Pelita Perkebunan, Coffee and Cocoa Research Journal (CCRJ): ISSN:0215-0212 Since its establishment in 1911, Indonesian Coffee and Cocoa Research Institute (ICCRI) formerly Besoekisch Proefstation, had published its research findings through a journal call Mededelingen van het Besoekisch Proefstation. Between 1948-1981 the research institute was under the supervision of Bogor Research Institute for Estate Crops, and published its research findings through De Bergcultures which was later changed to Menara Perkebunan. Since the institute held the national mandate for coffee and cocoa commodities, and due to rapid increase in the research findings, ICCRI published its first issue of Pelita Perkebunanjournal in April 1985. Pelita Perkebunanis an international journal providing rapid publication of peer-reviewed articles concerned with coffee and cocoa commodities based on the aspects of agronomy, plant breeding, soil science, crop protection, postharvest technology and social economy. Papers dealing with result of original research on the above aspects are welcome, with no page charge. Pelita Perkebunan is managed by Indonesian Coffee and Cocoa Research Institute (ICCRI), which publish the research findings not only for coffee and cocoa but also other commodities relevant with coffee and cocoa, i.e. shade trees, intercrops and wind breakers.

Abstract

One of the major diseases which limiting production in arabica coffee is the leaf rust caused by the fungus Hemileia vastatrix, B et Br. Selection and testing on thirteen promising arabica coffee clones were carried out at endemic area for leaf rust disease, Andungsari Research Station, for six fruiting times. The aims of these test were to find out superior planting material as clone with genetic resistance to leaf rust. As the beverage commodities, criterium selections for superiority clone besides resistant to leaf rust and yielding ability of more than 1,5 ton/ha also must be excellent in cup quality. Under this consideration, BP 416 A clone showed as the best high yielding ability i.e. 1,595 kg/ha and stable, besides resistant to leaf rust disease compared to the other clones tested. This clone had good cup quality, better than earlier released variety such as USDA 762 and S 795 although was not better than Andungsari 1 that were planted in the same location. The best physical bean characteristics was obtained on BP 513 A clone wich is derived from S 795 x Caturra red, having 89.2% of normal bean and 18.3% outurn characteristics. However, due to lower yielding and more susceptible to leaf rust, this clone could not be classified as superior clone.

Abstract

Differences in performance of grafting seedling were estimated from effect of using the root-stock and scion clones. This research aimed to study the differences of performance of some root-stock and scion clones on grafting seedling. The study used split plot design. First factor was root-stock consisted of ICS 60, Sca 6, Sca 12 and KW 165 and the second factor were 12 scions clones consisted of ICCRI 01, ICCRI 04, ICCRI 05, DR 2, Sulawesi 1, Sulawesi 2, KW 516, KW 514, KW 617, KW 570, KEE 2, and KW 604. There were three replication in each treatment and each replication consisted of 15 samples. Viability, lingkage diameter, ratio of root-stock and scion, hight of shoot, number of leaves and shoot were observed. The study showed that differences of root-stock just affected viability, lingkage diameter, and high of shoot. Scion clones factor affected to all of the characters. Their interaction caused the differences of all characters except of the number of shoot. Root-stock Sca 6 showed the lowest viability and number of the shoot, however root-stock KW 165 and ICS 60 showed the best performance to all parameters. The cluster analysis was done based on growth parameters and viability on lingkage distance of 15. There were three clusters; cluster 1 (KW 570), cluster 2 (ICCRI 04, ICCRI 05, Sulawesi 1, Sulawesi 2, KW 514, KW 165) and cluster 3 (ICCRI 01, DR 2, KW 516, KW 617, KW 604). Correlation analysis showed that main factor which affected the high viability root-stock diameter and diameter ratio of root-stock and scion were used.

Abstract

Plantlet that has developed shoots and roots will have a high level adaptation in the field. The objective of this experiment was to improve the ability of planlet in shooting and rooting so that it is ready for acclimatization in the field. The increase ability in shooting and rooting of the planlet were conducted by adding various types of auxin in the media. The arabica coffee embryo of clone AS 2K which has entered the phase of the cotyledons was transfered into the treatment media containing half-strength of MS (Murashige & Skoog) macro and micro nutrient, vitamin B5, 30 g/L glucose, 100 ml/L coconut water, 50 mg/L AgNO3 added with the combination of IAA, IBA and NAA. The research was conducted by using completely randomized design with seven combined treatment i.e. 0.1 mg/L IBA, 0.1 mg/L NAA, 0.1 mg/L IAA; 0 , 1 mg/L IBA + 0.1 mg/L NAA, 0.1 mg/L IBA + 0.1 mg/L IAA, 0.1 mg/L NAA + 0.1 mg/L IAA; without auxin. There were 12 replications in every treatment and each replication consisted of five cotyledonary embryos. The parameters of observation were the root length, leaf number, leaf area, stem diameter, and height of plantlets. The observations were conducted in eighth weeks after cotyledonary embryo had shoots. The results showed that in the number of leaves and height of planlet parameters, the treatment without auxin was the best result compared to planlet with auxin addition. The addition of auxin varians and their combination did not significantly influent leaf area, root length and stem diameter parameters. The medium tested was optimum for the growth of shoots and roots of AS 2K arabica coffee.

Abstract

Rice husk ash as desiccation material can be used to maintain seed viability in storage through its ability to absorb humidity during its storage. High relative humidity caused seed moisture content to increase so the respiration rate of seed increases and uses faster food stock. Finally the viability of seed is lost. A research on use of rice husk ash as desiccation material of cocoa seed storage was conducted in Agronomy Laboratory and Kaliwining Experimental Station, Indonesian Coffee and Cocoa Research Institute. Completely Randomized Design was used in this research with treatment of rice husk ash application on cocoa seed as follows: 0 g/100 seeds (A), 5 g/100 seeds (B), 10 g/100 seeds (C), 15 g/100 seeds (D), and 20 g/100 seeds (E). This experiment used four replications. Cocoa seeds were stored in plastic bag within carton box in ambient temperature. The storage periods were 1, 2, and 3 weeks, and parameters of observation consisted of electrical conductivity of dipped water of cocoa seeds, percentage of seed germination, percentage of seed emergence, early growth parameters at one month old including seedlings height of seedling, diameter, leaf number, root length, and dry weight. The result of the experiment showed that the use of rice husk ash at 5-10 g/100 seeds could maintain electrical conductivity of dipped water at low level, percentage of seed germination at 99-100 % and percentage of seed emergence at 79-91% after two weeks storage. The use of rice husk ash at 5-10 g/100 seeds after two weeks storage affected height of cacao seedling, but did not affected stem diameters, leaf numbers, root lengths, and dry weights

Abstract

A trial on cyantraniliprole 10% against coffee berry borer (Hypothenemus hampei) has been conducted on arabica coffee at Kalibendo Estate, in Banyuwangi regency, East Java. The altitude of the estate is about 650 m above sea level (asl.) and belongs to B type of climate classification according to Schmidt and Ferguson. Composite variety of arabica coffee at about four years old planted at the location was used as plant materials. Five levels of cyantranilprole dosage and two compared insecticides i.e: carbaryl 85% and lamda cyhalothrine 25 g/L have been applied as treatments and each treatment is replicated four times. Infestation of coffee berry borer (CBB) has been observed on berries in the field as well as on harvested berries and green coffee. The results revealed that cyantraniliprole 10% was very effective in suppressing infestation and population of CBB on coffee berries in the field as well as on harvested parchment and green coffee. The dosage of 2,000 ml/ha was the most effective and the highest level of efficacy against CBB until the last observation during 14 weeks trial. Application of cyantraniliprole 10% also has increased the production of green coffee harvested. The highest increase occurred on the treatment of cyantraniliprole 10% with a dosage of 1000 ml/ha, which it reached 62.87% higher compared to untreated treatment. Carbaryl and lamda cyhalothrine have effectiveness and efficacy level lower than the highest dosage of cyantraniliprole 10%.

Abstract

The approach to authenticate Luwak coffee is made through analysis of volatile compounds of luwak coffee. Luwak coffee bean from type of arabica obtained from Andungsari Plantation in Bondowoso district, East Java Province Indonesia, was wet processed and sundried prior to roasting step. As many as 120 g green bean was roasted at 170-220°C for 8-12 minutes until reached light brown colour (Agtron scale 65) and was ground prior to extraction. Volatile compounds of roasted Luwak arabica coffee bean were extracted by using solid phase microextraction (SPME) at 60°C for 30 minutes. The extracted analyte was subsequently transferred into GC-FID system by splitless injection at 260°C with five minutes sampling time, continued with separation through 50% phenyl 50% dimethylpolysiloxane capillary column and oven temperature programmed from 60°C to 180°C with rate of 5°C/min. Resulted chromatogram shows major peaks mainly in Rt 8.360-9.981, and Rt 9.705-14.778, and minor peaks identified before Rt 10 and after Rt 24. Varied sample quantity ranged within 0.5-2.5 g produced chromatograms which were not significantly different (p=0.08). This research also observed the use of γ-picoline (4-methylpyridine) as internal standard. It was showed that γ-picoline appeared at Rt 8.6~ without overlaying other peaks originated from sample. Concentration of γ-picoline at 0.05 μL/g, resulted separable peaks. These findings showed that the use of solid phase microextraction and GC-FID is capable to be apply for identification and quantification of Luwak coffee

Abstract

Bacteria live in wild in form of a consortium. Use of microbial consortium tends to give better results than single isolate, because the action of enzyme of each type of microbe can complement each other in order to survive. This study aimed to study the effectiveness of bacterial endophytic consortium from coffee plant on plant growth and mortality of parasitic nematodes in coffee. Isolation of bacteria is conducted  by growing the crushed roots, stems and leaves of coffee on 20% TSA media, then testing their hemolysis and hypersensitivity reaction. Selected isolates were tested on their effect on the growth of seedling and Pratylenchus coffeae mortality, as well as their chitinolytic, proteolytic, lipolytic, HCN production, dissolution of phosphate (P) and fixation of nitrogen (N2) abilities. The results showed that from 27 isolates of the consortium, 23 isolates showed negative reaction to hypersensitive test and 9 isolates to hemolysis test. The highest mortality rate was shown by K6 isolate (65.8%). The highest growth was shown by K15 and K 21 isolates while the highest root length by K21 isolate. Further analisys showed that 100% of the isolates could hydrolyze proteases, lipid, and produce HCN, while chitinolytic activity was shown by 78% isolates which could fix N2 and 11% of isolates could dissolve phosphate.

Abstract

Soil erosion is a serious problem in the field of ecology and environment. Providing rorak (small blocked ditches) and mulches as an alternative conservation action is expected to minimize water runoff. The purpose of this study was to examine the effects of combination of rorak and mulches in controlling water runoff in a cocoa farm. Location of this research was in Kaliwining Experimental Station, Indonesian Coffee and Cocoa Research Institute during rainy season in early 2015. This research used a nested design in which there is a complete factorial design of random groups that every combination treatment was repeated three times. Each experiment plot consisted of 16 cocoa trees of eight years old. The first factor was rorak treatment consisted of without rorak, 9 rorak per 16 trees (plot) and 16 rorak per 16 trees, whereas the second factor was mulch treatment consisted of control (without mulch), cocoa leaves as mulch and rice straw mixed with cocoa leaves as mulch. Rorak collecting runoff water was made of aluminum with a length of 40 cm, width 30 cm and  high 30 cm. Observation of runoff water was carried out early every morning. The results showed that increased number of rorak combined with cocoa leaf and rice straw significantly controlled runoff water compared to control. Rorak and mulch treatments were able to reduce runoff water compared to control. High number of rorak per plot increased the effectiveness in controlling runoff water when it was combined with mulch especially mixture of wide leaf (cocoa leaf) and needleshaped leaf (rice straw). Treatment of 16 rorak in every 16 trees with cocoa leaves and rice straw mulch could reduce runoff water by 82.8% compared to a control.

Abstract

Jember district is one of the centers for smallholder plantation of the Robusta coffee in East Java. However, there are still limited reports about environmental conditions, particularly soil fertility of existing smallholder plantations and its effect on coffee flavor. The purpose of this study was to investigate the effect of soil fertility on flavor of coffee in existing smallholder Robusta coffee plantations in Jember district. This research was carried out by survey method in several area of the smallholder Robusta coffee plantation in Jember. Evaluation of sensory profile was carried out using cup test organoleptic method based on Standard cupping method of the Specialty Coffee Association of America. Research locations as subdistricts were chosen by purposive sampling method, namely Panti, Silo, Rambipuji, Sukorambi, and Tanggul subdistricts. Results of this study showed that soil of the smallholder Robusta coffee plantation in research locations possessed low organic carbon content in Sukorambi subdistrict and medium concentration in other locations. Nitrogen content was low in all locations, potassium in Sumber Baru subdistrict was high whereas other locations were medium. Phosphorus availability was high in all locations. Calcium and magnesium were medium in Panti, Sukorambi, Sumber Baru, and Silo subdistricts. Soil acidity was low in all locations. Results of the sensory analysis of smallholder’s Robusta coffee was from divided into three groups. First, categorized as fine Robusta coffee was smallholder’s Robusta coffee samples obtained from Silo and Sukorambi subdistricts. Second, good quality Robusta coffee was from smallholder Robusta coffee samples from Panti and Sumber Baru subdistricts. Third, medium quality Robusta coffee was from smallholder’s Robusta coffee from Tanggul subdistrict. Total scores of coffee testing from samples of Panti, Sukorambi, Sumber Baru, Tanggul, and Silo subdistrict were 79.8, 82.0, 79.9, 76.0, and 82.5, respectively

Abstract

Cocoa powder is one of the main products of the cocoa grinding process which is frequently used in confectionery industries. Today, the utilization of cocoa powder has been derivatized not only it can be used as food ingredient, but also expanding in its utilization in pharmaceutical. The utilization of cocoa powder mainly due to its advantageous and healthy polyphenol content which should be extracted from cocoa powder. Generally, this process will result in significant amounts of underutilized cocoa powder grounds. This research was carried out to evaluate the utilization of ultrasound-assisted extraction method to extract antioxidative compound from cocoa powder and to study the possibility of utilizing its grounds for its antioxidative compound by the use of papain solution system. The extraction study was carried out using different concentration of ethanol ranged from 53.79, 60, 75, 90 and 96.21% for 23.39, 30, 45, 60 and 66.21 min extraction time. The evaluation was carried out using the analysis of total phenolic content and DPPH radical scavenging activity. Further, freshly prepared papain was used in different concentration ranged from 2, 4 and 6% to incubate the cocoa grounds for 2, 3, and 4 hours. The obtained cocoa hydrolysate solution was then analyzed for its protein content and its antioxidant activity. The optimization study of the process was carried out using response surface methodology. This study shows that the use of 60% ethanol for 30 min extraction was optimum to extract the phenolic compound from cocoa powder. On the other hand, the use of papain revealed that the underutilized cocoa grounds could be potentially used as the source of protein and antioxidant.
