Abstract

Phytoestrogens have some important biological effects and could be employed as medicinal resources, for example as cancer chemopreventive agents. Phytoestrogen is defined as a phytochemical that has estrogenic or anti estrogenic effects. However, there is no database providing comprehensive list of phytoestrogen structures. In computer-aided drug discovery, the database is required to perform virtual screening for retrospective validation and structure-based drug design. The research presented in this article attempted to collect a comprehensive list of phytoestrogen structures. Subsequently, the structures were prepared in their three dimensional structures using SPORES1.3 for molecular docking simulations using PLANTS1.2. The ready-to-dock structures were then stored online as phytoestrogens.zip and could be downloaded from http://molmod.org/phytoestrogens.zip. This database contains 30 ready-to-dock unique phytoestrogens with total of 53 different configurations.

Abstract

Piper betle leaf in extract form is more effective than crude drug. Eugenol is a component in the extract that has antibacterial activity but irritate. Patch of piper betle leaf extract was used on the mucosa to make oral cavity hygiene. Antibacterial activity was influenced by the release of eugenol from the patch. Release enhancer substances (RES) such as glycerin, propylen glicol and tween 80 were added in patch formulation to increase the release of active substances. The aim of the research was to investigate the physicochemical properties, eugenol release profiles, and local tolerance test of the patch. Extract of piper betle leaf was made using infundation method. Patch was made according to the variation concentration of extract (1, 2 and 4%) and RES (glycerine, propylen glycol and tween 80) using chitosan as vehicle. Patch produced solvent casting method. Patch obtained was tested for swelling index, folding endurance, surface pH, antibacterial activity, release of eugenol, and local tolerance. Data obtained were analyzed descriptively. The result showed that the addition of RES did not affect the surface pH but increase the water absorption with in inconsistent way except patch with tween 80. The flexibility (folding endurance value) increased, and the highest amount of eugenol released was achieved by patch using propylen glicol. Patch with tween 80 and glycerin for all extract concentration and patch with 1% extract concentration using propylen glycol showed medium sensation (local tolerance), and patch with 2 and 4% extract using propylen glycol showed severe sensation.

Abstract

Estrogen deficiency causes various health problems in postmenopausal women, including osteoporosis. Phytoestrogen emerged as a potential alternative of estrogen with minimum side effects. Green clover (Marsilea crenata Presl.) is a typical plant in East Java which suspected contains estrogen-like substances. The aim of this research was to report the phytochemical properties of M. crenata using GC-MS as a preliminary study. M. crenata leaves were dried and extracted with n-hexane, then separated using vacuum column chromatography to get four fractions, after that the n-hexane extract and four fractions were identified with GC-MS. The results of GC-MS analysis showed some compounds contained in M. crenata leaves like monoterpenoid, diterpenoid, fatty acid compounds, and other unknown compounds. The results obtained in this research indicated a promising potential of M. crenata as medicinal plants, especially as antiosteoporotic agent.

Abstract

Ficus carica L and Ficus parietalis Bl. (Moraceae) are closely related plants which also known in Indonesia as Figs L. Considering the wide therapeutic value of Figs, this research was aimed to evaluate the DPPH-radical scavenging activity of both plants as well as their total phenolic and flavonoids. Extracts were produced by using boiled water and diluted to gain the desired concentration. Analyses were performed by using UV-vis spectrophotometer. Radical scavenging activity testing was done by using radical of DPPH (2,2-diphenyl-1-pycrylhydrazyl) to determine the IC50s. The determination of total phenolic was conducted by using Folin-Ciocalteau method and calculated as Gallic Acid Equivalence (GAE). The total flavonoid was measured by using AlCl3-reagents, and calculated as Rutin Equivalence (RE). Afterwards, the radical scavenging activity was correlated to the total phenolic and flavonoids contents. The results showed that the water soluble extract of F. carica fruit had the best IC50 value of 33.38 mg/mL, followed successively by the F. parietalis fruit (35.69 mg/mL), F. parietalis leaves (44.01 mg/mL) and the F. carica leaves (76.38 mg/mL). The highest content of total phenolic was shown by the leaves of F. parietalis (1.46% w/w GAE) and the lowest was in the fruit or F. carica (0.36% w/w GAE). The highest flavonoid content was detected in the leaves of F. carica (1.42% w/w RE) and the lowest was in the F. parietalis fruit (0.20% w/w RE). Correlation analyses of the IC50 values vs. the total phenolic and the flavonoids contents resulted in a positive slope having R2 values as 0.5362 and 0.9895, respectively. As a conclusion, the total flavonoid content influenced the DPPH radical scavenging activity by 98.95%, while the total phenolic content influence was only 53.62%.

Abstract

The objective of this research was to evaluate the anthelmintic activity of ethanolic extract of nuts Veitchia merrillii against intestinal nematode, Ascaridia galli. Phytochemical tests were performed for testing different chemicals group present in the extract. The effect of these extract were determined by in vitro based on inhibition of motility and mortality of worms. Amount of four worms were exposed in triplicate to each of phosphate buffered saline, 25 mg/mL, and 75 mg/mL crude ethanolic extract of V. merrillii, and 15 mg/mL albendazole. The motility of worm was observed on 9, 18, 27, and 36 hours interval. The mortality of worms were recognized by their straight flat appearance with no movements at the head and tail regions of the body. We found that the ethanolic extract of V. merrillii contains tannins, alkaloids, flavonoids, triterpenoids, and saponins neither steroid. Based on the in vitro trials conducted using above extract at 25 and 75 mg/mL concentration, the extract of V. merrillii showed anthelmintic effect. The extract of V. merrillii was effective at 75 mg/mL concentration only. The study indicated that it is potential to develop herbal-based anthelmintic to control A. galli in poutry.

Abstract

Pomegranate seed oil has antioxidant, anti-inflammatory, and chemo preventive activities. Pomegranate seed oil is lipophilic substance suitable to be prepared in emulsion dosage forms. Long-chain triglyceride (LCT) and medium-chain triglyceride (MCT) are commonly used as oil phase in emulsion dosage forms. This research aimed to compare the use of LCT and MCT in the Nano emulsion formula of pomegranate seed oil dosage forms. Formulation of pomegranate seed oil Nano emulsion was conducted using high energy emulsification. Parameters observed were pH, Nano emulsion type, percent transmittance, viscosity, turbidity, and droplet size before and after 3 cycles of freeze-thaw. The result showed that there was no significant difference between physical properties of pomegranate oil Nano emulsion with LCT as oil phase and pomegranate oil Nano emulsion with MCT as oil phase. Moreover, physical stability of pomegranate oil Nano emulsion with LCT as oil phase was better than pomegranate oil Nano emulsion with MCT as oil phase.

Abstract

Antidiabetic activities of Phaleria macrocarpa fruits and Annona muricata leaves through α-glucosidase inhibition have been reported. Each extract of the two medicinal plants showed weak α-glucosidase inhibitory activities. The effect of synergies is expected from combining the two extracts. The purpose of this research was to investigate α-glucosidase inhibitory activity of the combination of ethanolic extract of P. macrocarpa fruits and A. muricata leaves and to confirm the presence of different chemical constituents of the most active individual and extracts combination by thin layer chromatography (TLC). Extracts were obtained by maceration method using 96% ethanol. Various concentration of extracts combination was analyzed based on IC50 of the enzyme inhibition of each individual extract. The result showed that α-glucosidase inhibitory activity of each extracts was concentration dependent, with IC50 values of individual extracts of P. macrocarpa fruits and A. muricata leaves were 261.34 and 428.79 μg/mL, respectively. However, the extracts combination with the highest inhibition activity (84.51±0.79%) was obtained at its IC50 of P. macrocarpa fruits and two-third IC50 concentration of A. muricata leaves. The TLC profile of the most active extract combination showed a phytochemical constituent that is also detected in the profile of each individual extract. This can be assumed as a synergistic effect due to increased phytochemical contents of the combined extract.

Abstract

Pain is a sensory and emotional feelings associated with tissue damage. Leaves kerehau recognized by people Dayak and Kutai as postpartum pain medication. Kerehau leaf has long been known by the public because many have efficacy in medicine. This research to determine the analgesic activity of ethanolic extract of leaves kerehau (Callicarpa longifolia Lamk.) on male white mice (Mus musculus). This is experimental research, randomized complete unidirectional pattern. The animals test were divided into 5 groups with each treatment of 5 mice. Group I (Na CMC 0.5% negative control), Group II (ibuprofen as a positive control), group III - V the ethanol extract of leaves kerehau dose of 150 mg , 300 mg and 600 mg/ Kg BW). Inductor pain acetic acid 0.5 % (ip). Observed number of writhing in mice for 1 hour and calculated percent analgesic . The results obtained in this study has efficacy as an analgesic with Strength of power dose I: 51.65 %, dose II: 65.40 %, and dose III: 75.98 %.
