Abstract

Objectives of this study are to determine the genetic variability among progeny of intraspecific crossing between three strains of Oreochromis niloticus Bleeker (BEST, Nirwana, and Red NIFI) using Randomly Amplified Polymorphic DNA (RAPD) and truss morphometric. The results showed that hybridization enabled to increase genetic variability respectedly on progeny of Nirwana male crossed to BEST female (27,93%) as well as to Red NIFI female. An analysist of truss morphometric indicated that BEST population had the lowest similarity and largest genetic distance (0,6) to the Nirwana and Red NIFI which were closer to the hybrid populations.

Abstract

YUN INDIARTO, MULYADI & RAZALI YUSUF, 1989.The structure and composition of mangrove in Bikoen-Kupang,Timor.Berita Biologi 3(9): 455 - 457.Mangrove forests in EastNusa Tenggara cover a total areas of about 1930 ha(Darsidi, 1984). Little is known about the mangrove forest in that island, especially in Bikoen, Kupang Region.Therefore, the study has been conducted at the mangrove forest of Menipo Nature Reserve in order to analyse the structure and composition of the species.Four transects perpendicular to the coast line were used for data collection.At least, there were 12 mangrove species found in the forest, dominated by Ceriops tagal, Rhizophora stylosa and Bruguiera exaristata. Seedlings dominated the forest by 49,92%, where as the sapling were 45.43%.

Abstract

Saccharomycopsis sp. produced glucoamylase (a-1,4 glucanglucohydrolase; EC 3.2.1.3) shown by the presence of clear zone on media containing 0.2% yeast extract, 0.5% pepton, 0.3% KH2PO4, 0.05% MgSO4. 7H2O, 0.01% CaClr2H2O, 2% agar and 2% soluble starch. In liquid media containing soluble starch glucoamylase production reached a maximum at 2 days fermentation, with levels of 4.2 x 10 U/ml (one unit activity is define as micromoles of glucose produce per ml per minute).Studies on the glucoamylase characterization revealed that the optimum temperature for activity was 40°C - 50°C. The enzyme was stable for lh at 45"C - 55°C, while at 60°C to 65°C, 40% of the original activities were lost. The optimum pH of the enzyme was 6.0. After incubation of crude enzyme solution for 1 h at pH 8.0, a decrease of about 40% of its original activity was observed. Saccharomycopsis sp. produced high glucoamylase when it was grown in media containing rice flour as carbon source with the glucoamylase activity at 9,28 x 10 U/ml.

Abstract

Enzymes a-amylase is one of the enzymes used in process of starch degradation. This present study was aimed to characterize and identify of a-amylase producing strain isolated from bulk shrimp-paste in Samarinda, East Kalimantan was carried out. An experiment was conducted to examine influences of incubation, carbon source, substrate concentration temperature for incubation, pH of media, and addition of metal ions. Identification of strain C2 was carried out by using Wizard Genomic DNA Purification Kit (Promega). The result showed that optimum activity of a-amylase from C2 after six days incubation was 18.93 U/mL. Tests on the type of substrate , soluble starch was the best source to produce a- amylase (14.51 U/mL). However, at concentration of 2 % and incubation temperature at 40°C, enzymatic activity was decreased to 12.56 U/mL and 12.79 U/mL, respectively within residual activity of 74.75%. The enzyme activity was 16.43 U/mL and its residual activity was 39.14 % when it was assayed at pH 8.5. Addition of metal ions in the form of divalent and monovalent cations (1 mM) showed that the enzyme could be activated by ion Ca2+ while ion Cu2+, Co2+, Mg2+, Zn2+ , Na+  decrease the activity of the enzyme. Identification of strain C2 using molecular characterization demonstrated that partial sequences of 16S rDNA reffered to as Bacillus subtilis.

Abstract

H B.MUNAF 1986.The life cycle of tinks of Indonesia:Argas(Persigas)robertsi Hoogstraal,Kaiser&Kohls,1986(Acarina Argasidae).Berita Biologi 3(5): 199-202.in the sequence of laboratory works in orde to fulfil the laks of biological data of ticks of Indonesia,the life-cycle of a fowl tick,argas (pericargas) robertsi, was studied. Nymphs and adults were fed on pigeons using capsule system.The completion of the life-cycle required minimum 55 days.Other observable data in this study,such as amount of eggs, percent tages of edysized ticks survivals were given.

Abstract

Vegetation types in fresh water swampy areas: Mandor in West Kalimantan and Lahei in Central Kalimantan, were compared.There were three types: kerangas, kerangas shrub and peat swamp forest. Kerangas forests in both areas had similar flora in some extent.That of peat swamp was very different from kerangas though both vegetations distributed in adjacent area.

Abstract

National fisheries production is not proportional to the increase of the population now, so that the consumption of animal protein needs are not met.For the sake of national fisheries production there is a need for intensive cultivation technology breakthroughs to spur the density in nursery phase ‘rajadanu’ carp (Cyprinus carpio) strain. This experiment aim to determine the appropriate density of rajadanu carp growth in the pond at the nursery phase. The average length of tested fishes is 2.02 cm and the average initial weight is 0.25 g maintained in the out door plot pool of Research Installation for Germ Plasm of the Research and Development Division of Freshwater Fisheries, Cijeruk, Bogor measuring 1x1x1 m with density 100, 150 and 200 fishes/m3 .Each treatment was repeated 3 times. Feed used was a commercial feed containing 28% protein given in 2 times a day as much as 5% of the total weight of fish. The results showed that after 40 days, each treatment showed no significant difference (P> 0.05) with absolute length and weight growth of the absolute highest in nursery density of 200 fishes/m3 of 1.30 ± 0.10 cm and 1.14 ± 0, 18 cm with a daily growth rate of 1.30 ± 0.10% and 4.08 ± 0.55%, and the highest survival at densities 200 fishes/m3 of 92.5 ± 6.55%.

Abstract

The bacterial populations in the farming of spiny lobster could have either beneficial or harmful effects depending on the prevailing conditions. We designed this study to identify and to perform a pathogenicity test of some bacteria isolated from wild and farmed spiny lobsters Panulirus homarus. The adult farmed lobsters were obtained from Pangandaran and Lombok coastal areas, while the wild lobsters were collected from Lombok, with five lobsters for each location. The bacteria were isolated from the midgut, gill, hepatopancreas, and muscle tissues of the lobsters. The identification of the bacteria was carried out by molecular methods. Pathogenicity test was performed by intramuscular injection of 0.1 ml bacterial suspensions at the density of 7×106cfu/ml into each three adult apparently healthy lobsters for every eight bacterial isolates. Our study identified six bacterial isolates that exhibited high homology of a nucleotide sequence with Shewanella algae, Bacillus firmus, Vibrio alginolyticus, Tenacibaculum lutimaris, Pseudomonas sp. and Vibrio sp., while two isolates were reminded unidentified due to low nucleotide similarities (< 97%). The pathogenicity test showed that there was no mortality of lobsters injected with those bacterial isolates. This may because the dose of injection was too low to induce bacterial infection particularly for Vibrio, or the bacteria were not pathogenic for lobster or even have the potency as probiotic bacteria.

Abstract

Diversity on the lowland pitcher plants (Nepenthes spp.) in Central Kalimantan were conducted between August 2005 and September 2006 at three locations study. That is: Barito Ulu, Sebangau and Kasongan district. Survey inventories were used to determine the diversity of Nepenthes at the study site. We found sixteen Nepenthes at study area, divided nine Nepenthes species; N. albomarginata, N. ampullaria, N. gracilis, N. hirsuta, N. mirabilis, N. rafflesiana, N. reinwardtiana, N. stenophylla, one unidentified Nepenthes species and seven natural hybrid; N. xhookeriana (N. rafflesiana x N. ampullaria), N. xtrichocarpa (N. gracilis x N. ampullaria), N. xneglecta (N. gracilis x N. mirabilis), N. mirabilis x ampullaria, N. mirabilis x rafflesiana, N. reinwardtiana x N. mirabilis and N. reinwardtiana x gracilis. Heath forest and peat swamp forest are generaly the habitat of lowland Nepenthes at Central Kalimantan.

Abstract

Cempirit [Ervatamia svhaerocarva Burck.(Apocynaceae)] is a sticky plant found in Indonesia. This species had long been used as traditional medicine for skin diseases and ulceration of nose.Major biologically active compound was isolated from bark of E.sphaerocarDa by using combination of chromatography techniques (Thin Layer Chromatography,Colum Chromatography and High Performance Liquid Chromatography).The structure of this compound was elucidated by spectroscopic methods, mainly by NMR (Nuclear Magnetic Resonance) spectral. This compound was determined as indole alkaloide called Tabernaemontanine.
