Abstract

Abstract

Hypoglycemic activity of hexane, ethylacetate, ethanol and aqueous extract of Andrographis paniculata herbs had been tested using glucose tolerance test. Extraction was conducted consecutively using hexane, ethylacetate and ethanol. Aqueous extract was obtained directly from the crude drug similar to the traditional use. At the dose level of 0.5 g/kg bw, hexane and ethylacetate extract did not show the hypoglycemic effect. Significant activity was shown by ethanol and aqueous extract and the activity of the latter was higher than the former.

Abstract

The moving split-window (MSW) technique, a landscape ecological method to detect ecotone or landscape boundary location, is described as an alternative to the use of conventional vegetation analysis. The effectiveness of MSW was tested by applying it to vegetation transect data collected from the field. While MSW analysis is practical and easily interpretable, results indicate that this method tended to overestimate minor shifts in species composition at the community level. For more accurate results, MSW must still be supported by information at the community level (e.g., by ordination), and an alternative method must be developed which makes use of the strengths of available methods.

Abstract

Intracellular dehalogenase enzyme had been isolated from Pseudomonas cepacia UK7WS1 strain which was grown in MSM broth containing yeast extract (0.18 g/L) and MCPA (0,02 g/L) and incubated for 22-24 h at 30 oC. Enzyme purification was carried out by ammonium sulphate fractionation, DEAE-cellulose column chromatography and Sephadex G-75 gel filtration column chromatography. Protein purify factor achieved was 12.4 times and protein yield was 10 % w/w of the crude cell extract protein. The enzyme purity of each purification stage was monitored by SDS-polyacrylamide gel electrophoresis and resulted in one protein band for each denatured enzyme (dimeric protein of two identical subunits) and non-denatured enzyme (monomeric protein subunit).

Abstract

Complexes [Fe(3-pp)3][X]2.2H2O, where 3-pp = (2-(pyrazol-3-yl)pyridine, and X = I, Br, NO3, and CF3SO3, and X2 = [Fe(CN)5(NO)], have been isolated and characterized in terms of magnetic, Mössbauer, and electronic spectral properties. All the dihydrate salts which were isolated from aqueous solution are red brown, and heating up to 110 - 1600C results in decomposition for all salts except for the triflate which gives complete dehydration to the anhydrous yellow salt. At room temperature, the magnetic moment of the dihidrate salts are slightly higher than the purely low-spin value (2,0-2,5 BM). The magnetic moments change gradually parallel to the change in temperature within 90 - 363K, and this is associated with the continuous spin state - singlet (1A1)quintet (5T2) - transition in iron(II). For the yellow anhydrous triflate salt, the spin state transition occurs dramatically, being discontinuous and associated with hysteresis (Tc↓ = 229K, Tc↑= 241K ; ∆Tc = 12K ). The triflate salt also shows a termochromic nature, being yellow at room temperature and dark red-brown at low temperature.

Abstract

Shoot-tips of broccoli, Brassica oleracea var. italica cv. Green Valiant, were used as explant source in in vitro clonal propagation. The objective of this research is to examine the influence of growth regulators on axillary shoot induction and root formation. Shoot-tips were excised from sterile germinated seedlings and placed on shoot induction medium containing basal salts of Murashige and Skoog. Benzyl adenine (BA) and kinetin (KIN) at 0.10-10.0 μM were used either singly or in combination with naphthalene acetic acid (NAA) at 0.01-10.0 μM to induce shoot multiplication. For inducing root formation, NAA, indole acetic acid (IAA) or indole butyric acid (IBA) at 0.10-10.0 μM were applied. Experimental results showed that the addition of BA at 10.0 μM induced axillary shoot proliferation respectively. The addition of KIN induced less shoot multiplication than that of BA. During a four-week period of subculture, a single shoot-tip of broccoli produced 16-20 shoots on medium with the addition of 10.0 μM BA. Elongated shoots were mostly rooted on medium containing 1.0 μM NAA. Plantlets were successfully transferred to a soil-sand mixture medium (1:1) and grown naturally in the green house.

Abstract

Translation termination in Saccharomyces cerevisiae is mediated by eRF-1 and eRF-3 protein encoded by SUP45 and SUP35 gene respectively. To probe more detail concerning the mechanism of translation termination in this organisms, the structure-function of the eRF-1 protein was studied by analyzing of sup45 mutants. In this study, we used three paramomycin hyper sensitive mutants, namely sup45-23, sup45-24 and sup45-38. Phenotypic characterization and quantitation of allosuppresor level showed that the mutants were allele specific mutants. Cloning and sequencing of the sup45 genes from these mutants showed that the sup45-23 exhibited Met48ATG → Ile48Ala and Gly431GGT → Gly431GGA mutation. While sup45-24 and sup45-38 exhibited single silent mutation, Gly431GGT → Gly431GGA . The structure-function analysis of sup45 gene of these mutant suggested that phenotypic mutants were not only due to the alteration of amino acid of eRF-1 protein in the cells.

Abstract

The achievement in genetic transformation is not only determined by the success in integrating transgenes into the plant genome, but also by the ability of transgenes to be stably expressed in the following generations. Transgenes integrated into trangenic wheat obtained via microprojectile bombardment method could be transmitted and, in most cases, expressed until the third generation as demonstrated in this study. The expression of the GUS gene was stable as observed in leaves, microspores, anthers, ovaries and seeds. Nevertheless, the bar gene appeared to undergo a reduction or disappearance in expression in a number of plants from the first to the third generation.

Abstract

Gallium Arsenide (GaAs) film have been grown by Metalorganic Chemical Vapor Deposition (MOCVD) method on Semi Insulating-Gallium Arsenide (SI-GaAs) substrates using of Trisdimethylaminoarsenic (TDMAAs) and Trimethylgallium (TMGa) precursors. The characteristic of GaAs film strongly depends on growth temperature. The best crystallinity quality of film was obtained at growth temperature, V/III ratio, reactor pressure, N2 and H2 dilute are 580 oC, 4.8, 50 torr, 300 sccm, respectively. X-ray diffraction data indicate that the GaAs films grown at 580 oC show epitaxial layer with FWHM on (200) peak of about 0,477o. Hall effect measurement data indicate that the grown layer were p-type semiconductor, with Hall mobility and carrier concentration in the range of 346 cm2/V.s and 3.17 x 1017 cm-3, respectively. The band gap of GaAs films determined by photoluminiscense (PL) measurement was 1,42 eV. This value is same as the band gap of bulk GaAs.
