Abstract

Study on the productivity of two species of local crickets; Gryllus testaceus Walk and Gryllus mitratus Burn which are generally found in the local market have been conducted with the main aim at obtaining information on the capability of those species in producing eggs including the length of production cycles.Three levels of box square density (50 female/box,100 female/box and 150 female/box) and two levels of additional feed (green master and papay leaves) were used as treatments. The results showed that the number of eggs produced by G.mitratus were larger with longer production cycles compared to those of G.testaceus. Species and density has highly significant effects on egg production and feed consumption, whereas, additional feed significantly influencing egg production and feed consumption of the two species. The use of green master as additional feed resulted in a higher egg production than those given papay leaves.

Abstract

Fresh milk is a beverage with high protein contents that can be consumed either directly or as ingredient supplement into safely and healthy food. However, the milk also as a good media for development of pathogenic bacteria that also dangerous for human health. The aim of this research was to determine pathogenic bacteria contamination in fresh milk and its antibiotic resistance profiles to several antibiotics. Fresh milk samples were taken from milk cans belong to the farmers at 34 dairy cows centre in Cibungbulang, Bogor, West Java. The quantitative determination was conducted on 34 milk samples. Several parameters examined were based on the Indonesian Nasional Standard (Standar Nasional Indonesia/SNI) No. 01-3141-1998 for Fresh Dairy Milk and SNI No. 7833-2009, such as total bacteria and coliform. The qualitative examination result for isolation and identification of bacteria were found that the milk samples consisted of 41.18% E. coli, 23.53% Streptococcus Gorup B, 8.82% Staphylococcus aureus, and none for Salmonella. The antibiotic resistence profiles were tested to 5 antibiotics. It showed that Escherichia coli isolates were resitance to penicilline (14.3%), oxytetracycline (21.4%), chloramphenicole (57.1%), and streptomycin (28.6%), whereas those Streptococcus Group B isolates were resistance to penicilline (12.5%), Oxytetracycline (37.5%), chloramphenicole (25.0%), streptomucin (87.5%), and ciprofloxacin (87.5%). Multiresistance of E. coli were found against 2 antibiotics, whereas Streptococcus against 2-3 antibiotics. This research indicated that fresh milk samples taken from farmers at Cibungbungang, Bogor were contaminated with several pathogenic bacteria and mostly highly resistance to 5 antibiotics testing.

Abstract

The pyrene dioxygenase coded by gene is an indicator of bacterial isolates capabilities in pyrene degradation. The encoded gene of pyrene dioxygenase can be amplified and detected in Pseudomonas sp. PyrA2 and Burkholdoriasp. PyrA4 isolates, using primary specific Diox which designed based on PhdF gene sequence, the coding gene of pyrene dioxygenase in Mycobacterium vanbalenii PYR-1. The sequence alignment of pyrene dioxygenase putative gene in both of bacterial isolates with the sequence of pyrene dioxygenase coding gene in M. vanbalenii PYR-1 shows the similarity percentage of 41 % and 42% with Pseudomonas sp. PyrA2 and Burkholdoria sp.PyRA4.

Abstract

Caryota no Becc,(fish-tailpalm) is considered vulnerable palm, Its population is declining from the habitat by destructive exploitation but also to some extent by local people who remove the edible apex or 'cabbage 'as a vegetable and also obtain sago from the pith of the trunk,so kill the tree.A though its biological characteristics have not been fully studied, this species is also potentially for ornamental plant Thus conservation measures should be taken to improve the status of this threatened species. Tissue culture techniques may provide ways either to propagate or to conserve itin-vitro. Naturally Caryota no Becc.palm fruit after 20years and they can germinate more than 3 months after sowing with low germination value. In this experiment, the tissue culture response of this species are studied using the explant of embryo from (relatively) young fruit (juvenility fruit). The effect ofBA and 2,4D on the shoot growth, root growth and calluses was evaluated, while the evaluation of the effect ofactifated charcoal was discussed. The satisfactory results have not been achieved.

Abstract

Rice-stink bug (Leptocorisa acuta Thumb) that attack rice crop often causing heavy damage of panicle of rice as well as decreasing either quantity or quality of grain after harvest. The objective of this research was to select 14 entomopatogenic fungi of Beauveria spp. isolates, collected from rice stink bug (L. acuta) and their pathogenicity assay to insect host L. acuta. We also aimed to study diversity of Beuveria spp. which was isolated from insect host from Situgede, Bogor. The research was conducted at Laboratory, and Green house of Center for Agricultural Biotechnology and Genetic Resources Research and Development, Bogor, in 2014. Result of the study showed that isolates from Situgede Bogor were sucessfully isolated  as Beauveria spp. based on Koch Postulat assay.  The identified fourteen isolates were morphologically confirmed as Beauveria spp. The pathogenicity test was indicated by symptoms and mortality of rice stink bug after inoculation with these entomopathogenic fungi. The Beuveria isolates Stgd2(14)1, Stgd6(14)1, Stgd7(14)2, Stgd8(14)2, and Stgd0113 were the most virulent isolates. Stgd2(14)1 provided the fastest time to kill L. acuta with LT50 values of 6.9 days. The 13 isolates of Beauveria were well amplified by ITS primers. However, no diversity was found among isolates, presumably due its narrow host range tested.
