Abstract

Klebsiella pneumoniae causes several diseases such as chronic obstructive lung disease and community pneumonia. The antibiotics used for treating the infectious diseases cause several side effects. Therefore, alternative medicines which have minimal side effects are needed. This research aimed to investigate antibacterial activity of ten ethanolic extract of leaves against K. pneumoniae, identify compounds and active compounds in extract which has the highest antibacterial activity. Compounds in leaves were extracted using 70% ethanol with maceration method. The disk diffusion method was used for screening antibacterial activity against K. pneumoniae. The identification of group of compounds in extract with the largest inhibitory zone diameter was done using Thin Layer Chromatography (TLC) and bioautographic test was performed to determine the active compounds. The results of antibacterial activity screening tests showed that all extracts have antibacterial activity except bamboo leaf extract. Breadfruit extract (Artocarpus altilis) has the highest antibacterial activity with diameter of inhibition zone of 18,33 ± 5,57 mm. Breadfruit extract contains tannin, polyphenol, alkaloid, terpenoid, saponin, and the active antibacterial compounds were terpenoid and saponin.

Abstract

Iron was silvery white, clay, and can be formed.  it was usually obtained as hematite in nature. Iron was a chemical element that could be found in almost all places on the face of the earth, in all parts of the geological layer, and all bodies of water. In surface water, Fe was rarely found more than 1 mg / L, but in the water, Fe content could be much higher. When it accumulated in the body, iron could cause health problems, such as skin irritation, respiratory problems, and cause cancer in a long time. This study aims to determine the reduction of iron content in well water at the Universitas Muhammadiyah Surakarta (UMS) using the Spectrophotometry method with phenanthroline reagents. The samples were taken as many as five points in three different regions, namely Tanuragan (near the river), Menco and Nilasari (dense settlements), and Gonilan (near the rice fields) showed positive results containing iron with the highest levels in the sample without filtering at 9,478 ppm. These levels decreased after filtration using four filters, namely filters made of carbon, cotton, foam, carbon three layers in each sample. The iron content reduction was best to use cotton filters with 100% reduction in percent.

Abstract

Aloe vera L. contains antiseptic saponins and anthraquinone complexes as antibacterial. This research was conducted to formulate aloe vera powder into nanoemulgel, characterize and test its physical properties, and to know the effect of gelling agent concentration (carbopol and chitosan) on nanoemulgel. Emulsion type used is oil in water (O / W) with surfactant tween 80 and co-surfactant propylene glycol. Nanoemulsion is characterized by percent of transmittance with UV-Vis spectrophotometer, also potential droplet and zeta size distribution with Particle Size Analyzer. Hydrogels consist of 4 combination formulas of carbopol 0.5%; 1%; 1.5%; 2% and 0.3% chitosan. The formula is tested for physical properties including organoleptic, pH, spreadability, adhesion, viscosity and stability with freeze thaw method. The results showed that the nanoemulsion droplet size was 65.05nm ± 13.49 with zeta potential of -0.1mV and the percent of transmittance above 98%. The result of physical properties of the four formulas shows that the different gelling agent concentration gives different physical properties of viscosity and spreadability. Overall, formula 3 has the best physical properties and physical stability compared to formula 1, 2 and 4.

Abstract

Damaged skin due to free radicals from ultraviolet radiation can be prevented using antioxidants. Red dragon fruit (Hylocereus polyrhizus) containing antioxidants can be made into a peel off gel face mask using gelatin as a gelling agent to be applied easily to the skin. The purpose of this study is to determine the effect of various gelatin concentrations on the physical properties and antioxidant activity of peel off gels containing red dragon fruit juice. The peel off gels face mask were made using various gelatin concentration i.e 7.5% w/w (F1); 10% w/w (F2); and 12.5% w/w and containing 10% w/w red dragon fruit. The peel off gels then evaluated for physical properties and antioxidant activity using DPPH. The result showed that the increasing of the gelatin concentration would increase the drying time and adhesive ability of the peel off gels. The escalation of gelatin concentration from 7.5% w/w (F1) to 10% w/w (F2) did not affect the spreading ability of the peel off gel. However, the spreading ability of the peel off gel decreased with the increasing concentration of gelatin from 10% w/w (F2) to 12.5% w/w (F3). The antioxidant test of peel off gel had an average percentage of inhibition of 9.101% (F1); 12.469% (F2); and 15.109% (F3). Based on the results of physical properties and antioxidant activity showed that formula 2 was recomended.

Abstract

Streptomyces sp. GMR22 is local isolate from Wanagama 1 Forest in Yogyakarta. They have the potential to be developed to produce active compounds because have PKS and NRPS genes. The active compounds from isolation are strongly influenced by various factors, one of them is extraction techniques. Effect difference of extraction technique will be affected by the quality of secondary metabolites produced. The purpose of this study was to compare the cytotoxicity effects of secondary metabolites of Streptomyces sp. GMR22 which have extracted with different stages from previous studies. The extraction technique was carried out by multilevel separatory funnel extraction methods, which was first extracted using non-polar solvent (n-hexane) and then extracted using semi-polar solvent (ethyl acetate). This research is important because in previous studies (separatory funnel only extracted using ethyl acetate) with the use of the lowest concentration in the dengue virus antiviral test (further test) caused 100% of deaths in BHK-21 cells. This study indicate that multilevel extraction result in lower CC50 value than previous studies. There are 49.160 µl/ml (n-hexane extract) and 284.56 µl/ml (ethyl acetate extract) while water extract is 464,38 µl/ml. FTIR compound analysis show that the three extracts produced have different spectrum patterns, especially in the n-hexane and ethyl acetate extract. Value of CC50 is not too high, it is expected that the secondary metabolites contained in the extracts can be used for further analysis such as antiviral testing because it is safe for normal host cells such as BHK-21 cells
