Abstract

A total of 236 yeast isolates from mollusc, litter, and plant samples from Gunung Halimun National Park were screened for cellulolytic activity based on Smith method by using 0,2% (w/v) cellulose-azure for 30 days. The results showed that 12 isolates (9 isolates from plants, 2 isolates from molluscs, dan 1 isolate from litter) have cellulolytic activity. These isolates were further screened based on Teather and Wood method for six days to determine their cellulases components by using specific substrates. Carboxymethyl cellulose (CMC) 0,1% (w/v) was used as a specific substrate to determine endoglucanase activity. Avicel 0,1% (w/v) was used as a specific substrate to determine exoglucanase activity. Cellobiose 0,1% (w/v) was used as a specific substrate to determine β-glucosidase activity. The results showed that 6 isolates from plant have β-glucosidase activity, and 1 isolate from plant have β-glucosidase and endoglucanase activities. Five isolates (2 isolates from plants, 2 isolates from molluscs, and 1 isolate from litter) showed no cellulase activity on specific substrates after six days incubation.

Abstract

Dua studi lapangan yang terpisah telah dilakukan di Lembah Baliem (138030’– 139030’ BT dan 3400’ – 4200’LS) Papua untuk mengidentifikasi jenis-jenis tanaman berguna bagi suku Dani yang hidup di lembah tersebut. Studi lapangan pertama dilakukan selama lima bulan (Maret – Juli 1994) di Mume-Kuyawage dan Mapnduma. Studi ini merupakan bagian dari program kajian ekologi Taman Nasional Lorentz dari WWF. Studi kedua merupakan observasi singkat (21 - 26 Mei 2005) di sekitar kota Wamena yakni Kumima, Siapkosi, Napua, Sinakma, Pisugi, Wanima, Sunili, Tulem dan Woma. Studi ini bagian dari penelitian usaha peternakan tradisional kerjasama Dinas Peternakan Kabupaten Jayawijaya, International Potato Centre (CIP) Bogor and South Australian Research and Development Institute (SARDI). Investigasi langsung dilakukan diikuti dengan wawancara semi-struktural untuk menghimpun informasi tentang jenis tanaman berguna yang biasanya dimanfaatkan oleh suku Dani.

Abstract

Penelitian inventarisasi jenis Opisthobranchia telah dilakukan untuk menunjang ekowisata di daerah Taman Laut 17 Pulau Riung. Taman laut ini terletak di kecamatan Riung, Kabupaten Ngada, Flores, Nusa Tenggara Timur. Kawasan ini di sebelah barat dibatasi oleh kabupaten Manggarai dan disebelah timur oleh desa Nggolonio, di utara dibatasi dengan laut Flores dan di selatan oleh Desa Wangka, Alo Mamek, dan Ten Terong. Areanya meliputi lima buah desa yaitu desa Lengkosambi, Tadho, Benteng Tengah, Nangamese dan Sambinasi. Secara geografis terletak antara 8o20’30” – 8o28’30” LS hingga 90o55’30” – 90o09’00” BT. Luas daratan sekitar 5 ha dan terumbu karang seluas 7.500 ha (Anonim a dan b. 1993). Opisthobranchia amat disukai oleh penyelam dan penggemar fotografi bawah laut dari mancanegara, namun masih amat sedikit penelitiannya di Indonesia

Abstract

Daerah tropis memiliki keanekaragaman jenis yang tinggi, misalnya saja lebih kurang 80% jenis burung petengger (passerine bird) hidup dan berbiak di daerah tropis. Namun barangkali belum banyak yang mengetahui bahwa beberapa teori dalam biologi dan ekologi burung lebih banyak didasarkan pada pengamatan empirik dan pemodelan yang menggunakan jenis burung dari daerah beriklim sedang (temperate). Hal ini tidak terlepas dari persebaran ahli yang sebagian besar terkonsentrasi dan melakukan penelitian di daerah tersebut baik Eropa maupun Amerika Utara. Celakanya, pola perilaku burung, misalnya, yang ditemukan di daerah tersebut selama ini telah dianggap sebagai norma perilaku umum untuk semua jenis burung. Apakah jenis-jenis burung di daerah tropis mengikuti norma perilaku umum tersebut?

Abstract

Penelitian ini merupakan kegiatan inventarisasi mamalia kecil di Toraut, Taman Nasional Bogani Nani Wartabone,SulawesiUtara. Pada penelitian ini dilakukan pengamatan diversitas mamalia kecil pada dua habitat utama yaitu hutan dan perkebunan, yang masing-masing diamati selama enam hari. Empat puluh perangkap kawat digunakan untuk mendokumentasi jenis-jenis tikus dan empat buah jaring kabut untuk kelelawar, serta pengamatan langsung di lapangan. Dalam penelitian ini dapat didokumentasikan sebanyak 129 ekor mamalia kecil yang terdiri dari 12 jenis.Limajenis di antaranya endemikSulawesidan tiga jenis mempunyai status konservasi dari IUCN.

Abstract

The study about fruiting and flowering phenologies on various tuber weights of iles-iles has not been common yet. The fact was emphasized by Lingga et al. (1989) that most biologists and agronomists are not interested in doing research about the iles-iles, so the source is not easy to get. This research was aimed to studying the fruiting and flowering phenologies on various tuber weights of Iles-iles (Amorphophallus muelleri Blume). The experiment was done at Bogor Agricultural Institute experimental field, Darmaga at 250 m sea level height, from February 2000 to August 2002. The experimental design used was Completely Randomized Block Design with six replications. The treatment consist of four level of tuber weights, A = more than 1050 g; B = more than 700-1050 g; C = more than 350-700 g; D = less than or same as 350 g. The results showed that flowering of iles-iles occur on rainy season, after high rain intensity everydays. The higher tuber weights, compared with smaller tuber weights, produce bigger percentage of flower sprout, size of flower parts, flower size and fruit amount, and also total amount of embryo per stem. On the contrary, the percentage of vegetative sprout was produced more by lighter tuber. The flowering and fruiting on iles-iles, seem influenced by the interaction between tuber weight and the growth age of tuber.

Abstract

A study on the antioxidative properties of white saffron extracted with aceton, ethanol or methanol was undertaken. The purpose of this research is to have a white saffron extract with a high antioxidant activity. White saffron rhizome was peeled, washed, grated, and added within either aceton, ethanol or methanol (white saffron : solvent = 1:3 or 1:4) (W/V) then stirred for 1 hour. The residue was washed with solvent in the same volume. The antioxidant activity of the extract was measured by Ferrythiocyanate (FTC) or Thio Barbituric Acid (TBA) methods. The result showed that the antioxidant activity of white saffron extracted with ethanol was higher than that with methanol, and aceton, respectively. The highest antioxidant activity was obtained from the extract of white saffron extracted with ethanol.

Abstract

Kajian laboratorium ini dilakukan untuk mengetahui mekanisme pengaruh negatif hujan asam terhadap makrofauna tanah. Armadillidium vulgare yang merupakan salah satu makrofauna tanah pemakan seresah (saprofag) penting di Tokyo yang digunakan dalam penelitian ini. Pengaruh makanan, pengaruh langsung dan pengaruh medium yang direndam dengan cairan asam digunakan untuk mengetahui mekanisme ini. (1). Pengaruh makanan dilakukan dengan cara merendam seresah daun aoki (Aucuba japonica) dalam air atau air yang diberi cairan H2SO4 dengan pH 6,4, dan 2. Seresah ini diberikan pada A. vulgare sebagai makanan (2) Efek langsung dilakukan dengan cara meneteskan 1 ml air atau air yang diberi cairan H2SO4 langsung ke tubuh hewan setiap dua minggu. (3) Pengaruh medium dilakukan dengan merendam medium dalam air maupun air yang diberi cairan H2SO4. Pertumbuhan dan persen hidup A. vulgare dicatat. Pertumbuhan dan persen hidup A. vulgare yang diberi makan seresah yang rendam dalam larutan asam sangat rendah. Penetesan cairan asam pada tubuh A. vulgare secara langsung tidak berpengaruh negatif terhadap pertumbuhan dan persen hidup A. vulgare. Pertumbuhan A. vulgare tidak dipengaruhi oleh medium yang direndam dalam cairan asam, tetapi persen hidupnya sedikit berkurang. Pengaruh cairan asam secara tidak langsung melalui makanan lebih penting daripada pengaruh langsung. Hal ini disebabkan tubuh A. vulgare dilindungi kutikula yang tebal. Untuk taksa yang memiliki tubuh yang lunak pengaruh langsung mungkin juga penting.

Abstract

Industrial waste-water has become a serious problem nationwide. However, one type of waste-water, i.e. tofu waste-water (whey), can be utilized as the source of nata de soya production. This present research is attempts to reveal the effect of various concentrations of inocula and green-bean extracts on nata productions from tofu waste-water. Ten combinations of treatment were applied and coded as A1 – A10. The parameters observed included physical and chemical characters of nata and nitrogen (N) contents. The results show that the increase in concentration of N and inocula will follow by the increase of water content, fiber content, plasticity, and N content of nata de soya produced. Highest thickness of nata was gained at 200 g/L concentration of green-bean extract and 100 ml/L concentration of inoculum.

Abstract

The aim of this study was to evaluate the concentration, progressively motile, and percent live sperm in the various regions of epididymis and ductus deferens after preservation at 4oC. Epididymis and ductus deferens were collected from 21 epididymis and ductus deferens of domesticated cat (Felis catus) by castration. One testicle of pair (control testicle) was analyzed at the day of castration, while the other testicle of the pair was stored at 4oC up to 7 days. The sperm concentration, percentage of sperm motility and live sperm were examined daily until day-7 of preservation. The sperm concentration was higher (p<0.05) in cauda epididymis (23.99x106 sperm/ml) and ductus deferens (25.42x106 sperm/ml) than caput (11.51x106 sperm/ml) and corpus epididymis (14.82x106 sperm/ml). The percentage of sperm motility and live sperm decreased (p<0.05) during preservation period. However, the percentage of motile (11.33 to 16.00 %) and live (15.05 to 20.20 %) sperm could be found in preserved epididymis and ductus deferens up to day-7. These results show that motile and live sperm can be collected from cat’s epididymis and ductus deferens up to day 7 after preservation at 4oC.
