Noori M, Baghaeifar Z, Chehregani A, Faraki F. 2018. Seeds characters, pollen fertility and flavonoids of ten Brassicaceae collected near a kilns thermal power plant for air pollution bioindication. Nusantara Bioscience 10: 96-104. Shazand Steam Power Plant located on North-East of Shazand, Iran began to work from 2000. The power plant necessity fuel is natural gas and mostly heavy fuel oil. The most pollutant of power plant is sulfur compounds in addition to nitric and carbon mono oxide. Because environmental pollutants influence plant fertility and chemical compounds, therefore this study was done on ten wild Brassicaceae (Alyssum linifolium var. linifolium, Alyssum longistylum, Alyssum marginatum, Choriospora persica, Clypeola lappacea, Conringia perfoliata, Descurainia sophia, Goldbachia laevigata, Isatis kotschyana and Neslia apiculata) taxa collected from the thermal power plant area for bioindication of regional air pollution comparing to controls collected 40 km away from the power plant. Brassicaceae members are important for their ecological, pastoral, medicinal and edible points. Seed width and length max. and their ratio and abnormal seed percentage were calculated. Pollen abnormality and sterility percentages determined using Muntezing’s acetocarmine and light microscopy. Also, their pollen flavonoids were semi-quantitatively assessed using two-dimensional paper and thin layer chromatography. Results showed seeds health and their dimensions reduction in polluted samples in comparison with controls. In C. lappacea significant differences of seed and pollen abnormality and pollen sterility percentages, morin and kaempferol concentrations, between control and polluted samples were observed (P?0.05). Also, number and kind of pollen flavonoid changes especially increasing flavonoid contents were observed in polluted plants comparing to control. Studying seed and pollen characters can be used as air quality bioindicators.

Nusantara Bioscience 10: 105-110. Asthma is still a major health problem in the world. Antiasthma drugs raise the problem of side effects and medical expenses. Thus, it is necessary to develop medicinal plants based antiasthma drugs with fewer side effects and more affordable costs. The potential antiasthma medicinal plant came from the genus of Curcuma (Zingiberaceae family). Turmeric (Curcuma longa L.) is the most prominent species in this genus and has been extensively studied for the treatment of asthma. However, there are other Curcuma indigenous species that are also traditionally used for asthma, namely Curcuma aeruginosa Roxb. and Curcuma mangga Val. Zijp. Both indigenous species have not been studied pharmacological activity as an antiasthma. This research was conducted to know the tracheospasmolytic activity of ethanol extract of C. aeruginosa and C. mangga on the separate organ of guinea pig trachea to determine the antiasthma effect. The research was also conducted to know anti-inflammatory activity with membrane stabilization test and inhibition of lipoxygenase. Results showed that C. aeruginosa and C. mangga had lower tracheospasmolytic activity when they were compared with aminophylline as a positive control. However, the EC50 value of C. aeruginosa (0.020 ± 0.004) and C. mangga (0.039 ± 0.005) were not significantly different with aminophylline (0.016 ± 0.005) as positive controls. The result of membrane stabilization test showed that the EC50 value of C. aeruginosa (47.76 ± 1.57) and C. mangga (67.50 ± 0.97) were high when they were compared with indomethacin (26.39 ± 2.91) as the positive control. Results of lipoxygenase inhibition test showed that the EC50 value of C. aeruginosa (-111.11 ± 2.30) and C. mangga (-101.98 ± 1.32) were high than that of indomethacin (-282.84 ± 7.41) as the positive control. These results show the potential use of C. aeruginosa and C. mangga as antiasthma via tracheospasmolytic activity in separate organs of guinea pig. They also had anti-inflammatory effect based on membrane stabilization and inhibition of lipoxygenase test.

Joel KA, Runo S, Muchugi A. 2018. Genetic diversity and virulence study of Striga hermonthica from Kenya and Uganda on selected sorghum varieties. Nusantara Bioscience 10: 111-120. Parasitic weeds pose a severe problem in agricultural production, causing massive crop losses in many regions of the world and especially Africa. One example to be considered the most significant biological constraint to food production in sub-Saharan Africa (SSA) is Striga hermonthica, the most widespread among the Striga species in the semi-arid tropical African zones. The wide geographical distribution set conditions for genetically structured populations. The genetic variations among the weed populations allow for the quick breakdown of resistance in crops hence making control of the weed difficult. Efficient and effective control of S. hermonthica demands knowledge on inherent genetic variability within local and regional races of the weed. However, the genetic diversity and virulence of S. hermonthica ecotypes in Kenya and Uganda on selected sorghum varieties remain unknown. This study aimed at evaluating the genetic diversity among seven S. hermonthica populations from locations in Kenya and Uganda using 5 primer sets of Expressed Sequence Tags-Simple Sequence Repeats (EST-SSR). The genetic diversity was moderate as shown by the Nei’s genetic distance values. AMOVA measured low genetic differentiation among the populations. This study also investigated twelve sorghum varieties for their response to S. hermonthica infection. The result demonstrated that the varieties resistance responses to S. hermonthica varied widely. The phenotype of resistant interaction was characterized by the inability of the weeds haustoria to penetrate the sorghums root endodermis due to severe necrosis and in rare cases the parasites radicle growing away from the host root. The resistant sorghum varieties were the Asareca W2, Asareca AG3, N13 and the Wild-type which had low mean number of S. hermonthica plantlets growing on their roots, while the most susceptible varieties were Sap 027, Epurpur which had the highest mean number of S. hermonthica plantlets growing on their roots. There was a highly significant difference in the means of the number of Striga growing on the roots of sorghum varieties, Striga dry biomass and S. hermonthica length between the susceptible and resistant ones. This knowledge holds unique potentiality since resistant sorghum germplasm tested will be sourced and targeted to the seven specific geographical areas where virulence of the particular S. hermonthica populations was characterized.

Nusantara Bioscience 10: 121-125. The aim of this study was to investigate the effect of different levels of turmeric powder (0, 1, 2, 3 g/kg) on production performance and the number of Escherichia coli and Staphylococcus aureus bacteria in the intestine of Sentul chickens. The results showed that addition of 3 g/kg of turmeric powder to diet significantly reduced feed intake (FI) and improved feed conversion ratio (FCR) of Sentul Chickens aged 9-16 weeks. The birds fed with control diet had the highest number of S. aureus and E. coli, while the birds fed with the diets added with 2 and 3 g/kg turmeric powder had the lowest number of bacteria. The study reveals that increasing levels of turmeric powder in diets had positive effects on bird’s performance and the number of intestinal bacteria. It can be concluded that a level of 3 g/kg turmeric powder in diets gave the best result due to significantly improved FCR.

This research was conducted to obtain information of topographical tetrazolium staining pattern of pepper (Capsicum annum) seeds. Tetrazolium test was conducted to determine the seed viability and plant growth vigor. Laboratory test for standard germination and field performances were performed on four lots (A, B, C, D) of pepper seeds. The viability categories pattern were determined by Root Mean Square (RMS), regression, and correlation analyses. Nine topographical patterns were recognized. The laboratory test results and field performances were compared with the topographical pattern. Combination of patterns 1,2 (embryonic axis and cotyledon completely stained) selected as viable category as it gave the least RMS value, the highest determination (R²) and correlation (r) coefficient with standard germination (RMS = 4, 06; R² = 0,761; r = 0,872). Combination of patterns 1,2 also gave the highest determination (R²) and correlation (r) coefficient with field stand (R² = 0,921; r = 0,959). The combination of patterns 1,2 is recommended for estimating plant growth performance in the field.

Sadeghirad E, Majd A, Iranbakhsh A, Javanshah A. 2018. Male gametophyte development steps in Pistacia vera L. Nusantara Bioscience 10: 151-158. Salinity affects the growth and development of plant. It also affects the development steps of male gametophyte in pistachio plant. Pistachio (Pistachio vera) is a member of family Anacardiaceae and order Sapindales. To study the effects of salinity on those steps, an experiment was performed in two locations in Golshan Anar with commensurate circumstances, namely: a control area (A) which was well-irrigated with fresh water, and the other area (B) which was well-irrigated with salty water added with NaCl solution with EC values of 14 dS.m-1. The flowers sampling was done in two Golshan Anar regions on the springtime based on a completely randomized design with three replications. The development steps of male gametophyte in pistachio plant were observed using conventional cell histology techniques and light microscopy observations and were then contrasted with samples subjected to no salinity stress. The results represented that several steps of male gametophyte development are as follows: (1) the anther experiencing normal growth which is tetrasporangiate, (2) cytokinesis taking place simultaneously with meiosis in the microspore mother cell, the tetrahedral tetrads, (3) microspores being delivered after meiosis by microsporogenesis were more or less irregular in shape during the contraction period. Finally, the abnormal shape and structure of the number of cases reviewed in three replicates of pollens can be one of the significant factors influencing the decline in the product.

Alfiah L, Zul D, Nelvia N. 2018. The effect of combination of indigenous phosphate solubilizing bacteria of Riau, Indonesia on the available phosphorus and phosphorus uptake of soybean. Nusantara Bioscience 10: 146-150. Despite the abundant amount of phosphorus (P) in the soil, P uptake by the plants is very limited. In acidic soil, phosphorus (P) is bound to aluminum (Al) and iron (Fe), whereas in the alkaline soil, phosphorus (P) is bound to calcium (Ca). The improvement of efficiency and availability of P to plants can be made by utilizing a group of solubilizing phosphate microorganisms. Potency test to investigate the P solubility by Phosphate Solubilizing Bacteria (PSB) has been conducted by isolating the bacteria from peat soil at Biosphere Reserves of Giam Siak Kecil Bukit Batu, Riau, Indonesia. The semi-quantitative test revealed that the PSB were able to dissolve Ca3 (PO4)2, FePo4 and phosphate rock. However, the adaptation ability and potency of PSB from indigenous Riau peat soil inoculated into soybean (Glycine max L. Merr) plants in the mineral soil have not yet been investigated. The present study was carried out from March to June 2015 on the alluvial soil in Babussalam Village, Rambah Sub-district, Rokan Hulu District, Riau. The aim of this study was to determine the effect of PSB inoculation on bacterial population and phosphatase activity. The study also aimed to determine the available P and P uptake and their impact on soybean growth and production. The study employed a factorial experiment laid out in a completely randomized design (CRD) consisted of two factors, i.e., soil treatment and PSB. The first factor comprised of two levels, i.e., T0: non-sterilized soil, T1: sterilized soil. The second factor consisted of 4 levels, i.e., B0: without PSB inoculation, B1: inoculation using 2 isolates of PSB (BB_UB6 and BB_K9), B2: inoculation using 3 isolates of PSB (BB_UB6, BB_K9 and BB_K2), and B3: inoculation using 4 isolates of PSB (BB_UB6, BB_K9, BB_K2, and BB_HS13). The results showed that inoculation of starter 3 had the highest phosphatase activity rate of 12.43 ?g p NP g-1 hour-1. The highest available P was produced by starter 2, while the P uptake on non-sterilized soil was higher than that on the sterilized soil at 2.63 mg plant-1. PSB inoculation and soil sterilization did not significantly affect the population of phosphate solubilizing bacteria.

Siregar HM, Purwantoro RS, Praptiwi, Agusta A. 2018. Antibacterial potency of simple fractions of ethyl acetate extract of Begonia baliensis. Nusantara Bioscience 10: 159-163. Balinese people utilized Begonia baliensis Girm. (Begoniaceae) as traditional medicine to relieve cough. It was applied in a unique way by inserting the plant material into a bamboo column and then burning it in the fire. The liquid produced by the combustion process was used as a cough medicine. Based on this traditional knowledge, it is expected that B. Baliensis has antibacterial activity. B. baliensis plant collected from Bukit Sangyang, Penebel, Tabanan-Bali has been used in this study. The chemical compounds of ethyl acetate extracts were isolated/ separated by column chromatography. The obtained fractions were analyzed for antibacterial activity by disc diffusion assay against Escherichia coli, Bacillus subtilis, Staphylococcus epidermidis and Staphylococcus aureus. Chromatographic column yielded 14 simple fractions, whereas antibacterial test results showed 5 active fractions. Fraction 3 (F3) was active against S. epidermidis, fraction 5 (F5) against E. coli and S. epidermidis, while fractions 10, 11 and 12 were active only against Bacillus subtilis. Isolation and purification of the active components likely increased their potential as antibacterials.

Noori M, Jafari M, Azimi H, Node-Farahani M. 2019. Effects of Ruta graveolens total and flavonoids extracts on rat blood glucose, cholesterol, triglycerides and urea comparing synthetic drugs. Nusantara Bioscience 11: 23-29. Rue (Ruta graveolens L.: Rutaceae) is a medicinal plant that is used to treat many diseases in America, Asia and Europe due to its pharmaceutical properties. So far more than 120 different phytochemicals such as flavonoids have been extracted and identified from the species, which make it valuable in traditional medicine and manufacturing of synthetic drugs. Due to its endemic and high potential pharmacological effects, this study examined the oral effect of each of its total, flavonoids and non-flavonoids extracts on reducing blood glucose, cholesterol, triglycerides and urea in STZ-diabetic rats comparing to control and Atorvastatin, Allopurinol, Metformin as synthetic drugs. Aerial part of the plant was collected from around Yazd, Iran and dried in shade, crushed and extracted in 70% ethanol in three steps. Total and flavonoid extracts were prepared after rotary evaporation, then were used for making required doses after calculation. The plant flavonoids were identified using two-dimensional, thin-layer chromatography and TLC Scanner3 methods. Atorvastatin, Allopurinol and Metformin were obtained and also appropriate doses were prepared. Thirty-six adult male Wistar rats were divided in 12 groups (n=3): control, synthetic drugs control, total and flavonoids control, STZ-diabetic control, diabetic treated with synthetic drugs, total and flavonoids extracts, then weighed. All rats were treated orally at a specific time for 2 weeks. Subsequently, the secondary rat weights were determined and blood donation was performed after anesthesia. Then, blood serum glucose, cholesterol, triglycerides and urea were determined by spectrophotometry using commercial enzymatic chlorometric assay kits. All data were analyzed using EXCEL and SPSS software. Results showed aerial part of R. graveolens contains flavones C and C-/O glycosides, flavonoid sulphates and had no aglycones. Apigenin, chrycin, isorhamnetin, kaempferol, myricetin, quercetin, rutin and vitexin were found in the species aerial part. This plant had 329/99 gr/kgDW of effective material and 257.7 g/kgDW of flavonoids. The lowest blood glucose levels were observed in treated Metformin and flavonoids control rats respectively. Diabetic rats treated with R. graveolens total extract had the lowest blood cholesterol, urea and glucose levels. The highest renal damage was observed in control diabetic rats and the least renal damage was observed in the STZ-diabetic rats treated with the plant total extract. These results indicate that R. graveolens extract and its rutin contained antihyperglycemia, antihyperlipidemia, insulinity and protective properties.

Wijayanti II, Budiharjo A, Pangastuti A, Prihapsara F, Artanti AN. 2018. Total phenolic content and antioxidant activity of ginger extract and SNEDDS with eel fish bone oil (Anguilla spp.). Nusantara Bioscience 10: 164-169. Aims of this study were to measure phenolic content of ginger (Zingiber officinale) and to observe the comparison of antioxidant activity between pure ginger extract and ginger extract in SNEDDS (Self Nanoemulsifying Drug Delivery System) with bone of eel fish oil (Anguilla spp.) as a conductor. Analysis was done based on IC50 value using DPPH (2,2 diphenyl-1-picrylhydrazyl) method and activity of Superoxide Dismutase (SOD) in vivo using Adrenochrome Assay. Determination of total phenolic content of ginger extract was measured by UV/Vis Spectrophotometric using Folin-Ciocalteau reagents then the absorbances were measured at wavelength 756 nm. Calculation of IC50 DPPH of the ginger extract and SNEDDS were measured using spectrophotometer UV/Vis at 517 nm wavelength. Measurements of SOD was based on the ability of SOD inhibit spontaneous auto reduction of epinephrine using Adrenochrome Assay at 12 male Wistar rats that divided into 4 treatment groups with 3 replicates for each group. Total phenolic content of ginger extract from 3 replications were 4,8519 ± 0,037 µg/mL equivalent with gallic acid standard solutions (GAE) or 485,1847 ± 3,7045 mgGAE/100 g dry weight. Antioxidant scavenging activity by DPPH assay of ginger was higher than SNEDDS. IC50 of ginger was 385.4 mg/mL, and IC50 of SNEDDS was 428.4 mg/mL. The result of descriptive analysis of Kruskal Wallis of SOD activity assay indicated significant differences between treatment groups (P = 0.001 (P <0.05)). SOD activity test performed in vivo showed that ginger extract made into SNEDDS system with eel fish oil used as the conductor has a significant influence compared with ginger extract group.
