Abstract. Philander M, Ragobeer P, Daniel R. 2019. Short communication: A comparison of the rate of polyethylene decomposition among Bacillus spp., Pseudomonas spp., and Escherichia coli bacteria. Nusantara Bioscience 11: 97-100. This study did a comparative analysis of the rate of polythene plastic decomposition among three species of bacteria namely, Bacillus spp., Pseudomonas spp., and Escherichia coli. The three bacterial species were cultured and grown in 2-liter polyethylene terephthalate bottles with nutrient broth separately in triplicates along with a control that did not include any bacteria. These bottles were left undisturbed at room temperature for three months (92 days) after which the containers were subjected to sterilization. Plastic weights were taken using an electronic scale and compared with original values. From the results, it showed that in the three months there was a relatively large amount of degradation that occurred. Bacillus spp. and E.coli bacteria yielded similar results and moved at a rate faster than the Pseudomonas spp., A daily rate for each bacterial species was calculated and a predictive figure based on rates was made. If the rates remained constant there would be total degradation of the polyethylene plastics by the Bacillus spp., E. coli and Pseudomonas spp., in 11.45, 11.45 and 12.35 years respectively. This is a significant finding since in nature plastics can take hundreds of years to decompose.

Abstract

Abstract. Rini DS. 2019. Short Communication: Sequence variation of DREB2 gene as a potential molecular marker for identifying resistant plants toward drought stress. Nusantara Bioscience 11: 35-43. Drought stress is the most destructive environmental factor affecting agricultural productions worldwide. Drought stress affects around 60 million hectares of agricultural land in Indonesia. Therefore, the selection of crops that can grow well and survive under drought is necessary to overcome the problem of decreased crop yield during stress conditions. DREB2 (Dehydration Responsive Element Binding2) belongs to the Transcription Factor (TF) gene families as a master regulator of plant responses under stress. Due to increased expression by stress, this gene can be used as a molecular marker to identify resistant plants toward drought. However, study the sequence variation of DREB2 is required to understand the character of this gene related to its function at a molecular level. The sequence of DREB2 in several plants was obtained from NCBI and aligned to determine the similarity of the regions. Even though variations were found in N-terminal of DREB2 sequences studied, there was a high similarity of the sequences in the DNA binding domain. It has been predicted that this domain is part of a conserved region in the sequence of DREB2 gene. This domain is then proposed to be used for primer designing purposes to study DREB2 gene expression between plant species.  Keywords: DREB2, drought stress, sequence variation

Short Communication: Biodegradable of bio-composites made from Polylactic Acid (PLA) and cellulose fibers from oil palm empty fruit bunch. Nusantara Bioscience 11: 8-11. Over the last two decades, plastic has become an integral part of people's everyday life. However, plastic is a material that difficult to be degraded in the environment. Due to environmental concern, it is indispensable to find environmentally friendly plastic materials. This study aimed to determine the biodegradable polymer composites manufacture from cellulose fibers of oil palm empty fruit bunches (OPEFB) and polylactic acid (PLA). Cellulose fibers from oil palm OPEFB were isolated by chemical methods. Polymer composites made by mixing cellulose fiber of oil palm OPEFB and PLA based on differences in the composition of the cellulose fiber of OPEFB. The composition used is 0 PHR, 5 PHR, 10 PHR, 15 PHR, and 20 PHR. Results showed that polymer composite with higher composition of cellulose fiber of OPEFB more easily degraded in the environment. There are no significant differences in biodegradable between 15 PHR and 20 PHR, neither among 10 PHR, 5 PHR and control. At higher cellulose fiber of OPEFB content, 20 PHR, polymer composite was the most degraded in the environment.

Abstract

Intara YI, Nusantara AD, Supanjani, Caniago Z, Ekawita R. 2019. Spatial patterns of evapotranspiration distribution in palm oil plantation. Nusantara Bioscience 11: 1-7. Evapotranspiration can directly calculate by some climatic factors which are processed in the form of simulation modeling of two-dimensional evaporation distribution on specified coordinates. This research had been conducted in palm oil plantation PT. Bio Nusantara Bengkulu, Indonesia. The sampling location covered some areas named as TM2, TM22, Palm oil-Field, Palm oil-Residential Area and Palm oil-Secondary Forest. The TM2 and TM22 were a location of palm oil plantation used for 2 and 22 years planting respectively. The three remaining locations dedicated to areas of which mix TM22 with soccer Field, Residential Area and Secondary Forest. The results showed that in palm oil plantation TM 2 which consisted of herbaceous plants and bushes produced more water vapor. At palm oil plantation TM 22 which had fully developed canopy or the entire soil area had been covered by canopy, so only transpiration occurred at this site, less water vapor was produced. This is due to the temperature condition is inversely proportional to the relative humidity. As a comparison of a palm oil plantation evapotranspiration measurement result, indicated on area of football field where only has grass in the field, with low intensity of light, wind, and humidity, affected directly to high amount of evapotranspiration, therefore, more water vapor produced. The measurement result of palm oil plantation nearby residential area showed that the area which consisted of herbaceous plants and bushes, also zinc-roofed residential with the intensity of light and wind directly hit the area, produced less water vapor. Moreover, the condition at the border area between TM 22 and the palm oil secondary forest contained mixed plantation. The measurement result showed that the palm oil secondary forest had the same height of canopy and almost had equal humidity (>76%), but secondary forest plants had more dense plantation density which produced less water vapor.

Nusantara Bioscience 11: 12-17.  The aim of this study was to determine the effect of combination treatment washing chicken meat with three ozone concentrations in the water during 10 min and stored at 2-7?C without ozonation and with ozonation, on the shelf life of chicken meat. The study was conducted with a complete random factorial design (CRD) factorial pattern 3 x 2 with 3 replications. The data were analyzed by using analysis of variance (ANOVA). If the results were significant, the analysis followed Duncan's New Multiple Range Test (DMRT). The results of statistical analysis showed that average pH values, color scores intensity b * for brightness (yellowness) and Peroxide value (PV) showed significant differences (P <0.05). H 2 S test showed that combination treatment washing chicken meat with ozone concentrations 3ppm in the water during 10 min and stored at 2-7?C with ozonation was able to inhibit the start process of decomposition chicken meat until the 10th days. The conclusion of these results that combination treatment washing chicken meat with ozone concentrations 3ppm in the water during 10 min and stored at 2-7?C with ozonation can extend the shelf life of chicken meat until the 10th days.

Wulandari YRE, Harjosudirjo MA. 2019. Micropropagation of Morus cathayana through in vitro culture from local Bogor, West Java, Indonesia. Nusantara Bioscience 11: 18-22. Mulberry (Morus spp.) is a dicotyledonous plant known for its medicinal benefits as well as silkworm breeding (Bombyx mori L.) to produce silk. Morus cathayana mainly cultivated due to its high content of 1-deoxynojirimycin, widely utilized as an anti-diabetic agent. However, common practice in planting and maintenance of mulberry generate less profit, contributed to its long juvenile period and high heterozygosity level. Hence the development and cultivation through plant tissue culture techniques are necessary. The purpose of this research was to obtain the best combination of thidiazuron as a plant growth regulator. M. cathayana branches with nodal segments were used as explants. Research stages were explants initiation into Murashige and Skoog media, shoot induction in MS media as control and MS+BAP 1 mg/L+NAA 0.25 mg/L+TDZ 0.1, 0.5, 1 mg/L as treatment media, and statistical data analysis. Greater increase in shoot growth was observed in MS+BAP 1 mg/L+NAA 0.25 mg/L+TDZ 0.5 mg/L media, while the formation of shoots and calluses on the explants grown in control and treatment media showed no significant growth.

Abstract

Abstract. Authors. 2019. The activity phagocytosis of isoeugenol-ester compound on Mus musculus macrophage cell. Nusantara Bioscience 11: 117-122. Chemotherapy is drugs therapy for cancer. However, chemotherapy could damage normal cells and decrease immune system. For this reason, it is necessary to find immunostimulant compound that is able to increase immune system. Isoeugenol-ester is unnatural compound that has been tested for its cytotoxicity against cancer cell and showed positive result. Nevertheless, ability of Isoeugenol-ester for inducing phagocytosis is still unknown. Therefore, bioassays study using in vitro phagocytosis were performed. Latex beads methods using antigen model in 5 isoeugenol-ester concentration series: 6, 25; 12, 5; 25; 50 and 100 ?g/mL were tested to Mus musculus peritoneal macrophage cell. The optimum concentration of Isoeugenol-ester compounds in inducing phagocytosis activity of M. musculus macrophage cell was 12.5 ?g/mL.

Sukma, Callina, and Caliso: Effect of loading treatment and immersion time in plant vitrification solution-2. Nusantara Bioscience 11: 71-78. Sukma, Callina, and Caliso are papaya cultivars released by the Center for Tropical Horticulture Studies, IPB University, Bogor, West Java, Indonesia. In general, papaya seeds cannot be stored for a long time with a conventional GenBank storage system, even though some of them consider as “orthodox” class of seed. Cryopreservation, storage at an ultra-low temperature (-196 0C) of liquid nitrogen, could be possible for long-term storage of papaya seeds. The experiment conducted to get the necessary of loading treatment and the best immersion time in PVS2 for cryopreservation of papaya seeds cv. Sukma, Callina, and Caliso, so they still had considerable viability. We conducted experiment as factorial in completely randomized design, with the first factor was loading treatment with two levels (with and without loading treatment), and the second factor was immersion time in PVS2 with three levels (15, 30 and 45 min). So, there was six treatment and we used 50 seeds for each treatment. Results showed that the three papaya cultivars gave different responses to the treatment before cryopreservation proved that papayas seed had different characteristics, depending on genotype. For papaya seeds cv. Sukma, loading treatment was not needed and the best immersion time in PVS2 was 15 min. The viability of papaya seeds cv. Callina was low, so we should try another treatment before cryopreservation. For papaya seeds cv. Caliso, loading treatment was needed and the best immersion time in PVS2 was 30 min.

MDA levels in the pancreas, testes, liver, and plasma of diabetic rats: The effect of snakehead (Channa striata) extract. Nusantara Bioscience 12: 50-54. There are several biomarkers of oxidative stress in diabetes mellitus; one of those biomarkers is Malondialdehyde (MDA). Increasing oxidative stress will cause increased tissue damage. This study was conducted to determine the effect of snakehead extract (SHE) on MDA level of pancreas, testes, liver, and plasma of alloxan-induced diabetic rats. There were 5 groups of treatments: non-diabetic rats/negative control (C-), diabetic rats/positive control (C+), and 3 SHE level of administration: 1 mL/day (low dose/LD), 1.6 mL/day (middle dose/MD) and 2.1 mL/day (high dose/HD). Alloxan-induced diabetic rats were administered with SHE extract orally every day for two weeks. The results showed that MDA levels in the testes, liver, pancreas, and plasma of diabetic rats administered with SHE and non-diabetic rats/negative controls (C-) were significantly lower (p <0.05) compared to MDA levels in the testes, liver, pancreas, and plasma of diabetic rats without SHE administration (positive control/ C +). The highest dose of SHE treatment (2.1 mL/day) results in decreasing MDA levels were not significantly different (p> 0.05) with the group of non-diabetic rats / negative controls (C-). The conclusion of this study was increasing SHE administration up to 2.1 mL/day result in reducing more of MDA levels in plasma, pancreas, liver, and testes of diabetic rats.

Anthocyanins content of methanol extract of parijoto (Medinilla speciosa) and its effect on serum malondialdehyde (MDA) level of hyperlipidemic rat. Nusantara Bioscience 11: 112-118. Parijoto (Medinilla speciosa) is an endemic plant in Indonesia that its fruit have a purplish red color, which indicated that parijoto fruits contain anthocyanins, pigments of the flavonoid class that responsible for the red, blue and purple coloration of many flowers and fruits. This study was aimed to determine the total yield of extract and anthocyanins content of methanol extract of parijoto (Medinilla speciosa) pericarp and whole fruit and the effect of parijoto whole fruit extract on serum malondialdehyde (MDA) level of the hyperlipidemic rats. Extraction of parijoto fruit and pericarp were carried out by the maceration method with methanol as a solvent. Determination of the total anthocyanin was done by pH difference method. MDA serum levels were done in a pre-test and post-test Randomized controlled group design was used in this study. Rats were divided into five groups: normal rats, hyperlipidemic rats, hyperlipidemic rats treated with methanol extract of parijoto (500 mg · kg ?1, 1000 mg · kg ?1, and 1500 mg · kg?1 body weight). Blood was collected at hyperlipidemia state and after treatment of methanol extract of parijoto for determination of MDA level in blood serum. The results showed that the total yield of parijoto pericarp extract and the whole fruits were 11.46 g (3.18% ) and 9.02 g (2.88%) respectively. Anthocyanins content of parijoto pericarp and whole fruit extracts  were 208.75 mg · L-1 and 173.7 mg · L-1. Administration of methanol extract of whole fruit parijoto  in hyperlipidemic rats reduced serum MDA levels significantly (p < 0.01), from 14.88 nmol · mL-1 to 8.63 nmol · mL-1.
