Abstract

This review aimed to provide a scientific and rational basis for developing the Melastoma malabathricum plant into an herbal medicine capable of providing effective, safe therapy for long-term use in diseases originating from oxidative stress processes and associated with inflammatory events, including diabetes, cardiovascular disorders, and neurodegenerative diseases. Electronic searches of the Scopus and Web of Science (WOS) databases identified relevant articles. The literature search and screening were conducted on March 2, 2025. Keywords such as (“Melastoma malabathricum”) AND (antioxidant* OR “anti-inflammatory” OR “anti-inflammatory” AND anti-inflammation* OR anti-inflammation) were used to obtain articles published between 2006 and 2024. During the process, only original, English-language studies reporting M. malabathricum anti-inflammatory or antioxidant properties were included, while reviews, conference abstracts, preprints, retracted, irrelevant, and duplicate papers were excluded. A total of 39 articles fulfilled the inclusion criteria and were subjected to critical evaluation. The results showed that M. malabatricum plant extract demonstrated consistent anti-inflammatory activity. Pathways of mechanisms and safety profiles were reported across in vitro and in vivo assays. In conclusion, this narrative review of the studies provides comprehensive, integrated information from the scientific literature on the antioxidant, anti-inflammatory, and safety activities of M. malabathricum plant extracts. The results offer a strong scientific foundation for the rational development of the plant extracts as an anti-inflammatory drug candidate.

Abstract

Cucurbita maxima (C. maxima), exhibits significant morphological diversity and is cultivated widely for both culinary and medicinal purposes globally including anti-diabetic, antioxidant, anti-carcinogenic, and anti-inflammatory properties. However comprehensive pharmacological properties like antioxidant and cytotoxic potential of C. maxima seeds are limited. This study aimed to evaluate the phytochemical composition, antioxidant activity, and cytotoxicity of methanolic extracts from C. maxima seeds. Seeds were dried, powdered, and extracted with methanol for phytochemical screening. The extracts were subjected to in vitro assays, including H2O2 and DPPH scavenging assays for antioxidant activity, and brine shrimp lethality bioassay for cytotoxicity evaluation. IC50 values were determined to assess antioxidant and cytotoxic potency. Phytochemical analysis revealed the presence of alkaloids, flavonoids, saponins, tannins, carbohydrates, steroids, and proteins in the methanolic extract. Quantitative analysis showed 5.09 mg of GAE/g phenols, 2.48 mg of QAE/g flavonoids, and 0.035 mg of tannins. The IC50 values for H2O2 and DPPH scavenging assays were 793.72 µg/ml and 606.25 µg/ml, respectively, indicating moderate antioxidant activity compared to standards. The LC50 value from the brine shrimp assay was 2.52, indicating cytotoxic activity. The study suggests that C. maxima seed extracts contain pharmacologically active phytochemicals with antioxidant and cytotoxic properties. The findings support its potential as a natural source for developing therapeutic agents. These results underscore the importance of C. maxima in traditional medicine and its potential for pharmaceutical applications. Further research could focus on elucidating the mechanisms of action, toxicity profiles, and clinical efficacy to harness its therapeutic potential effectively.

Abstract

Oxidants and free radicals play a crucial role in the skin aging process, driving the exploration of natural ingredients for cosmetics. While Passiflora edulis (passion fruit) has been studied for its antioxidant properties, comparative research on the phenolic content and antioxidant activity of its leaf and seed ethanol extracts remains limited. This study aims to compare the phenolic compounds and antioxidant activities of ethanolic extracts from purple passion fruit leaves and seeds. Extracts were obtained by maceration using 96% ethanol. Phenolic and flavonoid content were analyzed qualitatively and quantitatively using Folin-Ciocalteu and colorimetric methods, respectively. Antioxidant activity was determined using the DPPH radical scavenging assay, with data analyzed descriptively and expressed as mean ± SD. Qualitative analysis confirmed the presence of phenolic and flavonoid compounds in both extracts. The leaf extract contained total phenolics and flavonoids of 42.97 ± 2.6 mg GAE/g and 58.49 ± 4.05 mg QE/g, respectively. The seed extract contained 40.46 ± 4.9 mg GAE/g total phenolics and a higher flavonoid content of 71.79 ± 4.21 mg QE/g. The IC50 values for the leaf and seed extracts were 101.82 ± 8.44 ppm and 63.36 ± 8.22 ppm, respectively, indicating strong antioxidant activity for the seed extract and moderate-to-strong activity for the leaf extract. The ethanolic extracts of purple passion fruit leaves and seeds exhibit significant antioxidant activity, with the seed extract being particularly potent. These findings highlight their potential as promising natural ingredients for anti-aging cosmetic formulations.

Abstract

Diabetes mellitus (DM) is conditionally defined by chronic high blood sugar, oxidative stress, and inflammation, with tumor necrosis factor (TNF) and peroxisome proliferator-activated receptor gamma (PPARγ) acting as critical therapeutic targets. This research investigated the phytochemical profile, antioxidant potential, and molecular pathways of Ruellia tuberosa L. leaf extract in relation to diabetes using a dual experimental and computational framework. The total phenolic content (TPC) and total flavonoid content (TFC) were quantified using Folin-Ciocalteu and aluminum chloride colorimetry assays, respectively, while antioxidant efficacy was assessed using the DPPH radical scavenging assay. Furthermore, a Network pharmacology approach incorporating target prediction, protein-protein interaction (PPI) analysis, and molecular docking was used to uncover multi-target interactions. Results showed a TPC of 34.14 mg GAE/g and a TFC of 4.949 ± 0.089 mg QE/g, with the extract demonstrating strong antioxidant activity (IC₅₀ = 83.96 ppm). Key bioactive compounds identified included β-sitosterol, stigmasterol, betulinic acid, apigenin, and luteolin. Computational docking revealed high binding affinity of β-sitosterol and stigmasterol toward TNF (-8.7 and -8.5 kcal/mol) and PPARγ (-10.2 and -9.8 kcal/mol). Network modeling identified 87 protein targets with TNF (0.79) and PPARγ (0.77) showing the highest centrality, emphasizing their significance in the extract's antidiabetic effects.  This study provides molecular insights into the multi-target therapeutic potential of R. tuberosa L. leaf extract for diabetes treatment, addressing both inflammation and metabolic dysregulation.
