Abstract

This study was aimed to determine the fatty acids profile in fermented products of chicken breast meat (Musculus pectoralis superficialis) of spent hen layer using Gas Chromatography Mass Spectrophotometric technique. The chicken used was of Isa Brown strain after a period of laying of 76 weeks which was obtained from some local farms in Tabanan, Denpasar, and Gianyar - Bali. The chicken meat was cut into small pieces then mixed with ground spices concisting of galangal, coriander, garlic, brown sugar, salt, white pepper and coconut oil. The batter meat was mixed evenly and then wrapped with upih, in which both ends were tied with a rope followed by drying under the sun for 5 days (natural fermentation process). The profile of the fermented chicken meat was integrated and presented descriptively. The results showed that the fermented meat contained higher bioactive component of fatty acids such as oleic acid, palmitic acid and linoleic acid compared to that of the control, but lower stearic acid content. It was also evident that the cholesterol content was the lowest at the end of drying.

Abstract

This research aims to find out the decrease effectivity of dodecil benzene suphonate (DBS) concentrationfrom detergent waste treated using activated sludge. This research was carried out in two steps : the first step wasseeding process or preparation of the activated sludge. This was carried out on plastic container of 2 L. About 3 gsediment taken from Tukad Mati, 1 g NPK were mixed in a plastic container. The volume was made 1 L by addingaquadest, the mixture was aerated for 1 week. The second step was treatment of detergent waste which was carriedout using 2 plastic container of 5 L each. About 2 L detergent waste and 1 L activated sludge solution were mixed inthe plastic container. Mean while, 3 L detergent waste as a control were palced in the other plastic container. Bothcontainer were aerated for observation for decrease DBS concentration was carried out during 0, 3, 5, 7, 10 and 15days. DBS concentration was analyzed using Methylen blue method.The research result indicated that generally, there were decrease of BDS concentration both in control andtreated samples. The lowest DBS concentration was observed on day 15 both on control and treated samples withvalue, of 46.726 and 0.381 mg/L respectively. The decrease effectivity were 8.18% for control and 99,25% fortreated samples. From this research, it can be seen that treatment of waste using activated sludge was effective todecrease DBS concentration in detergent containing waste.

Abstract

DNA content in total honey microorganism can be used to study DNA fingerprint of honey. Metagenomic is a new approach to analyse complex genomes from unculturable total microorganism. The aim of this research was to get DNA metagenomic isolated directly from honey samples enriched with microorganisms growing media LB (Luria-Bertani) and without enrichment of microorganisms growing media LB in order to study DNA fingerprint of honey for the next research. The study was started with incubation of the LB enriched honey samples and honey without LB samples at variation time of 0, 24, 48, and 72 hours. Following the incubation process, the DNA metagenomic form both LB enriched and unenriched honey samples were isolated with direct cell lysis method. The DNA obtained was analyzed by spectrophotometer UV-Vis and agarose gel electrophoresis.  The result showed that DNA metagenomic can be isolated from LB enriched honey and without LB enriched honey samples. Analysis using the agarose gel electrophoresis indicated that the metagenomic DNA obtained from both honey samples was still contaminated with RNA. The purity ratio of metagenomic DNA from both honey samples analysed using UV-Vis Spectrophotometer at ? 260/280 nm was between 1.2 to 1.6. This ratio suggested that the metagenomic DNA isolated from both samples were contaminated with protein and fenol.

Abstract

This study aims to determine the concentration, contact time, and the optimum pH required by the ILS rice straw mushrooms to degrade azo dyes remazol red type. Subject in this study is ILS rice straware mushroom which is obtained from existing fields in Temukus Village, Banjar District, Buleleng Regency. The objecs are the concentration, contact time, and the optimum pH required by the ILS rice straw mushroom to degrade textile dyes remazol red type. Stage of data collection starts from the preliminary test biodegradability of dyes, mold growing on PDA medium (potato dextrosa agar) and liquid Czapex, qualitative test of lignolitic enzyme, and dye biodegradation test on the variation of environmental conditions. The concentrations of remazol red dye before and after degraded by the ILS rice straw mushroom was measured by using a Hach spectrophotometer. The results showed that the degradation of azo dyes type remazol red RB using the ILS rice straw mushroom was optimum at color concentration of 30 ppm, contact time of 10 days, and pH 5 with the efficiency of each of 57,77; 71,13; and 50,88%.

Abstract

Pada penelitian ini dipelajari efek ekstrak etanol biji jagung (Zea mays) terhadap profil lipid tikus wistar dengan diet tinggi lemak. Penelitian ini bertujuan untuk membuktikan efek antihiperkolesterolemia dari ekstrak etanol biji jagung dengan variasi dosis 50 mg/Kg BB; 100 mg/Kg BB dan 150 mg/Kg BB. Subjek penelitian ini ialah 24 ekor tikus Wistar yang dibagi menjadi 6 kelompok secara random dengan design penelitian posttest only control group design. Data hasil penelitian di analisis dengan uji tukey anova. Ekstrak etanol biji jagung dapat menurunkan kadar kolesterol total tikus wistar dengan diet tinggi lemak dengan dosis 50 mg/Kg BB, 100 mg/Kg BB dan 150 mg/Kg BB sebesar 5,44%; 12,89% dan 19,77%. Kadar kolesterol LDL dapat diturunkan dengan dosis 50 mg/Kg BB, 100 mg/Kg BB dan 150 mg/Kg BB sebesar 2,55%; 8,74% dan 15,66%. Kadar kolesterol HDL dapat ditingkatkan dengan dosis 50 mg/Kg BB, 100 mg/Kg BB dan 150 mg/Kg BB sebesar 1,5%; 2,08% dan 3,24%. Hasil uji fitokimia menunjukkan bahwa ekstrak etanol biji jagung positif mengandung senyawa golongan steroid dan fenol. Senyawa tersebut memiliki aktivitas antioksidan dan dapat meningkatkan antioksidan endogen di dalam tubuh, sehingga dapat menurunkan kolesterol darah.

Abstract

Mount Agung in Karangasem, Bali which erupted in September 2017 produced abundant volcanic ash. This ash can be used as a raw material for making silica gel. The stages of the research carried out included analysis of the content of silicon (Si), synthesis of silica gel using sol-gel technique, characterization using FTIR, X-RD instruments as well as surface acidity. The results of the analysis using the X-RD instrument showed the presence of silicon from SiO2 found at 2? = 28,07530. The silica gel yield obtained in this study was 15.05% w/w. The results of the characterization with the FTIR instrument showed that silica gel from volcanic ash of Mount Agung had the main Si-OH and Si-O-Si groups. The hydroxyl bond from Si-OH grup were found at ±3649.32 cm-1 for stretching vibrations and ±1687,71 cm-1 for bending vibrations. Si-O bond from Si-O-Si group were found at ±1207.44 cm-1 for asymmetric stretching vibrations and ±482.20 cm-1 for bending vibrations. Diffractogram showed that the silica gel had an amorphous structure with surface acidity of 18.0222 mmol/g and a surface area of 36.5514 m2/g. Abstract

The research was conducted to observe the photodegradation of methylene blue by UV light and Al2O3 catalyst. The research determinated the optimum Al2O3 catalyst weight, the optimum pH, the optimum radiation duration and the effectivity of photodegradation at optimum conditions. Degradation of methylene blue were analyzed by UV-Vis spectrophotometer. The results showed that the optimum Al2O3 catalyst weight was 40 mg the optimum of pH was 11, and the optimum of radiation duration was 4 hours. The effectivity of photodegradation was (30.22 ± 1.17)%.

Abstract

Gayam (Inocarpus fagiferus Fosb) is a tropical plant whose stem bark has been used as traditional medicine. This research aims to identify the flavonoides isolate from stem bark of gayam and identify the class of flavonoids compound as antioxidant. Extraction of 1000 g stem bark powder with 8500 mL of 96 % ethanol to yield 30 g concentrated ethanol extract. Partition of 15 g extract ethanol-water (7:3) with chloroform and n-buthanol to yield 1,5 g concentrated chloroform extract, 5,87 g concentrated n-buthanol extract and 5,63 g concentrated water extract. The flavonoids test using Wilstatter and Bate-Smith Matcalf reagent showed that n-buthanol and water extract contain flavonoids compounds. Antioxidant activity of flavonoids tested with DPPH method and lipid peroxide ammonium thiocyanate method showed that n-buthanol extract had antioxidant activity with IC50 of 24 ppm and n-buthanol extract has ability to inhibit the formation of lipid peroxide with inhibition value of 63,04 %. Separation of n-buthanol extract using thin layer chromatography preparative with chloroform as mobile phase and silica gel GF254 as stationary phase obtained four fractions. Flavonoids test showed that fraction F3 contained flavonoids compounds. FTIR analysis indicated that isolate had functional groups such as  -OH , C=O, C-O alcohol, C=C aromatic, C-H aromatic and C-H aliphatic. UV-vis spectra showed 2 peaks at ? 310 nm  and  261 nm which indicated flavonoids group of  isolfavone with hydroxyl groups at C-5 and  C-7. Antioxidant activity test with DPPH method showed that the isolate had antioxidant activity with IC50 of 1 ppm.

Abstract

The 8-isoprostane compound can be used for a biomarker of oxidative stress  formed by lipid peroxidation in the form of arachidonic acid with free radical in the body. The purpose of this study was to analysis  urinary 8-isoprostane in male Wistar rats after sub-acute exposer to ethanol and cigarette smoke. Extraction method used was solid phase extraction and the analysis was conducted by TLC and spectrophotodensitometry. The results showed that the 8-isoprostane compound was not detected in the male Wistar rat’s urine as control rats and in those exposed to cigarette smoke. The average concentrations of 8-Isoprostane in the male Wistar rat’s urine exposed to ethanol and combination of ethanol-cigarette were 1.519 ± 1,386 and 2.063 ± 1,308 ppm respectively.

Abstract

Decantation process was done to separate oil and water. Mostly, Small Medium Enterprises (SME) use lowtraditional technology as multiple drums. As a result, there is much oil dumped together with wastewater. A betterway to decant oil and water can be done by using an interceptor plate in decanter equipment. This research aims wasto test the performance of decanter with special design by using an interceptor plate. The observation focused ondistance between plates toward the oil content in wastewater, which was measured by its density. Three variables ofplate distance were 10 mm, 20 mm, and 30 mm. Material tests were nilam and clove oil with several concentrationsi.e. 1.96%, 3.85% and 5.66%, and the flow rate was adjusted at 2 liters/minute. The result shown that the interceptorplate with 10mm distance produced wastewater with lower content of oil. In addition, in industry scale test, thedecanter having 10mm distance plate interceptor could reduce oil loss up to 11.8% for nilam and 42.9% for clovecompared to that of multiple drum.
