Abstract

Parasetamol merupakan obat analgetik-antipiretik yang dikonsumsi secara luas oleh masyarakat. Parasetamol dapat berinteraksi dengan karbohidrat dan alkohol. Buah durian merupakan buah asli Indonesia yang mengandung karbohidrat dan alkohol. Tujuan penelitian ini adalah untuk mengetahui bagaimana pengaruh jus buah durian terhadap kinetika absorpsi dan eliminasi parasetamol serta mengetahui dosis jus buah durian yang dapat mempengaruhi kinetika absorpsi dan eliminasi parasetamol. Uji dilakukan dengan membagi 16 ekor tikus dalam 4 kelompok (tiap kelompok 4 ekor). Tiap kelompok diberi perlakuan sebagai berikut: kontrol parasetamol (Parasetamol 9 mg/200 gBB), kelompok dosis 1 (Parasetamol 9 mg/200 gBB dan jus buah durian 0,675 g/200 gBB), dosis 2 (Parasetamol 9 mg/200 gBB dan jus buah durian 1,350 g/200 gBB) dan dosis 3 (Parasetamol 9 mg/200 gBB dan jus buah durian 2,700 g/200 gBB). Pengambilan cuplikan darah dilakukan dari vena ekor tikus pada menit ke- 10, 20, 30, 40, 60, 90, 120, 180, 240, 300 dan 360. Kadar parasetamol dalam plasma diukur menggunakan spektrofotometer UV pada panjang gelombang 243 nm. Hasil penelitian menunjukkan bahwa jus buah durian dapat mempengaruhi kinetika absorbsi parasetamol dengan menurunkan nilai Ka dan Cpmaks serta meningkatkan Tmaks parasetamol. Sedangkan kinetika eliminasi parasetamol yang dipengaruhi yaitu menurunkan nilai Vd, Cl, Ke, sehingga meningkatkan nilai AUC dan T1/2 parasetamol. Dosis 1, 2 dan 3 jus buah durian dapat mempengaruhi kinetika absorpsi dan eliminasi parasetamol yang meliputi parameter Ka, Cpmaks, Tmaks, Vd, Cl, Ke,  T1/2 dan AUC pada tikus putih (Rattus norvegicus L.) jantan galur wistar.

Abstract

Prevention of diseases caused by bacteria can be carried out with the use of antibacterial soap. The use of antibacterial soaps derived from natural one is to plant a red ginger (Zingiber officinale Rosc. var rubrum) which proved to have antibacterial activity. Therefore, preparations to make it more acceptable liquid soap containing antibacterial compounds 6-shogaol, gingerol, and zingiberen from red ginger plant. Simplex Lattice Design is a method which to find a good formula of dosage from. The study aims to determine the optimum formulation liquid soap ethanolic extract of red ginger rhizome using the method of Simplex Lattice Design. Simplex Lattice Design Methods used to optimize the formulation of liquid soap with five formulas with variations in the amount of olive oil and potassium hydroxide (KOH), FI (0%:100%), FII (25%:75%), FIII (50%:50%), FIV (75%:25%), FV (100%:0%). Evaluation of liquid soap ethanol extract of red ginger rhizome (Zingiber officinale Rosc. var rubrum) include an examination viscosity, pH, and free alkali. Physical stability was analized to find higher total response value (R total) as parameter of the optimum formula by Simplex Lattice Design Methods with Design Expert software  version 8.0.7.1. Optimum formula liquid soap ethanolic extract of red ginger rhizome with the proportion of variation base olive oil 98,1%(27,167 g) and potassium hydroxide (KOH) 1,9%(3,99 g). Data obtained physical characteristics test compared with prediction value using Design Expert software version 8.0.7.1. The result showed that physical properties of liquid soap formula optimum didn’t different significantly for easy response.

Abstract

Paracetamol is widely used as analgesic and antipyretic agent in treatment of pain and fever. Paracetamol has interaction with carbohydrates and alcohol. Durian is the fruit native of Indonesia, that contain carbohydrates and alcohol. The aims of this research were to study the influence of durian fruit juice to the absorption and elimination kinetics of paracetamol and to know the dose of durian fruit juice that influence the absorption and elimination kinetics of paracetamol. The study was conducted using 16 rats, divided into 4 groups (n=4 per group). Each group was treated the following treatment : control paracetamol (paracetamol 9 mg/200 gBW), dose 1 group (paracetamol 9 mg/200 gBW and 0.675 g/200 gBW of durian fruit juice), dose 2 group (paracetamol 9 mg/200 gBW and 1.350 g/200 gBW of durian fruit juice) and dose 3 group (paracetamol 9 mg/200 gBW and 2.700 g/200 gBW of durian fruit juice). Blood sampling is done from the vein of rat’s tail at minutes 10, 20, 30, 40, 60, 90, 120, 180, 240, 300 and 360. The quantitation of paracetamol in plasma was determined by UV spectrophotometer at 243 nm. Result showed that durian fruit juice changed the absorption kinetics of paracetamol, durian fruit juice decreased Ka,  Cpmaks  and increased  Tmaks  parameter of paracetamol. Durian fruit juice also changed the elimination kinetics of paracetamol, its decreased Vd, Ke, Cl and increased the value of AUC and T1/2 paracetamol. All doses of durian fruit juice can influence the absorption and elimination kinetics of paracetamol includes Ka, Cpmaks, Tmaks, Vd, Cl, Ke, T1/2 and AUC parameters on wistar male rats (Rattus norvegicus L.).

Abstract

Pendahuluan: Metronidazol merupakan suatu antibiotik yang biasa digunakan dalam mengobati infeksi Helicobacter pylori. Metronidazol mempunyai kelarutan yang tinggi dalam asam.  Untuk mengoptimalkan kelarutan tersebut maka dibuatlah hidrogel mukoadhesif yang dapat mempertahankan waktu tinggal di lambung lebih lama sehingga terapi infeksi H.pylori lebih optimal. Tujuan: untuk mendapatkan formula optimum dari hidrogel dengan kombinasi polimer kitosan dan natrium karboksimetilselulosa yang dapat mengontrol pelepasan metronidazol secara in vitro. Metode: Metode yang digunakan untuk memprediksi formula optimum menggunakan desain faktorial. Rancangan formula awal untuk memprediksi formula optimum terdiri dari 4 formula dengan perbandingan polimer kitosan dan natrium karboksimetilselulosa berturut-turut 0,5:1,5; 1,0:1,5; 0,5:3,0; dan 1,0:3,0. Hasil: Variasi konsentrasi dari Na-CMC dan kitosan dapat mempengaruhi pelepasan obat, yakni pada formula I dengan kinetika pelepsan obat mengikuti kinetika higuchi dan mekanisme pelepasan mengikuti mekanisme difusi fickian. Sedangkan, pada formula 4 memiliki sediaan fisik yaitu kekuatan mukoadhesif yang paling tinggi dibandingkan formula yang lain karena adanya peningkatan konsentrasi kitosan dan natrium karboksimetilselulosa (Na-CMC). Sehingga kombinasi antara kitosan dan Na-CMC dapat menjadi model pelepasan metronidazol dari matriks hidrogel yakni pada rentang 55,633% - 68,319% dalam waktu 240 menit. Hasil observasi menunjukkan formula optimum dengan perbandingan polimer kitosan dan natrium karboksimetilselulosa sebesar 1,0% (kitosan) dan 3,0% (natrium karboksimetilselulosa), dengan respon untuk kemampuan mengembang sebesar 96,95%; kekuatan mukoadhesif sebesar 0,1911 N/cm2; pelepasan obat sebesar  67,457%. Berdasarkan uji independent sampel t-test pada program SPSS 22 trial menjelaskan bahwa formula hasil observasi tidak berbeda siginifikan dengan hasil prediksi (p-value > 0,05). Kesimpulan: Hasil menunjukkan bahwa metode desain faktorial dapat digunakan untuk menentukan formula optimum dari hidrogel mukoadhesif metronidazol.

Abstract

Coronavirus Disease 2019 telah dinyatakan sebagai pandemik oleh WHO. Jumlah kasus konfirmasi positif ini diperkirakan masih akan terus bertambah sampai tahun 2021. Dalam penanggulangan penularan Covid-19 masyarakat menjadi garda terdepan, sehingga kader di desa atau kecamatan mempunyai peranan yang sangat penting untuk meningkatkan pengetahuan masyarakat tentang virus ini. Namun di tengah situasi pandemik ini, protokol kesehatan seperti menjaga jarak wajib dipatuhi sehingga dalam rangka menghindari kerumunan masyarakat, maka peranan teknologi seperti Whatsaap memberikan kontribusi yang baik sebagai media komunikasi edukasi masyarakat sehingga terhindar dari berita Hoax. Oleh karena itu kegiatan pengabdian kepada masyarakat (PKM) ini bertujuan mengedukasi kader di Kecamatan Jongkat, Kabupaten Mempawah tentang virus Covid-19 melalui media Whatsapp. Dalam pelaksanaan kegiatan PKM ini terdiri dari beberapa kegiatan diantaranya adalah perancangan metode edukasi; pembuatan media, seleksi kader, pemberian pre-test, peningkatan pengetahuan kader melalui group Whatsapp, post-test serta evaluasi. Berdasarkan hasil kegiatan ini, didapatkan terjadi peningkatan pemahaman kader setelah dilakukan edukasi melalui media Whatsapp dalam materi yang disampaikan berbentuk video, leaflet dan brosur, buku saku. Kader lebih menyukai materi edukasi berupa video yang disampaikan melalui media WhatsApp.

Abstract

Penerapan perilaku sehat dalam upaya peningkatan kualitas hidup selama pandemik penting dalam upaya menekan penyebaran Kasus Corona Virus Disease (Covid-19). Baru 50% masyarakat Indonesia yang berperilaku hidup bersih dan sehat dengan benar. Tujuan Kegiatan Pengabdian ini adalah mengukur penerapan perilaku sehat dalam upaya peningkatan kualitas hidup masyarakat di desa Bintang Mas 2 Kabupaten Kubu Raya. Kegiatan ini dilakukan dalam beberapa kegiatan berupa sosialisasi penggunaan masker, mencuci tangan dengan sabun, makanan sehat peningkat imun, cara menggosok gigi, praktik senam sehat, dan medical check-up. Program-program tersebut dilaksanakan dengan metode sosialisasi dan praktik langsung. Hasil yang diperoleh dari kegiatan yaitu peningkatan pengetahuan perilaku hidup sehat dalam upaya peningkatan kualitas hidup hingga 96%. Hasil medical check-up dari 63 partisipan yang diperiksa sebanyak 58,73% dengan tekanan darah >120/80 mg/dL dan 41,27% dengan tekanan darah

Abstract

Drug interaction happens when drugs is given together with foods. This research was aimed to observe the effect of mangosteen juice to the paracetamol pharmacokinetic profile in Wistar male white rats. The study used Wistar rats weight of 200-350g. The animals were divided into three groups with the total of the male rats are 15. Group I (paracetamol) was given a single oral paracetamol 9 mg/200gBW. Group II (treatment 1) was given a single oral paracetamol 9 mg/200gBW together with mangosteen juice 2,88 g/200gBW. Group III(treatment 2) was given a single oral paracetamol 9 mg/200gBW together with mangosteen juice 0,66 g/200gBW. The serial blood was collected from tail vein for 9 hours and was analyzed using spectrophotometer UV for unchanged paracetamol. The pharmacokinetic parameters of paracetamol were calculated by recidual method and were analyzed by One Way ANOVA using 95% confidence interval. The difference between groups were analyzed using Post Hoc LSD-test. The results showed that the group II(paracetamol-mangosteen 1) and group III(paracetamol-mangosteen 2) increased of ka, Cpmaks, tmaks, t½ab, ke, t½el, CL, AUC and decreased of ka, ke, and CL. The research showed that mangosteen juice doses 2 give higher decreased metabolism and decreased elimination of paracetamol in rat.

Abstract

Onchidiid slug or Onchidium typhae is a marine animal commonly found in West Kalimantan waters. This gastropod is commonly used as medicine and cosmetic by the locals. This research aims to assure the content and quality of O. typhae methanol extract by determining its secondary metabolites and heavy metal contamination. Phytochemical screening of alkaloid, steroid/ triterpenoid, flavonoid, tannin, and saponin are done using reagent tests. Chromatography profile was completed using silica gel G60 F254 plate and hexane: ethyl acetate: methanol as mobile phase. Heavy metal contamination analysis was performed using atomic absorption spectrophotometry to determine Hg, As, Cd, and Pb content. The phytochemical screening and chromatography profile of O. typhae methanol extract confirm the presence of alkaloids and steroids. Heavy metal analysis stated Hg, As, Cd, and Pb content in O. typhae are < 0,0008; < 0,001; < 0,001; and 0,05 ppm respectively. Since heavy metal levels are below the maximum contamination limit, it qualifies as a natural product ingredient.

Abstract

The impact of the Covid-19 pandemic is felt by the people of Pontianak, in the economic, sociocultural, and health fields. One of the prime commodities in Pontianak is corn farming. The aim of the PKM Program is to innovate corn-based processed products. This PKM activity began with outreach to the heads and employees of the Puskesmas, as well as identifying health cadres as potential participants in training related to using corn silk as herbal tea drinks. Training on using corn silk as herbal tea drinks for cadres was carried out offline and interactively using leaflets, videos, and hands-on practice. Evaluation and monitoring activities are carried out using a Google form (pre-test and post-test). The results of the PKM activity showed success in understanding the cadres with good knowledge when the pre-test was carried out by only ten people (30.30%). After being given knowledge training, respondents in the good category amounted to 30 people (90.90%). PKM activities targeting UPT Puskesmas Perum II cadres increased the ability of cadres to utilize corn silk waste in herbal tea. The next activity is expected to be able to assist so that the resulting product obtains a permit.

Abstract

Alpha (a) and beta (β) asarone were identified as the main compounds of red Jeringau (Acorus calamus L.) and had antimicrobial properties. This study aimed to know these two compounds' antibacterial mechanism and toxicity prediction against the PBP 2 protein and 50S Ribosomal Protein of Shigella flexneri. Molecular docking protocol using PyRx device was performed with Exhaustiveness value= 106, grid x=38.738375, y=112.645792, z=46.926417 for PBP2, and grid x=71.721251, y=47.551601, z=9.663173 for 50S Ribosomal Protein. The molecular docking results on the α -Asarone compound obtained an affinity value of -5.7 kcal/mol for PBP2 and an affinity value of -5.6 kcal/mol for 50S Ribosomal Protein. In comparison, β-Asarone had an affinity value of -5.6 kcal/mol to PBP2 and an affinity value of -5.7 kcal/mol for 50S Ribosomal Protein. The α and β-Asarone affinity had better values than the control. Molecular docking of α and β-Asarone compounds results in ionic bonds to the TYR529 amino acid and polar bonds to the ASN552 amino acid of PBP2. However, only β-Asarone produces ionic bonds at the amino acid ILE17 and polar bonds at GLU13 from 50S Ribosomal Protein. Based on this study, the α and β-Asarone compounds were shown to have antibacterial activity by interfering with the permeability of the bacterial cell wall. Both compounds are also predicted to have carcinogenic and mutagen effects.
