Abstract

Kajian terhadap profil hormon-hormon steroid merupakan kunci penting dalam upaya memahami aspek fisiologis satwa. Beberapa dekade terakhir telah dikembangkan metode alternatif untuk mengetahui profil hormon steroid, yaitu melalui pengukuran metabolit hormon steroid yang diekskresikan melalui ekskreta tubuh seperti feses. Metode tersebut dikenal sebagai metode non-invasif. Metode ini memungkinkan pengumpulan sampel secara terus menerus dalam jangka panjang dengan meminimalisasi gangguan terutama pada satwa liar. Kajian terhadap profil metabolit hormon steroid yang terukur dapat diaplikasikan antara lain untuk mengetahui status reproduksi, penentuan jenis kelamin, studi perilaku hingga monitoring tingkat stres satwa. Berbagai kajian dengan memanfaatkan metabolit hormon steroid telah berhasil diaplikasikan pada berbagai taksa vertebrata. Tulisan ini memberikan gambaran terkini mengenai aplikasi metode non-invasif untuk kajian profil metabolit hormon steroid dari sampel feses.

Abstract

Pasak bumi is popular as an aphrodisiac in the traditional medical herb. Boiled drink water of the root powder of pasak bumi is believed can increase the secondary reproduction characters such as libido and the duration of erection. This article is part of the research report that focus on: the distribution of hypophysis acidophil and basophil cells caused of the pasak bumi optimal dose treatment. Research carried out in an experiment using Hemaktosilin Eosin(HE) staining method to study micromorphological distribution of the acidophil and basophil cells of the male rat hypophysis after pasak bumi treatment (the dose of 18 mg pasak bumi/200 g body weight during three days, and the control of 1 ml aquadest). Result of the research showed that: Basophyl cells increased significantly in the third days of pasak bumi administered. Meanwhile acidophyl cells decreased in the third days of pasak bumi administered.

Abstract

The aim of this study was to evaluate the concentration, progressively motile, and percent live sperm in the various regions of epididymis and ductus deferens after preservation at 4oC. Epididymis and ductus deferens were collected from 21 epididymis and ductus deferens of domesticated cat (Felis catus) by castration. One testicle of pair (control testicle) was analyzed at the day of castration, while the other testicle of the pair was stored at 4oC up to 7 days. The sperm concentration, percentage of sperm motility and live sperm were examined daily until day-7 of preservation. The sperm concentration was higher (p<0.05) in cauda epididymis (23.99x106 sperm/ml) and ductus deferens (25.42x106 sperm/ml) than caput (11.51x106 sperm/ml) and corpus epididymis (14.82x106 sperm/ml). The percentage of sperm motility and live sperm decreased (p<0.05) during preservation period. However, the percentage of motile (11.33 to 16.00 %) and live (15.05 to 20.20 %) sperm could be found in preserved epididymis and ductus deferens up to day-7. These results show that motile and live sperm can be collected from cat’s epididymis and ductus deferens up to day 7 after preservation at 4oC.

Abstract

Experiments were conducted on the morphology, fertilization and embryo development rate of vitrified ovine oocytes matured in vitro. Three vitrification solutions were used for vitrification. PBS supplemented with 1% BSA, 30% ethylene glycol was added by one of three different sucrose concentrations, 1.00 M (VS1), 0.50 M (VS2), and 0.25 M (VS3). The results showed that the percentages of normal vitrified oocytes after warming were 78 and 63% in VS1 and VS2, respectively, which was significantly higher as compared for VS3. The fertilization rates were 59 and 66% in VS1 and VS2, respectively, which were also significantly higher as compared with VS3 (35%). Zygote viability after 18 h was 57; 43; and 40%, for VS1,VS2, and VS3, respectively, which was not significantly different. The incidence of polyspermic penetration increased with increasing sucrose concentration, i.e 23, 11, and 9% in VS1, VS2, and VS3, respectively, as compared with unvitrified oocytes (4%). Abstract

Sumatran rhino is the most endangered rhino species. Its population is estimated less than 300 individuals remaining in the wild with highly declining rate to 50% in the last 15 years. The number of male rhinoceroses in the captivity are very few, therefore the assessment of its fertility is very important in order to support the breeding success since the captive breeding success is very poor. The objectives of this study were (i) to determine the male reproductive status, (ii) to establish a reliable semen collection method, and (iii) to assess semen parameters of the fresh collected sample. Three methods of semen collection were examined to determine its fertilizing potential, i.e. (i) stimulated combination of artificial vagina (AV), penile massage (PM) and accessory gland massage (AGM); (ii) AV and PM; and (iii) only with PM. The first method gave the best result with an ejaculation success of 85.71% (6/7, n = 7). The second and third methods obtained an ejaculation success rate of 50% (2/4, n = 4)) and 25% (1/4, n = 4), respectively. The collected ejaculates had a volume of 1.2-12.4 ml with whitish to cream turbid colour and pH 6.90-6.99. Sperm concentration was (143-333) x 103 sperm/ml. The quality of the sperm was low with only approximately 1% of them moved forward slowly. Approximately 80% of the spermatozoa were immature (prox. cytoplasmic droplet) with head (macro-, microcephalic) and tail abnormalities (broken tail). Semen quality increased after several collections and the amount of immature sperm decreased up to 5%. Electroejaculation procedure could produce 34 ml semen, but no sperm was found in the ejaculate. Hence, the combination of AV, PM, and AGM could get higher volume of ejaculate compared to other methods, but sperm concentration was better obtained using AV and PM only. Repeated semen collection increased semen quality, although the male has low fertilizing capacity due to low sperm concentration (oligozoospermia) and small volume of the ejaculate (oligospermia). Abstract

Heterotopic grafting sites can be useful in producing oocytes for in vitro Fertilization, therefore, maximising the oocyte yield from the graft by gonadotrophin stimulation would be advantageous. The aim of this study was to investigate the number and quality of oocytes collected from heterotopic autografted ovary after Pregnant Mare Serum Gonadothropin (PMSG) induction. Graft recipients were treated either with or without PMSG stimulation 48 hours prior to graft collection. Ovarian tissue from four weeks old mice (DDY strain) were autotransplanted under the kidney capsule of the same ovariectomized mice and the oocytes were collected 21 days after autotransplantation. The results showed that the average number of oocytes collected from autografted ovaries without PMSG induction were 9.0. + 2.8 not significantly different with those received PMSG induction, 10.9 + 5.1. The percentage of matured and fertilized oocytes and the developed embryos from the autografted ovaries without PMSG induction were 52.4, 33.4, and 26.0%, respectively not significantly different with those received PMSG induction, 53.2, 35.1, and 29.9%, respectively. The number of oocytes and the capacity to matured, fertilized and developed were significantly lower (P < 0.05) compared to the superovulated nongrafted (control) ovaries. In conclusion, PMSG induction on the graft recipients did not significantly increase oocytes yield from grafted heterotopic ovaries. The number and quality of oocytes produced from the autografted ovaries were lower than the superovulated nongrafted ovaries, but still can be used for in vitro embryo production after sequential in vitro maturation and fertilization.

Abstract

PO cattle have weaknesses to show clear estrus signs which cause difficulty in artificial insemination implementation. The present study was designed to obtain ovarian dynamic as effect of GnRH injection in ovsynch protocol based on progesterone intravaginal device. Heifers (18) and cows (n= 12) were allocated to one of three groups. Cuemate-PGF2α (CP) group inserted with a Cuemate on day 0-7 and injected with prostaglandin on day 7. Cuemate-PGF2α-GnRH (CPG) group was treated as CP group with the addition of GnRH injection on day 9. GnRH-Cuemate-PGF2α-GnRH (GCPG) group was treated as CPG group with addition of GnRH injection on day 0. Ultrasonography was performed on days 0-3, day 7 until ovulation and 7 days after ovulation. Percentage of ovulation synchronization increased significantly (P<0.01) between CP, CPG, and GCPG, respectively, both in heifers (16%, 50%, and 85%, respectively) and cows (0%, 60%, and 100%, respectively), on day 11. Preovulatory follicle diameters between CP, CPG, and GCPG treatments were not different significantly both in heifers (11.9±0.5, 11.9±0.5, and 12.1±0.6 mm, respectively) and cows (11.7±0.4, 11.8±0.7, and 11.1±0.6 mm, respectively). This study concluded that GCPG protocol increased the synchrony of ovulation rate both in cows and heifers, without affecting the follicle preovulatory and CL diameters.Key words: ovarian dynamic, GnRH, ovsych, PO Cattle

Abstract

An experiment was carried out to analyze the application of laparoscopic technique for oocyte collection, in vitro embryo production and embryo transfer in sheep. The first experiment was conducted to observe effect of gonadotropin stimulation on follicle development and laparoscopic technique for oocytes aspiration. In the second experiment, effect of culture system on the embryo development in vitro was assessed and in the third experiment, the application of laparoscopic for embryo transfer has been conducted. The result showed that single dose of gonadotrophin was sufficient to support follicle development significantly and it could help follicle visualization. It also showed that laparoscopic ovum-pick up could be conducted weekly without any restriction The second series experiment showed CR1aa culture system was better than TCM 199 (29.90°/o vs 8.00%) and the changing of media was required to ensure better metabolism process for embryos. The third experiment revealed that embryo transfer could be conducted with an aid from laparoscope. In conclusion, single dose PMSG stimulation is sufficient to support follicle development for /aparoscopic ovum-pick up, the culture media changing affects the successful rate of in vitro embryo production (8% vs 25.66%) and the laparoscopy technique can be used safely for embryo transfer on sheep.Keyword: laparoscopic, oocyte, embryo transfer, sheep

Abstract

Domba garut merupakan salah satu aset plasma nutfah Jawa Barat yang memiliki potensi untuk dikembangkan sebagai sumber daging. Domba garut memiliki keunikan tersendiri, terutama untuk jenis garut jantan tipe tangkas, sebagai daya tarik wisata daerah. Tujuan penelitian ini adalah untuk mengevaluasi hubungan antara ukuran fisik domba garut jantan tipe tangkas, yaitu bobot badan, lingkar skrotum, dan produksi semen dengan status reproduksi domba jantan. Penelitian ini dilakukan dengan mengukur hewan coba secara fisik pada kelompok umur 3 bulan, 6-12 bulan, 13-18 bulan, 19-24 bulan, dan 36-48 bulan. Evaluasi semen dilakukan pada semua kelompok umur. Pemilihan domba jantan garut tipe tangkas didasarkan pada sifat kualitatif dan kuantitatif. Sifat kualitatif (bentuk tanduk) yang dipilih oleh petani ialah gayor (44,44%), ngabendo (33,33%), leang (16,67%), dan ngagolong tali (5,56%). Sifat kuantitatif meliputi bobot badan, lingkar skrotum, dan panjang skrotum pada domba jantan, yang meningkat sampai kelompok usia 18-24 bulan. Evaluasi semen menunjukkan nilai tertinggi ditemukan pada kelompok usia 18-24 bulan. Bentuk tanduk tipe tangkas tidak memiliki korelasi dengan bobot badan, lingkar skrotum, dan panjang skrotum garut jantan secara statistik.

Abstract

Penelitian tentang sinkronisasi estrus pada sapi perah Friesian Holstein (FH) menggunakan cloprostenol telah dilakukan. Penelitian ini bertujuan untuk mengamati respons dan karakteristik estrus serta pengukuran nilai hambatan arus listrik lendir vagina. Penelitian dilakukan pada 20 ekor sapi FH dengan dua perlakuan injeksi cloprostenol, yaitu injeksi tunggal dengan keberadaan corpus luteum dan injeksi ganda dengan selang waktu injeksi 11 hari. Pengamatan estrus dan pengukuran lendir dilakukan lima hari berturut-turut setelah injeksi cloprostenol yang terakhir. Parameter pengukuran yang diamati meliputi respons, onset, dan durasi estrus, serta nilai hambatan arus listrik lendir vagina. Hasil penelitian menunjukkan bahwa injeksi ganda menghasilkan respons estrus yang lebih baik dibandingkan injeksi tunggal (90% vs 70%), dengan onset estrus lebih pendek (47,55 jam vs 53,28 jam), dan durasi estrus lebih pendek (24,85 jam vs 23,55 jam). Data hambatan arus listrik lendir vagina pada kelompok injeksi ganda lebih rendah dibandingkan dengan injeksi tunggal (187,77 vs 192,14), dengan pola tinggi sebelum estrus, rendah pada saat estrus, dan tinggi kembali setelah estrus, baik pada kelompok injeksi tunggal maupun injeksi ganda. Dapat disimpulkan bahwa respons estrus pada kelompok injeksi ganda lebih baik dibandingkan dengan kelompok injeksi tunggal.
