Abstract

ABSTRACTTo understand the function of KNAT1 gene Arabidopsis in the forming and developing of Indonesian origin orchid shoot Phalaenopsis amabilis through transform p35S::KNAT1 and pGreen to protocorm like bodies (plb) of the orchid mediated by Agrobacterium tumafaciens LBA4404. The plb transformants were grown on New Phalaenopsis selection medium containing 5 mg/l BAP, 0.15 mg/l NAA, 15 mg.l Kanamycin and addition of 300 mg/l Cefotaxim to eliminate the overgrowth of Agrobacterium. The analysis of positive transformant use Polimerase Chain Reaction (PCR) with the specific oligonucleotide primers for KNAT1 gene: KNAT1F1R1 and universal primers for pGreen: 35SO and Tnos. The result shows that 3 shoots of 1850 transformants positively carry out the 35S::KNAT1 construct (frequency of transformation was 0.16 %) while 5 shoot of 1850 transformants also positively carry out the pGreen vector, with the frequency of transformation was 0.27 %. The phenotype analysis of 35S::KNAT1 transformants show multiplication on forming of the leaf from a plb to + 10 shoots and forming of the leaf shape which has terompet like shapes and rectangular shape.Key words: Arabidopsis, KNAT1 gene, Phalaenopsis amabilis, shoot

Abstract

ABSTRACTThe objective of the research was to obtain information on inheritance of characters related seed soybean storability. Genetic material used were 10 varieties and 10 breeding lines. The research was arranged in randomized completely block design with three replicates. The results showed that all characters observed was significantly affected by genotypes. Electrical conductivity showed the highest heritability among the characters observed. Some genotypes (B.3749, B.3641, Tanggamus, and Burangrang) showed good storability after 2 month stored period under room temperature conditions.Key words: soybean, seed storability, heritability

Abstract

This study aims to observe the effect of various types of mulch on the growth and yield of seedless watermelon (Citrullus vulgaris Schard). Implementing research in farmers' fields Scales Village, District Indralaya, Ogan Ilir, with a height ranging from 4-22 m above sea level, from February to April 2012. Research design using randomized block design, consisting of five treatments and five replications. The treatments were P0 = control (without mulch), P1 = use black-silver plastic mulch, P2 = use black plastic mulch, mulch P3 = using reeds as mulch as 0.8 kg of dry weight per m², P4 = use corn stalks mulch 2.6 kg dry weight per m². Parameters observed that the length of plant, days to flowering, fruit diameter, mean fruit weight, fruit weight per plant, fruit weight ha-1 (yield), wet biomass, and weight of wet roots. Black-silver plastic mulch influenced better on the growth and yield of seedless watermelon, compared with the other mulch and can increase the production of fruit namely the fruit weight/plant of 10.15 kg or 20.3 tons ha-1.

Abstract

ABSTRACTRubber replanting in Jambi Province started from 2006 to 2010 need millions high quality of rubber clon every year. Assesment was aimed to describe a potency of rubber seedling in every year mucipal in Jambi Province. Assesment was done in three month Maret to June 2006 in Tebo, Merangin, Sarolangun and Muaro Jambi by survey, interview with questioner of state breeder of rubber estate. Respondences were selected by purposive sampling. Result show each municipal has already TRUP. Quantity rubber seed was enough produced to fulfill their own need. Treat was occurred due to aviabilitty of entress, hence it needs a supply from other breeding places.Key words: High quality of rubber clon, Breeder, Entres, TRUP

Abstract

ABSTRACT   The objective of this experiment was conducted to identification  of seed-borne fungi that infected or contaminated paddy seed. Paddy seed samples were collected from sub district of Ciruas, Serang, Banten. Eighty hundred seeds  used  by random sampling  method as the work sample in seed health testing were  obtain from 8200 gram of the sample that collected from 82 group of farmer.  Testing method that been used were blotter test and agar test.   Thirty genera of fungi were found from this experiment.  They were Rhizopus,  Fusarium,  Mucor,  Alternaria, Drechslera,  Pythium,  Botrytis,  Penicillium,  Aspergillus,  Cercospora,  Cercosporella,  Trichoderma,  Colletotrichum.Keywords : identification,  fungi,  paddy,  seed-borne

Abstract

ABSTRACTSolid and liquid waste are important environmental problems that if not managed well, it great potential to reduce the quality of the environment and health. Efforts to overcome the solid and liquid waste requires an approach. The objective of this research is to know the handling of environmental issues, especially solid and liquid waste handlers in Banten Province and provides an alternative waste management in overcoming it. Research done by taking a secondary data from various institutions related to research such as this BAPEDAL Province Banten, the BPS Banten Province and library literature. Handling environmental issues in Banten Province is stressed to technology and financial approach rather than through a social approach, partnership, cultural, political, legal and institutional arrangements. In connection with this effort will need, a new approach and change the paradigm in the management of solid and liquid waste. For example, the management of solid waste is concentrated to handle waste only and it should be changed to penyadaran process community and in the case of waste management.Key words: Solid Waste, Liquid Waste

Abstract

ABSTRACTThe objective of this experiment was conducted to identification of seed-borne microorganism that infected or contaminated soybean from Galunggung, Wilis, and Anjasmoro varieties. Soybean seed samples were collected from BB Biogen Cimanggu Bogor. One hundred seeds used by random sampling method as the work sample in seed health testing were obtain from 1000 gram of the sample. Testing method that been used were blotter test and agar test. Bacteria and twelve genera of fungi were found from this experiment. Abstract

ABSTRACTThe objective of this experiment were find out interaction effect between soil tillage and Compost dose to some chemistry properties of Ultisols Banten . The experiment was carried at the Kampung Baru Petir Serang Banten from June to September 2008, by using Randomized Block Design with factorial pattern and three replications. The first factor was soil tillage (one, two and three time tillage), and the second factor was compost dung (0; 10; 20; and ton ha-1). The data of experiment were analyzed by Fisher test and Duncan multiple range test =0,05 The result of experiment showed there was interaction effect between soil tillage and compost dosages only on the soil pH, but independently soil tillage and compost dosages application gave significant effect chemical properties ( C-Organic, and Nitrogen).Key words: Soil Tillage, Compost dung, Ultisols

Abstract

ABSTRACTThis research aimed at knowing the potentials of composed siam weed (Chromolaena odorata) for enhancement of crops performance and its effect to fruitflies population’s to chili crops. The research was carried out at the experimental plantation of Agriculture Faculty, Gadjah Mada University, Banguntapan, Bantul, Yogyakarta from Oktober 2009 up to Maret 2010.This research used Randomize Complete Block Design used with 4 treatments, i.e., composed siam weed 40 ha-1 (G), dunk manure of cow 40 ha-1 (S), NPK (N) (Urea N 46% 40 kg/1000 m2, SP 36 20 kg/1000 m2, and KCL 20 kg/1000 m2), and without fertilizer as control (K) were 4 treatmens to repeated 6 time. Observation of arthropods population was taken with interval of 1 week. Crops samples observed were consisted of 2 plants per treatment. Crops performances observed were fruit number, fresh weight and dry weight of fruit, fresh weight and dry weight of crops. Totals N-leaf was analyzed on generative phases of the crops.Composed siam weed can not increased damage of herbivores if compared with other treatment, it’s indicated that highest of weigh of fruits and weight of crops. The result of research indicated that composed siam weed if compared with NPK and dunk manureof cow, that composed siam weed can not increased frutflies population. It suggest fruitflies prefer nutrition on fruit to secondary compound in chili crop.Key words: fruitflies, population, siam weed, chili.

Abstract

This experiments objective to determine the effect of the used of various growth substrates to make stock culture of oyster musroom spawn on the growt of mycelium. The experiments was conducted at the Faculty of Agriculture Agroekoteknologi Universitas Sultan Ageng Tirtayasa Serang Banten Province for the manufacture of pure cultures and Laboratory Pemuda Aktif Sejahtera, Village Sukamekarsari, Kalanganyar, Lebak, for making spawn in May until July 2011. The experiments used Randomized Control Group Design with seven treatments (A: corn , B: green beans, C: grain, D: millet, E: albasiah sawdust, F: mahoni sawdust, G: kecapi sawdust) and five replications. Results obtained from research the use of various growth substrates were significantly different age parameter begins to form mycelium, mycelium growth and percentage growth rete of mycelium.
