and 7.5% (w/v) sago hampas produced ethanol at only 8.38 g/L (75.00% TEY) and 23.28 g/L (69.48% TEY), respectively. Total biomass reduction was recorded between 66.3% to 71.3% by the end of the SSF period. This study demonstrated that fresh sago hampas is a promising feedstock for bioethanol production as yields are generally high for all the substrate loadings tested. Moreover, bioethanol production using fresh sago hampas may assist in reducing pollution caused by sago waste accumulation.

The agricultural crops and horticultural plants are always under constant assault caused by diseases, insect pests, viruses, and other pathogens which may substantially reduce yield. Chili Pepper (Capsicum annum L.) is a popular horticultural plant of the Solanaceae family in Indonesia. Some serious diseases widely found in chili peppers, are anthracnose (Colletotrichum capsici), leaf spot (Cercospora capsici) and fruit rot (Phytophthora capsici). To manage the potential problems, this study is aimed to provide an explanatory knowledge of the use of plant-based pesticide to control the diseases in chili peppers. The information is meant to fill the knowledge gaps in the use of plant-based pesticide to control the chili diseases. The use of nonchemical pesticide benefits not only the environment but also as an organic strategy for disease management. This research used a randomized complete block design (RCBD) categorized into four groups and six different treatments. The organic pesticides were prepared from the extract of neem leaves, soursop leaves, lemongrass extract, tuba root extract, and kenikir/Cosmos caudatus extract). The result indicates that neem leaves are the most effective organic pesticides to control the chili pepper disease in Indonesia.

Phenolic compounds have attracted much attention because of their role in some oxidative diseases. Therefore, the study aimed to investigate the production of antioxidant compound from tissue culture of A. annua. The callus were developed on solid MS medium treated with four combination of plant growth regulators: NAA 0.5 mg/L + BA 0.5 mg/L; NAA 0.5 mg/L + kinetin 0.5 mg/L; 2,4-D 0.5 mg/L + BA 0.5 mg/L; and 2,4-D 0.5 mg/L+ kinetin 0.5 mg/L. The callus extracts were evaluated their antioxidant activity and were analyzed the content of phenolic content. Furthermore, detection and quantification of phenolic compound were investigated. Combination of NAA 0.5 mg/L + BA 0.5 mg/L showed the highest DPPH radical scavenging activity and total phenolic content. GCMS analysis also revealed that the combination also showed the highest content of caffeic acid, a phenolic compound known as an antioxidant agent. Furthermore, the callus extract treated with NAA 0.5 mg/L + BA 0.5 mg/L increased both the antioxidant activity and caffeic acid content compared with A. annua leaf extract. In conclusion, plant tissue culture enhanced pharmaceutical properties of A. annua. Moreover, the study suggested that the treated callus extract of A. annua is potential to be further developed for production of antioxidant compound.

Abstract. Oksari AA, Susanty D, Wanda IF. 2019. Allelopathic effect of invasive species air potato (Dioscorea bulbifera) on seeds germination of Polyalthia littoralis. Nusantara Bioscience 11: 63-70. Dioscorea bulbifera L. (Dioscoreaceae) is included in the Global Weed Compendium and one of the invasive plants that cause environmental problems in many parts of the world. This study aims to determine the effect of allelopathy from various concentrations of leaf and tuber extract from D. bulbifera on seed germination of Polyalthia littoralis (Blume) Boerl. The method used was a Completely Randomized Design (CRD) with seven treatments and three replications. The treatment given was the concentration of D. bulbifera extract 10%, 20%, 40%, 60%, 80%, 100% and aquadest solvent control. The results of phytochemical screening analysis showed that the extract of leaves and tuber of D. bulbifera did not contain alkaloids, but specifically on leaves extract contained tannins. Both extracts contained steroids, flavonoids, and saponins. Based on the results of the study, total phenolic obtained in the leaf extract as much as 28.81 ± 2.51 mg TAE/g extract, while the tuber extract had a total phenolic of 6.74 ± 1.19 mg TAE/g. The results showed that allelopathic substances of D. bulbifera affected the germination of P. littoralis seeds. Based on DMRT test at 5% level, it was known that allelopathy concentration of D. bulbifera significantly affected the germination rate at concentrations of 20%, 60%, 80%, and 100% and significantly affected the coefficient of velocity of germination at a concentration of 20%. But, concentration allelopathy of D. bulbifera had no significant effect on the coefficient of uniformity of germination. The concentration that reduced the lowest seed viability was at a concentration of 80% with an average germination power of 56.67 ± 7.45%.

Stability of black grass jelly (Mesona chinensis) probioticated by encapsulation of Lactobacillus plantarum Mar8 with agar and gum Arabic. Nusantara Bioscience 11: 84-88. Black grass jelly [Mesona chinensis (Benth)] is one of the traditional food ingredients that can be used as the encapsulation material. However, syneresis easily occurs so that becomes a problem during storage. This study was conducted to evaluate the effect of type of stabilizers (agar and gum Arabic) and storage temperature (cold/4oC and ambient temperature/37oC) on black grass jelly stability and the viability of encapsulated Lactobacillus plantarum Mar8 in probiotication. The concentrations of stabilizers were 0.5%; 1.0%; 1.5% and 2.0% (w/v). The result showed that the syneresis rate was stable up to 2% of agar concentration at low storage temperature, while gum Arabic in the concentration of >1% resulted in increasing syneresis rate. Ambient temperature storage increased the syneresis rate. This study showed that agar or gum Arabic was supporting the probiotication as good encapsulant. Moreover, it was suitable for stabilizing or gelling agent at 0.5%-1% concentration to produce black grass jelly as RTD.

Abstract

Abstract. David D, Daniel R, Seecharran D, 2019. Antimicrobial properties of Chlorocardium rodiei on Pseudomonas aeruginosa, Bacillus spp. and Candida albicans. Nusantara Bioscience 11: 128-132. This research seeks to investigate the antimicrobial properties of Chlorocardium rodiei leaves and seeds on Pseudomonas aeruginosa, Bacillus spp. and Candida albicans. It also seeks to extract and identify the phytochemical compounds presented in leaves and seeds. The crude plant extract was obtained using the maceration method of extraction (ethanol and acetone as solvent) followed by rotary evaporation. Phytochemicals were analyzed from the crude extract using standard phytochemical procedures. 1%, 50% and 100% concentrations of each extract were prepared and used in the disk diffusion assay against C. albicans, P. aeruginosa, and Bacillus spp. and compared against Ciprofloxacin and Erythromycin for the bacteria and Fluconazole for the fungus. The results showed that the extract from the leaves possesses weak (?12 mm) to moderate (>12 ?15mm) inhibitory effect, while the extract from the seeds had weak (<12mm) inhibitory effect on the microorganisms. The phytochemical compounds tannins, essential oils, saponins, and phenols were tested positive from the crude extract. For the first time, the antimicrobial properties of C. rodiei’s leaves and seeds were evaluated and showed to possessed antimicrobial properties.
