Abstract

Food poisoning cases need to be examinated intensively. There have been five poisoning cases reported between 2003-2004 in Sleman area. Three of them were caused by contaminated tuna fish. The objective of this research was to determine bacteriological quality of salted tuna fish from several markets in Yogyakarta. Salted tuna fish sample were collected from five markets, namely Beringharjo, Condongcatur, Demangan, Kranggan and Sentul. In each market, four vendors were selected. Bacteriological test were done including total microbe cells utilizing PCA medium, Coliform test utilizing VRBA medium. Identification and isolation of Vibrio parahaemolyticus use pre-enrichment selective medium of Alkaline Petone Water and selective Chromogenic medium agar for Vibrio. Salmonella test utilizing Chromogenic agar continued by utilizing TSI and LIA medium. The result showed that bacteriological quality of salted tuna fish from markets in Yogyakarta did not fulfill the requirements based on SNI 01-2717-1992. It was shown that total microbial count were high, i.e. 2.4 x 105 – 5.27 x 107 CFU/ g. Coliform count ranged from < 2.5 x 101 - > 6.5 x 106 CFU/g. From 20 samples that were brought to analyzed, 60% was positively presumed contaminated by V. parahaemolyticus and 50% was positively presumed contaminated by Salmonella.

Abstract

Protease is one of the leather commercial enzymes which is widely used such in food processing, medicine and leather industry. Clostridium sp T11-3 was isolated from Tiu Jeruk River in Nusa Tenggara Barat. Sequence analysis of 16S rRNA indicated that Clostridium spp T11-3 was closely related to C. bifermentans. This isolate produced maximum protease activity after 18 hours of cultivation in liquid media. Protease of Clostridium spp 11-3 displayed maximum activity at pH 5 and 60oC with casein as substrate. In the presence of 1 mM divalent ion Mg2+ the enzym activity increased to 141 %, while others ion divalent (Ca2+, Zn2+, Cu2+, Fe2+, and Co2+) inhibited protease activity.

Abstract

The influence of fed Orange II containing medium on the growth of Enterococcus faecalis ID 6017 and its decolorization ability was studied in this research. A fed batch growth was compared with the batch growth. Total Orange II added to the both was 120 mg/L. In the fed batch, the Orange II was given into three steps. The first addition was at the initial culture medium. The second and third were added after the almost total decolorization or clearance of the Orange II given in the first or the second addition respectively. The culture was incubated under static condition and room temperature. The fed batch growth was better than the batch growth, seen from both aspects; their growth parameters and the decolorization ability. The biomass yield and specific growth rates of the fed batch was higher than the batch growth. Under fed batch growth, the decolorization was 85-94%, while the batch growth was only 54%. For achieving these performances, the fed batch growth was needed to consume more glucose.

Abstract

Many people in West Papua value forest for the benefits obtained from the extraction of plants and animals. Hunting and gathering of wild animals has always been and continues to be an important aspect of life in rural West Papuan communities. Even in modern days, some ethnic groups in West Papua depend almost entirely on hunting as a part of their tradition (Pattiselanno, 2003). In other words, hunting is one of the main ways of living.

Abstract

Karakter morfologi 8 spesies yang merupakan koleksi dari Museum Zoologi Bogor (satu spesies merupakan record koleksi baru berasal dari Taman Nasional Gunung Harimun) genus Amphidromus telah dianalisa Cladistik. Koleksi Museum ini Amphidromus banksi, A. furcillatus, A. filozonatus, A. heerianus, A. inversus, A. javanicus, dan A. palaceus dimana koleksi terbarunya adalah A. alticola.

Abstract

The research of insecticide effect to the growth of genus Chlamydomonas had been done.  Research was experimental study with block random design to 9 concentrations of chlorpiriphos containing insecticide in Beneck and soil water media i.e. 0 (control), 0.05, 0.10, 0.15, 0.20, 0.25, 0.30, 0.35, and 0.40%.  Observations were done for 35 days.  Friedman test showed that there were some effects of media and insecticide concentrations on cell numbers of Chlamydomonas (cell/ml) in culture (p>0.05).  Dunnets test showed that mean of cell numbers of Chlamydomonas (cell/ml) differ (on p>0.05) and very differ (on p>0.01) on every concentrations in Beneck and soil water media.  Insecticide could reduced the cell size of Chlamydomonas, yet increased the number of cells on 0.05--0.30% concentrations in Beneck medium, and 0.05--0.20% concentrations in soil water medium.  On peak culture, insecticide with 0.10% concentration produced the highest cell numbers i.e. 21,179,167 cell/ml in Beneck medium, and 4,087,500 cell/ml in soil water medium.

Abstract

Penyusunan pustaka dari open reading frames (ORF) buatan yang tersusun atas 798 bp (pasangan basa), 576  di antaranya tersusun secara acak, yang mampu menyandi 266 asam amino telah berhasil dilakukan. Dalam upaya penyusunan tersebut diperoleh 32 transforman,  lima di antaranya membawa ORF buatan. Dari kelima transforman yang membawa ORF buatan tersebut, hanya satu transforman yang mampu berekspresi dan menyandi suatu protein. Protein yang dihasilkan memiliki ukuran 17 kDa, berukuran lebih kecil daripada ukuran yang diharapkan yaitu 29 kDa.

Abstract

Enterococcus faecalis ID 6017 can utilize glycerol as the source of carbon and energy for its growth. The present of glycerol in the medium containing Reactive Red-2 not only influenced its growth but also its ability to decolorize Reactive Red-2. The aim of this study was to investigate the growth of  Enterococcus faecalis (E. faecalis) and its ability to decolorize Reactive Red-2. The microbe was grown in batch system with three different growth medium, i.e. medium which contained (i) 1.643 g/l glycerol and 0.08 g/l reactive red-2, (ii) 1.643 g/l glycerol, and (iii) 0.08 g/l Reactive Red 2. The result of this study showed that the growth of  E. faecalis and its ability to decolorize Reactive Red-2 on medium contained glycerol was better than without glycerol. E. faecalis could not growth and decolorized Reactive Red-2 on medium without glycerol.

Abstract

Polyculture cultivation improvement of Aloe vera Linn (A. vera Linn) and Vigna unguiculata (V. unguiculata) has been done using Rhizobium strains inoculation onto V. unguiculata seeds to increase their growth and production. The study was done in Ciomas district, Bogor, using rice paddy field with a total of 24th plots of 1 x 2,5m2 width. A. vera Linn was planted until  two months old using an interval of  60 x 60 cm2. The Rhizobium used were strains number 171, 172, 173, and their combination of those three, which was planted using an interval of  20 x 35 cm2. The negative controls were uninoculated, without N (K1) and the positive controls were uninoculated, with urea fertilizer equal to 100 kg/ha (K2). The experiment design was using Randomized Complete Block Design (RCBD) with four replications.  The parameters used were leaves numbers, dry weight of plants, roots, root nodules, and pods production of V. unguiculata and leaves number, leaves length and width of A. vera.  Results showed that isolate number of 172 (isolate of Glycine max Linn.) had given the best results on the production of  V. unguiculata in the polyculture system with A. vera.
