Abstract

Tujuan penelitian ini adalah untuk mempelajari karakter pertumbuhan, daya hasil lateks dan heritabilitas karakter kuantitatif  beberapa genotipe karet  PP/07/04. Sebanyak lima belas  genotipe karet dan dua klon pembanding PB 260 dan RRIC 100 diuji pada penelitian ini. Pengujian genotipe tersebut dibangun di Kebun Percobaan Balai Penelitian Sungei Putih, Pusat Penelitian Karet pada tahun 2004, yang terletak di Kabupaten Deli Serdang - Provinsi Sumatera Utara. Rancangan yang digunakan pada penelitian ini adalah Rancangan Acak Lengkap (RAL) dengan tiga ulangan. Hasil penelitian menunjukkan bahwa genotipe berpengaruh nyata terhadap dua belas karakter yang diamati yaitu lilit batang, tebal kulit, jumlah cincin pembuluh lateks, diameter sel pembuluh lateks, panjang alur sadap, kecepatan aliran lateks, indeks penyumbatan, kadar fosfat anorganik, kadar sukrosa, kadar thiol, kadar karet kering, dan  daya hasil lateks. Berdasarkan karakter pertumbuhan dan daya hasil lateks menunjukkan bahwa genotipe HP 92/309 memiliki potensi hasil lateks tinggi, sedangkan genotipe HP 92/542 memiliki hasil lateks tinggi dan pertumbuhan jagur. Dua belas karakter yang diamati memiliki nilai heritabilitas tinggi dengan nilai h2bs antara 0,52 – 0,95. Abstract

The objective of this research was to study the inheritance of Al tolerance of sweet sorghum at seedling stage. Thefour populations in this research were UPCA S1 (P1, susceptible parent) and Numbu (P2, tolerant parent), F1 and F1reciprocal and F2. The crosses of parental, F1, F1R and F2 generation were grown in University Farm of IPB and UPTDTenjo from June 2008 to June 2009. The study of Al tolerance inheritance in nutrient solution was conducted in greenhouse of University Farm of IPB from July-August 2009. Based on reciprocal analysis, all of characters are controlled bynuclear genes and their inheritance in not affected by maternal cytoplasm. The distribution of F2 genotype indicatedcontinuous with skewnesses which indicate that all characters are polygenics. All characters of Al tolerance in seedlingstage i.e. root length, root and shoot dry weight were controlled by additive gene action with contribution bycomplementary epistasis gene except shoot length that is controlled by additive gene action with duplicate epistasisgene. All characters had broad sense heritability from medium to high. Based on heritability value, pedigree selectionis the most effective selection method in breeding program for sorghum tolerance to Al.

Abstract

This research used gamma ray irradiation on low doses (micro mutation). The aim of this research was to know the respons of doses level bymicro mutation on gamma ray irridation to the growing and development of Argomulyo variety of Glycine max (L) Merr (M1), the seeds isirradiated by gamma ray by micro mutation was 0 Gy, 50 Gy, 100 Gy, 150 Gy, and 200 Gy. Variation that obtained of each characters atgeneration M1 and M2 influences plants growth and development either through qualitative and quantitative that finally will influence plantsproduction. The average highest genetic variation at M 2 generation of soybean was on 200 Gy doses. Results of the research indicated thatgamma ray irradiation on 200 Gy doses was effective caused of plant variation genetic.

Abstract

Global warming becomes a pressure in food production sustainability because it affected crop growth and development. The purpose of this study was to obtain information on the effect of high-temperature stress on the growth and development phase of rice and to evaluate the genotype for tolerance to high-temperature stress. Two environment conditions were used in the field and greenhouse of IPB Cikabayan experimental field, IPB University from August 2016 until February 2017. The study used varieties of IPB 4S, IPB 6R, Mekongga, and Situ Patenggang. High-temperature treatment was done by transferring the rice plants to the greenhouse at 50 days after transplanting. Observations were made on the generative phase in two different environmental conditions. The results showed that the total tillers number, filled grain number per panicle, unfilled grain number per panicle, total grain number per panicle, grain filling rate, percentage of filled grain and filled grain weight per plant had different responses among rice genotypes due to high-temperature stress. High-temperature decreased pollen fertility in all genotypes, which classified IPB 4S as a sensitive genotype and Mekongga as a tolerant genotype. Abstract

Sorghum has a growing popularity for food, feed, biofuel, and therefore needs to be developed. The research aimed to compare the effectiveness of pedigree and modified bulk selection methods for improving yield in three sorghum populations. The genetic materials used were 60 F6 lines from three populations, namely PI 10-90-A x Numbu, PI 150-20-A x Numbu and PI 150-20-A x Kawali which had been selected using pedigree and modified bulk methods. The experiment was conducted from October 2018 to February 2019 at the Cikabayan Experimental Station of IPB University, Dramaga, Bogor. The experiment was arranged in an augmented design with six checks, namely Kawali, Numbu, PI 10-90-A, PI 150-20-A, Samurai 1, and Samurai 2 which were replicated four times. Observations were made on agronomic traits and yield. Plant height and grain weight per panicle had high heritability and large genotypic coefficients of variation. The contrast test results showed significant differences between populations in plant height, panicle length, days of harvesting and 100-seed weight. On the other hand, no significant difference was observed among selection methods, indicating that both methods were equally effective for increasing sorghum yield. The pedigree selection and modified bulk selection increased grain yield per panicle by 14.1 g and 18.2 g respectively. Bulk of the best genotypes in early generation could be an alternative of bulk selection method. Keywords: contrast test, differential selection, genotypic coefficient of variation, heritability

Abstract

Penggunaan bibit sebagai bahan tanam maupun batang bawah untuk perbanyakan klonal sering dilakukan. Bahan tanam berupa bibit merupakan bahan tanam unggul yang berasal dari tetua unggul maupun persilangan antar tetua unggul. Penelitian ini bertujuan untuk mengetahui nilai duga komponen genetik melalui desain persilangan North Carolina II pada karakter morfologi tanaman kakao fase bibit. Persilangan dilakukan dengan menggunakan lima genotipe tetua betina, yaitu; KW 516, Sulawesi 3, TSH 858 dan dua genotipe tetua jantan, yaitu; ICCRI 09, Scavina 6. Pengamatan dilakukan pada tanaman umur 16 MST pada karakter morfologi tanaman yaitu tinggi tanaman, diameter batang, jumlah daun, panjang akar, volume akar, bobot basah tajuk, bobot basah akar, bobot kering tajuk, bobot kering akar, dan rasio akar/tajuk. Genotipe Sulawesi 3 merupakan genotipe tetua betina terbaik, sedangkan Scavina 6 merupakan genotipe tetua jantan terbaik. Genotipe persilangan terbaik berdasarkan nilai DGK nya adalah genotipe TSH 858 x Scavina 6, KW 516 x ICCRI 09, dan Sulawesi 3 x ICCRI 09. Genotipe persilangan Sulawesi 3 x ICCRI 09 merupakan genotipe dengan vigor yang baik yang memenuhi kriteria bibit unggul dan siap untuk ditanam di kebun. Karakter morfologi pada tanaman kakao fase bibit dikendalikan oleh gen dominan pada semua karakter kecuali pada karakter volume akar. Abstract

The challenges in upland rice cultivation are phosphorous (P) deficiency and aluminum (Al) toxicity, each controlled by Pup1 and Alt loci, respectively. Pyramiding the two genes into Indonesian rice varieties were previously done through Marker-Assisted Backcrossing method to obtain BC3F2 populations. The aims of this study were to analyze the BC3F2 upland rice lines containing the Alt and Pup1 loci molecularly (foreground and background analyses) and to test their phenotypic performances. Genetic materials tested included Dupa variety (donor of Alt) and three improved Indonesian genotypes (Dodokan-Pup1, Situ Bagendit-Pup1, and Batur-Pup1) as recurrent parents, Kasalath (donor of Pup1), and 300 BC3F2 lines from Dodokan-Pup1+Alt, Situ Bagendit-Pup1+Alt, and Batur-Pup1+Alt, respectively. The rice genotypes were selected individually using modified Yoshida nutrient solution, followed by foreground and background analyses. 150 out of 300 seedlings were selected and maintained until harvest in the greenhouse. Foreground analysis using markers (RM1361, RM12031, and Kas46-2) and tiller number performances resulted in 18 plants from BC3F2 Dodokan-Pup1+Alt, 30 plants from BC3F2 Situ Bagendit-Pup1+Alt, and 25 plants from BC3F2 Batur-Pup1+Alt still carrying Alt and Pup1 loci. Background analysis using molecular markers showed that the best individual lines of BC3F2 were number 56 for BC3F2 Dodokan-Pup1+Alt, number 32 or 70 for BC3F2 Situ Bagendit-Pup1+Alt, and number 20 for BC3F2 Batur-Pup1+Alt. The selected lines having both both Alt and Pup1 loci in homozygote condition with highest number of tiller per plant which are useful genetic materials for developing upland rice variety tolerance to low P and Al toxicity.

Abstract

Nicotine is the major alkaloid compound in cultivated tobacco (Nicotiana tabacum) that could potentially be converted into carcinogenic compound (nor-nicotine). The PMT gene encoding putrescine N-methyltransferase (PMT) is one of the two key genes that play a prominent role in nicotine biosynthesis. The aimed of this study was to isolate and characterize the cDNA sequence originated from Indonesian local tobacco cv. Sindoro1 (Ntpmt_Sindoro1). The results showed that the Ntpmt_Sindoro1 was 1124 bp in length. This cDNA fragment encodes for 374 amino acid residues. The predicted polypeptide from the cDNA is a hidrophilic protein, and has a predicted molecular weight of 40.95 kDa. The predicted amino acids sequence also showed high similarity to the PMT gene product Nicotiana sp. available in the GenBank data base. The amino acid sequences also exert conserved residues specifically exhibited only by PMT gene originated from N. tabacum. Clustering analysis revealed that Ntpmt_Sindoro1 belongs to the same clade as the PMT3 gene, a member of the N. tabacum PMT gene family. The Ntpmt_Sindoro1 cDNA sequence covering exon1-exon8 of the PMT gene fragment has been registered in the GenBank data base, under the accession number JX978277.

Abstract

Indeks seleksi merupakan salah satu prosedur seleksi sifat berganda yang penting dalam prog­ram pemuliaan tanaman. Tulisan ini bertujuan untuk mendeskripsi dan menduga parameter-parameter skor indeks dari metode-metode indeks seleksi nilai fenotipe, dan kemungkinan perluasannya bagi metode indeks seleksi nilai pemuliaan BLUP. Peragam fenotipe dan genotipe dari indeks basis dan Smith-Hazel digunakan sebagai landasan statistik untuk pendugaan parameter dan perluasan pengguna­an indeks. Penurunan rumus menghasilkan peragam aditif dan fenotipe bagi indeks seleksi berbasis nilai eigen, dan memperoleh parameter indeks seperti heritabilitas, koefisien deter­minasi, respons seleksi, respons seleksi tak langsung dan respons komponen sifat-sifat kuantitatif bagi semua metode indeks nilai fenotipe. Parameter peragam nilai pemuliaan sebenarnya dapat diduga dari analisis eigen dan keakuratan nilai pemuliaan BLUP yang memungkinkan metode indeks seleksi nilai fenotipe dapat diterapkan dengan prosedur yang sama pada nilai pemuliaan BLUP. Terakhir, heritabilitas dan koefisien determinasi indeks dapat digunakan dalam simulasi pembobot ekonomis untuk menghasilkan indeks terbaik.

Abstract

Tujuan dari penelitian ini adalah untuk mengevaluasi potensi hasil dari 8 galur harapan kedelai hitam (Glycine max (L.) Merr.). Penelitian ini dilakukan di Bogor (Jawa Barat) pada bulan Oktober 2011 sampai Januari 2012 sebagai bagian dari uji coba multilokasi. Rancangan percobaan yang digunakan adalah rancangan kelompok lengkap teracak dengan pengulangan dan tiga varietas nasional sebagai kontrol, yaitu Cikuray, Malika dan Willis. Hasil penelitian menunjukkan bahwa tinggi tanaman, waktu berbunga dan waktu panen, jumlah cabang produktif dan jumlah polong hampa sceara statistik berbeda. SSD-82, SSD-91, dan SC-68-2 memiliki produktivitas tinggi dibandingkan varietas pembanding (Cikuray).
