Abstract

Plant Diversity in the Mount Celering Nature Reserve. Titi Kalima and Nur M. Heriyanto. Study of the plant species diversity in the Nature Reserve of Mount Celering (CAGC) Jepara, Central Java, was carried out from April to May 2008, with aimed to get information about plant diversity after natural disasters and encroachment. The method used was the checkered path with length of 500 m and width of 20 m, made of three lines cut slopes. The research was found 32 species, 29 genera, and 21 families. Tree level was dominated by Hibiscus macrophyllus Roxb. (IVI = 26.75%), Artocarpus elasticus Blume (IVI = 26.53%), and Alseodaphne umbelliflora Blume. (IVI = 22.75%). Saplings by Syzygium acuminatissimum (Blume) A.DC. (IVI = 23.86%), Dipterocarpus hasseltii Blume (IVI = 18.71%), and Artocarpus elasticus Blume (IVI = 18.52%). Six species of endangered trees were Alstonia scholaris (L.) R.Br., Alstonia angustifolia Wall, D. hasseltii Blume, Parkia javanica (Lamk.) Merr., Stelechocarpus burahol Hk.f.et Th., Sterculia cordata Blume. AbstrakPenelitian keragaman jenis tumbuhan di Cagar Alam Gunung Celering (CAGC) Jepara, Jawa Tengah, dilakukan bulan April sampai dengan Mei 2008 dengan tujuan untuk mendapatkan informasi tentang keragaman jenis tumbuhan pasca bencana alam dan perambahan. Metode yang digunakan adalah jalur berpetak dengan panjang 500 m dan lebar 20 m, dibuat tiga jalur memotong lereng. Dari hasil penelitian ditemukan 32 jenis, 29 genera, dan 21 famili. Tingkat pohon didominasi Hibiscus macrophyllus Roxb. (INP = 26,75%), Artocarpus elasticus Blume (INP = 26,53%), dan Alseodaphne umbelliflora Blume. (INP = 22,75%). Anakan pohon oleh Syzygium acuminatissimum (Blume) A.DC. (INP = 23,86%), Dipterocarpus hasseltii Blume (INP = 18,71%), dan Artocarpus elasticus Blume (INP = 18,52%). Enam jenis pohon yang termasuk katagori kritis dan terancam punah yaitu Alstonia scholaris (L.) R.Br., Alstonia angustifolia Wall, D. hasseltii Blume, Parkia javanica (Lamk.) Merr., Stelechocarpus burahol Hk.f.et Th., Sterculia cordata Blume.

Abstract

Cashew and cocoa are main commodities in South-East Sulawesi. Main problem of cocoa is cocoa pod borer (CPB) which to date, no effective control was found. Used of resistant variety is recommended control measure. The result of individual identification in field were: BPTP.R1.97, BPTP.R2.97, BPTP.R3.97, BPTP.W1.97, BPTP.W2.97, and BPTP.W3.97 resistant to CPB. Others having high yield i.e.: GC 7, ICS 60, ICS 13, UIT 1, Pa 300, TSH 858, RCC 70, RCC 71, RCC 72, and RCC 73. The result of side grafting trial at smallholder plantation the best clones were UIT-1, Na-32, ICS-13, Pa-7, RCC-70, RCC71, and BR-25. It is recommended for enters to side grafting the control to CBB. Problem of Cashew farming is low productivity and rainfall fluctuation. Individual selection on muna type 95 accessions were high productivity and 28 of the other were tolerant to rainfall fluctuation. Sixty two cashew accessions were collected. Six accessions were planted in commercial plantation. Superior clones of cocoa and cashew nut will be used for new planting and rehabilitation of old crop. AbstrakJambu mete dan kakao merupakan komoditas unggulan di Sulawesi Tenggara. Masalah utama pada kakao adalah serangan hama penggerek buah kakao (PBK) yang sampai saat ini belum ditemukan cara penanggulangan yang efektif dan efisien. Pilihan utama yang prospektif adalah menggunakan bahan tanam yang tahan. Hasil identifikasi di lapang ditemukan individu BPTP.R1.97, BPTP.R2.97, BPTP.R3.97, BPTP.W1.97. BPTP.W2.97, dan BPTP.W3.97 yang tahan terhadap hama PBK. Selain itu, telah dianjurkan menanam GC 7, ICS 60, ICS 13, UIT 1, Pa 300, TSH 858, RCC 70, RCC 71, RCC 72, dan RCC 73. Uji sambung samping kakao di pertanaman rakyat menghasilkan 7 nomor yang memiliki daya sambung amat baik dan baik yaitu UIT-1, Na-32, ICS-13, Pa-7, RCC-70, RCC71, dan BR-25. Masalah pada usahatani mete adalah produktivitas rendah dan berfluktuasi karena curah hujan yang tinggi. Dari seleksi individu pada tipe muna ditemukan 95 nomor berproduksi tinggi dan 28 nomor di antaranya toleran terhadap fluktuasi curah hujan. Koleksi Jambu mete mengsilkan 62 nomor aksesi. Enam nomor yang diunggulkan ditanam dalam kebun entres komersil. Klon-klon unggul kakao dan mete tersebut layak digunakan sebagai bahan tanam untuk pengembangan baru dan merehabilitasi tanaman tua.

Abstract

Study of SL-8-SHS hybrid rice was conducted in Watang Sawitto District, Pinrang Regency, to evaluate its performance and productivity in Wet Season (WS) 2007 June-November. This was planted on 2 ha of irrigated Riceland. Eighteen day old seedlings of SL-8-SHS and Ciherang variety (as control) was planted at a distance of 25 x 25 cm with two plants per hill. Fertilizers used were 300 kg/ha Urea which was applied at 10, 25, and 45 days after planting (dap) and 125 kg/ha SP36 applied at 10 day. Integrated pest management was applied to control pests and disease. Weeds were controlled manually as well as by managing irrigation water. Both hybrid and inbred variety showed good performance. However yield of hybrid rice was 8,5 t/ha which 39% higher than the inbred variety whose yield was 6, 1 t/ha. AbstrakKajian padi hibrida varietas SL-8-SHS di Kecamatan Watang Sawitto, Kabupaten Pinrang, Sulawesi Selatan, bertujuan untuk mengetahui penampilan dan produktivitas. Pengkajian dilaksanakan pada Juni-November 2007 pada lahan sawah irigasi seluas 2 ha. Benih padi yang digunakan adalah hibrida varietas SL-8-SHS dan sebagai pembanding digunakan padi inbrida varietas Ciherang. Tanam pindah dilaksanakan setelah tanah diolah sempurna dengan umur bibit 18 hari setelah semai, jarak tanam 25 cm x 25 cm, dua tanaman per rumpun. Tanaman diberi pupuk urea 300 kg + SP36 125 kg/ha. Pemberian urea dilakukan tiga tahap, yaitu pada umur 10, 25, dan 45 hari setelah tanam (HST). Pupuk SP36 diberikan satu kali pada saat tanaman berumur 10 HST. Pengendalian hama dan penyakit dilakukan berdasarkan pinsip pengelolaan hama terpadu (PHT). Gulma dikendalikan dengan cara manual dan genangan air diatur sedemikian rupa selama pertumbuhan tanaman. Tanaman padi hibrida menampilkan pertumbuhan yang baik dan memberikan produktivitas 8,5 t/ha gabah kering giling (gkg) atau 39% lebih tinggi dibandingkan dengan hasil padi inbrida varietas Ciherang

Abstract

The Phenotypic Characteristics of Alabio Duck (Anas platyrhynchos Borneo) in South Kalimantan. A study on phenotypic characters was carried out to identify Alabio duck (Anas platyrhynchos Borneo) being kept by smallholder. This research was conducted at Hulu Sungai Selatan (HSS), Hulu Sungai Tengah (HST) and Hulu Sungai Utara (HSU), South Kalimantan from May until November 2009. Six hundred (75 males and 525 females) the duck used in this study was Alabio duck ranged from 5-5.5 months old. The observed parameters were plumage color, color feature, plumage shine, the color of bill, feet and shank. The results showed that the ducks from three locations (HSS, HST and HSU) have same color patterns. The dominant color of the male duck is grayish white, grayish, brownish grey, blue-green and black. Whereas the female ducks of brown spotted and blackish grey. Twinkle Alabio duck plumage on males and females have the highest percentage is the glint of silver and shiny blue-green. The color of bill, feet and shank of male and female ducks were pale yellow until bright orange. AbstrakPenelitian mengenai karakteristik fenotipe untuk mengindentifikasi itik alabio (Anas platyrhynchos Borneo) di peternakan rakyat telah dilakukan di Kabupaten Hulu Sungai Selatan (HSS), Hulu Sungai Tengah (HST), dan Hulu Sungai Utara (HSU), Kalimantan Selatan mulai Mei sampai November 2009. Materi yang digunakan adalah itik alabio dara dengan kisaran umur 5-5,5 bulan sebanyak 600, ekor terdiri atas 75 ekor jantan dan 525 betina. Parameter yang diamati meliputi warna bulu, pola dan kerlip bulu, warna paruh, kaki, dan shank. Hasil penelitian menunjukkan bahwa itik alabio dari Kabupaten HSS, HST, dan HSU memiliki karakter bulu yang sama. Warna bulu dominan pada itik alabio jantan maupun betina adalah putih keabuan, abu kehitaman, coklat keabuan, hijau kebiruan, dan hitam. Itik alabio betina memiliki corak bulu coklat totol-totol sedangkan itik jantan hitam dan polos. Kerlip bulu pada itik alabio jantan maupun betina adalah kerlip perak dan hijau kebiruan mengkilap. Warna paruh, kaki, dan shank itik alabio jantan maupun betina adalah kuning gading pucat sampai kuning gading tua.

Abstract

Resistance Test of Promising Rice Lines Against Bacterial Leaf Blight (Xanthomonas oryzae pv. oryzae) Race III, IV, and VIII. Siti Yuriyah, Dwinita W. Utami, and Ida Hanarida. Development of new superior rice varieties resistant to bacterial leaf blight (BLB) has been conducted through utilizing of a wide rice germplasm, from crossing between IR64 and Oryza rufipogon. The aim of this study is to get promising rice lines that resistant to BLB race III, IV, and VIII. The experiments were conducted at greenhouse and Laboratory of Molecular Biology, ICABIOGRAD Bogor, using of 13 promising rice lines that have different levels of resistance to inoculum from pure cultures of BLB race III, IV and VIII. Of these 13 rice lines, six lines showed resistance to race III (Bio5-AC-Blas/BLB-03, Bio62-AC-Blas/BLB-03, Bio111-BC-PIR7, Bio129-BC-WBC, Bio148-Mamol, and Bio154-Mamol-Dro), one line showed resistance to race IV (Bio154-Mamol-Dro), and one line showed resistance to race VIII (Bio5-AC-Blas/BLB-03), with severity rate 1.8 to 8.1%. Of these improve lines Bio5-AC-Blas/BLB-3 and Bio 111-BCPir- 7, were released as new rice varieties, namely Inpari HDB and Inpari Blas, respectively. AbstrakPerakitan varietas padi unggul baru tahan terhadap hawar daun bakteri (HDB) terus dilakukan melalui pemanfaatan sumber daya genetik yang luas. Saat ini telah dirakit galur-galur unggul yang berasal dari persilangan IR64 dan padi liar Rupifogon. Penelitian ini bertujuan untuk mengevaluasi ketahanan galur harapan padi terhadap penyakit HDB dari Ras III, IV, dan VIII. Percobaan dilakukan di rumah kaca dan Laboratorium Kelti Biologi Molekuler, BB Biogen, Bogor. Materi genetik yang digunakan adalah 13 galur harapan padi dan biakan murni dari Ras III, IV, dan VIII. Metode inokulasi dengan pengguntingan dan skoring ketahanan dilakukan sesuai dengan sistem penilaian dari Yashitola et al. (1997). Dari 13 galur padi harapan yang diuji, diperoleh enam galur tahan terhadap Ras III (Bio5-AC-Blas/BLB-03, Bio62-AC-Blas/BLB-03, Bio111- BC-PIR7, Bio129-BC-WBC, Bio148-Mamol-Dro, dan Bio154-Mamol-Dro), satu galur tahan terhadap Ras IV (Bio154-Mamol-Dro) dan satu galur tahan terhadap Ras VIII (Bio5-AC-Blas/BLB-03), dengan tingkat keparahan sebesar 1,8-8,1%. Dalam perkembangannya, dua dari galur harapan, yaitu galur Bio5-AC-Blas/BLB-03 dan Bio111-BC-Pir-7 disetujui untuk dilepas sebagai varietas Inpari HDB dan Inpari Blas.

Abstract

The effect of freeze-drying process and preserving in a vacuum at room temperature against viability and pathogenicity of veterinary microbe germ plasma of Pasteuerella multocida BCC 2331 was investigated at Balitvet. The aim of this study was to find out the most effective and efficient conservation method. As much as 5,2 x 1011 colony forming unit (CFU)/ml of bacteria suspension in 7.5% glucose serum as the preservation medium being pathogenic in mice with LD50 of 9,8 CFU/ml was freeze dried then stored at room temperature (±27oC) until the study was completed. Viability and pathogenicity test were done immediately after the process, 1 and 2 months after storage. The results showed that there were viability decreases amounted 1,3 x 101 CFU/ml, 102 CFU/ml and 8,2 x l02 CFU/m1 due to the effects of the process, one month and two-month storage respectively. The decreases of pathogenicity on mice were shown by the increases of LD50 amounting log 1, log 2, and log 3 a day after the process, one month and two-month storage respectively. AbstrakPengaruh proses kering beku dan penyimpanan hasil proses pada suhu kamar 27oC terhadap viabilitas dan patogenisitas plasma nutfah mikroba veteriner telah dipelajari di Balitvet untuk menentukan cara pelestarian yang efektif dan efisien. Dalam kegiatan ini dipakai bakteri Pasteurella multocida koleksi Balitvet Culture Collection nomor koleksi B2331. Suspensi bakteri sebanyak 5,2 x l011 coloni forming unit (CFU)/ml dalam medium preservan 7,5% glukosa, serum dan bersifat patogen pada mencit dengan LD50 9,8 CFU/ml diproses kering beku, kemudian disimpan pada suhu kamar (+27oC) sampai penelitian selesai. Uji viabilitas dan patogenisitas dilakukan langsung setelah proses dan pada 1 serta 2 bulan setelah penyimpanan. Hasil penelitian menunjukkan terjadi penurunan viabilitas sebanyak 1,3 x 101 CFU, dan 8,2 x 102 CFU/ml masingmasing karena pengaruh proses, pengaruh penyimpanan selama 1 dan 2 bulan. Patogenisitas pada mencit menurun yang ditandai oleh adanya peningkatan LD50 sebanyak log 1, log 2, dan log 3 masing-masing 1 hari setelah proses, 1 dan 2 bulan setelah penyimpanan.

Abstract

Microbe germplasm of Bordetella bronchiseptica has been found in pigs in Indonesia and recognized as the causal agent of atrophic rhinitis and pneumonia, and also one of the agents involved in porcine respiratory disease complex. Those microbes have been characterized and conserved ex situ at Balitvet Culture Collection. The aim of this research was to find out seroepidemiological of B. bronchiseptica infections at four pig herds in two regencies in Central Java, by utilizing the germ plasma as antibody detection kit. The ELISA technique used lypopolysaccharide and sera hypperimmune of B. bronchiseptica local isolate as an antigen and positive sera respectively. Referring to cut off level of 0.404, the survey results showed that of 25.4% of 63 pig serum samples examine by ELISA were positively infected by B. bronchiseptica; 14.29% and 11, 11% from Karanganyar regencies and Sragen regencies respectively. Base on pig ages it showed that 10.35%, 24%, and 77.8% to be seropositive from less than three-month age group, three-month until five-month age group, and more than five-month age group respectively. Base on pig farm it was showed that 28.60; 30; 13.64; and 40% were seropositive collected from Farm 1, Farm 2, Farm 3 and Farm 4 respectively. The results indicated that B. bronchiseptica infection was spread on four pig herds in two regencies in Central Java with the higest seropositive (77,8%) in more than five-month age group. AbstrakPlasma nutfah Bordetella bronchiseptica telah ditemukan pada babi di Indonesia, selain merupakan agen penyebab atrophic rhinitis dan pneumoni juga salah satu agen porcine respiratory disease complex. Mikroba tersebut telah dikarakterisasi dan dikonservasi ex situ di Balitvet Culture Collection (BCC). Tujuan penelitian adalah untuk mengetahui seroepidemiologi infeksi B. bronchiseptica pada empat peternakan babi di dua kabupaten di Jawa Tengah, dengan memanfaatkan plasma nutfah tersebut sebagai bahan baku perangkat deteksi antibodi. Teknik Enzyme-linked Immunosorbent Assay (ELISA) menggunakan lypopolysaccharide dan serum imun dari isolat lokal B. bronchiseptica BS(BCC 2455) masing-masing sebagai antigen dan serum kontrol positif. Mengacu pada nilai cut off 0,404, hasil survei menunjukkan bahwa 25,4% dari 63 sampel serum yang diperiksa positif terinfeksi oleh B. bronchiseptica, 14,3% di antaranya berasal dari Kabupaten Karanganyar dan 11,1% dari Kabupaten Sragen. Berdasarkan pengelompokan umur diketahui bahwa seropositif sebanyak 10,35; 24; dan 77,8% masing-masing berasal dari kelompok umur di bawah 3 bulan, kelompok umur 3 sampai 5 bulan, dan kelompok umur di atas 5 bulan. Berdasarkan peternak sebaran seropositif sebanyak 28,60; 30; 13,64; dan 40% masing-masing pada peternakan 1, 2, 3, dan 4. Hal ini menunjukkan infeksi B. bronchiseptica telah tersebar di empat peternakan babi di dua kabupaten di Jawa Tengah dengan seropositif tertinggi (77,8%) pada kelompok babi umur di atas 5 bulan.

Abstract

Orang utan (Pongo pygmaeus) is a protected animal with restricted distribution was only in Sumatera and Kalimantan. With its restricted habitat and population in the conservation area, the occurrence of forest degradation would have significant impact to the habitat and population of this species, therefore forest area outside conservation area became important for conservation of orang utan. In this matter, production forest had been known as essential ecosystem for orang utan conservation objective. Population of orang utan was studied in Muara Lesan, former Concession Area of PT Alas Helau, Berau, East Kalimantan that covered area of 12,228 ha, with method of nest counting. Nest counting was carried out in transect lines 500-1000 m length. Total length of lines within transect was 28 km, or equal with 5.7 percent of study area. Population density of orang utan in Muara Lesan was between 1.92-7.13 individuals/km² (average of 3.69 individuals) with total population of 365-450 individuals. Estimation of population by nest counting method was influenced by age of nest (new to 285 days old), potency of food trees, movement behavior, including migration and condition of habitat. Based on its total population, orang utan in this area was categorized as critical. Protection of orang utan habitat and population in Forest Concession Areas of Kalimantan, in its management should determine sufficient conservation area, or should propose essential ecosystem for habitat and population of orang utan in former area of forest concession that was not managed as conservation area. AbstrakOrang utan (Pongo pygmaeus) adalah satwa langka yang dilindungi dengan penyebaran yang sangat terbatas di Sumatera dan Kalimantan. Dengan terbatasnya habitat dan populasi orang utan yang termasuk dalam kawasan konservasi, terjadinya degradasi hutan yang berdampak penting bagi habitat dan populasi, maka kawasan hutan di luar kawasan konservasi menjadi penting untuk pelestarian orang utan. Dalam hal ini hutan produksi telah diketahui sebagai ekosistem esensial untuk tujuan pelestarian. Populasi orang utan yang diteliti di kawasan Muara Lesan eks HPH PT Alas Helau seluas 12.228 ha dilakukan dengan metode penghitungan sarang. Penghitungan sarang dilakukan dalam jalur yang dibuat pada transek 500-1000 m. Dengan panjang total jalur 28 km, areal survei setara dengan 5,7% luas kawasan. Kerapatan populasi orang utan di Muara Lesan berkisar antara 1,92-7,13 individu/km² (rata-rata 3,69 individu) dengan jumlah total populasi 365-450 individu. Estimasi populasi dengan metode penghitungan sarang ini dipengaruhi oleh umur sarang yang mencapai 285 hari, potensi pohon pakan, perilaku pergerakan, termasuk migrasi serta kondisi habitat. Berdasarkan jumlah total populasi, orang utan di kawasan ini tergolong dalam populasi kritis. Perlindungan habitat dan populasi orang utan di kawasan HPH di Kalimantan, dalam pengelolaannya harus menetapkan wilayah konservasi yang cukup atau mengusulkan ekosistem esensial bagi habitat dan populasi orang utan di areal eks HPH yang tidak dikelola menjadi kawasan konservasi.

Abstract

Salacca Varietal Development of Sari Intan 48. Sri Hadiati, Agus Susiloadi, and Tri Budiyanti. The availability of new superior salacca varieties, which fulfills consumer’s preference, will increase fruit consumption and farmer’s income as well. Generally, consumers prefer salacca fruit which has the following characteristics: sweet taste, thick flesh and spineless peel. Crossing method is hoped to meet demand. This study aimed to obtain a new superior salacca variety through crossing within salacca varieties which were ready to release. The study was conducted in Solok district of West Sumatera Province and Tanjung Pinang district of Riau province and started from year 2002 to 2008. The method used in this research was crossing between Gula Pasir x Pondoh salacca, then their seeds were germinated at Indonesian Tropical Fruit Research Institute, located in Solok. The seedlings were then planted in Tanjung Pinang. Selection and evaluation activities were done over two years. The results showed that compared with Pondoh, Gula Pasir, and Bali varieties, the salacca of Sari Intan 48 had the following superior characters : thick flesh (0.5-1.8 cm), sweet taste (TSS: 19-20.8° Brix), no sour taste and non astringent, high vitamin C content(58.65 mg/100 g), and strong aroma. AbstrakTersedianya varietas unggul baru salak yang sesuai dengan selera konsumen dapat meningkatkan konsumsi buah dan pendapatan petani. Konsumen buah salak umumnya menyukai buah salak yang manis, berdaging tebal, dan sisik buah tidak berduri. Ideotipe ini dapat diperoleh antara lain melalui persilangan. Penelitian ini bertujuan untuk mendapatkan varietas unggul baru salak hasil persilangan antara beberapa varietas salak yang siap untuk dilepas. Penelitian dilakukan di Solok dan Tanjung Pinang, mulai tahun 2002-2008. Metode yang digunakan adalah persilangan antara salak Bali Gula Pasir x salak Pondoh, kemudian biji dipanen dan dikecambahkan di Balitbu Tropika sampai siap tanam ke lapang. Bibit ditanam di Tanjung Pinang dan selanjutnya diseleksi serta dievaluasi produktivitasnya selama dua tahun berturut-turut. Hasil penelitian menunjukkan bahwa salak Sari Intan 48 mempunyai beberapa keunggulan, antara lain daging buah tebal (0,5-1,8 cm), rasa manis (TSS : 19-20,8° Brix), tidak ada rasa asam dan sepet, kandungan vitamin C tinggi (58,65 mg/100 g), dan beraroma sangat harum dibandingkan dengan varietas pembanding (salak Pondoh, Gula Pasir, dan salak Bali).

Abstract

To broaden the genetic bases of indigenous vegetable, the country indigenous vegetable collecting mission to Karawang, Purwakarta, and Subang district West Java was conducted from June 17 to June 27, 2003. The Collector Team lead by IVEGRI scientist had successfully collected a total 111 accessions consisted of 74 accessions from seeds and 37 accessions from stems. This accession collecting of strains from farmers, seed shops, and markets. The indigenous vegetables also have some characteristic which promising, namely: good adaptation in various environment condition, as well as protein, vitamin, mineral and fiber resources which relative cheap, traditionally is one of cropping pattern especially to utilize home garden as well as tolerant to environmental stress. AbstakUntuk memperluas keragaman sumber genetik sayuran indigenous telah dilakukan eksplorasi dan koleksi ke Kabupaten Karawang, Purwakarta, dan Subang, Jawa Barat dari tanggal 17-27 Juni 2003. Tim kolektor yang dipimpin oleh peneliti Balitsa berhasil mengumpulkan 111 aksesi koleksi sayuran indigenous yang terdiri dari 74 aksesi berupa benih dan 37 aksesi berupa setek. Aksesi-aksesi koleksi ini diperoleh dari petani, toko benih, dan pasar. Sayuran indigenous juga mempunyai beberapa karakteristik yang cukup menjanjikan, di antaranya beradaptasi baik dalam kondisi lingkungan yang relatif beragam, merupakan alternatif sumber protein, vitamin, mineral, dan serat yang relatif murah, serta secara tradisional sudah merupakan salah satu komponen pola tanam, khususnya dalam pemanfaatan pekarangan dan relatif tahan cekaman lingkungan
