Abstract

The objective of this research was to evaluate the anthelmintic activity of ethanolic extract of nuts Veitchia merrillii against intestinal nematode, Ascaridia galli. Phytochemical tests were performed for testing different chemicals group present in the extract. The effect of these extract were determined by in vitro based on inhibition of motility and mortality of worms. Amount of four worms were exposed in triplicate to each of phosphate buffered saline, 25 mg/mL, and 75 mg/mL crude ethanolic extract of V. merrillii, and 15 mg/mL albendazole. The motility of worm was observed on 9, 18, 27, and 36 hours interval. The mortality of worms were recognized by their straight flat appearance with no movements at the head and tail regions of the body. We found that the ethanolic extract of V. merrillii contains tannins, alkaloids, flavonoids, triterpenoids, and saponins neither steroid. Based on the in vitro trials conducted using above extract at 25 and 75 mg/mL concentration, the extract of V. merrillii showed anthelmintic effect. The extract of V. merrillii was effective at 75 mg/mL concentration only. The study indicated that it is potential to develop herbal-based anthelmintic to control A. galli in poutry.

Abstract

The aim of the study was to observe the histopathological changes of the entire egg cells in the uterine of Ascaridia galli worms treated with crude ethanolic extract of Veitchia merrillii nuts. Histopathological changes of the entire egg cells were observed to describe that the activity of extract V. merrillii nuts interfered the reproduction process could decrease the quantity and quality of A. galli eggs. The experiments consisted of 16 head A. galli female adult worms divided into four groups, contained four worms of each, and maintained in 0.9% phosphate buffered saline (PBS). First group, a negative control experiment consisted of A. galli in 0.9% PBS. Second group, concentration of 15 mg/mL albendazole was used as a positive control. Third group and fourth group, in vitro exposure of the worms to the crude ethanolic extract V. merrillii nuts at a concentration of 25 mg/mL and 75 mg/mL, respectively. The histopathological changes of egg cells in the uterine region of stained worms was evaluated by light microscopic examination at 40x magnification. The results showed that shrinkage and disintegration of the entire egg cells in the breakage of uterus. The results indicate that the possible use of the plant as a potential anthelmintic against A. galli, the intestinal nematode parasite of domestic fowl.

Abstract

ABSTRAKSumber pencemaran Escherichia coli pada produk pangan salah satunya yang penting yaitu diakibatkan oleh kontaminasi tangan. Penelitian ini bertujuan untuk mengetahui cemaran dari tangan pedagang dan angka prevalensi cemaran bakteri Escherichia coli pada tangan pedagang daging ayam broiler di dua pasar tradisional Kota Banda Aceh dengan metode Replicate Organism Direct Agar Contact (RODAC).Sampel yang digunakanadalah 20 tangan pedagang daging ayam broiler. Tangan langsung ditempelkan pada media Eosin Methylen Blue Agar (EMBA) dan selanjutnya diinkubasi dengan suhu 37˚C selama 24 jam. Jumlah koloni bakteri yang tumbuh dihitung untuk mengetahui angkap revalensi. Dari 20 sampel tangan pedagang yang diperiksa, adanya cemaran Escherichia coli pada 9 sampel tangan pedagang. Dapat disimpulkan bahwa tangan pedagang daging ayam broiler tercemar Escherichia coli dengan angka prevalensi keseluruhan 45%.Kata Kunci : Escherichia coli, cemaran, tangan pedagang ABSTRACTOne important source of Escherichia coli pollution in food products is caused by hand contamination. The purpose of this study to determine the contamination from the hands of traders and the prevalence rate of Escherichia coli contamination in the hands of broiler meat traders in two traditional markets in Banda Aceh City with the Replicate Organism Direct Agar Contact (RODAC) method. The sample used was 20 hands of broiler meat traders. The hands were placed on Eosin Methylen Blue Agar (EMBA) media and then incubated at temperature 37˚C for 24 hours. The number of bacterial colonies that grow is calculated to determine the prevalence rate. From the 20 samples of traders hands examined, there were Escherichia coli contaminations in 9 samples of traders hands. It can be concluded that the hands of broiler chicken meat traders are contaminated Escherichia coli with an overall prevalence rate of 45%.Keywords: Escherichia coli, contamination, traders hand

Abstract

Penelitian ini bertujuan untuk mengetahui jumlah total plate count (TPC) dan kontaminasi Escherichia coli pada susu kambing segar yang berasal dari usaha peternakan kambing perah di kecamatan Syiah Kuala, Banda Aceh. Sampel susu yang digunakan adalah susu kambing segar  hasil pemerahan pada pagi hari yang diperoleh dari 2 usaha ternak kambing perah di kecamatan Syiah Kuala, Banda Aceh dengan 2 kali pengulangan masing-masing sebanyak 250 ml. Selanjutnya, sesegera mungkin dibawa kelaboratorium untuk dilakukan pengujian guna mengetahui adanya bakteri dalam air susu tersebut. Pengujian jumlah cemaran TPC dan E.coli dilakukan dengan metode tuang. Berdasarkan hasil penelitian ternyata jumlah total bakteri pada peternakan A adalah 4,9x102cfu/ml dan dari peternakan B adalah 9,2x103 cfu/ml. Hasil penelitian E.coli di usaha ternak kambing perah A  adalah 0 dan hasil penelitian E.coli usaha ternak kambing perah B adalah 1x101 cfu/ml. Kesimpulannya adalah jumlah TPC pada uasaha ternak kambing perah KecamatanSyiah Kuala tidak melebihi standar SNI sedangkan cemaran E.coli pada usaha ternak kambing perah A adalah 0 sedangkan pada usaha ternak kambing perah B adalah melebihi SNI. study aims to determine the total plate count (TPC) number and escherichia coli contamination on fresh goat milk from a dairy goat farm in Syiah Kuala district Banda Aceh. The milk samples used is the fresh goat milk  from morning milking from 2 dairy goat farm in Syiah Kuala district Banda Aceh  with two repeatation each repetition as much as 250 ml. The milk sample were taken to the laboratory as soon as possible to test the presence of bacteria in the milk. Total plate count (TPC) and E. coli test were done using pour methode.  Based on the results of the study showed that the total number of bacteria farm A  were 4, 9x102 cfu/ml and farm B were 9, 2x103 cfu/ml. The E.  coli result in farm A were 0 and in farm B were 1x101 cfu/ml. The conclusion is that the number of TPC in dairy goat farm in Syiah Kuala district banda Aceh do not exceed the SNI level while the E. coli contamination in farm A were 0 and farm B exceed the SNI level.

Abstract

ABSTRAKPenelitian bertujuan mengetahui keberadaan dan tingkat pencemaran Staphylococcus aureus pada daging ayam yang dijual di pasar tradisional Ulee Kareng. Penelitian ini menggunakan 8 sampel daging pada bagian dada ayam yang diperoleh dari 8 pedagang daging ayam di pasar tradisional Ulee Kareng. 8 sampel dibagi menjadi 2 bagian, 4 sampel diperoleh pada pagi hari dan 4 sampel berikutnya diperoleh pada siang hari. Pengkoleksian sampel dilakukan selama 2 hari berturut-turut. Seluruh sampel yang telah diperoleh diidentifikasi untuk mengetahui keberadaan bakteri S. aureus dengan cara dibiakan dalam media spesifik Mannitol Salt Agar (MSA) dan dilakukan pemeriksaan tingkat cemaran bakteri menggunakan metode Total Plate Count (TPC). Hasil penghitungan bakteri S. aureus pada daging ayam di hari pertama menunjukkan angka dengan rata-rata 15,2x104 cfu/g (pagi hari) dan 13,9x104 cfu/g (siang hari), sedangkan pada hari kedua menunjukkan tingkat cemaran dengan rata-rata 17,7x104 cfu/g (pagi hari) dan 7,1x104 cfu/g (siang hari). Dapat disimpulkan bahwa tingkat cemaran bakteri S. aureuspada daging ayam yang dijual di pasar tradisional Ulee Kareng digolongkan tidak higienis dikarenakan melebihi nilai standar SNI-2897-2008. ABSTRACT This study aims to know the existence and level of Staphylococcus aureus contamination in chicken meat sold in Ulee Kareng traditional market. This study used 8 samples of chicken breast meat obtained from 8 chicken meat traders in  Ulee Kareng traditional markets. 8 samples were divided into 2 parts, 4 samples were obtained in the morning and 4 subsequent samples were obtained during the afternoon. The sample collecting is done for 2 consecutive days. All samples obtained were identified to determine the presence of S. aureus bacteria by culturing in a specific medium of Mannitol Salt Agar (MSA) and the level of bacterial contamination were counted using Total Plate Count (TPC) method. The results of TPC calculation of S. aureus bacteria on chicken meat on the first day showed a number with an average of 15.2x104 cfu/g (morning) and 13.9x104 cfu/g (afternoon). Followed by TPC counting on the second day showed a number with an average of 17.7x104 cfu/g (morning) and 7.1x104 cfu/g (daytime). Can be concluded that the level contamination of S. aureus bacteria in chicken meat sold by traders in the Ulee Kareng traditional market exceed the SNI-2897-2008 standard value.

Abstract

ABSTRAKEscherichia coli dikenal sebagai bakteri indikator sanitasi, keberadaanya dalam pangan menunjukkan bahwa pangan tercemar oleh lingkungan. Penelitian ini bertujuan mengetahui angka prevalensi bakteri E. coli pada meja dan peralatan pedagang daging ayam broiler. Sampel yang digunakan yaitu 20 swab meja, pisau, dan talenan pedagang daging ayam broiler di dua Pasar Tradisional Kota Banda Aceh. Penelitian ini menggunakan metode Total Plate Count (TPC) dengan larutan Buffer Peptone Water (BPW) dan mengidentifikasi E. coli menggunakan media selektif diferensial, yaitu dengan Eosin Methylene Blue Agar (EMBA). Hasil penelitian menunjukkan bahwa 2 meja, 2 pisau, dan 3 talenan di Pasar Tradisional Seutui positif tercemar E. coli dengan angka prevalensi cemaran E. coli pada meja (40%), pisau (40%), dan talenan (60%), sedangkan di Pasar Tradisional Peunayong terdapat 2 meja dan 2 pisau positif tercemar E. coli dengan angka prevalensi cemaran  E. coli pada meja (13%) dan pisau (13%). Hasil kesimpulan yang didapat yaitu rata-rata angka prevalensi cemaran E. coli secara keseluruhan pada peralatan pedagang dari dua Pasar Tradisional di Kota Banda Aceh yaitu 18,33%.Kata kunci : Escherichia coli, prevalensi, peralatan pedagang, pasar tradisional  ABSTRACTEscherichia coli is known as an indicator of sanitation bacteria, its presence in food indicates that food is polluted by the environment. This study aims to determine the prevalence of  E. coli bacteria on the table and equipment of broiler chicken meat traders. The samples used were 20 table swabs, knives, and cutting boards of broiler traders in two Traditional Markets in Banda Aceh City. This study uses the Total Plate Count (TPC) method with Buffered Peptone Water (BPW) solution and identifies E. coli using differential selective media, namely Eosin Methylene Blue Agar (EMBA). The results showed that 2 tables, 2 knives, and 3 cutting boards in the Seutui Traditional Market were positively contaminated with E. coli with the prevalence of E. coli contamination on the table (40%), knives (40%), and cutting boards (60%), while in the Peunayong Tradisional Market there are 2 tables and 2 positive knives contaminated with E. coli with the prevalence rate of E. coli contamination on the table (13%) and knives (13%). The conclusion obtained is the average prevalence rate of E. coli contamination as a whole on the equipment of traders from two Traditional Markets in the City of Banda Aceh is 18.33%. Key words : Escherichia coli, prevalence, equipment traders, traditional markets

Abstract

Penelitian ini bertujuan mengetahui jumlah cemaran mikrob pada telur asin mentah yang dijual di pasar Ulee Kareng. Penelitian ini menggunakan 20 butir telur asin yang diperoleh dari 10 pedagang. Jumlah cemaran mikrob dilakukan dengan pour plate method (Total Plate Count) dengan pengenceran berseri 10-1–10-4untuk isi telur. Data hasil penelitian ini disajikan secara deskriptif. Jumlah cemaran mikrob terkecil terdapat pada 3 dari 20 telur asin yang dijual oleh 10 pedagang yaitu berkisar dari 101 – 9,8×104 CFU/g dan jumlah cemaran mikrob tertinggi pada pedagang 5, 9 dan 10 yaitu 1,8×105 CFU/g,  1,1×106 CFU/g,  dan 1,2×105 CFU/g. Berdasarkan hasil penelitian dapat disimpulkan bahwa terdapat 15% telur asin mentah yang isi telur tercemar mikrob melebihi batas SNI ( 1×105 CFU/g).(This study was aimed to determine the amount of microbial contamination in raw salted eggs sold at Ulee Kareng market. This study used 20 salted eggs obtained from 10 traders. The amount of microbial contamination was not by pour plate method (Total Plate Count) with 10-1-10-4 dilution for egg content. The data of this research were presented descriptively. The smallest amount of microbial contamination was found in 3 of 20 salted eggs sold by 10 traders, ranging from 101-9.8 × 104 CFU / g and the highest number of microbial contaminant in traders 5, 9 and 10, namely 1.8×105 CFU /g, 1.1×106 CFU / g, and 1.2 × 105 CFU / g. Based on the results of the study it can be concluded that there were 15% of raw salted eggs which contain microbial contaminated egg exceeding the SNI limit (1×105CFU / g))

Abstract

Ikan kakap putih merupakan bahan pangan yang sangat dibutuhkan manusia karena mengandung asam amino esensial seperti lisin, metionin dan histidin. Penelitian ini bertujuan untuk melihat kadar lemak ikan kakap putih (Lates calcarifer) segar dan kukus. Penelitian ini menggunakan sampel ikan kakap segar yang dibagi menjadi 2 kelompok perlakuan yaitu segar dan dikukus. Uji Laboratorium yang dilakukan adalah menghitung jumlah kadar lemak menggunakan metode Soxhlet. Hasil penelitian dianalisa menggunakan independent sampel T tes, analisa statistik menunjukkan perbedaan nyata antara kadar lemak ikan kakap putih segar dengan ikan kakap putih kukus. Berdasarkan penelitian ini lemak yang terkandung dalam ikan kakap putih yang kukus adalah 1,27 ± 0,09 dan segar 1,78 ± 0,08. Kesimpulan dari penelitian ini adalah kadar lemak ikan kakap putih segar lebih tinggi daripada kadar lemak ikan kakap kukus.Kata kunci: Ikan kakap putih, kadar lemak, soxhlet

Abstract

Milkfish merupakan ikan yang mampu mentolelir perubahan sanilitas yang cukup luas atau biasa disebut dengan euryhalin. Penelitian ini bertujuan untuk mengidentifikasi endoparasit pada Ikan bandeng di TPI Pasar Peunayong Kota Banda Aceh dengan pengambilan sampel secara acak, sedangkan pemeriksaan sampel dilakukan di laboratorium FKH Unsyiah. Dari 30 sampel ikan bandeng yang digunakan terdapat 2 jenis endoparasit pada ikan tersebut yaitu Anisakis sp dan Radhinorhynchus laterospinosis. Terdapat 15 ekor (50%) sampel yang terinfeksi Anisakis sp dan 3 ekor sampel (10%) yang terinfeksi Rhadinorhynchus laterospinosis. Dapat disimpulkan bahwa ikan yang di ambil di TPI Pasar Peunayong Kota Banda Aceh terinfeksi endoparasit Anisakis sp dan Rhadinorhynchus laterosinosis terdapat pada usus dan eshopagus. Kata kunci: Endoparasit, Anisakis sp, Rhadinorhynchus laterosinosis.

Abstract

Many people favor quail eggs because they contain good nutrients, and are easy to obtain at an affordable price. This research was conducted to study the effect of temperature and storage time on the quality of quail eggs. The samples in this study were 66 quail eggs which were divided into two groups, the first group (33 eggs) was stored at 27oC, and the second group (33 eggs) was kept at 5oC. This study used a completely randomized design with two factors, temperature and storage time. The observation was carried out on days 1, 7, 13, 19, 25, and 31 with triplicates. The parameters used in this study were air sac height, egg yolk index, white egg index, and Haugh Unit (HU). This study showed that temperature and storage time affected the quality of quail eggs. Eggs stored at 27oC showed a quality decrease indicated by higher air sac position, and lower egg yolk index and egg white index. Eggs kept at 5oC showed no significant degradation. The HU for both groups ranged between 76-81 which implied that the eggs had AA quality even after 31 days of storage.
