Abstract

Infeksi Malassezia furfur pada kulit bisa karena faktor buruk . Salah satu bahan yang dapat digunakan sebagai anti jamur adalah ekstrak rimpang Lengkuas (Alpinia galanga L.). Penelitian ini bertujuan untuk mengetahui aktivitas antijamur dan efektivitas gel ekstrak rimpang lengkuas pada jamur patogen menggunakan metode maserasi dengan etanol 96 %. Pembuatan gel digunakan HPMC dan basis Carbopol. Pengujian aktivitas ekstrak dan menguji efektivitas gel dengan metode difusi cakram (uji Kirby - Bauer ). Hasil skrining menunjukkan bahwa ekstrak mengandung triterpenoid, flavonoid dan minyak esensial . Berdasarkan hasil uji terhadap jamur Malassezia furfur aktivitas antijamur dari ekstrak dalam gel meningkat dibandingkan ekstrak tanpa dirumuskan menjadi gel . Namun peningkatan aktivitas tidak signifikan berdasarkan analisis statistik dengan satu - way ANOVA diperoleh signifikansi sebesar 0,234 (p>0,05).

Abstract

Phalerin in extract of Phaleria macrocarpa fruit has activity as a sunscreen. The aims of this research was to discover the physical stability of extract in lotion  and primary irritation test. The optimal formulation o/w was determined by Simplex Lattice Design (LSD) method using software Design Expert® version 8.0.7 with variety of cetyl alcohol, stearic acid, and triethanolamine and lotion w/o with variety of cera alba, span 80 and cetyl alcohol. Lotion was made in 10 optimum formula then the physical characteristics were measured. The phycical characteristics was employed to create the optimum formula. The difference of physical characteristic between experimental and software was analyzed by one sample t-test analysis statistics. Optimum formula of lotion was determined on skin white rabbit. The optimum formula lotion o/w were cetyl alcohol 2,690%, stearic acid 4,146%, and triethanolamine 3,164% and lotion w/o were cera alba 12,136%; span 80 7,181%; and cetyl alkohol 3,683%. The result of one sample t-test gave no signifficant differences on physical characteristics between experimental and software. Optimum formula of lotion o/w was not irritant at topical on white rabbit skin.

Abstract

Quorum sensing is a communication system among bacterial cells which correlates with biofilm formation. Biofilm can protect bacteria from environment including antibiotic of which can cause higher antibiotic concentration of 100 up to 1000 times. Inhibition of quorum sensing is expected to inhibit the biofilm formation. The cinnamon bark (Cinnamomum burmanii Ness. Ex Bl.) has been known to have antibacterial and antibiofilm activities. Leaves are available abundantly which urges a research to find out the activity as anti quorum sensing against Pseudomonas aeruginosa. The succesive maceration of dried pulverized leaves produced hexane, ethyl acetate, and methanol extracts.  Antibacterial activity was observed by microdillution method with MTT assay. Afterwards, the active extract was examined for anti quorum sensing activity by diffusion method in cetrimide Agar. Quorum sensing activity was shown by dark zone (opaque) growth around sample application, observed under UV light of 366 nm. TLC bioautography method was done to observe the active spots by using silica gel F254 as the stationary phase, chloroform-methanol (6:1 v/v) as the mobile phase, loading sample used was 1.25 mg and 30 min of plate contact duration. The ethyl acetate extract inhibited growth of  P. aeruginosa with shown by MIC at 8 µg/µL Quorum sensing as well as growth inhibition activities were observed at loading samples 12,5 and 25 mg/wells, while at 6,25 mg the extract only exhibited quorum sensing inhibition. Presences of substances having phenolic, flavonoid, alkaloid and aldehyde/keton as functional groups were detected by TLC method of the extract but no active spot identified on bioautography.

Abstract

Premature aging skin condition characterized by dry, scaly, rough and accompanied by the appearance of wrinkles and dark spots or blemishes has become to be a women fearing thing, especially among women in reproductive age. Factors causing premature aging are internal factors (health, stamina, stress and hormonal changes) and external factors (free radicals, sunlight and pollutants). Free radicals can be prevented by antioxidant compounds either synthetic or natural materials. Tomato is one of the natural antioxidants. The most antioxidant content in tomato is lycopene. Tomato is used in the form of tomato juice made by extraction method. Tomato in their use directly is less effective so that tomato is made in the form of cream. Cream compositions were made by variation concentration of tomato juice (5%, 10%, 15%, and 20%). The antioxidant activity was tested by DPPH method. Physical stability parameters tested in this research were homogenity, viscosity, dispersive power, adhesion, cream separation ratio and pH. Data were analyzed by Kolmogorov-Smirnov test, one way ANOVA test and Tukey test. Organoleptic test data, the physical properties of the cream, and the type of emulsion were analyzed descriptively .The results showed that tomato juice has antioxidant activity with IC50 value 2,69%. The increase in the concentration of tomato juice gave significantly different results in all formulas except the formulas III and IV that were not significantly different. The IC50 values cream of tomato juice were 9,12%; 708%; 3,61%; and 2,85% respectively for the cream with tomato extract concentration of 5%, 10%, 15%, 20%. This showed that the higher the concentration of tomato juice, the higher the antioxidant activity as indicated by the lower of IC50 value. The increase in the concentration of tomato juice as active ingredients provided significantly different results for difference antioxidant activity, color, and smell, but did not cause significant differences in the consistency, viscosity, dispersive power, time and pH.

Abstract

The aim of the study was the determination of glycaemic index (GI) of local tubers such as Uwi (Dioscorea alata), Gadung (Dioscorea hispida), dan Talas (Colocasia esculenta). Tubers were prepared as raw flour which were administered orally to the rats. Blood were taken at 0; 0,5;  1 and 2 hour after tubers administration. The dose was 2,5 kg/kg body weight (BW) whereas glucose was administered as a standart treatment. Area Under Curve (AUC) was calculated after plotting the graph of time versus glucose levels. Glucose levels were determined using GOD-PAP kit. GI values were calculated as a ratio of AUC (0-2hr) of tubers to glucose. Porang was moderate while the other tubers were low GI.

Abstract

Disorders of carbohydrate metabolism can lead to diabetes mellitus. Carbohydrates  are metabolized in the gastrointestinal tract  into simple glucose and absorbed into the bloodstream and affected blood glucose levels. The absorption process is catalyzed by α- 1 ,4 - glycoside breaking bond  enzyme , namely α - amylase and α -1 ,6 - glycoside breaking bond  enzyme, namely α – glucosidase. They are found in the intestinal cells. Research had been conducted in an effort to develop an alternative treatment of diabetes mellitus by testing the ability of isolates of andrographolide in inhibiting α-amylase activity and α-glucosidase in vitro. Andrographolide isolates showed fairly good activity in inhibiting α-amylase ( IC50 = 1,.49 mg/mL) and weak in inhibiting α-glucosidase (IC50 = 38,86 mg/mL). Inhibition of α-amylase activity is evidence of one mechanism of andrographolide in reducing carbohydrate metabolism that can affect blood glucose levels and indicates that andrographolide is  a potential alternative medicine in addressing diabetes mellitus .

Abstract

Mosquitoes are one of the insect vector-borne diseases such as dengue, malaria, filariasis, etc. The use of materials whose got repellent activity such as lemongrass, cloves, and orange leaves are one of the best way to be protected from mosquito bites. Combustible insence known as aromatherapy preparations that can be used at any time, which is commonly used in religious events. This research is intended to make preparations of combustible insence which has dual functions, serve as room fresheners and also as repellent against Aedes aegypti. The research is conducted by making preparations of the active ingredient of lemongrass, cloves, and orange leaves, each with 5 different concentrations. Those preparats are tested on 25 mosquitoes contained in double trap mosquito cages. The number of mosquitoes in box A and B will be calculated at 5, 10, 15, 30, 45, and 60 minutes respectively. Thus, the repellent activity can be determined. Chromatogram profile testing is also being conducted using TAS microdestilation metode with several reagents for identification. Result shown that all preparations have mosquito repellent activity with 40% of effective concentration for lemongrass, 20% for orange leaves, 10% for cloves leaves. Chromatogram profile test shows orange leaves have the most spots for terpenoid and fenolic compound, followed by cloves and lemongrass. Spots as compounds with carbonyl groups (aldehydes and ketones) is only found in lemongrass.

Abstract

Hibiscus flower (Hibiscus rosa-sinensis L.) is efficacious in cough remedies. This flower has many variations , one of which is a red flower with a crown subject.  Ethanolic extract of the hibiscus flower has been studied and shown to have mucolytic activity. Fractionation will be closer in the process of finding active compounds. This study aimed to determine the effect of fractions containing alkaloids of red hibiscus flower varieties subject as mucolytics in vitro and determine the range of concentrations of the fractions equivalent to acetylcysteine effect of 0,1 %. This study included maceration, fractionation by VLC, the identification of fractions containing alkaloids, and mucolytic activity test fractions containing alkaloids. Mucolytic activity assay performed in vitro to decrease the viscosity of mucus cow. Test solutions were made with concentrations of fractions 0,4;0,6; and 0,8 %. Acetylcysteine 0,1 % was used as a positive control. Viscosity values were analyzed statistically using one -way ANOVA test , followed by the LSD test level of 95 % to determine differences between treatment groups. The results showed that the fraction containing alkaloids of hibiscus flower with a concentration of 0,4 % has not shown mucolytic activity, whereas at a concentration of 0,6 % and 0,8 % has been demonstrated in vitro mucolytic activity. Fractions containing alkaloids of hibiscus flower with a concentration of 0,6 % and 0,8 % have mucolytic activity equivalent to 0,1 % acetylcysteine in vitro.

Abstract

Infection of Malassezia furfur on skin can be caused by bad factor. One of the materials that can be used as an antifungal is Lengkuas rhizome extract (Alpinia galanga L.). This study had purpose to determine the antifungal activity and the effectiveness of gels Lengkuas rhizome extract on pathogenic fungi using maseration method with etanol 96%. Gel making is used HPMC and carbopol base. Testing activity of extract and testing the effectiveness of the gel by disc diffusion method (Kirby-Bauer test). Screening result  showed that the extract contains triterpenoids, flavonoids and essential oil. Based on the test results against Malassezia furfur, antifungal activity of the extract in the gel increased compared to extracts without formulated into a gel. But the activity increasing was not significant based on statistical analysis with one-way ANOVA test obtained a significance of 0.234 (p> 0.05).

Abstract

Paracetamol is widely used as analgesic and antipyretic agent in treatment of pain and fever. Paracetamol has interaction with carbohydrates and alcohol. Durian is the fruit native of Indonesia, that contain carbohydrates and alcohol. The aims of this research were to study the influence of durian fruit juice to the absorption and elimination kinetics of paracetamol and to know the dose of durian fruit juice that influence the absorption and elimination kinetics of paracetamol. The study was conducted using 16 rats, divided into 4 groups (n=4 per group). Each group was treated the following treatment : control paracetamol (paracetamol 9 mg/200 gBW), dose 1 group (paracetamol 9 mg/200 gBW and 0.675 g/200 gBW of durian fruit juice), dose 2 group (paracetamol 9 mg/200 gBW and 1.350 g/200 gBW of durian fruit juice) and dose 3 group (paracetamol 9 mg/200 gBW and 2.700 g/200 gBW of durian fruit juice). Blood sampling is done from the vein of rat’s tail at minutes 10, 20, 30, 40, 60, 90, 120, 180, 240, 300 and 360. The quantitation of paracetamol in plasma was determined by UV spectrophotometer at 243 nm. Result showed that durian fruit juice changed the absorption kinetics of paracetamol, durian fruit juice decreased Ka,  Cpmaks  and increased  Tmaks  parameter of paracetamol. Durian fruit juice also changed the elimination kinetics of paracetamol, its decreased Vd, Ke, Cl and increased the value of AUC and T1/2 paracetamol. All doses of durian fruit juice can influence the absorption and elimination kinetics of paracetamol includes Ka, Cpmaks, Tmaks, Vd, Cl, Ke, T1/2 and AUC parameters on wistar male rats (Rattus norvegicus L.).
