Abstract

Cocoa plant generally is classified as cross pollinated plant and self-incompatible plant, then there is indication the presence of pollen influence on cocoa bean quality. This study was aimed to determine the effect of pollen sources (xenia) on the colour and size characters of fresh cocoa beans (Theobroma cacao L.). This study was done on two stages. The first stage, a treatment of hand pollination of cocoa clones of DR 1, DR 2, DRC 16, KW 264 and ICCRI 03 was applied in this study. There were five treatments of hand pollination: DR 2 x DR 1, DRC 16 x DR 1, DRC 16 X KW 264, DR 1 x DR 2 and ICCRI 03 x DRC 16. Each treatment was repeated four times. Colour parameters of fresh cocoa beans were observed. The second stage consisted of nine hand pollination combinations: TSH 858 x Sulawesi 1, TSH 858 x ICCRI 03, TSH 858 x KW 264, TSH 858 x KW 570, TSH 858 x Sca 6, Sulawesi 1 x TSH 858, Sulawesi 1 x Sca 6, Sulawesi 1 x KW 570 and KW 516 x Sca 6. Each treatment was repeated three times. Size parameters of fresh cocoa pod and beans were observed. The result showed that hybridization using pollen source from bulk cocoa and DR 1 contributed dark colour to fineflavored cocoa. This colour contribution was showed by the increase in the dark fresh percentage of cocoa beans. On the other hand, hybridization using pollen source from fine-flavored cocoa contributed white and pale colour to bulk cocoa. This light colour contribution was showed by the decrease in the percentage of the dark fresh beans. Xenia effect occured on pod weight, pod length, pod girth, bean weight with pulp per pod and bean weight without pulp per pod which was shown by the hybridization with TSH 858 as female parent and weight bean without pulp per pod shown by the hybridization with THS 858 and Sulawesi 1 as female parent. Abstract

Photosynthesis is one of the physiological process that influence the bean weight and this process related with the efectiveness of the stomata character and chlorophyll content in the leaves. The research was conducted at Kaliwining Research Station, Indonesian Coffee and Cocoa Research Institute. Design of experiment was randomized complete block design (RCBD) consisted of six clones as treatment were Sulawesi 1, Sulawesi 2, Sca 6, ICS 60, TSH 858, ICCRI 03, PA 300. Each treatment was replicated three times.  Stomata resistance diffusion, tranpiration,the content of chlorophyll a, chlorophyll b, chlorophyll total (a+b), bean number and bean weight were observed. The resuts  of experiment showed that  difference in chlorophyll a, chlorophyll b, chlorophyll total (a+b), stomata resistance diffusion, bean number and bean weight existed within  six clones tested. Transpiration rate did not show the significantly different between six clones tested. Sulawesi 1 showed the highest content of chlorophyll a and  ICS 60 and ICCRI 03 showed higher content of chlorophyll b than the other clones. Chlorophyll a, b and total (a+b) showed positively influence on bean number and bean weight. Transpiration rate had negatively influence to bean number per pod, on the otherhand it showed positively influence to bean weight. Chlorophyll total (a+b) showed high genetic variance (σg2), high phenotypic variance (σf2)  and high estimated value of heritability (H).  The chlorophyll a,b had moderate genetic variance, moderate phenotypic variance and high of estimated value of heritability.  Chlorophyll total (a+b) could be used a selection criteria based on the value of correlation, genetic variance, phenotypic variance and estimated value of heritability would give high opportunity in selection process.

Abstract

Differences in performance of grafting seedling were estimated from effect of using the root-stock and scion clones. This research aimed to study the differences of performance of some root-stock and scion clones on grafting seedling. The study used split plot design. First factor was root-stock consisted of ICS 60, Sca 6, Sca 12 and KW 165 and the second factor were 12 scions clones consisted of ICCRI 01, ICCRI 04, ICCRI 05, DR 2, Sulawesi 1, Sulawesi 2, KW 516, KW 514, KW 617, KW 570, KEE 2, and KW 604. There were three replication in each treatment and each replication consisted of 15 samples. Viability, lingkage diameter, ratio of root-stock and scion, hight of shoot, number of leaves and shoot were observed. The study showed that differences of root-stock just affected viability, lingkage diameter, and high of shoot. Scion clones factor affected to all of the characters. Their interaction caused the differences of all characters except of the number of shoot. Root-stock Sca 6 showed the lowest viability and number of the shoot, however root-stock KW 165 and ICS 60 showed the best performance to all parameters. The cluster analysis was done based on growth parameters and viability on lingkage distance of 15. There were three clusters; cluster 1 (KW 570), cluster 2 (ICCRI 04, ICCRI 05, Sulawesi 1, Sulawesi 2, KW 514, KW 165) and cluster 3 (ICCRI 01, DR 2, KW 516, KW 617, KW 604). Correlation analysis showed that main factor which affected the high viability root-stock diameter and diameter ratio of root-stock and scion were used.

Abstract

Level of anthocyanin content in flush is different between fine flavor cocoa and bulk cocoa. This study was aimed to determine the potential of anthocyanin and chlorophyll content in flush as one of the selection indicator for fine flavor cocoa. Flush sampling was carried out at Cocoa Germplasm Collections in Kaliwining Experimental Station, Indonesian Coffee and Cocoa Research Institute, while anthocyanin and chlorophyll analysis was conducted at Laboratory of Ma Chung Research Center for Photosynthetic Pigments, Ma Chung University, Malang.The research utilized a completely randomized design consisted of four genotypes of fine flavor cocoa, namely DR 2, ICCRI 02, DRC 16, PNT 16 and five genotypes of bulk cocoa, namely Sulawesi 1, MCC 01, MCC 02, KW 617 and PA 191.Each treatment was repeated three times. Anthocyanin and chlorophyll content in flush was measured. Anthocyanin analysis was done by destructive methods, while flush color was observed based on Munsell Color Chart for Plant Tissues Data analysis used orthogonal contrast, multivariate and simple correlation. The results showed that there were differences in flush color on fine flavor cocoa and that of bulk cocoa. The flush of the fine flavor cocoa had a tendency towards yellow and that of bulk cocoa had a tendency toward red with different levels of variation among genotypes. The analysis results of chlorophyll content in flush showed that chlorophyll content of fine flavor cocoa was higher than bulk cocoa with a range of 2376-3692 µmol/L for fine flavor cocoa and 1567-2628 µmol/L for bulk cocoa. On the contrary, anthocyanin content in fine flavor cocoa was lower than bulk cocoa with a range of 3.82-5.72 µmol/L for fine flavor cocoa and 6.01-11.88 µmol/L for bulk cocoa. The analysis results of orthogonal contrasts showed that chlorophyll content value was more stable than anthocyanin. There was negative relationship between anthocyanin and chlorophyll in flush with the regression Y = -221,1X + 3935 with a value of correlation r = -0.50. The anthocyanin and chlorophyll content in flush had a potential to be used as selection markers for fine flavor cocoa hybrid

Abstract

Self-incompatibility is an important factor in limiting the yield of cacao (Theobroma cacao L.). The research aimed to study the effect of self-compatibility on cocoa related to pod and bean characteristics and seedling performance. The research was conducted at Kaliwining Research Station, Indonesian Coffee and Cocoa Research Institute, Jember and Bogor Agriculture University in Bogor. The research used completely randomized design with three clones as treatment and each treatment repeated three times. The three clones used had a different self-compatibility characteristics; NA 32 (self-incompatible), DR 2 and Sulawesi 2 (self-compatible). Fruit set, pod characteristics, bean characteristics and seedling performance were observed. The results showed that self compatible cocoa clones had higher of fruit set per cushion (4,7-7,9 fruit set per tree) than selfincompatible (1,9 fruit set per cushion). Sulawesi 2 as a self compatible cocoa clone showed the lowest of fallen fruit set per cushion after two and four weeks. Self compatible cocoa clones (Sulawesi 2 and DR 2) had greater pod and better bean quality than self incompatible (NA 32) included pod length, pod weight, pod girth, weight of wet beans per pod, number of beans per pod, volume per wet bean, weight of dry bean and weight of nib. While number of poorly beans per pod in self incompatible showed higher than self compatible clones. Seedling performance of self compatible showed better than self incompatible for plant height and stem diameter. Visually the seedling of self compatible showed homogeny than self incompatible and variant analysis of plant height showed that self compatible were lower than self incompatible.

Abstract

Global warming impact in increasing temperature and becomes threat for cocoa production because growth of cocoa depends on the amount of water available in the environment. The objective of this study was to describe cocoa hybrid that may potentially be developed as drought-resistant cocoa hybrid. The study used 14 types of cocoa hybrids and ICCRI 06H as control. Those plants were approximately 4 years old and the hybrids were planted in Sumber Asin Experimental Station and Kendeng Lembu Plantation. The variables being observed to evaluate 2015 El-Nino effect were percentage of growing plants, tree diameter, jorquette height, flush and flower intensity in 2016. Analysis of variance, correlation and additive main effect and multiplicative interaction (AMMI) biplot were used to find out which hybrids were potentially drought-resistant. The finding showed that there was significant interaction between the genotypes (hybrids) and environment towards the percentage of growing plants, tree diameter, flush and flower intensity. The correlation between the variables showed that there was significant, negative correlation between the percentage of growing plants and height; even though there was significant correlation between the two, it was quite low. There was significant correlation between the tree diameter and the jourquette height while there was not any between the tree diameter and the percentage of growing plants. The AMMI analysis classified the drought tolerant hybrids into two groups, (1) cocoa hybrids adaptive to both normal as well as the temperature and water scarcity (drought), was ICCRI 03 x KW 617, and (2) drought tolerance cocoa hybrids, was KW 516 x KW 617.

Abstract

ENSO (El Nino Southern Oscillation) occurring in 2015 in Indonesia caused drought stress and the decrease in the percentage of production and even death of plants. One effective and relatively inexpensive way of reducing damage and economic losses due to drought is the availibility of tolerant cocoa. The objective of this research was to obtain tolerant clones based on morpho-physiological characters under drought stress. The experiment was conducted in Kaliwining Experimental Station of Indonesian Coffee and Cocoa Research Institute (ICCRI) during dry season (El Nino period with 5 consecutive dry months, from May to October 2015). Stomata, proline, leaf water status was measured at the peak of dry season on tested cocoa clones. The cocoa clones that predicted tolerant against to drought stress were KW 641, KW 514, KW 535, KW 619, and KW 516, whereas the cocoa clones that predicted susceptible under drought stress were KW 609, KW 614, KW 635, KW 606, and KW 651. The treatment clones had higher values of RWC, leaf proline content, narrower width and length of stomata openings, but smaller value of WSD compared to susceptible clones. Meanwhile, the character of leaf thickness might not significanly different to the character of drought tolerance.

Abstract

Natural enemy is an important factor in management of cacao pests. One way to increase its diversity and abundance is through agroecosystem management techniques that support. The study was conducted for one year from February 2014 to February 2015 in Banjarsari Plantation and a cocoa farm of Kaliwining Experimental Station, Jember, which applied different cropping patterns and use of insecticides. The purpose of this research was to study the effect of cropping pattern and use of insecticide on diversity and abundance of predators, parasitoids, abundance and intensity of Helopeltis antonii attack. Observation of pests and natural enemies was done visually, using sweep net, yellow pan traps, malaise trap and pifall trap. Intensity of the attack used Pedigo & Buntin equations. Diversity index used Shannon Diversity Index (H’) and Simpson’s dominance index (C). Differences in predator abundance, parasitoid, air temperature, relative humidity, light intensity and rainfall used Wilcoxon test. Results of this study showed that monoculture and intensive use of synthetic insecticides caused lower diversity and abundance of predators and parasitoids, while the abundance and intensity of H. antonii attacks were higher compared with polyculture without insecticide. Banjarsari cocoa plantation that applied monoculture and intensive use of synthetic insecticides, had lower diversity and abundance predators and parasitoids compared with cocoa plantation in Kaliwining that applied polyculture planting without using insecticide. Dominant predators in Banjarsari and in Kaliwining cocoa plantations were Dolichoderus thoracicus and Araneus diadematus, while dominant parasitoid in Banjarsari and in Kaliwining cocoa plantations was Araneus diadematus.

Abstract

Breeding for cocoa resistance to VSD is the main goal in breeding program in Indonesia. Research for selection criteria on cocoa (Theobroma cacaoL.) resistance to vascular-streak dieback (VSD) has being carried out by exploring leaf stomatal characteristics corresponding to VSD damages. This research had objectives to select best crosses by observing the estimated value of heterosis and heterobeltiosis on F1 hybrid populations based on leaf stomata characteristics related to VSD resistance. Trial was established at Kaliwining Experimental Station of Indonesian Coffee and Cocoa Research Institute (ICCRI) in a VSD endemic area. Treatments consisted of 5 F1 hybrid populations generated by biparental crosses of TSH 858(S) x Sulawesi 1 (R), TSH 858 (S) x KEE 2 (R), TSH 858 (S) x Sulawesi 3 (R), Sca 6 (R) x KW 264 (S) and KW 617 (R) x KW 264 (S). Each cross consisted of 100-300 genotypes planted in high planting density of 0.5 m x 0.5 m. Theresistance was evaluated by scoring the plant damage in the scale of 0-3 at three years after planting. Stomata characteristics were assessed microscopically based on the variables of density, length, width and diameter of stomata and width of opening stomata. Analysis of correlation indicated that most of stomatal charac-teristicssignificantly correlated to VSD damage but the width of opening stomata performedthe highest coefficient of correlation (r = 0.49) and contributed to VSD damage (0.45). Meanwhile, path analysis indicated higher value of the char-acteristics performed less resistance of the tested hybrids, KW 617 x KW 264 showed lower value of heterosis and heterobeltiosis for the characteristics which of the best crosses for generating VSD-resistant genotype.

Abstract

Cocoa is a sensitive plant to availability of soil water. The availability of water affects the formation of cocoa pods and beans. The aim of this research was to determine the genetic diversity and the influence of season on the diversity of cocoa beans quality as well as determining the pattern of genotype and season interaction on the quality of physical physiology of cacao beans. The research was conducted in Kaliwining Experimental Station, Indonesian Coffee and Cocoa Research Institute, Jember during dry season (2015) and rainy season (2016). This study used a complete randomized block design 2 x 12 factorial and three replicates as blocks. The first factor was the season consisting of two levels namely, dry season and rainy season. The second factor was cocoa clones, TSH 858, KW 084, KEE 2, Sulawesi 1, Sulawesi 2, BAL 209, KW 215, JTC 5A, JTC 5B, KC 2, KKM 22,and KJ 2. Observation variables included pod diameter, pod length, pod weight, dry bean weight, number of good beans/pod, number of empty beans/pod, numberof beans, and number of pods/tree/season. Data was analyzed using ANOVA fixedfactor. The bean dry weight characteristics possesed a low genetic diversity (0.27%) whereas the physical quality character of other beans, i.e bean count had moderategenetic diversity (14.20%). Meanwhile, the real difference was shown on the dry weight of bean characteristics. The best dry bean weight was observed during the dry season. Clones KW 215 and Sulawesi 1 in the dry season was catego-rized in grade A, while the lowest quality JTC 5A with grade D. The interaction of genotype (clone) with the environment (season) resulted in a significant effect onpod diameter character, pod weight, bean count, dry bean weight, and numberof pods/tree. Based on the biplot AMMI graph it was known that the TSH 858 clone showed genetic stability in bean count character. As for the character ofthe number of pods/tree, clones KKM 22, BAL 209, and KW 084 had the stability of the number of pods in rainy and dry seasons. Clones KJ 2 and Sulawesi 2 showed season-specific in the dry season for the number of pods/tree characters.
