Abstract

The aim of study was to identify the association of FSH beta-subunit gene polymorphisms withsperm quality traits. A total of 470 samples of normal mature bull from several breeds were used forpopulation study and 127 bulls from National and Regional AI centre of Indonesia for association study.To amplify, a PCR-RFLP method was used and digested with Pst1 restriction enzyme. The allelefrequency of the A and B in Bali cattle were (0.000) and (1.000), respectively. The absence of otherallele A suggested that the Bali cattle was monomorphic, while Brahman, FH, Simmental and Limousinewere polymorphic. The highest observed heterozygosity were found in Limousine (0.318) and thehighest expected heterozygosity were in Simmental (0.420). The higher incident of percentage of spermabnormalities were found in Simmental, Limousin, Brahman compared to Bali and FH. Among all typesof sperm abnormalities, the abaxial and microcephalus were found in highest number.

Abstract

The present study was conducted to analyze the broiler farmers’ capacity level in the partnership pattern and to analyze the factors which affect it. The current study was conducted in Bogor and Sukabumi, West Java Province using a survey approach on 247 farmers who ran farm businesses with the partnership pattern. Data were collected through questionnaire filling by farmers and in-depth interviews with key informants. The data were collected from July to October 2018. The data were analyzed descriptively and with inferential analysis (regression). The results of the study revealed that the farmers’ business capacity was weak. This was evident in the managerial and partnership capability which was categorized as low, the entrepreneurship and problem-solving capability which was categorized as medium, and only the technical capability which was categorized as high.  The regression results demonstrated that business capacity was influenced positively by the formal education level, social environment characteristics (organizational support, access to experts), and the role of private extension agents (their role as educators, communicators, consultants, dynamizators, and organizers).

Abstract

This research was aimed to investigate the different growth and to identify growth hormone gene polymorphism in Muscovy ducks. Two hundred Muscovy day-old ducks consisting of white-plumed male and female duck, black and white-plumed male and female ducks. Body weight was recorded weekly and the obtained data were subject to T test. Primer design used the Custal X Program based on a database from the GeneBank Cairina moschata GH gene, partial cds (AB158762). Primer base sequence of GH gene was forward/Sequence: 5’-CTGGGGTTGTTTAGCTTGGA-3’ and reverse/Sequence: 5’-TAAACCTTCCCTGGCACAAC-3’. The DNA sequences were aligned by using the BioEdit version 7.7 for identification of the single nucleotide polymorphism. The result showed that male Muscovy duck produced higher an average body weight gain and more relative growth than those of females. The highest body weight gain was at three weeks old, and then it started to decrease at four weeks old. The sequencing PCR product obtained nucleotide polymorphism. AA genotype was observed at 136 t of black female Muscovy duck, CC in black and white male Muscovy duck, and white female Muscovy duck. Conclusively, a body weight gain of 3-week-old male Muscovy ducks was higher than that of females and GH gene polymorphism was observed in Muscovy ducks.

Abstract

The objectives of this study were to obtain the fiber-digesting bacteria isolates from termitedigestive tract and to determine the optimum conditions of growth and production of cellulase, xylanaseand ligninase enzyme of isolate. The first study was conducted to isolate and select the fiber-digestingbacteria from the digestive tract of termites based on the highest activity of cellulolytic (S), xylanolytic(X) and lignolytic (L). The second study was optimation of the growth conditions of bacteria and theenzyme production due to effect of rice straw substrate and nitrogen. The material used were dry woodtermites, rice straw, and culture medium. The design used was a completely randomized factorial design,in which the first factor was rice straw substrate (1, 2, and 3% W/V), while the second factor wasnitrogen (0.1, 0.2 and 0.3% W/V). Variables measured were cellulase, xylanase and ligninase activities.Results of the first sudy showed that the isolates obtained consisted of 3 types, those were cellulolyticbacteria (S1, S2, and S3), 3 types of bacteria xylanolytic (X1, X2, and X3) and 3 types of bacteria lignolytic(L1, L2, and L3). Meanwhile, results of the second study showed that isolates of S2, X3, and L1 had thehighest activity, those were 1.894 U/mL, 1.722 U/mL and 0.314 U/mL, respectively. In conclusion, the addition of 1% level of rice straw substrate and 0.3% of nitrogen showed the highest enzyme activity oncellulase, xylanase and ligninase.

Abstract

This research was aimed to evaluate the effect of incubation time and level of urea on dry matter,organic matter and crude protein digestibility of Passion fruit hulls. The research was conducted by twophases. The first, the research was conducted to analyze of Passion fruit hulls before and afterammoniation, and then conducting digestibility of Passion fruit hulls using rumen fluid. FactorialRandomized Block design 2x3, with factor A was incubation time of ammoniated Passion fruit hulls (2weeks, 3 weeks) and factor B was the level of urea used (4%, 6% and 8%) was used in this study. Theresults showed that there was no significantly effects among treatments on dry matter and organic matterdigestibilities, but significant effect (P<0.05) on crude protein digestibility by level of urea treatment,even those there was no interaction between each treatment. The research showed that increasing levelof urea could increase in-vitro digestibility of crude protein nutrients. In conclusion, the best treatmentwas 8% level of urea with 2 weeks of incubation length.

Abstract

The study was aimed to improve the physical characteristics of culled duck meat using marinationin ginger extract and aging. The material used were Magelang culled ducks (2.5 years old). Gingerextract was derived from the rhizome of fresh ginger (Zingiber officinale Roscoe). The experimentalused was a factorial design (3x3), with two main factors, namely the concentration of ginger extract andaging time in completely randomized design. Level of ginger extract concentrations were 0, 5, 10% andlong times of aging were 24, 48, 72 hours as combination treatments, and replication for each treatmentwere 3 times. There was a significant interaction between the factors of aging time with ginger extractconcentration in influencing moisture, pH , hardness and the water holding capacity of culled Magelangduck meat. The best treatment to improve the physical characteristics of culled duck meat weremarination in 5% ginger extract and aging for 48 hours.

Abstract

An experiment on the use of Complete Rumen Modifier (CRM) to improve dairy cowproductivity and to mitigate enteric methane production has been conducted. Sixteen lactating dairycows were distributed into 4 groups by using compelete randomized design (CRD). Group I (Control)fed by basal diet consisted of elephant grass and concentrate 7.5 kg/hd/dy (CP 16% and TDN 70%),Group II (Pro. Woodii) fed by basal diet + probiotic Woodii, Group III (Pro.Noterae) fed by basal diet +probiotic Noterae; Group IV (CRM-Noterae) fed by basal diet + CRM + Pro.Noterae. Measurementswere conducted on body weight gain, average daily gain, feed conversion ratio, milk and methaneproduction. Results showed that CRM-Noterae increased ADG by 72% (1.29 vs 0.75 kg) and improvedFCR (9.2 vs 15.6). Probiotic noterae as single treatment or combined with CRM increased fat and totalsolid content of milk from 3.18% and 10.58% in control group to become 3.91%; 11.31% and 3.55%;11.02%, respectively. The lowest methane production was recorded in Group IV. The combination ofCRM and Noterae reduced percentage of methane production by 14%. It is concluded that combinationof CRM and Noterae can improve dairy cow performance and decrease methane production. ProbioticNoterae improved milk quality.

Abstract

Livestock including duck as one of crucial sector in agriculture, contributes a large proportion to Vietnam’s economic and to Mekong delta’s economic as well. Recently, herbal supplementation in feed is common which has brought many benefits for farmers and lemongrass is one of herbs is commonly used in many studies. A total of 96 ducks aged from 1 to 8-week-old was conducted through four treatments and three replicates per treatment. The experiment was a completely random design which aimed to estimate the effect of lemongrass powder on duck’s performance and carcass characteristics. The birds in the treatments were fed by the same feed but different supplement ratio of lemongrass powder (0; 0,5; 1; 1,5%). Besides, the birds were fed and watered ad libitum in all of the treatments. The conclusion showed that there was a linear decrease in feed intake, linear improvements in live weight gain and feed conversion as the proportion of lemongrass in the diet was increased from zero to 1.5%, inclusion of 1.5% lemongrass in feed will help the ducks increase their daily weight gain, improve feed intake and feed conversion ratio (P<0.05). However, lemongrass powder had no effect on carcass characteristics (P>0.05).
