Abstract

Research has been carried out to detect pathogenic gene markers of Avian Pathogenic Eschericia coli (APEC) iroN, iutA, and hlyF in Escherichia coli bacteria isolated from organs of sick chickens in Bali, and to determine phylogenetic relationships between those marker genes in Bali and in the other countries in the world. Six isolates of E. coli bacteria with codes E2, E3, E7, E8, E9, and E10 were used in this study. The isolates were isolated from domestic chicken in 2018. All genes were detected using the Polymerase Chain Reaction (PCR) method. The genes of iroN, iutA, and hlyF could be detected from all isolates. Well readable sequence of iroN, hlyF, and iutA was 659 bp, 518 bp, and 250 bp, respectively. All three genes were homogenous. Phylogical analysis shows that all pathogenic markers share same cluster with the pathogenic E. coli from all countries in the world.

Abstract

Duck is one of the potential egg poultry production in Indonesia. General farming systemof duck was carried out an intensive and semi-intensive. The effect of difference farmingsystem may be contribute to differences in the duck’s egg quality, especially in relation toaspects of Egg White Index (EWI), Egg Yolk Index (EYI) and Haugh Unit (HU) of duck’segg, as well as to the long storage at room temperature (±28°C). The research usedCompletely Randomized Design (CRD), the pattern of 2 (farming system) x 4 (0,7,14 and21 day) two factor treatments farming system as intensive farming and semi-intensive andfour factor that is a long storage at room temperature. Each treatment combination wasreplicated 4 times. Data were analyzed with ANOVA, and followed by Duncan multiplerange test. Based on these results it can be concluded, farming system intensive and semiintensiveto produce duck’s egg quality with EWI, EYI and HU are not significantlydifferent (P>0.05). EWI, EYI and HU were significantly (P <0.01) influenced by longstorage; if the longer stored, the duck’s egg quality to be decreased.

Abstract

Ulcerative dermatitis is a disease that most commonly affects turtles and/or hatchlings that are kept in ponds, including in Turtle Conservation and Education Center (TCEC), Serangan. A study was conducted to determine the prevalence, pathology, and lesion location of ulcerative dermatitis in olive ridley hatchlings reared in TCEC Serangan, also the differences of the body condition index of olive ridley hatchlings with ulcerative dermatitis and healthy ones. Prevalence was determined by counting sick hatchlings compared with the total number of hatchlings at TCEC. Morphometry (straight carapace length, straight carapace width, curved carapace length, curved carapace width) and body weights were measured to determine the body condition index, and then body index differences were compared with unpaired T-Test. Skin tissue samples were processed into pathology slides and routinely staining of hematoxylin eosin (HE). The prevalence of ulcerative dermatitis in olive ridley hatchlings at TCEC was 16.2%. Crusty-yellow wound lesions of 2 mm to 2 cm in diameter were found and microscopically there were heterophils and mononuclear cells infiltration in skin dermis accompanied by erosion, parakeratosis, and necrotic material containing cell debris. Lesions were found mostly on neck area (63.04%), followed by flippers, head, skin near carapace, eyelids, and neck area with flippers combined. There was a very significant difference in body condition index between suffered hatchlings with healthy ones, where the average body condition index of suffered hatchlings were greater than healthy ones. Rearing management should be fixed.

Abstract

Bali cattle females to be one of the priorities in repairing genetic local cattle because of its superiority in the field of production and reproduction compared to other cattles. This research aims to determine the respiration profile on a bali cattle adult female at Bali cattle breeding centers Sobangan village, Mengwi sub district, Badung regency, which has an average altitude of 350 meters above sea level and rainfall reached around 2029.0 mm. This research used 20 bali cattle adult female age 1.5 years to 4 years. Data collected by examination and observation were performed in the morning, afternoon and evening. The observed parameters are the frequency of respiration. The results obtained by the average frequency of respiration morning of 16.05±0.82 at 25°C ambient temperature, daylight 20.95±0.88 at 32°C ambient temperature, and the afternoon of19.80±1.57 at 29°C ambient temperature. These results indicate that the frequency of respiration in Bali cattle females can be changed according to environmental conditions and other contributing factors.

Abstract

Sapi bali(Bos sondaicus) merupakan salah satu ternak unggulan sebagai penghasil daging di Indonesia yang harus terus dikembangkan dan ditingkatkan populasinya. Dari tahun ke tahun permintaan akan daging semakin meningkat, data Bappenas menunjukkan untuk memenuhi kebutuhan daging nasional,pada tahun 2002-2003 telah diimpor sapi bakalan dari Australia sebanyak 325.000 – 375.000, sedangkan impor daging pertahun setara dengan 100.000 ekor sapi (Apfindo 2005).Berdasarkan data tersebut sapi bali sangat berpeluangdikembangkan secara makimum untuk memenuhi kebutuhan daging di IndonesiaFenomena adanya dua bovine lentivirus pada sapi bali yakni Jembrana Disease Virus danBovine Immunodeficiency Virus, segera harus dibuktikan keberadaannya. Selanjutnya dapat dikembangkan penelitian yang bertujuan kearah produksi vaksin heterolog untuk mencegah penyakit jembrana, danakhirnya membantu peningkatan populasi sapi bali.

Abstract

This study aims to determine the sensitivity pattern pathogenic E. coli against antibiotics: oxytetracycline, ampicillin and sulfamethoxazole, in broilers in the Mangesta Village, Penebel, Tabanan Regency. The samples used in this research were 15 samples of colisepticaemia infected broiler’s organs . Sampels were isolated on media EMBA then identified with a Gram staining and biochemical tests with a TSIA, SIM, SCA and MRVP. Positive isolates of the bacterium E. coli were tested sensitivity to antibiotics oxytetracycline, ampicillin and sulfamethoxazole. The results showed that the sensitivity test of E. coli bacteria were isolated from the organs broilers suffered from colisepticaemia: 73.3% of E. coli resistant, 20% intermediate, and 6.7% was sensitive to the antibiotic oxytetracycline, 100% resistant to the antibiotic ampicillin, 53.3% and 46.7% intermediate resistance against sulphamethoxazole.

Abstract

The codeine has been used as a marker for illicit heroin uses, that for increasing differentiating illicit heroin uses, it needs the codeine pharmacokinetic data. Eleven rabbits have been injected 11.04 mg codeine (with body weight: ± 1.8 kg) via marginal vena. Blood has taken at every 5, 15, 30, 60, 90, and 120 minutes. Serum has stored at -20 0C before analyzed. Blood protein was predicated by isoprophanol. Codeine was extracted with chloroform-isopropanol (3:1) and buffer phosphate 0.2N pH 9.3 then separated by phase toluene-acetone-ethanol-ammonia (45:45:7:3) as mobile phase and aluminium sheet of silica thin layer chromatography Gel 60 F254 as the stationary phase. Codeine determined by spectrophotodensitometry at 215 nm. Limit of detection and limit of quantification of codeine was 184.43 ng and 563.62 ng per spot, respectively the percentage recovery of in vitro was 60.60%. Profile concentration of codeine after IV bolus showed decreasing concentration at the first terminal and increasing codeine concentration at the last terminal that indicated an enterohepatic cycle. Codeine pharmacokinetic followed the first compartment with the half time was 30.61 minutes and the distribution volume was 7760.50 mL. Enterohepatic cycle caused a prolonged lifetime of codeine at the rabbit

Abstract

Telah dilakukan penelitian yang bertujuan untuk melakukan analisis faktor tipe lahan pemeliharaan dengan kadar mineral (Mg, K, Na, Ca, Cu, Co, Zn, Fe) serum sapi bali.  Sampel berupa serum diambil masing-masing 15 ekor, berasal dari tipe lahan tegalan, kebun, sawah dan hutan. Analisis mineral menggunakan metode Apriyantono, dkk. (1989) dengan pengabuan basah, dan kadar mineral dibaca dengan alat AAS (Atomic Absorpsion Spectrophotometry). Hasil penelitian menunjukkan rerata mineral  magnesium sapi bali yang dipelihara pada lahan tegalan : 22,36 mg/l sedangkan tembaga : 0,09 mg/l mg/l. Di lahan perkebunan, kalium : 24,84 mg/l dan kobalt : 1,80 mg/l. Serum sapi bali di lahan hutan, kadar natrium : 19,63 mg/l dan seng: 0,33 mg/l.  Kalsium : 6,23 mg/lt, besi : 8,39 mg/lt diperoleh pada sapi bali yang dipelihara pada lahan persawahan. Hasil analisis membuktikan faktor tipe lahan pemeliharaan sangat terkait dengan kadar mineral serum sapi bali. Sapi bali yang dipelihara pada lahan pemeliharaan dengan ketersediaan mineral yang kurang menyebabkan terjadinya defisiensi mineral.

Abstract

This study aimed to determine influence of collecting frequency against concentration and spermatozoa abnormalities of quail. This study used a completely randomized design (CRD) with 3 treatment groups, is collecting sement at 2, 3 and 4 days intervals. Each treatment be repeated 9 times. Observations were made on the concentration of spermatozoa (107sel / ml) and abnormalities of spermatozoa (%). The data was analyzed using analysis of variance (ANOVA), if there are significantly different (P <0.05) followed by Duncan test. The results showed the concentration of spermatozoa in each treatment respectively 39.00 ± 5.148 x 107 cells / ml; 59.49 ± 1.878 x 107 cells / ml; 59.11 ± 2.028 x 107 cells / ml. Abnormality in each treatment was 14.22 ± 1.394%; 7.22 ± 1.202%; 7.49 ± 1.130%. The frequency of ejaculation was statistically significant (P <0.05) against the concentration and spermatozoa abnormalities of quail.
