Abstract

Patchouli mottle virus (PatMoV) is the most severe disease pathogen and causes substantial losses in many patchouli-producing regions in Indonesia. Serological detection tool  for the disease was developed in this  research. Virus isolation was conducted on Chenopodium amaraticolor resulted on  the homogenous local lesions 6 days after  inoculation. Virus  purification was obtained from 200g inoculated leaves resulted on 2 ml virus solution with the concentration of 1 mg/ml. Polyclonal antibodies were produced on rabbits. Harvested antiserum was used for further virus detection by Indirect-Enzyme linked Immunosorbent assay (I-ELISA) and dot-immunobanding assay (DIBA) techniques. The antibodies were  positively  reacted with purified  viruses, infected field collection of patchouli, and  inoculated C. amaranticolor. On the other hand un-inoculated C. amaranticolor samples and healthy patchouli generated from tissue cultures gave negative reaction with the antibodies. This is the first report of cheap practical antibody production for PatMoV detection in Indonesia.

Abstract

Pepper (Piper nigrum), known as the “King of Spices” is one of the most important spices. In the international market, Indonesian pepper has high selling value, due to its flavor characteristics. Pepper yellowing disease is one of the most important disease that caused the decrease of pepper production and become the main problem in the cultivation of pepper in West Kalimantan. This research was conducted to determine the major causal agent of leaf yellowing disease of pepper. The Fusarium associated with diseased plant were isolated from the symptomatic plant and nematodes were isolated from the root with leaf yellowing symptom. The Fusarium isolates were cultured on agar medium, and the nematode was cultured on tomato plant. From diseased pepper in West Kalimantan, it was isolated 4 Fusarium isolates and plant parasitic nematode Meloidogyne. The result showed that H isolate of Fusarium was the most virulent isolate and identified asFusarium solani. The Meloidogyne was identified by the female perenial patern.The nematode was identified as Meloidogyne incognita.INTISARILada (Piper nigrum L.) merupakan salah satu jenis rempah penting yang telah dikenal sebagai “King of Spices”. Di pasar internasional, lada Indonesia mempunyai daya jual tinggi karena cita rasanya yang khas. Salah satu kendala dalam budidaya lada adalah adanya penyakit kuning lada dan sampai saat ini menjadi masalah utama pada pertanaman lada di Kalimantan Barat. Informasi tentang patogen utama yang berinteraksi dengan penyakit kuning lada masih sangat terbatas, sehingga penelitian ini bertujuan untuk mengidentifikasi patogen utama yang berasosiasi dengan penyakit kuning lada. Isolasi Fusarium dilakukan dari batang lada dan isolasi nematoda dilakukan dari akar lada yang bergejala penyakit kuning di Kalimantan Barat. Fusarium hasil isolasi dikulturkan dalam medium agar, sedangkan nematoda hasil isolasi dikulturkan dalam akar tomat. Dari hasil isolasi berhasil didapatkan empat isolat Fusarium dan nematoda Meloidogyne. Identifikasi Fusarium dilakukan secara morfologis dan molekuler, dan identifikasi Meloidogyne dilakukan dengan menggunakan irisan bagian posterior nematoda betina. Dari hasil identifikasi diketahui bahwa patogen yang berasosiasi dengan penyakit kuning lada adalah Fusarium solani dan Meloidogyne incognita.

Abstract

Banana Bunchy Top Disease (BBTD) is the most economically important virus disease of bananas. The disease is widespread in Asia and South Pacific regions. In Indonesia, it was first reported in 1979 in the regency of Bandung in West Java, and now becoming widespread in several growing areas. A survey on disease incidence was conducted in five main banana growing districts in the provinces of West Java and Banten. BBTD was prevalent in four of five main banana growing districts with disease incidence varied from 0 to 38.6%. Most of the local cultivars grown was infected with various level of intensity. Cultivars Muli, Nangka, Raja, Kepok and Ambon were among the cultivars infected. ELISA confirmed that banana bunchy top virus is the causal pathogen and showed the relative susceptibility of the cultivars. Most of growers was not aware of the disease and tended to keep the infected banana. Some growers cut the infected pseudostem, but did not uproot the stem, allowing the disease to develop into the new plantings.

Abstract

Vorstenlanden tobacco is the best product of PT Perkebunan Nusantara X (PTPN X) Klaten, Central Java that is commonly produced into high economic value cigars. During the planting season of 2010/2011, there was an epidemy of leaf curl disease that caused billion rupiahs financial loss. Several efforts had been done, including the use of pesticides, but the result had not been satisfactory. Therefore, this research aimed to identify the pathogen of tobacco leaf curl disease and conduct integrated control system using three combinations in the nursery [A (biological agents treatment, plastic screenhouse, without physical barrier); B (biological agents treatment, plastic screenhouse + screen plot, with physical barrier of single screen, and sprayed insecticide); C (biological agents treatment, plastic screenhouse + single screen plot, with physical barrier of double screens, and fogging using white oil + insecticide)] and planting area [A (physical barrier of single screen and sprayed insecticide); B (physical barrier of single screen, sprayed insecticide, and sanitation of broad leaf weed + eradication of infected plant); C (physical barrier of double screens, fogging white oil + insecticide, and sanitation of broad leaf weed + eradication of infected plant)]. The result showed that leaf curl disease that occured at PTPN X Klaten was caused by Begomovirus, based on PCR result using Krusty and Hommer primers for Begomovirus Coat Protein gene. The most effective integrated disease control of leaf curl disease in the nursery was C combination, that consisted of biological agents treatment on the seedlings medium, screenhouse covered by plastic and double screens that was combined with physical barrier of double screens around the field, and fogging using white oil+insecticide of pyrethroid active ingredient. The most effective integrated disease control of leaf curl disease in the field was the same C combination, that consisted of the use of physical barrier of double screens, environmental sanitation of weeds around the field and eradication of infected plants and fogging using white oil + insecticide of pyretroid active agent.   IntisariTembakau vorstenlanden merupakan produk andalan PT Perkebunan Nusantara X (PTPN X) Klaten, Jawa Tengah sebagai bahan baku cerutu yang bernilai ekonomi tinggi. Pada musim tanam 2010/2011 telah terjadi epidemi penyakit keriting atau kerupuk tembakau dengan kerugian mencapai milyaran rupiah. Upaya pengendalian yag telah dilakukan dengan mengandalkan pestisida tidak memberikan hasil yang memuaskan. Oleh karena itu, penelitian ini ditujukan untuk mengidentifikasi patogen penyebab penyakit kerupuk tembakau dan melakukan pengendalian secara terpadu dengan menggunakan tiga macam kombinasi yang dilakukan di pembibitan [A (perlakuan agens hayati, penyungkupan plastik, tanpa barier fisik ); B (perlakuan agens hayati, penyungkupan, barier fisik tunggal, dan penyemprotan insektisida); C (perlakuan agens hayati, penyungkupan, barier fisik ganda, dan aplikasi white oil)] dan di lahan pertanaman [A (barier fisik tunggal dan insekstisida); B (barier fisik tunggal, insektisida, dan sanitasi gulma); C (barier fisik ganda, white oil + insektisida, dan sanitasi gulma)]. Hasil penelitian menunjukkan bahwa penyakit kerupuk tembakau di PTPN X Klaten disebabkan oleh Begomovirus berdasarkan hasil PCR menggunakan primer Krusty dan Hommer untuk gen Coat Protein Begomovirus. Pengendalian terpadu penyakit kerupuk di pembibitan yang paling efektif adalah kombinasi C yang terdiri dari perlakuan agens hayati pada media bibit, penyungkupan rangkap plastik + waring ganda dipadukan dengan barier fisik berupa waring ganda sekeliling lahan dan aplikasi white oil + insektisida berbahan aktif piretroid dengan cara fogging. Demikian juga pengendalian terpadu penyakit kerupuk di lapangan yang paling efektif adalah kombinasi C yang terdiri dari perlakuan agens hayati pada media bibit, sanitasi gulma di sekitar lingkungan pertanaman dan eradikasi tanaman sakit, penyungkupan rangkap plastik + waring ganda dipadukan dengan barier fisik berupa waring ganda sekeliling lahan dan aplikasi white oil + insektisida berbahan aktif piretroid dengan cara fogging.

Abstract

Recent reviews on Dorsalis and Dacine fruit flies had implication on host-insect inventory, including in Indonesia. Update is needed because of these changes and due to the rising of fruit and vegetable trade within islands, as well as with neighboring countries. Fruits were collected over a 3-year period from 24 provinces of Indonesia, covering areas from Aceh to Papua. This work was done specifically to obtain detailed information regarding the host range of the important species of Bactrocera spp., Atherigona orientalis, and Dacus longicornis, as well as Adrama determinata. Male lure Steiner traps to attract fruit flies were used in this study as an additional method to support species variation.Atotal of 1125 fruit samples were collected from many locations. Thirty five plant species from 18 families were recorded as fruit fly hosts and 27 plant species were new report as specific host for certain species of fruit fly. A larger array of host plants for fruit flies in Indonesia than reported before suggests the importance of effective quarantine measures to limit the spread of harmful fruit fly and to prevent the accidental introduction of invasive alien fruit fly species from other countries.

Abstract

Aloe (Aloe vera) planted in West Borneo peat soil is well known as having the best product quality in Indonesia. Soft rot disease is one of the constraints of Aloe cultivation on peat soil. Many methods have no significant result for controlling this disease. This research objectives are to obtain Plant Growth Promoting Fungi (PGPF) for controlling Aloe bacterial soft rot in peat soil. The research was conducted in Clinical Laboratory of Plant Pathology and glass house of Faculty of Agriculture, Gadjah Mada University, Yogyakarta from October 2008 to September 2009. The methods included fungal isolation from peat land, hypovirulent and PGPF ability test, and biological control test in the glass house. Among 42 peat soil fungi tested, 28 isolates were hypovirulent and only 2 isolates i.e. SNTH001 (Penicillium sp.) and SNTH003 (Aspergillus sp.) showed the PGPF ability. Glass house trial showed that single application of SNTH001 and SNTH003 isolates were able to increase the growth of Aloe. In the biological control of Aloe soft rot disease test showed that the lowest intensity (25%) might be obtained by using SNTH001 isolate.

Abstract

The leaf aphid Aphis gossypii Glover is an important vector of virus disease CVMV (Chilli veinal mottle virus) on chillies. In Malaysia the disease could cause the yield loss up to 60% and the predaceous coccinellid Menochilus sexmaculatus Fabricius was observed a dominant predator in the chilli's ecosystem. The studies had revealed that the coccinellid exhibited a strong numerical response to its prey, A.gossypii. The predator population {Y in log(Y-1)) was determined strongly by the aphid population {X in log(X+ 1)}. The immediate numerical response model was Y = -0,1148 + 0.3046X with r^2 = 0.81 and P<0.001, while the 3-day delayed numerical response model was Y = -0,1076 + 0.2962X with r^2= 0.75 and P<0.001. The coccinellid showed its power to stabilise A. gossypii population as indicated by the anti-clockwise direction of cyclic form of their delayed density-dependent relationship.

Abstract

This research aimed to study growth ability and virulence of several Fusarium oxysporum f.sp. zingiberi isolates originated from Boyolali and Temanggung after being preserved for six years in sterile soils media. Completely Randomized Design was used with 12 treatments and 3 replications. F. oxysporum f.sp. zingiberi originated from Temanggung consisted of 7 isolates, i.e., TKO1, TKO3, TKO4, TKO6, TKO7, TPO1, TPO5; and from Boyolali consisted of 4 isolates, i.e., BAO2, BAO7, BAC, and BAP. Variables observed were growth on PDA, colony color, colony diameter, macroconidia and microconidia, mycelial dry weight, incubation period, attack area, and difference of fresh weight of rhizome. The result showed that all isolates of F. oxysporum f.sp. zingiberi both from Temanggung and Boyolali were able to grow well on PDA and fully covered the Petridish at 4.75–7.5 days. The most virulent isolate was TKO6 from Temanggung showing the fastest incubation period of 5 days after inoculation and the highest attack area of 420 mm2 or increase for 107.6%.

Abstract

Root-knot nematodes Meloidogyne spp. are sedentary endoparasitic that attacks various economically important plants. Utilization of nematode’s fungal egg parasite as biocontrol agents of sedentary endoparasitic nematodes have a good possibility of potential success to be applied in the field level, because this fungi is able to colonize in and causes damage to eggs as well as female nematodes inside the root. The purpose of this research are to know the parasitism ability of this parasitic fungi to Meloidogyne incognita eggs, and its effects on second stage larvae hatching rate and the development of galls number in the host. The result shows that the parasitic fungi, those of Trichoderma, Penicillium, Talaromyces, Fusarium genera were able to parasitize root-knot nematode eggs (25.09 to 89.79%), caused root-knot nematode egg hatching to decrease, suppressed the formation of galls, and reduced the population of second stage nematode larvae in the greenhouse.Nematoda puru-akar Meloidogyne spp. adalah nematoda endoparasitik sedentari, bersifat polifag, dan mempunyai nilai ekonomi tinggi. Pemanfaatan jamur parasit telur sebagai agens hayati pengendali nematoda endoparasitik sedentari mempunyai potensi tingkat keberhasilan tinggi untuk diterapkan pada aras lapangan karena mampu mengoloni dan merusak telur maupun stadium nematoda betina yang terlindungi jaringan tanaman. Tujuan penelitian adalah untuk mengetahui kemampuan parasitasi isolat-isolat jamur parasit telur terhadap telur nematoda Meloidogyne incognita, dan pengaruhnya terhadap tingkat penetasan telur menjadi L-2, serta pembentukan jumlah puru pada tanaman terserang. Hasil penelitian didapatkan bahwa jamur parasit telur yang termasuk genera Tricoderma, Penicillium, Talaromyces, dan Fusarium mampu memarasit telur M. incognita berkisar antara 25,09–89,79%, mengakibatkan penurunan persentase jumlah L-2 nematoda yang bersangkutan, serta menekan pembentukan puru akar pada aplikasi aras rumah kaca.

Abstract

The objective of this study was to evaluate the insecticidal activity of bark extracts of 35 plant species belonging to four families (Clusiaceae, Lecythidaceae Meliaceae, and Sapindaceae) against the cabbage head caterpillar Crocidolomia pavonana. Extraction of the test plant materials were performed with maceration method using ethanol and methanol continued by counter-current distribution separation in ethylacetate and water. Bark extract preparation of each plant species was tested against C. pavonana larvae by leaf-feeding method at concentration of 0.25% and 0.5%. Second-instar C. pavonana larvae were fed extract-treated broccoli leaves for 48 hours, then were presented with untreated leaves until the surviving larvae reached the fourth-instar larvae. The number of dead larvae was recorded. The phytochemical test of bark extract of Calophyllum soulattri was done by qualitative method. The results showed that the bark extracts of C. soulattri (Clusiaceae), Barringtonia sarcostachys (Lecythidaceae), and Aglaia tomentosa (Meliaceae) exhibited good insecticidal activity. Methanol extract of C. soulattri barks possessed strong insecticidal activity against C. pavonana larvae with LC95 of 0.21%. This extract showed positive response in alkaloid and triterpenoid qualitative tests. Further studies are needed to identify insecticidal compounds in those active extracts.
