Abstract

ABSTRACTThe Newcastle Disease (ND) virus causes the most important disease in chicken, and very contagiuos in poultry in many countries. In Indonesia the Newcastle Disease or is called tetelo became endemic in chicken currently, the ND virus of genotype VII caused morbidity 100% and mortality 80% in chicken. The fusion and haemaglutinin-neuraminidase protein are involved in adhesion to the surface of hert cells to antibodies. HN and F protein induced by the ND vaccines have neutralizing effects. The aim of this study was to compare the antibody level and protection of several ND vaccine genotypes (genotype I, II, VI and VII) in chicken against ND virus chicken / Indonesia/GTT/11 genotype VII. Our result showed that all genotype ND vaccines (I, II, VI dan VII) produce 100% protection in chicken from clinical and mortality, but the challenge virus excretion in chickens vaccinated with ND vaccine VII genotypes were significantly different (p <0.05) to other groups. Response of antibody in chicken vaccinated ND genotype VII show highest mean titer (GMT 122) compered other ND vaccine. In onclusion, the ND virus chicken/Indonesia/ GTT/11 genotype VII is good to be used as inactive vaccine seed, which appropriate to the field virus circulating at this time.Keywords: virus, Newcastle Disease, genotype, vaccine

Abstract

ABSTRACTDibenzothiophene is one of polycyclic aromatic hydrocarbon (PAH) compound containing sulfur element. This compound has toxicity, mutagenic and quiet persistent in environment. From sreening test, it was known that isolate LBF-1-0061 was potential to degrade dibenzothiophene. The objectives of this study are to study dibenzotiophene degrading capability by marine bacteria isolate LBF-1-0061 using screening test; analysis of dibenzothiophene residue by GC/MS and identifiy the isolate by molecular identification. The result of this research shown that LBF-1-0061 isolate could grow up to 100 ppm of dibenzotiophene. This isolate also presented degrading capability approximately 37.5% of dibenzotiophene in 14 days incubation. Based on partial 16S rRNA gene analysis, LBF-1-0061 was identified 99% as Novosphingobium mathurense strain SM117.Keywords: sea bacteria, biodegradation, dibenzotiofen, hydrocarbon aromatic polisiclic

Abstract

The objective of this study were to obtain information about genetic parameters of agronomic characters of sorghum populations derived from Single Seed Descent (SSD) method, and to estimate selection differensial and also to select of F4 population that have a high yield and medium plant height . The experiment was carried out at Leuwikopo Farm, Darmaga, Bogor from January to April 2014. The genetic material were F4 population derived from SSD. The results showed that based on the skewness and kurtosis values, all of the agronomy characters observed in this study were poligenic controlled and influenced by additive gene action. The genetic coefficient of variability values for all parameters varied from intermediate to high in F4 population. Agronomic characters in F4 responded positively to selection because of high broad sense heritability estimates. Correlation analysis showed that plant height, number of leaves, stem diameter, panicle length, panicle weight and 100 seed weight have significant and positive correlation to grain yield/plant. Differential value with selection intensity 10 % based on grain yield/plant will increase grain yield/plant 83.89 % and plant height 8.91 % in the next generation, whereas differensial value based on grain yield/plant and plant height will increase grain yield/plants 68.33 % and plant height 0.26 % in the next generation. It means that selected plant were expected increasing yield 83.89 % or 68.33 % in next generation.Keywords: heritability, cefficient genetic variability, correlation analysis, selection differential

Abstract

ABSTRACTSoybean is an important legume crop in Indonesia and local varieties from different regions are diversed. Accurate identification of local varieties is needed to determine the genetic identity in order to protect them to indicate the geographical origin and registration purpose. This study aimed to analyze DNA fingerprints of local varieties of soybean in Indonesia to develop a specific identity using SSR markers along with the marker set. A total of 29 local varieties/accessions was analyzed using 9 selected SSR markers with semi automated genetic analyzer on the basis of capillary electrophoresis. Total markers successfully detected 50 alleles in all varieties, with the average polymorphism information content (PIC) of each locus (0.579) and the genetic diversity index (0.629) were quite high as a reflection of diversity of the local varieties. Based on the analysis of the genetic diversity index, PIC, rare alleles, dominant allele frequency, analysis of genetic distance, 6 SSRs (Satt009, Satt038, Satt177, Satt242, Satt308 and Satt114) were selected as a set of markers for variety identification. Although three varieties can not be distinguished because of their close genetic relationship, but the digital coding indicates that local varieties of soybeans have possessed specific identities (ID) representing variation of SSR alleles. The selected SSR loci with additional identifier can be used to develop ID of Indonesian local soybean in the germplasm collection. Overall, the marker set is a powerful tool to complement morphological markers for the protection of local soybean varieties that contribute in accelerating local varieties improvement in the future agriculture.Keywords: specific identity, SSR, Indonesian local soybean variety, DNA fingerprint.

Abstract

ABSTRACTSuccession dynamics (as a complex interaction and relationship between environment and species) is a crucial ecological process for managing and restoring habitats and ecosystems. This research was conducted at several locations based on the damage levels occurred due to the 2010 eruption of Mount Merapi: heavily damaged site was chosen at the Cangkringan Resort (consisting of both open and covered areas) and the Kemalang Resort, medium damage site was located at the Dukun Resort, and minor damage/relatively intact site was situated at the Selo Resort. A total of 135 plant species belonging to 64 families were recorded from all locations during the study. The Mount Merapi eruption occurred in 2010 had significantly impacted on the floristic community structure and condition. Canonical Correspondence Analysis (CCA) was used to analyze the relationships between environmental variables and the existing plant species conditions. The results indicated that different abiotic environment conditions (variables) significantly influenced species compositions and conditions. The covered area (?= 0:49; p= 0.002; F= 10:35) and the elevation factor (?= 0:32; p= 0.002; F= 7:08) provided the highest impact on vegetation conditions. Meanwhile, the relationships and correlations between edaphic factors and floristic community conditions varied from site to site.Key words: Abiotic environment, floristic community, succession, Mount Merapi, restoration

Abstract

ABSTRACTThis experiment aims to determine the effect of the addition of potato biscuit on digestibility and feed efficiency in Sumatran slow loris (Nycticebus coucang). Research has been conducted on Small Mammals Captivity Breeding of Zoology Division, Research Center for Biology - LIPI for 66 days consisting of a 10 days adaptation period of feed and 56 days (8 weeks) data collection period. The material used is four Sumatran slow lorises (N. coucang) and two Javan slow lorises (N. javanicus). Feed given during the study are banana, papaya, dragon fruit, guava, passion fruit, boiled sweet potato, boiled egg potato biscuits, crickets, and meal worm. Feed treatment to Sumatran slow loris consisting of feed control (T0) and T0 plus potato biscuits (T1), while Javan slow loris was only fed T1 as a comparison. Parameters measured were consumption, digestibility, and feed efficiency. The most palatable feed types for Sumatran slow loris and Javan slow loris are banana, cricket, and meal worm. Mean of feed intake at T0 and T1 treatment was 38.63 and 37.42 g / head / day, and that of Javan slow loris is 42.51 g / head / day. Mean of dry matter digestibility of Javan slow loris> T1> T0, namely 92.02%, 91.21%, and 88.95% respectively; whereas the highest average feed efficiency (EPP) is at 12.06% for Sumatran Slow loris and 9.10% in Javan slow loris. The average of total digestible nutrients (TDN) of Javan slow loris> T1> T0, namely 87.04%, 85.34%, and 83.54% respectively.Keywords: consumption, digestibility, feed utilization, Nycticebus coucang, Nycticebus javanicus

Abstract

ABSTRACTPlant Growth Promoting Rhizobacteria (PGPR) are a group of bacteria known to influence plant growth by direct or indirect mechanisms. In search of efficient PGPR which potential as biofertilizers, a total of 26 isolates of rhizobacteria were isolated from various plant rhizosphere. These 26 isolates were screened through in vitro methods for PGPR properties like phosphate solubilization, siderophore, IAA and amonia production, and catalase activity. The result revealed that 9 isolates showed positive activities for phosphate solubilization 17 isolates for IAA production 18 isolates for siderophores 19 isolates for ammonia production and all of the isolates have catalase activities . Only four isolates (Az.KT.CSC, Az.D.8B, Az.D.8A and Az.Lo.10B) exhibited multiple plant growth promoting traits viz., phosphate solubilization, siderophore, IAA, amonia production, and catalase activity. Ten isolates that have different characters were further investigated for quantitative analysis of IAA production, HCN production and its effect on germination and seedling growth of Zea mays. The range of IAA production was 3,12? 134,27 ppm, among ten isolates, Az.D.8B isolate produced the highest IAA (134,27 ppm). Production of HCN was detected in eight isolates. Four isolates positively affected the germination of Zea mays seeds. Highest root elongation was recorded when seeds were treated with Az.Lo.5 isolate. Whereas, the highest chlorophyl content and plant high were recorded when seeds were treated with Az.B.8B isolate.Keywords: N-fixing bacteria, PGPR, IAA, siderophore, catalase

Abstract

ABSTRACTAnabaena azollae is an heterocyst Cyanophyceaean as symbiont of Azolla sp. Anabaena azollae is able to fix N2 from atmosphere and transform it into ammonium by its heterocyst cell. This research was conducted to study the influence of different concentration level of nitrogen to the dynamic of Anabaena azollae heterocyst cell. Research was conducted by observing heterocyst cell and variation nitrogen concentration in growing media in 21 days. In each sampling of Azolla sp., heterocyst observation was conducted for every 1000 cells in Anabaena azollae filament colonies. Result showed that media with 0 mg/L nitrogen has the highest number of heterocyst cell. In early observation, there was increasing of ammonium concentration in media 0 mg/L and 5 mg/L. Media without nitrogen addition has generated the highest number of heterocyst cell.Keywords: Anabaena azollae, Azolla sp., heterocyst

Abstract

ABSTRACTThe study of carbon stock and biomasa of agricultural commodities was conducted in the Bodogol village, Gunung Gede Pangrango National Park - West Java. The purpose of this research to determine the biomasa, carbon stock and the rate of decomposition of agricultural commodities by using destructive sample and litterbag method. Six of agricultural commodities: green beans/buncis (Phaseolus vulgaris), chilli (Capsicum annum), corn (Zea mays), bean (Vigna cylindrica), peanuts (Arachis hypogaea) and cassava (Manihot esculenta), were calculated their biomasa and carbon stocks. The result showed that the biomasa of agriculture commodities in the range of 0.152 to 4.216 t ha-1, with a carbon stock ranging from 0.01 to 1.83 t ha-1. The decomposition rate (k) of those commodities were k = 5.6 y-1; 5.48 y-1, 5.18 y-1, 5.04 y-1, 4.42 y-1, and k = 1.21 y-1, for Manihot esculenta, Vigna cylindrica, Arachis hypogea, Zea mays, Capsicum annum and Phaseolus vulgaris, respectively.Keywords: Biomasa, Gede Pangrango, Carbon, Agricultural commodities
